Abstract:Normal-phase HPLC and hydrophilic interaction HPLC (HILIC) were investigated for the separation of selenometabolites in a water extract of Se-rich yeast prior to their detection by ICP-MS and identification by electrospray MS/MS. The targeted fraction was a low-abundant fraction co-eluting with salt and sulfur analogues in size-exclusion chromatography which has so far been inaccessible to Se speciation studies. The optimization of the separation conditions resulted in the highest separation efficiency when HILIC was used and elution was carried out isocratically with a low concentration ammonium acetate buffer (1 mM ammonium acetate/10 mM acetic acid) in 80% acetonitrile. Out of 15 peaks observed with the Se-specific ICP-MS detection 12 was identified by electrospray Q-TOF MS/MS (2,3-dihydroxypropionyl (DHP)-Se-methylselenocysteine [M+H]+: 272, Se-methyl-gamma-glutamyl-selenocysteinylglycine dioxide [M+H]+: 402, gamma-glutamyl-Se-methylselenocysteine [M+H]+: 313; isomers of gamma-glutamylselenocystathionine [M+H]+: 400; Se-methyl-selenoglutathione [M+H]+: 370, isomers of N-acetylselenocystathionine [M+H]+: 313, 2,3-DHP-selenohomolanthionine [M+H]+: 373, isomers of 2,3-DHP-selenocystathionine [M+H]+: 359, 2,3-DHP-selenolanthionine [M+H]+: 345 and selenohomolanthionine [M+H]+: 285).
Separation of selenium species in plant tissues by high performance liquid chromatography-ultraviolet treatment-hydride generation atomic fluorescence spectrometry using various mobile phases

Abstract:Anion-exchange chromatography (Hamilton, PRP-X100) with hydride-generation atomic fluorescence spectrometry (HG-AFS) is commonly used for selenium (Se) speciation analysis in environmental and biological samples. However, retentions of Se species are often different with different mobile phases. To address it, this study systematically investigated mobile phases based on ammonium salts to obtain good resolution of Se species. Various mobile phases, including NH4H2PO4, NH4Ac, NH4NO3 and (NH4)2HPO4, were tested. The best result was obtained with a mobile phase containing 60 mmol L−1 (NH4)2HPO4 at pH 6.0, where the separation of Se species, including selenite [Se(IV)], selenate [Se(VI)], selenocystine (SeCys) and selenomethionine (SeMet), was achieved within 10 min with reasonable resolution. In addition, the recoveries of SeCys, Se (IV), SeMet and Se (VI) in the Se-enriched samples were 95.72 ± 1.60%, 106.89 ± 2.20%, 93.28 ± 2.82% and 91.38 ± 1.27%, respectively. Finally, the proposed method was validated for the determination of Se species in Nicotiana tabacum tissues.

Abstract:There are many kinds of micronutrient elements contained in foods that are vital to human health. The strong dependence of the toxicity and bioavailability of microelement upon their chemical forms has made the speciation and bioavailability vital to the food quality control and nutrition evaluation. A brief introduction to the high performance liquid chromatography hyphenated with inductive coupled plasma mass spectrometry ( HPLC-ICP-MS) is given. The hyphenated technique which based on several chromatographic separation mechanisms plays an important role in speciation analysis for its advantage of high sensitivity, high selectivity, wide linearity, low detection and multi-microelement determination simultaneously. The application of HPLC-ICP-MS in microelements speciation analysis and bioavailability research in foods concerning the selenium speciation analysis in selenized yeast, selenium-enriched garlic, selenium nutritional supplement and other foods, the arsenic speciation analysis in sea-products and farm products, the other microelements speciation analysis and the investigation of metabolic and bioactivity of microelement in organism is reviewed in this paper.
Separation and Detection of Arsenic and Selenium Species in Environmental Samples by HPLC-ICP-MS

