Abstract:Treatment of hyphae of Neurospora crassa with antifungal plant defensins, i.e. Rs-AFP2 and Dm-AMP1 isolated from radish and dahlia seed, respectively, induced a rapid K+ efflux, Ca2+ uptake, and alkalinization of the incubation medium. The Rs-AFP2-induced alkalinization of the incubation medium could be inhibited with G-protein inhibitors. alpha-Hordothionin, an antifungal thionin from barley seed, caused a sustained increased Ca2+ uptake at subinhibitory concentrations but only a transient increased uptake at inhibitory concentrations. alpha-Hordothionin also caused increased K+ efflux and alkalinization of the medium, but these fluxes occurred more rapidly compared to those caused by plant defensins. Furthermore, alpha-hordothionin caused permeabilization of fungal hyphae to the non-metabolite alpha-aminoisobutyric acid and, in addition, altered the electrical properties of artificial lipid bilayers, consistently leading to rupture of the lipid bilayers. The plant defensins did not form ion-permeable pores in artificial membranes and did not exhibit substantial hyphal membrane permeabilization activity. Our results are consistent with the notion that thionins inhibit fungal growth as a result of direct protein-membrane interactions, whereas plant defensins might act via a different, possibly receptor-mediated, mechanism.
The YSIRK-G/S Motif of Staphylococcal Protein A and Its Role in Efficiency of Signal Peptide Processing

Abstract:ABSTRACT Many surface proteins of pathogenic gram-positive bacteria are linked to the cell wall envelope by a mechanism requiring a C-terminal sorting signal with an LPXTG motif. Surface proteins of Streptococcus pneumoniae harbor another motif, YSIRK-G/S, which is positioned within signal peptides. The signal peptides of some, but not all, of the 20 surface proteins of Staphylococcus aureus carry a YSIRK-G/S motif, whereas those of surface proteins of Listeria monocytogenes and Bacillus anthracis do not. To determine whether the YSIRK-G/S motif is required for the secretion or cell wall anchoring of surface proteins, we analyzed variants of staphylococcal protein A, an immunoglobulin binding protein with an LPXTG sorting signal. Deletion of the YSIR sequence or replacement of G or S significantly reduced the rate of signal peptide processing of protein A precursors. In contrast, cell wall anchoring or the functional display of protein A was not affected. The fusion of cell wall sorting signals to reporter proteins bearing N-terminal signal peptides with or without the YSIRK-G/S motif resulted in hybrid proteins that were anchored in a manner similar to that of wild-type protein A. The requirement of the YSIRK-G/S motif for efficient secretion implies the existence of a specialized mode of substrate recognition by the secretion pathway of gram-positive cocci. It seems, however, that this mechanism is not essential for surface protein anchoring to the cell wall envelope.

Abstract:A Medicago truncatula antifungal defensin MtDef4 binds to ribosomes of Botrytis cinerea and inhibits translation in vitro. Due to rapidly emerging resistance to single‐site fungicides in fungal pathogens of plants, there is a burgeoning need for safe and multisite fungicides. Plant antifungal peptides with multisite modes of action (MoA) have potential as bioinspired fungicides. Medicago truncatula defensin MtDef4 was previously reported to exhibit potent antifungal activity against fungal pathogens. Its MoA involves plasma membrane disruption and binding to intracellular targets. However, specific biochemical processes inhibited by this defensin and causing cell death have not been determined. Here, we show that MtDef4 exhibited potent antifungal activity against Botrytis cinerea. It induced severe plasma membrane and organelle irregularities in the germlings of this pathogen. It bound to fungal ribosomes and inhibited protein translation in vitro. A MtDef4 variant lacking antifungal activity exhibited greatly reduced protein translation inhibitory activity. A cation‐tolerant MtDef4 variant was generated that bound to β‐glucan of the fungal cell wall with higher affinity than MtDef4. It also conferred a greater reduction in the grey mould disease symptoms than MtDef4 when applied exogenously on Nicotiana benthamiana plants, tomato fruits and rose petals. Our findings revealed inhibition of protein synthesis as a likely target of MtDef4 and the potential of its cation‐tolerant variant as a peptide‐based fungicide.

