Abstract:Here, an ultrasensitive electrochemiluminescence (ECL) biosensor was developed to detect exosomes down to single particles based on multiple signal amplification. In this strategy, NiFe-tris(2,2'-bipyridyl) ruthenium(II) (Ru(bpy)(3)(2+)) on the electrode exhibited an excellent catalytic effect on the ECL reaction, significantly enhancing the ECL signal by approximately six-fold. In addition, the high affinity between the aptamer and CD 63 protein on the exosome surface released the RNA assembled into the DNA-RNA heteroduplex on the electrode. Duplex -specific nucleases activated DNA cleavage on DNA-RNA heteroduplex and initiated RNA recycling. Conse-quently, massive ferrocene (Fc)-DNA was detached from the electrode surface to recover the ECL emission of the NiFe-Ru(bpy)(3)(2+) emitter. Benefiting from the multiple signal amplification, the fabricated ECL biosensor achieved a limit of detection for exosomes as low as 5 particles/mu L and was successfully used for exosomes determination in serum. This sensor fabrication strategy will provide a universal and sensitive nano-platform for exosomes detection and enable the early diagnosis of related diseases.

Abstract:Exosomes have been reported to play an important role in the anti-tumor immune response, tumor diagnosis and other processes, and are promising biomarkers for early cancer diagnosis. In this work, a sensitive electrogenerated chemiluminescence (ECL) biosensor was developed for detection of exosomes using aptamer modified two-dimensional material Ti3C2 MXenes nanosheets as the ECL nanoprobe because of its large surface area, the excellent conductivity and catalytic properties. The exosomes can be high efficiently captured onto the electrode surface by an EpCAM protein recognized aptamer modified on the electrode surface. In addition, the ECL nanoprobe can also recognize the exosomes, and significantly enhanced the ECL signals of luminol. Based on this strategy, a highly sensitive ECL biosensor for MCF-7 exosomes detection was obtained. The detection limit is 125 particles μL-1, which was over 100 times lower than that of conventional ELISA method. The as prepared ECL biosensor was performed successfully for MCF-7 exosomes detection in the serum. This strategy provided a feasible, sensitive and reliable tool for the exosomes detection in exosomes-related clinical diagnostics.

Abstract:The marriage of energy transfer with electrochemiluminescence has produced a new technology named electrochemiluminescence energy transfer (ECL-ET), which can realize effective and sensitive detection of biomolecules. To obtain optimal ECL-ET efficiency, perfect energy overlapped donor/acceptor pair is of great importance. Herein, we present a sensitive ECL-ET based immunosensor for the detection of tumor markers, using energy tunable CdSeTe/CdS/ZnS double shell quantum dots (QDs) and gold nanorods (GNRs) as the donor and acceptor, respectively. Firstly a facile microwave-assisted strategy for the synthesis of green- to near-infrared-emitting CdSeTe/CdS/ZnS QDs with time- and component-tunable photoluminescence was proposed. And, on the basis of the adjustable optical properties of both CdSeTe/CdS/ZnS QDs and GNRs, excellent overlap between donor emission and acceptor absorption can be obtained to ensure effective ECL-ET quenching, thus improving the sensing sensitivity. This method represents a novel approach for versatile detection of biomolecules at low concentrations.
Electrochemical micro-aptasensors for exosome detection based on hybridization chain reaction amplification

Abstract:Abstract Exosomes are cell-derived nanovesicles that have recently gained popularity as potential biomarkers in liquid biopsies due to the large amounts of molecular cargo they carry, such as nucleic acids and proteins. However, most existing exosome-based analytical sensing methods struggle to achieve high sensitivity and high selectivity simultaneously. In this work, we present an electrochemical micro-aptasensor for the highly sensitive detection of exosomes by integrating a micropatterned electrochemical aptasensor and a hybridization chain reaction (HCR) signal amplification method. Specifically, exosomes are enriched on CD63 aptamer-functionalized electrodes and then recognized by HCR products with avidin-horseradish peroxidase (HRP) attached using EpCAM aptamers as bridges. Subsequently, the current signal that is generated through the enzyme reaction between the HRP enzyme and 3,3’,5,5’-tetramethylbenzidine (TMB)/H 2 O 2 directly correlates to the amount of bound HRP on the HCR products and thus to the number of target exosomes. By introducing anti-EpCAM aptamers, micro-aptasensors can detect cancerous exosomes with high specificity. Due to the micropatterned electrodes and HCR dual-amplification strategy, the micro-aptasensors achieve a linear detection response for a wide range of exosome concentrations from 2.5×10 3 to 1×10 7 exosomes/mL, with a detection limit of 5×10 2 exosomes/mL. Moreover, our method successfully detects lung cancer exosomes in serum samples of early-stage and late-stage lung cancer patients, showcasing the great potential for early cancer diagnosis.

