Abstract:BackgroundPopulus (poplar) tree species including hybrid varieties are considered as promising biomass feedstock for biofuels and biochemicals production due to their fast growing, short vegetative cycle, and widely distribution. In this work, poplar was pretreated with acetic acid (AC) to produce xylooligosaccharides (XOS), and hydrogen peroxide–acetic acid (HPAC) was used to remove residual lignin in AC-pretreated poplar for enzymatic hydrolysis. The aim of this work is to produce XOS and monosaccharides from poplar by a two-step pretreatment method.ResultsThe optimal conditions for the AC pretreatment were 170 °C, 5% AC, and 30 min, giving a XOS yield of 55.8%. The optimal HPAC pretreatment conditions were 60 °C, 2 h, and 80% HPAC, resulting in 92.7% delignification and 87.8% cellulose retention in the AC-pretreated poplar. The two step-treated poplar presented 86.6% glucose yield and 89.0% xylose yield by enzymatic hydrolysis with a cellulases loading of 7.2 m/g dry mass. Very high glucose (93.8%) and xylose (94.6%) yields were obtained with 14.3 mg cellulases/g dry mass. Both Tween 80 and β-glucosidase enhanced glucose yield of HPAC-pretreated poplar by alleviating the accumulation of cellobiose. Under the optimal conditions, 6.9 g XOS, 40.3 g glucose, and 8.9 g xylose were produced from 100 g poplar.ConclusionsThe AC and HPAC pretreatment of poplar represented an efficient strategy to produce XOS and fermentable sugars with high yields. This two-step pretreatment was a recyclable benign and advantageous scheme for biorefinery of the poplar into XOS and monosaccharides.

Abstract:Herein, a short-time alkaline enhanced aqueous DES (AaDES) pretreatment using choline chloride/ethylene glycol was reported, aiming at enhancing cellulose and xylan enzymatic digestibility. Simultaneously, saccharification efficiency of cellulose and xylan was reached to 91.2% and 99.0%, respectively, ∼4 and ∼24 times that of raw poplar. Pretreatment time was substantially shortened from 15-24 h to 4 h. Notably, 43.00 kg fermentable sugars (73% of the theoretical maximum) and 12.98 kg lignin with rich β-O-4' linkages were obtained based on 100 kg poplar. The complete removal of acetyl and partial removal of lignin and mannan contributed to excellent pretreatment performance. It was found that enzymatic digestibility of xylan/cellulose was positively correlated with removal of mannan (R2 = 0.9719; R2 = 0.9010) and delignification (R2 = 0.6888; R2 = 0.8293). Drastic reduction in pretreatment time along with high-yield sugars in AaDES system will provide strength towards industrial level biorefinery.

Abstract:BACKGROUND: Future biorefineries will focus on converting low value waste streams to chemical products that are derived from petroleum or refined sugars. Feedstock pretreatment in a simple, cost effective, agnostic manner is a major challenge.METHODS: In this work, beechwood sawdust was delignified via an organosolv process, assisted by homogeneous inorganic acid catalysis. Mixtures of water and several organic solvents were evaluated for their performance. Specifically, ethanol (EtOH), acetone (AC), and methyl- isobutyl- ketone (MIBK) were tested with or without the use of homogeneous acid catalysis employing sulfuric, phosphoric, and oxalic acids under relatively mild temperature of 175 °C for one hour.RESULTS: Delignification degrees (DD) higher than 90% were achieved, where both AC and EtOH proved to be suitable solvents for this process. Both oxalic and especially phosphoric acid proved to be good alternative catalysts for replacing sulfuric acid. High gravity simultaneous saccharification and fermentation with an enzyme loading of 8.4 mg/gsolids at 20 wt.% initial solids content reached an ethanol yield of 8.0 w/v%.CONCLUSIONS: Efficient delignification combining common volatile solvents and mild acid catalysis allowed for the production of ethanol at high concentration in an efficient manner.

Abstract:Cellulosic ethanol is the key technology to alleviate the pressure of energy supply and climate change. However, the ethanol production process, which is close to industrial production and has a high saccharification rate and ethanol yield, still needs to be developed. This study demonstrates the effective conversion of poplar wood waste into fuel-grade ethanol. By employing a two-step pretreatment using sodium chlorite (SC)-dilute sulfuric acid (DSA), the raw material achieved a sugar conversion rate exceeding 85% of the theoretical value. Under optimized conditions, brewing yeast co-utilizing C6/C5 enabled a yield of 35 g/L ethanol from 10% solid loading delignified poplar hydrolysate. We increased the solid loading to enhance the final ethanol concentration and optimized both the hydrolysis and fermentation stages. With 20% solid loading delignified poplar hydrolysate, the final ethanol concentration reached 60 g/L, a 71.4% increase from the 10% solid loading. Our work incorporates the pretreatment, enzymatic hydrolysis, and fermentation stages to establish a simple, crude poplar waste fuel ethanol process, expanding the range of feedstocks for second-generation fuel ethanol production.