Abstract:Aesculus hippocastanum antimicrobial protein 1 (Ah-AMP1) is a plant defensin isolated from horse chestnuts. The plant defensins have been divided in several subfamilies according to their amino acid sequence homology. Ah-AMP1, belonging to subfamily A2, inhibits growth of a broad range of fungi. So far, a three-dimensional structure has been determined only for members of subfamilies A3 and B2. In order to understand activity and specificity of these plant defensins, the structure of a protein belonging to subfamily A2 is needed. We report the three-dimensional solution structure of Ah-AMP1 as determined from two-dimensional 1H nuclear magnetic resonance data. The structure features all the characteristics of the "cysteine-stabilized alpha beta-motif." A comparison of the structure, the electrostatic potential surface and regions important for interaction with the fungal receptor, is made with Rs-AFP1 (plant defensin of subfamily A3). Thus, residues important for activity and specificity have been assigned.

Abstract:An antifungal peptide with a defensin-like sequence and exhibiting a molecular mass of 7.3kDa was purified from dried seeds of Phaseolus vulgaris 'Cloud Bean'. The isolation procedure entailed anion exchange chromatography on DEAE-cellulose, affinity chromatography an Affi-gel blue gel, cation exchange chromatography on SP-Sepharose, and gel filtration by fast protein liquid chromatography on Superdex 75. Although the antifungal peptide was unadsorbed on DEAE-cellulose, it was adsorbed on both Affi-gel blue gel and SP-Sepharose. The antifungal peptide exerted antifungal activity against Mycosphaerella arachidicola with an IC(50) value of 1.8 μM. It was also active against Fusarium oxysporum with an IC(50) value of 2.2 μM. It had no inhibitory effect on HIV-1 reverse transcriptase when tested up to 100 μM. Proliferation of L1210 mouse leukemia cells and MBL2 lymphoma cells was inhibited by the antifungal peptide with an IC(50) of 10 μM and 40 μM, respectively.

Overexpression and evaluation of an antimicrobial peptide from Heuchera sanguinea (Hs-AFP1) for inhibition of fungal pathogens in transgenic tabacco

Abstract:A plant defensin from Heuchera sanguinea, Hs-AFP1, shows high levels of in vitro activity against a broad range of fungi. In an effort to evaluate the possible application of Hs-AFP1 in the genetic transformation of crop plants to improve disease resistance, the Hs-AFP1 peptide was studied in transgenic tobacco. The gene encoding mature Hs-AFP1 was fused to a signal peptide from Mirabilis jalapa and was introduced into tobacco via Agrobacterium transformation. Southern blot analysis conducted on the resulting transgenic tobacco lines confirmed Hs-AFP1 gene integration. Hs-AFP1 was also transcribed actively and the presence of the transgenic protein was confirmed by Western blot analysis. Due to the welldocumented instability of small peptides on the plant surface, a fusion between the Hs-AFP1 peptide and the antifungal exoglucanase gene, EXG1, from Saccharomyces cerevisiae was prepared and also introduced into tobacco. This fusion protein was also targeted to the apoplastic region with the signal peptide from M. jalapa. Northern blot analyses conducted on the resulting transgenic tobacco lines confirmed transcription of the fusion construct, whereas Western blot analyses confirmed the presence of the fusion protein. The exoglucanase activity of the fusion construct was evaluated with a l3-glucosidase assay. All the transgenic lines that showed transcription of the various constructs were subjected to Botrytis cinerea inhibition and infection studies. Elevated levels of resistance towards B. cinerea were observed in several of the transgenic lines, reaching inhibition levels of 35-45%.