Abstract:In this work, an ultrasensitive electrogenerated chemiluminescence (ECL) biosensor for exosomes and their surface proteins was developed by the in situ formation of gold nanoparticles (AuNPs) decorated Ti3C2 MXenes hybrid with aptamer modification (AuNPs-MXenes-Apt). In this strategy, the exosomes were efficiently captured on an exosome recognized CD63 aptamer modified electrode interface. Meanwhile, in situ formation of gold nanoparticles on single layer Ti3C2 MXenes with aptamer (MXenes-Apt) modification was obtained, in which MXenes acted as both reductants and stabilizer, and no additional reductant and stabilizer involved. The in situ formed AuNPsMXenes-Apt hybrid not only presented highly efficient recognition of exosomes specifically, but also provide naked catalytic surface with high electrocatalytic activity of gold nanoparticles with predominated (111) facets that significantly improved the ECL signal of luminol. In this way, a highly sensitive ECL biosensor for exosomes detection was constructed ascribing to the synergistic effects of large surface area, excellent conductivity, and catalytic effects of the AuNPs-MXenes-Apt. The detection limit is 30 particles pL(-1) for exosomes derived from HeLa cell line, which was over 1000 times lower than that of conventional ELISA method and the linear range was from 10(2) particles mu L-1 to 10(5) particles mu L-1. This ECL sensing platform possessed high selectivity toward exosomes and their surface proteins derived different kinds of tumor cell lines (HeLa cells, OVCAR cells and HepG2 cells), and enabled sensitive and accurate detection of exosomes from human serum, which implied that the ECL biosensor provided a feasible, sensitive, and reliable tool for exosomes detection in exosomes-related clinical diagnostic.
High-Performance Detection of Exosomes Based on Synergistic Amplification of Amino-Functionalized Fe3O4 Nanoparticles and Two-Dimensional MXene Nanosheets

Abstract:Exosomes derived from cancer cells have been recognized as a promising biomarker for minimally invasive liquid biopsy. Herein, a novel sandwich-type biosensor was fabricated for highly sensitive detection of exosomes. Amino-functionalized Fe3O4 nanoparticles were synthesized as a sensing interface with a large surface area and rapid enrichment capacity, while two-dimensional MXene nanosheets were used as signal amplifiers with excellent electrical properties. Specifically, CD63 aptamer attached Fe3O4 nanoprobes capture the target exosomes. MXene nanosheets modified with epithelial cell adhesion molecule (EpCAM) aptamer were tethered on the electrode surface to enhance the quantification of exosomes captured with the detection of remaining protein sites. With such a design, the proposed biosensor showed a wide linear range from 102 particles μL−1 to 107 particles μL−1 for sensing 4T1 exosomes, with a low detection limit of 43 particles μL−1. In addition, this sensing platform can determine four different tumor cell types (4T1, Hela, HepG2, and A549) using surface proteins corresponding to aptamers 1 and 2 (CD63 and EpCAM) and showcases good specificity in serum samples. These preliminary results demonstrate the feasibility of establishing a sensitive, accurate, and inexpensive electrochemical sensor for detecting exosome concentrations and species. Moreover, they provide a significant reference for exosome applications in clinical settings, such as liquid biopsy and early cancer diagnosis.