Abstract:Lignocellulosic hydrolysates contain acetic acid derived from hemicellulose. Acetate released during acid pretreatment interferes with enzymatic saccharification of the biomass and inhibits yeast fermentation, caused by cellular oxidation and stress. This study utilized the alkaline deacetylation prior to acid pretreatment and evaluated its influence on the fermentation performance of Saccharomyces cerevisiae using two different lignocellulosic biomasses, hemp (Cannabis sativa) and kenaf (Hibiscus cannabinus). They are valuable for biofuel production. The maximal acetate removal with minimal sugar loss was obtained with 1% (w/v) NaOH treatment for both biomasses. Deacetylation pretreatment under this condition was effective both in enzymatic saccharification and simultaneous saccharification & fermentation. This resulted in improved ethanol production by xylose‐consuming yeast (S. cerevisiae SR8) by removing 4.7 g/L of acetate from hemp and 6.3 g/L from kenaf, thus resulting in the ethanol productivities of 0.79 and 0.67 g/L‐hr from hemp and kenaf, respectively. This suggests that strategies to overcome acetate inhibition are necessary to efficiently utilize the lignocellulosic biomass as a source for bioenergy production.

Abstract:The large quantities of Lycoris radiata Herbert (Amarylllidaceae) residues, left after the extraction of alkaloids from the bulbs, could threaten the environment if not properly disposed. Therefore, the aim of this research is to investigate the feasibility of bioconversion of L. radiata Herbert residues to ethanol through batch fermentation. In L. radiata Herbert residues, the average contents (g kg(-1)) of non-structural carbohydrates, crude fiber, crude protein, ash, and lignin are 485.1, 177.3, 124.7, 108.9, and 91.0, respectively. Five commercial enzymes (beta-glucanase, pectinase, xylanase, phytase and cellulase) were employed in pretreatment process and ethanol bioconversion was evaluated with three yeasts (Saccharomyces cerevisiae, osmotolerant S. cerevisiae and genetically engineered S. cerevisiae constructed to use cellobiose). The results showed that pretreament by beta-glucanase successfully facilitated the penetration of alpha-amylase into ground material. After residues pretreatment by 10 g kg (1) of beta-glucanase for 14 h at 50 degrees C, the viscosity decreased from 1135 to 59 Pas which was equivalent to that obtained by the combined whole five enzymes. Further experiments proved that osmotolerant S. cerevisiae was desirable for ethanol production from Lycoris radiate Herbert residues. Abstract:Choosing hydrolyzate of poplar as fermentation material,which was obtained by two-step super-low acids hydrolysis,fuel ethanol production was studied by using Candida tropicalis 2.637 and Saccharomyces cerevisiae 2.699 respectively.Simul-taneously the influences to the ethanol yield by various pretreatment methods,such as Ca(OH)2 neutralization,over-neutralization,active carbon adsorption and yeast-training were carried out.The experimental results show that C.tropicalis 2.637 is suitable to the first-step hydrolyzate rich in pentose to produce single cell protein.Yeast-training can improve fermentation speed and increase ethanol yield,especially to the second step hydrolyzate through which ethanol yield was increased from 14.45 % to 39.37 % by S.cerevisiae 2.699.The over-neutralization of hydrolyzate by Ca(OH)2 improved ethanol yield,too.Therefore,for the hydrolysis of the first-step hydrolyzate,which contains mainly pentose and accompanied with hexose,more ethanol can be obtained by C.tropicalis 2.637 than by S.cerevisiae 2.699.

Abstract:An integrated pretreatment that realizing effective fractionation and targeted valorization can guarantee the implementability to future biorefinery scenario. In the present study, a stepwise approach using acetic acid-catalyzed hydrothermal pretreatment (AAP) and rapid ball-milling process was successfully developed for producing high-yield of xylo-oligosaccharides (XOS), fermentable glucose and native lignin from poplar wood. It was found that the AAP removed the most hemicelluloses, releasing the highest yield (36.16%) of XOS at 160 °C 、for 1 h. Subsequent rapid ball-milling pretreatment for 10 min led to a remarkable increase in the saccharification efficiency from 40.68% to 91.35%. Meanwhile, the residual lignin fractions exhibited abundant β- O -4 linkages, which are beneficial to catalytic hydrogenolysis of these lignin fractions into the aromatic chemicals. Considering the integrated process could effectively produce XOS, fermentable glucose, and the residual lignin with well-preserved structures, the proposed process is a promising and efficient deconstruction strategy in current biorefinery scenario.