Fungicidal and binding properties of three plant peptides

Abstract:The fungicidal properties of plant seed peptides from Heuchera sanginea (Hs-AFP1), Raphanus sativus (EA-AFP2), and Impatiens balsamina (Ib-AMP3) were determined for the non-germinated and germinated conidia of Aspergillus flavus and Fusarium moniliforme. These peptides were weakly lethal for germinated but not for non-germinated conidia of A. flavus. Both non-germinated and germinated conidia of F. moniliforme were susceptible to these peptides. Overall, F. moniliforme was more susceptible than A. flavus to the peptides. The peptides bound strongly to chitin, mannan, galactocerebrosides, and sphingomyelin. Binding results varied for ergosterol, cholesterol, and beta1,3-glucan.

Abstract:Plant defensins are a large family of small cationic proteins with diverse functions and mechanisms of action, most of which assert antifungal activity against a broad spectrum of fungi. The partial mechanism of action has been resolved for a small number of members of plant defensins, and studies have revealed that many act by more than one mechanism. The plant defensin Ppdef1 has a unique sequence and long loop 5 with fungicidal activity against a range of human fungal pathogens, but little is known about its mechanism of action. We screened the S. cerevisiae non-essential gene deletion library and identified the involvement of the mitochondria in the mechanism of action of Ppdef1. Further analysis revealed that the hyperpolarisation of the mitochondrial membrane potential (MMP) activates ROS production, vacuolar fusion and cell death and is an important step in the mechanism of action of Ppdef1, and it is likely that a similar mechanism acts in Trichophyton rubrum.

Abstract:The 5'AMP-activated protein kinase (AMPK) is a potential antidiabetic drug target. Here we show that the pharmacological activation of AMPK by 5-aminoimidazole-1-beta-4-carboxamide ribofuranoside (AICAR) leads to inactivation of glycogen synthase (GS) and phosphorylation of GS at Ser 7 (site 2). In muscle of mice with targeted deletion of the alpha2-AMPK gene, phosphorylation of GS site 2 was decreased under basal conditions and unchanged by AICAR treatment. In contrast, in alpha1-AMPK knockout mice, the response to AICAR was normal. Fuel surplus (glucose loading) decreased AMPK activation by AICAR, but the phosphorylation of the downstream targets acetyl-CoA carboxylase-beta and GS was normal. Fractionation studies suggest that this suppression of AMPK activation was not a direct consequence of AMPK association with membranes or glycogen, because AMPK was phosphorylated to a greater extent in response to AICAR in the membrane/glycogen fraction than in the cytosolic fraction. Thus, the downstream action of AMPK in response to AICAR was unaffected by glucose loading, whereas the action of the kinase upstream of AMPK, as judged by AMPK phosphorylation, was decreased. The fact that alpha2-AMPK is a GS kinase that inactivates GS while simultaneously activating glucose transport suggests that a balanced view on the suitability for AMPK as an antidiabetic drug target should be taken.

However, mounting fungal resistance to many of the single-site chemical fungicides calls for the development of new antifungal agents with novel modes of action (MoA). The sequence-divergent cysteine-rich antifungal defensins with multi-site MoA are promising starting templates for design of novel peptide-based fungicides. Here, we experimentally tested such a set of 17-amino acid peptides containing the γ-core motif of the antifungal plant defensin MtDef4. These designed peptides exhibited antifungal properties different from those of MtDef4. Focused analysis of a lead peptide, GMA4CG_V6, showed it was a random coil in solution with little or no secondary structure elements. Additionally, it exhibited potent cation-tolerant antifungal activity against the plant fungal pathogen Botrytis cinerea, causal agent of gray mold disease in fruits and vegetables. Its multi-site MoA involved localization predominantly to the plasma membrane, permeabilization of the plasma membrane, rapid internalization into the vacuole and cytoplasm, and affinity for bioactive phosphoinositides phosphatidylinositol 3-phosphate (PI3P), PI4P, and PI5P. The sequence motif RRRW was identified as a major determinant of the antifungal activity of this peptide. While topical spray-application of GMA4CG_V6 on Nicotiana benthamiana and tomato plants provided preventative and curative suppression of gray mold disease symptoms, the peptide was not internalized into plant cells. Our findings open the possibility that truncated and modified defensin-derived peptides containing the γ-core sequence could serve as promising candidates for further development as bioinspired fungicides.

