Abstract:Palladium-catalyzed Suzuki-Miyaura cross-coupling or aryl halides is widely employed in the synthesis of many important molecules in synthetic chemistry, including pharmaceuticals, polymers and functional materials. Herein, we disclose the first palladium-catalyzed decarbonylative Suzuki-Miyaura cross-coupling of amides for the synthesis of biaryls through the selective activation of the N-C(O) bond of amides. This new method relies on the precise sequence engineering of the catalytic cycle, wherein decarbonylation occurs prior to the transmetallation step. The reaction is compatible with a wide range of boronic acids and amides, providing valuable biaryls in high yields (>60 examples). DFT studies support a mechanism involving oxidative addition, decarbonylation and transmetallation and provide insight into high N-C(O) bond activation selectivity. Most crucially, the reaction establishes the use of palladium catalysis in the biaryl Suzuki-Miyaura cross-coupling of the amide bond and should enable the design of a wide variety of cross-coupling methods in which palladium rivals the traditional biaryl synthesis from aryl halides and pseudohalides.

Abstract:An iodine-promoted, metal-, base-, and solvent-free cross-coupling reaction was developed for the synthesis of various useful secondary amides via an aryl N-addition reaction of aryl groups to cyano groups. This aryl transfer reaction proceeds with arylhydrazine hydrochlorides serving as the aryl donors. A labelling experiment shows that the N atom in the product comes from the cyano group of the nitriles, which are low in cost. A plausible radical-driven mechanism is also proposed.

Abstract:Given the natural abundance and industrial accessibility, inert amides have gained considerable attention in homologous amine synthesis and the development of novel catalytic strategies for their deoxygenative functionalization is highly desirable. Although organic electrosynthesis has become an uprising research area by virtue of its sustainability and atom economy, the combination of amide reduction with an electrochemical process in value-added amine synthesis is still undeveloped. Herein, a strategy merging iridium-catalyzed activation of amides with electrochemical radical generation to access deoxygenative arylation of secondary amides is developed. This methodology is featured by broad substrate scope, mild reaction conditions, and operational simplicity, giving corresponding benzylic amines with high efficiency. Notably, the successful application of electrochemical method in the deoxygenative functionalization of amides would provide an efficient strategy for quick access to value-added amines from inert amides.

Abstract:A new strategy for the sequential formation of aryl and amidyl C-N bonds is reported. Using trichloroethoxysulfonyl azide as a bifunctional nitrogen source, Ir-catalyzed aryl C-H sulfonamidation and subsequent desulfonative amide formation proceed effectively without any need of oxidants or coupling reagents. This protocol is suitable for readily available benzamides and stable carboxylates including primary, secondary, and tertiary alkyl, alkenyl, and phenyl carboxylates, thereby providing a direct and efficient method for the synthesis of biologically and chemically useful N-arylamides.

Abstract:Herein, we report a catalyst system for Pd-catalyzed decarbonylative Suzuki-Miyaura cross-coupling of aroyl chlorides with boronic acids to furnish biaryls. This strategy is suitable for a broad range of common aroyl chlorides and boronic acids. The synthetic utility is highlighted in the direct late-stage functionalization of pharmaceuticals and natural products capitalizing on the presence of carboxylic acid moiety. Extensive mechanistic and DFT studies provide key insight into the reaction mechanism and high decarbonylative cross-coupling selectivity.

Abstract:Robust NHC-palladacycles (NHC = N-heterocyclic carbene) were synthesized and exhibited high catalytic activity towards Suzuki−Miyaura cross-coupling reactions between inactive amides with N-acetyl/benzyl substituents and aryl boronic acids, producing diverse ketones in good to excellent yields. This unprecedented and practical palladacycles-catalyzed Suzuki−Miyaura cross-coupling of amides with boronic acids via selective C-N bond activation was attributed to the strong σ-donor and weak π-acceptor properties of acenaphthoimidazolylidene, which may highlight their potential in other challenging coupling transformations involving inactive amides.