Abstract:Exosomes derived vesicles and their surface anchored proteins by cells are extremely vital in intercellular communication, immuno-stimulation, and so on, which are promising in potential tumor biomarkers for disease diagnosis. However, the highly sensitive detection of exosomes and their surface proteins is still challenging. Herein, we combined the g-C3N4 conjugated polydopamine coated Galinstan liquid metal shell-core nanohybrids (g-C3N4@Galinstan-PDA) nanoprobes and multivalent PAMAM-AuNPs electrode interface to realize a highly sensitive detection of exosomes and their surface proteins by electrogenerated chemiluminescence (ECL) biosensor. The antibody-modified PAMAM-Au nanoparticles (NPs) electrode interface provided a multivalent recognition platform for highly effective capture of exosomes. Meanwhile, the Galinstan NPs were applied as the nanoprobe. The antibody modified g-C3N4@Galinstan-PDA can recognize the exosomes specifically and exhibit stable and strong ECL signals due to the excellent features of the Galinstan NPs in facilitating electron transfer and suppressing the g-C3N4 passivation during electrochemical reduction procedures. In this way, high sensitivity for HeLa cell derived exosomes analysis was obtained with the limit of detection (LOD) of 31 particles μL-1. Moreover, we operated the exosomes analysis in the real samples including serum, urine, and blood, and identified the multiple biomarkers (GPC1, CD9, CEA, and AFP) on the exosome surface derived from different kinds of cell lines (HeLa cell, OVCAR-3 cell, and BT474 cell). These consequences suggest that the proposed ECL biosensor has the potential to be a powerful tool for exosomes study and clinical diagnostic as well as wearable devices.

Abstract:Exosomes are non-invasive biomarkers for cancer diagnosis. Herein, we describe an electrochemiluminescent (ECL) aptasensor for the detection of exosomes from breast tumor cells. Mercaptopropionic acid (MPA)-modified Eu3+-doped CdS nanocrystals (MPA-CdS:Eu NCs) and H2O2 were used as ECL emitters and coreactant, respectively. The exosomes are recognized and captured by the CD63 aptamer, and then form a G-quadruplex/hemin DNAzyme, which efficiently catalyzes the decomposition of H2O2, resulting in the decreased ECL signal of MPA-CdS: Eu NCs. The exosomes from breast tumor cells (MCF-7 cells) can be detected in the range of 3.4 x 10(5) to 1.7 x 10(8) particles per mL. The limit of detection (LOD) was estimated to be 7.41 x 10(4) particles per mL at a signal-to-noise ratio of 3. The aptasensor has been successfully used to detect exosomes in the serum.

Abstract:Exosomes, involved in cancer-specific biological processes, are promising noninvasive biomarkers for early diagnosis of cancer. Herein, an immobilization-free dual-aptamer-based photoelectrochemical (PEC) biosensor was proposed for the enrichment and quantification of cancer exosome based on photoactive bismuch oxyiodide/gold/cadmium sulfide (BiOI/Au/CdS) composites, nucleic acid-based recognition and signal amplification. In this biosensor, the recognition of exosome by two aptamers would trigger the deoxyribonucleotidyl transferase (TdT) enzyme-aided polymerization, leading to the enrichment of alkaline phosphatase (ALP) on Fe 3 O 4 surface. After magnetic separation, ALP could catalyze the generation of ascorbic acid (AA) as electron donor and initiate the following redox cycle reaction for further signal amplification. Furthermore, all the above processes were performed in solution, the recognition and signal amplification efficiency would be superior than the heterogeneous strategy owing to the avoidance of steric hindrance effect. As a result, the proposed PEC biosensor was capable of enriching and detecting of cancer exosomes with high sensitivity and selectivity. The linear range of the biosensor was from 1.0 × 10 2 particles·μL −1 to 1.0 × 10 6 particles·μL −1 and the detection limit was estimated to be 21 particles·μL −1 . Therefore, the proposed PEC biosensor holds great promise in quantifying tumor exosome for nondestructive early clinical cancer diagnosis and various other bioassay applications.