Plant defensin antibacterial mode of action against Pseudomonas species

Abstract:Abstract Background Though many plant defensins exhibit antibacterial activity, little is known about their antibacterial mode of action (MOA). Antimicrobial peptides with a characterized MOA induce the expression of multiple bacterial outer membrane modifications, which are required for resistance to these membrane-targeting peptides. Mini-Tn 5-lux mutant strains of Pseudomonas aeruginosa with Tn insertions disrupting outer membrane protective modifications were assessed for sensitivity against plant defensin peptides. These transcriptional lux reporter strains were also evaluated for lux gene expression in response to sublethal plant defensin exposure. Also, a plant pathogen, Pseudomonas syringae pv. syringae was modified through transposon mutagenesis to create mutants that are resistant to in vitro MtDef4 treatments. Results Plant defensins displayed specific and potent antibacterial activity against strains of P. aeruginosa . A defensin from Medicago truncatula , MtDef4, induced dose-dependent gene expression of the aminoarabinose modification of LPS and surface polycation spermidine production operons. The ability for MtDef4 to damage bacterial outer membranes was also verified visually through fluorescent microscopy. Another defensin from M. truncatula , MtDef5, failed to induce lux gene expression and limited outer membrane damage was detected with fluorescent microscopy. The transposon insertion site on MtDef4 resistant P. syringae pv. syringae mutants was sequenced, and modifications of ribosomal genes were identified to contribute to enhanced resistance to plant defensin treatments. Conclusions MtDef4 damages the outer membrane similar to polymyxin B, which stimulates antimicrobial peptide resistance mechanisms to plant defensins. MtDef5, appears to have a different antibacterial MOA. Additionally, the MtDef4 antibacterial mode of action may also involve inhibition of translation.

Abstract:Antibiotic resistance.is one of the biggest challenges sciences faces today. It is extremely urgent to develop new antimicrobial compounds to control infections in the "post-antibiotic era”. Plant defensins belong to a large family of small cationic antimicrobial peptides and are an integral part of the innate immune system of plants. The gene coding for the mature peptide NmDef02 (isolated from Nicotiana megalosiphon) was cloned into a pSMT3 vector, generating the plasmid pSMT3-DEF02, used to express the protein SUMO-NmDef02 in Escherichia coli, strain Shuffle T7 Express lysY. The soluble chimeric protein was purified by Ni–NTA affinity chromatography and cleaved by the UPL1 protease. The sample was re-applied to a Ni–NTA and approximately 20 mg of NmDef02 was obtained from the fermentation of 1 L of E. coli culture. The purified proteins were analyzed by SDS-PAGE under reduction condition and its identity was confirmed by Western blotting, using anti-histidine and anti-NmDef02 antibodies. NmDef02 defensin showed antimicrobial activity against plant and human pathogens. The recombinant fusion strategy could be an approach to produce bioactive recombinant NmDef02.

Fractal topology of hand-crumpled paper.

Abstract:We study the statistical topology of folding configurations of hand folded paper balls. Specifically, we are studying the distribution of two sides of the sheet along the ball surface and the distribution of sheet fragments when the ball is cut in half. We found that patterns obtained by mapping of ball surface into unfolded flat sheet exhibit the fractal properties characterized by two fractal dimensions which are independent on the sheet size and the ball diameter. The mosaic patterns obtained by sheet reconstruction from fragments of two parts (painted in two different colors) of the ball cut in half also possess a fractal scale invariance characterized by the box fractal dimension DBF=1.68 ± 0.04 , which is independent on the sheet size. Furthermore, we noted that DBF, at least numerically, coincide with the universal fractal dimension of the intersection of hand folded paper ball with a plane. Some other fractal properties of folding configurations are recognized.

