What problem does this paper attempt to address?
### What problem does this paper attempt to solve? This paper aims to solve the problem of unexpected large - scale genome rearrangements (such as large deletions, LDs) often caused during the CRISPR - Cas9 gene - editing process. These large deletions may lead to potential safety risks, especially in clinical applications. However, there is currently a lack of effective methods to mitigate these risks. Specifically, the authors found through research that the microhomology - mediated end joining (MMEJ) DNA repair pathway plays a key role in a large number of large deletions caused by CRISPR - Cas9. They regulated genes related to MMEJ through genetic and pharmacological means and analyzed the impact of these regulations on the frequency of large deletions induced by CRISPR - Cas9. Eventually, they proposed several new strategies to reduce large deletions and enhance homology - directed repair (HDR), thus providing a new direction for safer and more precise gene editing. ### Main findings 1. **The role of MMEJ in large deletions**: Through long - read single - molecule sequencing (IDMseq), the authors found that the MMEJ pathway dominates in large deletions induced by CRISPR - Cas9. 2. **The role of POLQ**: Reducing the level or activity of DNA polymerase θ (POLQ) significantly reduces the occurrence of large deletions. 3. **The role of RPA**: Reducing the expression of replication protein A (RPA) increases the frequency of large deletions, while over - expressing RPA reduces the frequency of large deletions. 4. **The effect of small - molecule inhibitors**: Using small - molecule inhibitors to inhibit POLQ or delivering recombinant RPA protein can significantly promote HDR efficiency. 5. **Safety verification**: These strategies not only reduce large deletions but also do not affect the overall editing efficiency, ensuring the safety and accuracy of gene editing. ### Conclusion By regulating key factors in the MMEJ pathway (such as POLQ and RPA), large deletions in the CRISPR - Cas9 gene - editing process can be effectively reduced and HDR efficiency can be improved, thus providing a new method for safer and more precise gene editing. ### Key formulas - **Calculation of large - deletion frequency**: \[ \text{Large - deletion frequency}=\frac{\text{Number of large - deletion events}}{\text{Total number of events}} \] - **Calculation of MH frequency**: \[ \text{MH frequency}=\frac{\text{Number of deletion events containing MH}}{\text{Total number of deletion events}} \] These findings provide important theoretical support and technical improvement directions for the application of CRISPR - Cas9 gene - editing technology.