Abstract:The convenient cross-coupling of sp2 or sp3 carbons with a specific boron vertex on carborane cage represents significant synthetic values and insurmountable challenges. In this work, we report an Rh-catalyzed reaction between o-carborane and N-acyl-glutarimides to construct various Bcage -C bonds. Under the optimized condition, the removable imine directing group (DG) leads to B(3)- or B(3,6)-C couplings, while the pyridyl DG leads to B(3,5)-Ar coupling. In particular, an unexpected rearrangement of amide reagent is observed in pyridyl directed B(4)-C(sp3 ) formation. This scalable protocol has many advantages, including easy access, the use of cheap and widely available coupling agents, no requirement of an external ligand, base or oxidant, high efficiency, and a broad substrate scope. Leveraging the RhI dimer and twisted amides, this method enables straightforward access to diversely substituted and therapeutically important carborane derivatives at boron site, and provides a highly valuable vista for carborane-based drug screening.

Notably, the new approach employed active thioureas as coupling partners in the presence of arylsilanes to give amides in good yield. Abstract:Suzuki–Miyaura coupling is a practical and attractive carbon−carbon bond formation reaction due to its high efficiency and wide functional group compatibility, but its industrial applications are limited because it is typically catalysed by expensive palladium-containing transition-metal complexes. Here we show a robust and chemoselective organocatalytic Suzuki−Miyaura-type coupling of aryl halides with arylboronic acids catalysed by amines. The utility and scope of this reaction were demonstrated by the synthesis of several commercially relevant small molecules and a selection of derivatives of pharmaceutical drugs. The coupling of aryl halides and arylboronic acids is generally performed by metal-catalysed Suzuki–Miyaura reactions while metal-free approaches remain elusive. Here an organocatalytic approach based on amine catalysts is introduced for the preparation of commercially relevant asymmetric biaryls.

Abstract:Abstract Amides are among the most important and ubiquitous functional groups in organic chemistry and process development. In this Practical Synthetic Procedure, a protocol for the Suzuki–Miyaura cross-coupling of amides by selective N–C(O) bond activation catalyzed by commercially available, air- and moisture-stable palladium/N-heterocyclic carbene (NHC) complexes is described. The procedure described involves [Pd(IPr)(cin)Cl] [IPr = 2,6-(diisopropylphenyl)imidazol-2-ylidene, cin = cinnamyl] at 0.10 mol% at room temperature and is performed on decagram scale. Furthermore, a procedure for the synthesis of amide starting materials is accomplished via selective N-tert-butoxycarbonylation, which is the preferred method over N-acylation. The present protocol carries advantages of operational simplicity, commercial availability of catalysts, and excellent conversions at low catalyst loadings. The method is generally useful for activation of N–C(O) amide bonds in a broad spectrum of amide precursors. The protocol should facilitate the implementation of amide cross-coupling reactions.

Abstract:The tandem inert α-C-H and C-N bond activation of amides represents a highly valuable but challenging transformation in organic synthesis. Herein, a simple rare earth metal amido complex has been shown to catalyse unprecedented cyclization of amides with ynones to form trisubstituted 2-pyrones. This protocol significantly enables the selective merger of inert α-C-H and C-N bond activations of amides and indicates a particular role of rare earth catalysts in enhancing the selectivity for the α-C-H bond of amides in the presence of N-H bonds.

Abstract:The first palladium-catalyzed Suzuki-Miyaura cross-coupling of amides with boronic acids for the synthesis of ketones by sterically-controlled N-C bond activation is reported. The transformation is characterized by operational simplicity using bench-stable, commercial reagents and catalysts, and a broad substrate scope, including substrates with electron-donating and withdrawing groups on both coupling partners, steric-hindrance, heterocycles, halides, esters and ketones. The scope and limitations are presented in the synthesis of >60 functionalized ketones. Mechanistic studies provide insight into the catalytic cycle of the cross-coupling, including the first experimental evidence for Pd insertion into the amide N-C bond. The synthetic utility is showcased by a gram-scale cross-coupling and cross-coupling at room temperature. Most importantly, this process provides a blueprint for the development of a plethora of metal catalyzed reactions of typically inert amide bonds via acyl-metal intermediates. A unified strategy for amide bond activation to enable metal insertion into N-C amide bond is outlined ().

