Abstract:The progression of cardiovascular diseases is accompanied by myocardial injury and necrosis, heart failure, and inflammatory response. Accordingly, ultrasensitive and rapid detection of multiple biomarkers plays a vital role in clinical diagnosis and timely treatment. Here, we developed a novel 2 nanocomposite self-assembled microfluidic immunoassay biochip with digital signal output and applied it to the simultaneous detection of multiple serum biomarkers including inflammatory factors and cardiovascular biomarkers, PCT, CRP, IL6, cTnI, cTnT, and NT-BNP, with high throughput and sensitivity. The digital output signal was collected in the solid phase on the chip surface with two-dimensional distribution of targets. 2 nanocomposites self-assembled biochips could simultaneously detect all six biomarkers in 60 samples in 40 min with detection limit of a few to tens of pg/mL for all serum biomarkers. The microfluidic biochip based on 2 nanocomposites provides a promising method in applications for clinical diagnosis. The Supporting Information is available free of charge at https://pubs.acs.org/doi/10.1021/acs.analchem.1c05061.Self-assembly process of 2 substrate schematics of biosensing platform and detailed chip fabrication process; two functional components of microchip platform for multiplexed profiling of biomarkers; UV–vis spectra of MoS2, HAuCl4·4H2O, and 2; detection mechanism in construction process of sensor biochip; uniformity of substrate; fluorescence scan under different pH conditions; quenching efficiency; fluorescence mapping of detecting markers in patients and healthy people; statistical comparison and fluorescence intensity of biomarkers in serum of healthy and patients; biomarkers detection results of real serum samples from acute coronary syndrome (ACS) patients on biochip in comparison with the clinical results by chemiluminescence immunoassay; calculated detection limit of biomarkers; normal range of different biomarkers in human serum; and properties of different representative biomarker detection platforms (PDF)This article has not yet been cited by other publications.