Abstract:A method is presented for arsenic speciation analysis of an oyster sample using ion chromatography coupled with an inductively coupled plasma mass spectrometry (ICP-MS) instrument. A strong anion exchange resin was employed with a step gradient elution of 0.1 mM/0.1 M K 2 SO 4 at pH 10.2. Arsenobetaine and dimethylarsinic acid were determined following extraction based on trypsin enzymolysis with 95-100% extraction efficiency. Limits of detection in the range 0.1-0.3 mg kg m 1 of arsenic were obtained for organic arsenic species. No inorganic arsenic was detected. Validation was performed using TORT-2 as a certified reference material. Although high performance liquid chromatography (HPLC) coupled to ICP-MS is an effective method for speciation analysis it is not always necessary to obtain such a detailed picture. A simple liquid chromatographic separation technique based upon mini-column technology is presented. It was developed to obtain a fast, efficient and reliable separation of inorganic from organic, i.e. assumed toxic from non-toxic, arsenic and selenium species suitable for use as an initial screening method for environmental analysis. Two types of strong anion exchange resin were tested. Excellent separation was obtained for both min-column resins and analysis times were within 7 min. Limits of detection obtained for inorganic arsenic, organic arsenic, selenomethionine, Se IV and Se VI were 1.6, 1.8, 66, 32 and 22 µg kg m 1 , respectively.

Abstract:A high efficiency chromatographic separation on a porous graphitic carbon stationary phase was developed for a large-scale separation of selenium metabolites in Se-rich yeast prior to their identification by electrospray hybrid quadrupole trap/Orbitrap mass spectrometry (Orbitrap MS(n)). The reversed-phase (RP) separation mode offered distinctly higher separation efficiency than the hydrophilic ion interaction (HILIC) mode. The latter was nevertheless complementary and useful to validate the detection of several compounds. The method allowed the detection of 64 metabolites including 30 SeSe or SeS conjugates (3 triple S/Se/S ones) and 14 selenoethers. 21 previously unreported metabolites were detected on the basis of the selenium isotopic pattern usually matched with the sub-ppm mass accuracy. 9 of these metabolites were subsequently identified using the multi-stage high mass accuracy (<5ppm) mass spectrometry. The identified metabolites (and their groups) were quantified on-line by ICP-MS fitted with a frequency-matching generator allowing a quasi-uniform response over the large (20-90%) acetonitrile mobile phase concentration range. The morphology of HPLC-ICP-MS chromatograms was remarkably similar to that of HPLC multi-ion extracted ESI-MS chromatograms. The detection limits obtained by ICP MS and ESI MS were 1 and 2ppb, respectively.

Abstract:The biological activities of selenium (Se) compounds vary according to speciation. The identification of Se compounds is crucial to evaluate the potential health benefits of Se-enriched foods. Molecular recognition and quantification analysis of selenoamino acids in Se-enriched millets were developed by high-performance liquid chromatography/electrospray ionization tandem mass spectrometry (HPLC-ESI-MS/MS). Ultrasonic bath pretreatments were used to extract free Se, while the combinative ones were released using an enzymatic hydrolysis process with protease XIV and α-amylase. Derivatization by carbamidomethylation was applied to enhance the detection sensitivity of trace selenocystine and selenocysteine. Molecular structure analysis of free and combinative selenoamino acids was performed according to the MS2 cleavage rules and isotope property with the product ion scan mode. Only combinative forms of selenoamino acids were detected via the multiple reaction monitoring (MRM) mode. Combinative selenoamino acids as Se-methyselenocysteine and selenomethionine were found, occupying more than 75.74 % of the total Se. The spiked recoveries ranged from 79.76 % to 90.79 % in the matrices. The detection limits for carbamidomethyl-selenocystine, Se-methyselenocysteine, and selenomethionine were 4.759, 0.699, and 1.084 ng mL−1 in the hydrolysate matrix, respectively. The HPLC-ESI-MS/MS method was established to simultaneously analyze the free and combinative forms of selenoamino acids in millets.

Abstract:The quantitative speciation of selenium in biological systems is highly important for evaluating health status and elucidating transformations of Se species in physiological and pathological processes. Hyphenation of capillary electrophoresis with inductively coupled plasma mass spectrometry (CE-ICPMS) is promising for this purpose. However, the unfavorable or insufficient sensitivity for selenium analysis with CE-ICPMS seriously limits its practical applications in biological analysis, e.g., cell analysis. Therefore, it is crucial to improve the detection sensitivity for Se species. In this study, CE-ICPMS sensitivities for five selenium species (selenocystamine (SeA), methyl-2-acetamido-2-deoxy-1-seleno-β- d -galactopyranoside (SeSug 1), selenomethionine (SeMet), Se-Methylselenocysteine (MeSeCys) and selenocystine (SeCys)) were improved by in-capillary stacking via pH gradient between the zones of sample-leading buffer and the incorporation of isopropanol. The improvement on sensitivity of up to 9.9 folds was achieved in different biological samples, with LODs of 0.29–0.52 μg L −1 . This approach was further applied for Se speciation in cell lysate, urine and culture medium. It showed that SeMet was more readily reduced in the medium and favorably accumulated by HepG2, HuH-7 and HCCLM3 cells with respect to SeSug 1 and MeSeCys. In cells, all the three Se species were largely transformed into other Se species. Furthermore, more than 70 % of SeMet reduced in medium was transformed into unknown Se species after 48-h interaction with cells.