Abstract:A new recoverable and reusable bifunctional catalyst, MNP@IAH@Cu‐IL, was synthesized by combining an ionic liquid and linking Cu(II) on the surface of magnetic nanoparticles. The catalyst was successfully applied in the synthesis of primary and secondary amines via C(sp2)‐N coupling reaction, including unsymmetrical secondary amine derivatives with high yields. Copper, an abundant metal on Earth, has re‐emerged as a suitable alternative to versatile Palladium‐catalyzed C–N coupling. Herein, a new recoverable and reusable bifunctional catalyst was synthesized by combining an ionic liquid and linking Cu(II) on the surface of Fe3O4 nanoparticles. New nanocatalyst (MNP@IAH@Cu‐IL) was completely characterized using different techniques. Continuing, MNP@IAH@Cu‐IL was applied in the synthesis of primary and secondary amines via C(sp2)‐N coupling reaction successfully. This catalyst worked well in the synthesis of anilines derivatives in high yields and excellent quality. Further, we described a novel and one‐pot process for the synthesis of unsymmetrical secondary amine derivatives from two different aryl(alkyl) halides. This procedure shows attractive features such as 100% atom economy, high yields of the desired products under base‐free conditions, excellent functional group tolerance, work‐up simplicity, and recyclability.

Abstract:Reported herein is an unprecedented photocatalytic asymmetric cross-dehydrogenative coupling reaction between tertiary amines and simple ketones, and it proceeds by synergistic multiple catalysis with substoichiometric amounts of a hydrogen acceptor. This process is enabled by a simple chiral primary amine catalyst through the coupling of a catalytic enamine intermediate and an iminium cation intermediate in situ generated from tetrahydroisoquinoline derivatives by coupled Ru/Co catalysis.

Abstract:This work describes the first Ni-catalyzed cross-electrophile coupling of alkylpyridinium salts, derived from the corresponding amines, with aryl iodide, bromoalkyne or bromoalkyl coupling partners. C(sp)-C(sp 3 ), C(sp 2 )-C(sp 3 ) and C(sp 3 )-C(sp 3 ) bond formation was achieved to afford a variety of synthetically useful arenes, alkynes and alkanes in good yields from 2 - 33 . The advantages of the methodology are showcased in the two-step synthesis of the key lactonic moiety of (+)-Compactin and (+)-Mevinolin from commercially available starting materials. A one-pot procedure without isolation of alkylpyridinium tetrafluoroborate salt was also demonstrated to be successful. This work represents a new strategy for the cross-coupling reaction of two electrophiles, and also provides a complementary and highly valuable vista for the current methodologies of alkyl arene/alkyne/alkane synthesis.

Abstract:Direct dehydrogenative synthesis of α, β-unsaturated secondary amide still represents an elusive transformation. Herein we describe a palladium-catalyzed redox-neutral desaturation to prepare α, β-conjugated secondary amides. Without external oxidants, this approach relies on the N–O bond cleavage as the driving force to achieve the formal dehydrogenation. Complementary to known protocols, this transformation is enabled by the unique reactivity of hydroxamate, thereby representing a novel strategy to accomplish carbonyl desaturation. Desired conjugated secondary amides can be efficiently synthesized in presence of more reactive esters and even ketones, thus providing a solution to the long-standing issue of α, β-unsaturated secondary amides via C–C desaturation.

Abstract:We report the Pd-catalyzed Suzuki–Miyaura cross-coupling of aliphatic amides. Although tremendous advances have been made in the cross-coupling of aromatic amides, C–C bond formation from aliphatic amides by selective N–C(O) cleavage has remained a major challenge. This longstanding problem in Pd catalysis has been addressed herein by a combination of (1) the discovery of N,N-pym/Boc amides as a class of readily accessible amide-based reagents for cross-coupling and (2) steric tuning of well-defined Pd(II)-NHC catalysts for cross-coupling. The methodology is effective for the cross-coupling of an array of 3°, 2°, and 1° aliphatic amide derivatives. The catalyst system is user-friendly, since the catalysts are readily available and are air- and bench-stable. Mechanistic studies strongly support an amide bond twist and external nN → π*C═O/Ar delocalization as a unified enabling feature of N,N-pym/Boc amides in selective N–C(O) bond activation.
