Abstract:A strategy for rapidly and effectively assembling oligosaccharides using a novel dimeric imidazole-type ionic liquid has been developed. This method incorporating the advantages of both solid- and liquid-phase synthesis, can not only maintain the homogeneous reaction conditions, but also facilitate the phase separation process with extremely high recovery of products. The effectiveness of this approach was verified through the successful synthesis of β-(1→6)-linked triglucosamine with an overall yield of 61.6%.

Abstract:SW-1 is a well-known natural saponin with potent antitumor activities. We have designed and prepared a small library of 22 OSW-1 analogues with a variety of p-acylamino-benzoyl groups installed at C2 '' of the xylose residue, wherein a regioselective (1 -> 3)-glycosylation of arabinoside 3,4-diol has been achieved by manipulation of the protecting groups on the imidate donors. Bioassays lead to new structure-activity relationships as well as two applicable fluorescent probes, which are found to localize to lysosomes in HeLa cells and could be used in further antitumor mechanism studies of OSW-1 in living cells.

Abstract:C-oligosaccharides are pharmacologically relevant because they are more hydrolysis-resistant than O-oligosaccharides. Despite indisputable advances, C-oligosaccharides continue to be underdeveloped, likely due to a lack of efficient and selective strategies for the assembly of the interglycosidic C-C linkages. In contrast, we, herein, report a versatile and robust strategy for the synthesis of structurally complex C-oligosaccharides via catalyzed C(sp3 )-H activations. Thus, a wealth of complex interglycosidic (2→1)- and (1→1)-C-oligosaccharides becomes readily available by palladium-catalyzed C(sp3 )-H glycoside glycosylation. The isolation of key palladacycle intermediates and experiments with isotopically-labeled compounds identified a trans-stereoselectivity for the C(sp3 )-H glycosylation. The glycoside C(sp3 )-H activation manifold was likewise exploited for the diversification of furanoses, pyranoses and disaccharides.

Abstract:Herein, we present the first synthesis of hybrid molecules combining the pharmacophores of two natural compounds, schweinfurthin E (SW-E) and the glycosidic moiety of OSW-1. These hybrids were designed leveraging the complementary binding of SW-E and OSW-1 to their biological target. The synthetic process highlights, in particular, one-pot functionalization and glycosylation of an L-arabinose unit using a D-xyloside donor and a CuAAC click reaction involving a polyfunctionalized prenylated stilbene derived from SW-E.

Synthesis of a cholestane glycoside OSW-1 with potent cytostatic activity

Abstract:The potent antitumor agent OSW-1 was synthesized from the protected aglycone in different ways. It was proven that direct glycosylation of the aglycone in its hemiketal form could be achieved, affording the protected OSW-1 in a moderate yield. Alternatively, regioselective protection of the triol obtained by reduction of the aglycone, followed by glycosylation, deprotection and oxidation yielded the same OSW-1 derivative. The third approach to this compound consisted of glycosylation of the previously described lactol [Morzycki, J. W.; Gryszkiewicz, A. Polish J. Chem. 2001, 75, 983-989], reaction of the resulting aldehyde with a Grignard reagent, and oxidation. OSW-1 obtained on removal of the protective groups was identical with the natural product.

Abstract:Comprehensive SummaryA convenient and highly efficient strategy was developed for the simplification of enzymatic synthesis and purification of oligosaccharides. Glycosyl acceptors terminated with a thiol tag were extended by enzymatic modular assembly (EMA) in the aqueous phase, and the desired products were captured during solid‐phase workup (SPW) using commercially available thiopropyl Sepharose 6B resin through a reversible disulfide linkage. Finally, the products were released from solid‐phase resin in the presence of dithiothreitol (DTT) as reducing agent and the resin could be recovered and reused. This strategy overcomes the uncertain influence of polymer‐ or ionic‐supports commonly used in enzymatic glycosylation, and showed perfect compatibility with all 10 enzyme modules for the rapid assembly of a series of complex oligosaccharides.