Abstract:Exosomes, a class of extracellular vesicles carrying the biological information of their parent cells, have emerged as one of the most promising biomarkers for the early screening and prognostic monitoring of cancer. However, the effective capture and sensitive detection of cancer cell-derived exosomes pose significant challenges due to the inevitable interference from internal environment and the limited exosome abundance in clinical samples. Herein, we proposed an aptamer-based electrochemical biosensor combining with Au nanoprobe-mediated dual signal amplification for sensitive detection of tumor-related exosomes. Benefiting from the specific recognition of CD63 aptamer (CD63 Apt), the target exosomes were efficiently captured on the gold electrode, followed by trapping the iDNA-modified Au nanoprobe, which could induce the generation of hybridization chain reaction (HCR). The long double-stranded DNA produced by HCR procedure provided numerous insertion sites to RuHex, achieving the high sensitivity for exosome detection. Under the optimal experimental conditions, this aptamer-based electrochemical biosensor for MCF-7 cancer cell-derived exosome detection exhibited a good linear range within 5 orders of magnitude (from 1.29 × 10 4 to 1.29 × 10 8 particle/mL) with a low limit of detection (LOD) to 1.65 × 10 3 particle/mL. Furthermore, this biosensor possessed good stability and anti-interference ability. Therefore, this analysis platform is considered as a promising candidate for the exosome-based diagnosis and prognosis of cancer in clinical applications.

Abstract:Quantification of trace exosomes in biofluids is helpful for the early diagnosis of cancers. Electrochemical biosensors have attracted special attentions for exosome detection, while the employment of nanoconjugates in the construction of electrochemical biosensors can achieve better performance. So, in this work, we have designed and fabricated HRP-DNA-COF nanoconjugates (HDCs) that involve covalent organic frameworks (COFs) as the scaffold, cholesterol-tagged DNA to recognize exosomes and horseradish peroxidase (HRP) to amplify the signal readout. The successful preparation of HDCs have been verified experimentally, based on which an electrochemical biosensor has been constructed for exosome detection. Attributed to the specific recognition ability and excellent catalytic ability of HDCs, the biosensor achieves good performance to detect exosomes with simplicity and sensitivity in the linear range from 104 to 107 particles/mu L. Besides, the strategy proposed in this work to construct COF-based nanoconjugates may promote the wider application of COFs in biomedical research.

Abstract:Exosome analysis is emerging as an attractive noninvasive approach for disease diagnosis and treatment monitoring in the field of liquid biopsy. Aptamer is considered as a promising molecular probe for exosomes detection because of the high binding affinity, remarkable specificity, and low cost. Recently, many approaches have been developed to further improve the performance of electrochemical aptamer based(E-AB) sensors with a lower limit of detection. In this review, we focus on the development of using aptamer as a specific recognition element for exosomes detection in electrochemical sensors. We first introduce recent advances in evolving aptamers against exosomes. Then, we review methods of immobilization aptamers on electrode surfaces, followed by a summary of the main strategies of signal amplification. Finally, we present the insights of the challenges and future directions of E-AB sensors for exosomes analysis.

Abstract:Exosomes with diameters of 30-150 nm are small membrane-bound vesicles secreted by a variety of cells. They play an important role in many biological processes, such as tumor-related immune response and intercellular signal transduction. Exosomes have been considered as emerging and noninvasive biomarkers for cancer diagnosis. Recently, a large number of optical and electrochemical biosensors have been proposed for sensitive detection of exosomes. To meet the increasing demands for ultrasensitive detection, nanomaterials have been integrated with various techniques as powerful components. Because of their intrinsic merits of biological compatibility, excellent physicochemical features and unique catalytic ability, nanomaterials have significantly improved the analytical performances of exosome biosensors. In this review, we summarized the recent progress in nanomaterials-based biosensors for the detection of cancer-derived exosomes, including fluorescence, colorimetry, surface plasmon resonance spectroscopy, surface enhanced Raman scattering spectroscopy, electrochemistry, electrochemiluminescence and so on.