Abstract:The efficient extraction of the cryoablation iceball from a time series of 3D images is crucial during cryoablation to assist the interventionalist in determining the coverage of the tumor by the ablated volume. Conventional semi-automatic segmentation tools such as ITK-SNAP and 3D Slicer's Fast Marching Segmentation can attain accurate iceball segmentation in retrospective studies, however, they are not ideal for intraprocedure real time segmentation, as they require time-consuming manual operations, such as the input of fiducials and the extent of the segmented region growth. In this paper, we present an innovative approach for the segmentation of the iceball during cryoablation, that executes a fully automatic computation. Our approach is based on the graph cuts segmentation framework, and incorporates prior information of iceball shape evolving in time, modeled using experimentally-derived iceball growth parameters. Modeling yields a shape prior mask image at each timepoint of the imaging time series for use in the segmentation. Segmentation results of our method and the ITK-SNAP method are compared for 8 timepoints in 2 cases. The results indicate that our fully automatic approach is accurate, robust and highly efficient compared to manual and semi-automatic approaches.

Abstract:Different ionic liquids (ILs) were assayed as mobile phase modifiers for the separation and determination of selenite [Se(IV)], selenate [Se(VI)], selenomethionine (SeMet) and Se-methylselenocysteine (SeMeSeCys) by reversed-phase high-performance liquid chromatography coupled to hydride generation atomic fluorescence spectrometry (RP-HPLC-HG-AFS). The use of several ILs: 1-butyl-3-methylimidazolium chloride, 1-hexyl-3-methylimidazolium chloride ([C6mim]Cl), 1-octyl-3-methylimidazolium chloride, 1-dodecyl-3-methylimidazolium bromide, 1-hexadecyl-3-methylimidazolium bromide and tributyl(methyl)phosphonium methylsulphate was evaluated. Also, the effect of pH, buffer type and IL concentration on the separation of Se species was studied. Complete separation was attained within 12min using a C8 column and a gradient performed with a mobile phase containing 0.1% (v/v) [C6mim]Cl at pH 6.0. The proposed method allows the separation of inorganic and organic Se species in a single chromatographic run, adding further benefits over already reported methods based on RP-HPLC. In addition, the influence of ILs on the AFS signals of each Se species was evaluated and a multivariate methodology was used for optimization of AFS sensitivity. The limits of detection were 0.92, 0.86, 1.41 and 1.19μgL-1 for Se(IV), Se(VI), SeMet and SeMeSeCys, respectively. The method was successfully applied for speciation analysis of Se in complex samples, such as wine, beer, yeast and garlic.

Abstract:This paper presents a new method for the simultaneous speciation analysis of arsenic (As(III)-arsenite, As(V)-arsenate, DMA-dimethylarsinic acid, MMA-methylarsonic acid, and AsB-arsenobetaine) and selenium (Se(IV)-selenite, Se(VI)-selenate, Se-Methionine, and Se-Cystine), which was applied to a variety of seafood and onion samples. The determination of the forms of arsenic and selenium was undertaken using the High-Performance Liquid Chromatography Inductively Coupled Plasma Mass Spectrometry (HPLC–ICP–MS) analytical technique. The separation of both organic and inorganic forms of arsenic and selenium was performed using two analytical columns: an anion exchange column, Dionex IonPac AS22, containing an alkanol quaternary ammonium ion, and a double bed cation–anion exchange guard column, Dionex Ion Pac CG5A, containing, as a first layer, fully sulfonated latex for cation exchange and a fully aminated layer for anion exchange as the second layer. The ammonium nitrate, at pH = 9.0, was used as a mobile phase. The method presented here allowed us to separate the As and Se species within 10 min with a suitable resolution. The applicability was presented with different sample matrix types: seafood and onion.