Abstract:A collective synthesis of 3,6-dideoxysugars, including seven naturally known congeners, has been reported using commercially available methyl lactates in five steps. The essential tandem process involving the olefin cross-metathesis and isomerization steps was enabled by the dual function of Grubbs-II catalyst, affording the products in good yields and providing concise and practical access to a class of biologically important deoxysugars.

Abstract:An expeditious three-component, one-pot sequential glycosylation protocol has been developed for the preparation of 3,6-branched unsymmetrical mannopentaose (Man5), employing a mannose trisaccharide donor, a mannose monosaccharide donor and a mannose monosaccharide acceptor. The high efficiency of this one-pot procedure was demonstrated by comparison study with a stepwise synthesis using the same three building blocks.

Abstract:A new carrier containing three hydrophobic octadecyls was employed to improve the yield and purity of the synthesized oligosaccharides, and the applicability of hydrophobic carrier for HASP（hydrophobically assisted switching phase） was also explored. High performance liquid chromatography（HPLC） was used to analyze the purity of oligosaccharides and improve the elution efficiency. Elution system was optimized and replaced by isopropyl alcohol and water, in which the excess donor and other impurities had a better solubility than that in methanol.

Abstract:The oligosaccharide residue in glycoconjugates located in cell membranes is responsible for intercellular recognition and interaction: it acts as a receptor for proteins, hormones, and microorganisms and governs immune reactions. These significant activities have stimulated great interest in the field of oligosaccharides and glycoconjugates. Although many advances have been made in the synthesis of oligosaccharides, more convenient and efficient methods are still needed. This review describes one of these new methods-the one-pot sequential glycosylation approach as a potent tool for oligosaccharide assembly. The oligosaccharide library construction in a one-pot fashion is also summarized.

Abstract:The multifaceted biological importance of oligosaccharides and glycoconjugates has made them very popular synthetic targets in modem synthetic chemistry. Great progress has been made in the construction of complex oligosaccharides based on a variety of synthetic strategies. Among these strategies, the one-pot glycosylation is very attractive and shows great promise for assembly of oligosaccharide libraries and automated synthesis. Traditional one-pot glycosylation approach based on the chemoselective principle or orthogonal strategy needs extensive protecting group manipulations and/or aglycon adjustments for designing building blocks with suitable anomeric reactivities. The recently established pre-activation strategy has some advantages over the traditional one-pot glycosylation approach, since it does not require the tuning of the building blocks in an armed-disarmed fashion. The latest advances in one-pot oligosaccharide synthesis by pre-activation strategy are summarized in this review.

Abstract:Developing artificial "CO2-sugar" platforms is meaningful for addressing challenges posed by land scarcity and climate change to the supply of dietary sugar. However, upcycling CO2 into complex polyoxygenated carbohydrates involves several major challenges, including achieving enantioselective and thermodynamically driven transformation and expanding product repertoires while reducing energy consumption. We present a versatile chemoenzymatic roadmap based on aldol condensation, iso/epimerization, and dephosphorylation reactions for asymmetric CO2 and H2 assembly into sugars with perfect stereocontrol. In particular, we developed a minimum ATP consumption and the shortest pathway for bottom-up biosynthesis of the fundamental precursor, fructose-6-phosphate, which is valuable for synthesizing structure-diverse sugars and derivatives. Engineering bottleneck-associated enzyme catalysts aided in the thermodynamically driven synthesis of several energy-dense and functional hexoses, such as glucose and D-allulose, featuring higher titer (63 mmol L-1) and CO2-product conversion rates (25 mmol C L-1 h-1) compared to established in vitro CO2-fixing pathways. This chemical-biological platform demonstrated greater carbon conversion yield than the conventional "CO2-bioresource-sugar" process and could be easily extended to precisely synthesize other high-order sugars from CO2.
