Abstract:Background: LipoxinA4(LXA4) is an endogenous anti-inflammatory material and provided with strong ability to fight inflammation. The oxidative stress and NLRP3 inflammasome activation induced by MSU crystals show an essential role in the process of gouty arthritis, however, whether LXA4 can suppress this pathological response is still unexplored. The present study sought to investigate the inhibitory effect of LXA4 on the oxidative stress and NLRP3 inflammasome activation induced by MSU crystals and the specific mechanisms based on negatively regulating Nrf2. Methods: We cultured RAW264.7 cells and differentiated THP-1 macrophages to establish inflammatory model induced by MSU crystals. Then, ELISA, Western blotting, Fluorescent staining and Immunocoprecipitation were adopted to explore the oxidative stress and NLRP3 inflammasome activation. In addition, Immunofluorescence staining, qRT-PCR, Gene silencing, Molecular docking and Drug affinity responsive target stability were employed to study the specific mechanisms of negatively regulating Nrf2. In the end, the animal experiments were performed to confirm the therapeutic effect of LXA4 on gouty arthritis.Findings: We found that LXA4 can inhibit NLRP3 inflammasome activation and oxidative stress in macrophages stimulated with MSU crystals. More specifically, LXA4 suppressed the IL-1β production, oligomerization and speck formation of ASC and ASC-NLRP3 interaction as well as ruined the ROS generation from activated NADPH oxidase and damaged mitochondrial. Importantly, further mechanistic studies demonstrated that LXA4 negatively regulate Nrf2 via directly interacting with Keap1 and Klf9/TXNRD2 signal axis is the downstream molecules after LXA4 suppressing Nrf2, and LXA4 alleviates MSU crystal-induced oxidative stress and NLRP3 inflammasome activation via regulating Nrf2/Klf9/TXNRD2 signaling pathway. Furthermore, LXA4 could alleviate joint inflammation and cleaved Caspase-1 and matured IL-1β production in gouty arthritis rats.Interpretation: Overall, our study illuminate the inhibitory effect of LXA4 on the oxidative stress and NLRP3 inflammasome activation induced by MSU crystals and the regulatory mechanisms.