Selective delignification of poplar wood with a newly isolated white-rot basidiomycete Peniophora incarnata T-7 by submerged fermentation to enhance saccharification

Abstract:Background: Pretreatment is a critical step required for efficient conversion of woody biomass into biofuels and platform chemicals. Fungal pretreatment is regarded as one of the most promising technology for woody biomass conversion but remains challenging for industrial application. The exploration of potential fungus strain with high efficient delignification and less processing time for woody biomass pretreatment will be valuable for development of biorefinery industry. Here, a newly isolated white-rot basidiomycete Peniophora incarnate T-7 was employed for poplar wood pretreatment. Results: The chemical component analysis showed that cellulose, hemicellulose and lignin from poplar wood declined by 16%, 48% and 70%, respectively, after 7 days submerged fermentation by P. incarnate T-7. Enzymatic saccharification analysis revealed that the maximum yields of glucose and xylose from 7 days of P. incarnate T-7 treated poplar wood reached 33.4% and 27.6%, respectively, both of which were enhanced by sevenfold relative to the untreated group. Fourier transform infrared spectroscopy (FTIR), scanning electron microscope (SEM), X-ray diffraction (XRD) and pyrolysis gas chromatography-mass spectrometry (Py-GC/MS) characterization confirmed that lignocellulosic structure of poplar wood was largely broken by P. incarnate T-7, including delignification and de-crystalline of cellulose. Meanwhile, lignin component of poplar wood was selectively degraded by P. incarnate T-7, and G-type unit of lignin was preferentially attacked by the strain. Furthermore, quantitative proteomic analysis revealed that a considerable amount of lignocellulolytic enzymes were detected in the secretory proteins of P. incarnate T-7, especially with high abundance of lignin-degrading enzymes and hemicellulases. Combination of quantitative proteomic with transcriptomic analysis results showed that most of those lignocellulolytic enzymes were highly upregulated on poplar wood substrate compared to glucose substrate. Conclusions: This study showed that P. incarnate T-7 could selectively delignify poplar wood by submerged fermentation with short time of 7 days, which greatly improved its enzymatic saccharification efficiency. Our results suggested that P. incarnate T-7 might be a promising candidate for industrial woody biomass pretreatment.
Conversion of control and genetically-modified poplar into multi-scale products using integrated pretreatments

Abstract:In this work, a green and robust pretreatment which integrated acetic acid-catalyzed hydrothermal and wet mechanical pretreatment, was developed to efficiently produce high yield (up to 40.12%) of xylooligosaccharides and digestible substrates from Caffeoyl Shikimate Esterase down-regulated and control poplar wood. Subsequently, superhigh yield (more than 95%) of glucose and residual lignin were obtained after a moderate enzymatic hydrolysis. The residual lignin fraction exhibited a well-preserved β-O-4 linkages (42.06/100Ar) and high S/G ratio (6.42). Subsequently, lignin-derived porous carbon was successfully synthesized, and it exhibited a high specific capacitance of 273.8 F g-1 at 1.0 A g-1 and long cycling stability (remained 98.5% after 10,000 cycles at 5.0 A g-1) compared to control poplar wood, demonstrating that special advantage of this genetically-modified poplar in this integrated process. This work developed an energy-saving and eco-friendly pretreatment technology as a waste-free route for converting different lignocellulosic biomass to multiple products.

Abstract:The work in this article describes a spectral signature for the detection of a C(α) radical damaged peptide, which should enable the use of infrared spectroscopic methods to directly monitor oxidative events. Spectra for radical damaged peptides are computed with ab initio methods. The amide bands A, I, II, and III are analyzed for trends in the damage site. The spectral signature is found in a region (i.e., 1700-1620 cm(-1)) normally void of vibrational absorption bands from stable undamaged beta peptides. An analysis of the vibrational motions of the spectral signature is described. The uniqueness of the spectral signature is explored by an examination and comparison with C(α) monoradicals and polyradicals, as well as with other bioradicals that could act as spectral interferences. The identification of unique infrared spectral features for C(α) damage could have important implications in diagnostics for beta conformational peptides damaged by oxidative stress processes.
Enhancing biomass conversion: Efficient hemicellulose removal and cellulose saccharification in poplar with FeCl 3 coupled with acidic electrolyzed water pretreatment