Abstract:IMPORTANCE The bone-anchored hearing device (BAHD) was not introduced in China until 2010. To our knowledge, this is the first study to assess the efficacy of Chinese Mandarin-speaking patients with bilateral aural atresia. OBJECTIVE To evaluate the speech recognition of Chinese Mandarin-speaking patients with BAHDs as well as patients' satisfaction using 2 questionnaires. DESIGN, SETTING, AND PARTICIPANTS A retrospective case review of 16 patients with bilateral aural atresia conducted at a tertiary referral center. INTERVENTION A BAHD was implanted during auricle reconstruction surgery or after the auricle was rebuilt. A surgical method to combine the BAHD implantation with the second stage of ear reconstruction was introduced. MAIN OUTCOMES AND MEASURES Speech audiometry test and mean pure-tone threshold results were compared among patients with unaided hearing and those with BAHDs. Scores from the BAHD user questionnaire and Glasgow Children's Benefit Inventory (GCBI) were used to measure patients' satisfaction and subjective health benefit. RESULTS The mean (SD) speech discrimination scores measured in a sound field with a presentation level of 45 dB HL (hearing level) were 6.7% (7.4%) unaided and 86.5% (4.4%) with a BAHD. Scores with a presentation level of 65 dB HL were 56.5% (7.4%) unaided and 90.1% (3.4%) with a BAHD. The speech reception threshold was 60.6 (7.5) dB HL unaided and 24.7 (5.0) dB HL with a BAHD. The mean (SD) pure-tone threshold of the patients was 61.6 (7.8) dB HL unaided and 23.8 (5.9) dB HL with a BAHD. The BAHD application questionnaire demonstrated excellent patient satisfaction. The mean (SD) benefit score of GCBI was 45.6 (14.4). CONCLUSIONS AND RELEVANCE For aural atresia, the BAHD has been one of the most reliable methods of auditory rehabilitation. It can improve the patient's word recognition performance and quality of life. The technique of BAHD implantation combined with auricular reconstruction in a 2-stages-in-1 surgery and the modified incision of patients with reconstructed auricle proved to be safe and effective.
Stability of selenium and its speciation analysis in water using automatic system separation and HR-ICP-MS measurement

Abstract:A simple and convenient method has been developed for the pre-concentration and separation of inorganic selenium species from environmental water samples using anion exchange chromatographic column combined with high resolution inductively coupled plasma mass spectrometry (HR-ICP-MS) measurement. 75Se(IV) and 75Se(VI) were prepared and used as tracers during the experiments. The volatility of selenium during solution evaporation was investigated to establish a reliable water samples pretreatment procedure. The parameters which affect the uptake of Se(IV) and Se(VI) on Dowex1 × 8 resin was optimized and the procedure for Se(IV) and Se(VI) separation was proposed. Both Se(IV) and Se(VI) are retained on the column in natural or alkaline solution with high distribution coefficient. The successive gradient elution of pre-concentrated species of selenium with HNO3 solution allows to differentiate between them. Se(IV) and Se(VI) finally were eluted with 0.05 mol/L HNO3 and 5.0 mol/L HNO3, respectively. The proposed method has been successfully verified using the certified reference materials (CRMs) of real water samples, and spiked recoveries for real samples were 98%-104% with 5% relative standard deviations (RSDs). The developed procedure is proved to be reliable and can be used for the rapid determination of selenium species in environmental water samples.

Abstract:The role of calcium ions in the process by which nutrients affect glucagon secretion by pancreatic islet α-cells remains the matter of an apparently endless debate. In the prolongation of recent articles dealing with this matter, the present review draws attention to the dual role of Ca2+ as revealed by prior publications. In such a perspective, emphasis is placed on the increase in glucagon output in response to the omission of extracellular Ca2+ as recorded in the presence of D-glucose or 2-ketoisocaproate, the permissive role of extracellular Ca2+ in the positive secretory response to arginine or a mixture of fumarate, glutamate and pyruvate, and the effects of an organic calcium-antagonist on glucagon output. Considering the role currently ascribed to Ca2+ in the activation of motile events involved in stimulus-secretion coupling, attention is also given to the effects of cytochalasin B, D2O and mitotic-spindle inhibitors upon the secretory response of α-cells exposed to D-glucose in the absence or presence of arginine.
Speciation of selenomethionine and selenocystine using online micro-column containing Cu(II) loaded nanometer-sized Al2O3 coupled with ICP-MS detection