Abstract:In this work, we firstly enunciated the presence of synergy effect between the electron and energy transfer in an electrochemiluminescence (ECL) system involving the CdS NCs and Au NPs, based on which an amplified ECL biosensor was constructed for sensitive detection of DNA-protein interactions. Specifically, Au NPs modified on electrode, following by assembled probe DNA. Then the modified GCE was hybridized with SiO(2)@CdS/DNA conjugates. With DNA duplex as a rigid spacer, Au NPs could not only accelerate the electron transfer but also produce the surface plasma resonance upon excited ECL of CdS NCs. However, the enhanced ECL could be greatly suppressed by the binding of target protein. The prepared biosensor possesses excellent analytical performance with the linear range of target protein from 0.015 to 150 nM.
Ultrasound-pretreatment combined with Ti3C2-TiO2-AuNPs enhancing the electrogenerated chemiluminescence of the air-saturated luminol for exosomes detection

Abstract:It is still a great challenge to develop effective strategies to improve the low electrogenerated chemiluminescence (ECL) of air-saturated luminol. Herein, the synergistic effects of Ti3C2-TiO2-AuNPs nano hybrid and high-intensity focused ultrasound pretreatment (ultrasound-pretreatment) were used to significantly improve the ECL emission of the air-saturated luminol, and the mechanism was proposed. The ultrasound-pretreatment as a green method with the cavitation effect could form O2-• and H2O2 in situ as an initiator. TiO2 and Au nanoparticles (AuNPs) were in situ decorated on the Ti3C2 surface to form Ti3C2-TiO2-AuNPs, and it was proved as a highly efficient booster which could catalyze and aggregate H2O2 to the O2-•. The utilization rate of intermediates has been greatly improved. Exosomes as model targets can be sensitively detected by the ECL sensor. The detection limit was 195 particles μL-1. The detection results of exosomes in actual samples are satisfactory. We believe that the ultrasound-pretreatment strategy could be extended to the sensitive detection in the biological sample.

Abstract:Exosomes are considered potential biomarkers for early screening and accurate non-invasive diagnosis of cancer, so development of innovatively facile approaches for the detection of cancer cell-derived exosomes has become increasingly important. Herein, we propose a facile electrochemical biosensor based on divalent aptamer-functionalized nanochannels for highly efficient detection of cancer cell-derived exosomes. The aptamer against transmembrane receptor protein CD63 and the aptamer targeting membrane protein EpCAM are simultaneously immobilized on the nanochannels to construct the divalent aptamer-functionalized nanochannels. Thus, the target exosomes can be recognized and selectively captured by the functionalized nanochannels in a divalent collaborative manner. The combined exosomes overlay the ion channel effectively and hinder the ionic flow through the nanochannels, resulting in an evidently varied ionic transport behavior corresponding to the abundance of exosomes. The divalent aptamer-functionalized nanochannels can substantially promote the binding stability and enhance the detection specificity, while the sensitivity of detection is improved greatly by virtue of the amplified response of array channels synergized with the electrochemical technique. Therefore, the developed biosensor provides a highly specific, sensitive, and accurate approach for the detection of cancer cell-derived exosomes, which may hold great potential for application in early clinical cancer diagnosis.

Abstract:Zwitterionic electrochemiluminescence biointerface is incorporated into a gas diffusion electrode and a fluidic microreaction device, showing minimal fouling in complex biological fluids while maintaining high analytical performance in exosomes checking and dynamics tracking. The competence to construct sensing platforms capable of selective manipulation in complex biological fluids undoubtedly underpins critical future advances in healthcare. Despite the fact that electrochemiluminescence (ECL) has long been an influential technology for clinical diagnosis worldwide, ECL interface that optimizes fouling resistance has been mimicked less often, especially in an integrated platform. Herein, ECL transducer is prepared by the integration of protonated g‐C3N4 and Ti3C2Tx MXene nanosheets, displaying enhanced charge injection/transfer, and inherent catalytic capacities for coreactant ECL. Mussel‐bioinspired polydopamine was exploited as a thin, surface‐adherent substrate to coat the solid‐state transducer and further initiate secondary reactions via Michael Addition for tailing recognition element and zwitterionic segment. This architecture guarantees not only the least suppression of ECL performance but also desired antifouling properties, ensuring < 7.45% of ECL loss after 96 h of exposure to complex biological fluids. Creatively, a highly integrated platform is equipped with the established biointerface, gas diffusion electrode, and fluidic ECL microreactor, affording high‐performance exosome checking and dynamics tracking in a non‐label manner. Our study provides a general design strategy to obtain a robust antifouling ECL sensing interface based on zwitterionic chemistries and provides a fresh perspective in developing point‐of‐care and implantable ECL devices.