Abstract:Two antifungal peptides, named EAFP1 and EAFP2, have been purified from the bark of Eucommia ulmoides Oliv. Each of the sequences consists of 41 residues with a N-terminal blockage by pyroglutamic acid determined by automated Edman degradation in combination with the tandem mass spectroscopy and the C-terminal ladder sequencing analysis. The primary structurs all contain 10 cysteines, which are cross-linked to form five disulfide bridges with a pairing pattern (C1–C5, C2–C9, C3–C6, C4–C7, C8–C10). This is the first finding of a plant antifungal peptide with a five-disulfide motif. EAFP1 and EAFP2 show characteristics of hevein domain and exhibit chitin-binding properties similar to the previously identified hevein-like peptides. They exhibit relatively broad spectra of antifungal activities against eight pathogenic fungi from cotton, wheat, potato, tomato and tobacco. The inhibition activity of EAFP1 and EAFP2 can be effective on both chitin-containing and chitin-free fungi. The values of IC50 range from 35 to 155 μg/ml for EAFP1 and 18 to 109 μg/ml for EAFP2. Their antifungal effects are strongly antagonized by calcium ions.
Salt-Tolerant Antifungal and Antibacterial Activities of the Corn Defensin ZmD32

Abstract:Pathogenic microbes are developing resistance to established antibiotics, making the development of novel antimicrobial molecules paramount. One major resource for discovery of antimicrobials is the arsenal of innate immunity molecules that are part of the first line of pathogen defense in many organisms. Gene encoded cationic antimicrobial peptides are a major constituent of innate immune arsenals. Many of these peptides exhibit potent antimicrobial activity in vitro. However, a major hurdle that has impeded their development for use in the clinic is the loss of activity at physiological salt concentrations, attributed to weakening of the electrostatic interactions between the cationic peptide and anionic surfaces of the microbial cells in the presence of salt. Using plant defensins we have investigated the relationship between the charge of an antimicrobial peptide and its activity in media with elevated salt concentrations. Plant defensins are a large class of antifungal peptides that have remarkable stability at extremes of pH and temperature as well as resistance to protease digestion. A search of a database of over 1200 plant defensins identified ZmD32, a defensin from Zea mays, with a predicted charge of +10.1 at pH 7, the highest of any defensin in the database. Recombinant ZmD32 retained activity against a range of fungal species in media containing elevated concentrations of salt. In addition, ZmD32 was active against Candida albicans biofilms as well as both Gram negative and Gram-positive bacteria. This broad spectrum antimicrobial activity, combined with a low toxicity on human cells make ZmD32 an attractive lead for development of future antimicrobial molecules.

Abstract:AbstractThe wide use of antibiotics can lead to antimicrobial resistance. Biosurfactant rhamnolipids are not prone to induce antimicrobial resistance due to their potential to target microbial membranes in both bacteria and fungi. However, the membrane interaction of anionic rhamnolipids with microbials has never been visually observed. This study applied previously synthesized anionic fluorescent Trisodium 8‐hydroxypyrene‐1,3,6‐trisulfonate (HPTS)‐based surfactants, with hydrophobic tails from C8 to C18, to examine their interaction with the plant pathogen Alternaria alternata. The antifungal potency of the surfactants exhibited a positive correlation with the increase in hydrophobic chain length, culminating in C16, which demonstrated the highest activity. Subsequently, a modest reduction in potency was noted for the C18. Fluorescence microscopy confirmed the membrane targeting of HPTS‐C16, and molecular dynamics simulations supported its specific membrane interaction. HPTS‐C16 was notably more effective against spores than mycelium, consistent with its binding affinity. This research provides a fluorescent method to dissect the relationship between surfactant properties and microbial membrane characteristics, guiding the development of antimicrobials to combat resistance.
Aspergillus fumigatus-Related Species in Clinical Practice