Abstract:A flow injection online speciation procedure by using micro-column packed with Cu(II) loaded nanometer-sized Al(2)O(3) coupled to inductively coupled plasma mass spectrometry (ICP-MS) for the separation and determination of selenomethionine (SeMet) and selenocystine (SeCys(2)) has been developed. The main factors affecting the separation and preconcentration of SeMet and SeCys(2) including pH value, sample flow rate, eluent concentration, eluent volume and flow rate, and interfering ions have been investigated. It was found that SeCys(2) could be selectively retained by micro-column packed with Cu(II) loaded nanometer-sized Al(2)O(3) at pH 4.0, and the retained SeCys(2) could be eluted by 1.0 mol L(-1) HNO(3), while SeMet was not retained and passed through the micro-column directly at this pH. Both SeMet and SeCys(2) could be quantitatively adsorbed by the micro-column at pH 9.0, and the retained SeMet and SeCys(2) could be easily eluted with 1.0 mol L(-1) HNO(3). The content of SeMet was obtained by subtracting the SeCys(2) from the total content of seleno amino acids. With the enrichment factor of 7.8 and 7.7, the limits of detection (LODs) for SeMet and SeCys(2) were found to be 24 pg Se mL(-1) and 21 pg Se mL(-1), respectively. The relative standard deviations (RSDs) for SeCys(2) and SeMet with seven replicate determinations of 1.0 ng mL(-1) SeMet and SeCys(2), were 2.1% and 1.6%, respectively, the sampling frequency of 8h(-1) was obtained. The proposed method was applied to the speciation of SeMet and SeCys(2) in selenized yeast, human urine and serum with satisfactory results.

Abstract:A hyphenated method for mercury speciation analysis by the coupling of high performance liquid chromatography and inductively coupled plasma mass spectrometry with the online strong anion exchange column (SAX) preconcentration was developed. The Hg analytes (Hg(+), MeHg, EtHg and Hg(2+)) were absorbed on the SAX column preconditioned with sodium 3-mercapto-1-propanesulfonate, and then rapidly eluted (less than 16s) by 5μL 3% (v/v) 2-mercaptoethanol. The enrichment factors of 1025 for Hg(+), 1084 for MeHg, 1108 for EtHg and 1046 for Hg(2+) were obtained using 6mL sample in a 1.5-min enrichment procedure. Rapid separation of the four mercurial compounds was achieved within 5min on a 50-mm C18 column using 0.5% (v/v) 2-mercaptoethanol as the mobile phase. The detection limits for Hg(+), MeHg, EtHg and Hg(2+) were 0.015, 0.010, 0.009 and 0.016ng L(-1), each, and the relative standard deviations of peak height and peak area (5ng L(-1) for each Hg species) were all below 5%. Mercury speciation in three freshwater, two drinking water and two seawater samples were then analyzed by the proposed method. MeHg and Hg(2+) concentrations down to 0.14 and 0.56ng L(-1) were detected in the drinking waters.

Abstract:Matrix metalloproteinases (MMPs) are a family of enzymes that may be implicated in the pathogenesis of inflammatory demyelinating disorders such as multiple sclerosis. The present study investigated the expression of 92‐kd gelatinase (MMP‐9) and five other MMPs in sciatic nerve from Lewis rats with autoimmune experimental neuritis (EAN), an experimental model of the Guillain‐Barre syndrome(GBS). Quantitative polymerase chain reaction analysis revealed an up‐regulation of MMP‐9 mRNA with peak levels concurrent with maximal disease severity. Increased mRNA expression was associated with enhaced enzyme activity, as detected by gelatin zymography. Immunohistochemically, MMP‐9 could be localized primarily around blood vessels within the epineurium and enhoneurium in diseased but not normal sciatic nerve. Among all other MMPs investigated, mRNA levels of matrilysin (MMP‐7) were found to be up‐regulated at the peak of the disorder, remaining at high levels throughout the clinical recovery phase of the disease. To apply these findings to human disease, sural nerve biopsies from GBS patients were examined. By using immunohistochemistry, positive immunoreactivity against MMP‐9 and MMP‐7 was noted and corroborated by demonstrating augmented mRNA expression in comparison with noninflammatory neuropathies. Furthermore, increased MMP‐9 activity was detected by zymography. These findings indicate that 92‐kd gelatinase and matrilysin are selectively up‐regulated during EAN and expressed in nerves of GBS patients and thus may contribute to the pathogenesis of inflammatory demyelination of the peripheral nervous system.