Abstract:Aspergillus fumigatus is the main etiologic agent of invasive aspergillosis (IA). Other Aspergillus species belonging to the section Fumigati (A. fumigatus complex) may occasionally be the cause of IA. These strains are often misidentified, as they cannot be distinguished from A. fumigatus by conventional morphological analysis and sequencing methods. This lack of recognition may have important consequences as these A. fumigatus-related species often display some level of intrinsic resistance to azoles and other antifungal drugs. A. lentulus, A. udagawae, A. viridinutans, and A. thermomutatus (Neosartorya pseudofischeri) have been associated with refractory cases of IA. Microbiologists should be able to suspect the presence of these cryptic species behind a putative A. fumigatus isolate on the basis of some simple characteristics, such as defect in sporulation and/or unusual antifungal susceptibility profile. However, definitive species identification requires specific sequencing analyses of the beta-tubulin or calmodulin genes, which are not available in most laboratories. Multiplex PCR assays or matrix-assisted laser desorption ionization – time-of-flight mass spectrometry (MALDI-TOF MS) gave promising results for rapid and accurate distinction between A. fumigatus and other Aspergillus spp. of the section Fumigati in clinical practice. Improved diagnostic procedures and antifungal susceptibility testing may be helpful for the early detection and management of these particular IA cases.
Solution Structure of PAFP-S: A New Knottin-Type Antifungal Peptide from the Seeds of Phytolacca Americana,

Abstract:The three-dimensional solution structure of PAFP-S, an antifungal peptide extracted from the seeds of Phytolacca americana, was determined using 1H NMR spectroscopy. This cationic peptide contains 38 amino acid residues. Its structure was determined from 302 distance restraints and 36 dihedral restraints derived from NOEs and coupling constants. The peptide has six cysteines involved in three disulfide bonds. The previously unassigned parings have now been determined from NMR data. The solution structure of PAFP-S is presented as a set of 20 structures using ab initio dynamic simulated annealing, with an average RMS deviation of 1.68 A for the backbone heavy atoms and 2.19 A for all heavy atoms, respectively. For the well-defined triple-stranded beta-sheet involving residues 8-10, 23-27, and 32-36, the corresponding values were 0.39 and 1.25 A. The global fold involves a cystine-knotted three-stranded antiparallel beta-sheet (residues 8-10, 23-27, 32-36), a flexible loop (residues 14-19), and four beta-reverse turns (residues 4-8, 11-14, 19-22, 28-32). This structure features all the characteristics of the knottin fold. It is the first structural model of an antifungal peptide that adopts a knottin-type structure. PAFP-S has an extended hydrophobic surface comprised of residues Tyr23, Phe25, Ile27, Tyr32, and Val34. The side chains of these residues are well-defined in the NMR structure. Several hydrophilic and positively charged residues (Arg9, Arg38, and Lys36) surround the hydrophobic surface, giving PAFP-S an amphiphilic character which would be the main structural basis of its biological function.

Abstract:Defensins are small cysteine-rich antimicrobial peptides that are widely distributed in plants, insects and mammals. As potent defenders in protecting plants from pathogenic fungal attack, plant defensins are presumed to play an important role in the innate immunity of plants and are expected to find applications in the production of transgenic crops. A novel plant defensin protein SPE10 from Pachyrrhizus erosus seeds was purified and partially sequenced. Crystallization screening using the hanging-drop vapour-diffusion method resulted in two crystal forms, from one of which a diffraction data set was collected to 0.98 A resolution. The crystal belongs to space group P2(1), with unit-cell parameters a = 32.71, b = 28.11, c = 54.85 angstroms, beta = 103.78 degrees. Preliminary crystallographic studies revealed two subunits in the asymmetric unit.