Abstract:Due to the recalcitrant structure of woody biomass such as poplar, the efficient disassembly and separation of hemicellulose component from woody biomass is crucial for green biomass processing and full component utilization. This study presented an environmentally friendly approach to utilize acidic electrolyzed water (AEW) combined with metal salts and investigated its pretreatment effects on hemicellulose removal and cellulose and lignin retention under different conditions. Meanwhile, the structural properties and enzymatic hydrolysis performance of the pretreated residues were also characterized. As a result, under the optimized pretreatment conditions (0.03 mol/L FeCl 3 with AEW at 180 °C for 10 min), hemicellulose removal from poplar wood reached 98.64 %, accompanied by xylose recovery rate of 98.46 %, cellulose retention rate of 93.43 % and lignin retention rate of 94.29 %. Enzymatic hydrolysis rate of the pretreated cellulose-enriched substrate reached 97.65 %. Furthermore, comprehensive structural characterizations revealed that FeCl 3 coupled with AEW pretreatment resulted in surface damage to the poplar wood, effective removal of the amorphous hemicellulose component, and partial destruction of the cellulose crystallinity. In conclusion, FeCl 3 coupled with AEW pretreatment effectively separates hemicellulose, leading to significant alterations in biomass composition and structure, ultimately resulting in improved enzymatic digestion. These results provide theoretical support for targeted dissociation of hemicellulose and full component utilization of woody biomass.

Abstract:Concentrated acid hydrolysis (CAH) of lignocellulosic biomass (LCB) is an established method to obtain a near theoretical yield of monomeric sugars from the biomass. The CAH process produced 83.23 % of glucose from pretreated wheat straw (PWS) at optimum conditions (i.e., stage I = 74 % w/w acid, 1:2 biomass loading, and 50 °C for 1 h; and stage II = 30 % w/w acid, 90 °C for 2 h). However, the separation of sugar from acidic hydrolysate is a major constraint of this technology. In this study, an electro-electrodialysis (EED) process using a novel anion exchange membrane (AEM) has been developed to recover sulfuric acid from biomass hydrolysate. The synthetic hydrolysate (i.e., feed acid concentration = 2 M; glucose = 80 g L −1 ) was used to optimize the EED process parameters. The acid was successfully recovered from alkali pretreated wheat straw (PWS) hydrolysate using the most effective EED process parameters. This resulted in an 82.72 % acid recovery in the permeate under the following conditions: current density of 88 mA cm −2 , flow rate of 3 L h −1 , and feed acid content of 2 M. During the EED process, the highest glucose recovery in the feed was 90.98 %, achieved with a maximum current efficiency of 93.57 % and the lowest energy consumption of 0.052 kW h mol −1 at optimal conditions using PWS Hydrolysate. Saccharomyces cerevisiae fermented the acid-free PWS hydrolysate, resulting in a maximum ethanol yield of 92.3 %. The EED process cost of $0.049 kg H 2 SO 4 −1 and total revenue was $0.02 kg biomass −1 from the second batch onwards suggest the feasibility of the EED process for large-scale 2G bioethanol productions.

Abstract:The biorefinery concept makes use of renewable lignocellulosic biomass to produce commodities sustainably. A synthetic microbial consortium can enable the simultaneous utilization of sugars such as glucose and xylose to produce biochemicals, where each consortium member converts one sugar into the target product. In this study, woody biomass was used to generate glucose and xylose after pretreatment with 20% (w/v) sulfuric acid and 60-min reaction time. We compared several strategies for detoxification with charcoal and sodium borohydride treatments to improve the fermentability of this hydrolysate in a defined medium for the production of the growth-associated product pyruvate. In shake flask culture, the highest pyruvate yield on xylose of 0.8 g/g was found using pH 6 charcoal-treated hydrolysate. In bioreactor studies, a consortium of two engineered E. coli strains converted the mixture of glucose and xylose in batch studies to 12.8 ± 2.7 g/L pyruvate in 13 h. These results demonstrate that lignocellulosic biomass as the sole carbon source can be used to produce growth-related products after employing suitable detoxification strategies.

Abstract:As green and effective depolymerization processes, CrCl 3 and CrCl 3 -catalyzed ethanol pretreatment were comprehensively investigated to improve cellulose conversion from pretreated residues in poplar in the present study. The influence of pretreatment temperature on delignification , structural characteristics of lignin, and the yield of glucose and ethanol was comparatively analyzed to emphasize the synergy effect of CrCl 3 and ethanol solvent. Particularly, CrCl 3 -catalyzed ethanol pretreatment at 200 °C resulted in the pretreated substrates with >94% removal of hemicellulose and lignin, consequently reaching 88.88% and 56.11% yields of glucose and ethanol, respectively. Lignin's structural change was observed by Gel permeation chromatography and 2D nuclear magnetic resonance spectroscopy and its relationship with the efficiency of enzymatic hydrolysis and fermentation was determined. Results indicated that the increased pretreatment temperature enhanced the lignin depolymerization with smaller molecular weight, fewer β-O-4 linkages, and higher S/G ratio, especially in CrCl 3 -catalyzed ethanol pretreatment, which was positively associated with poplar cellulose conversion. In addition to offering a comprehensive illustration of the sugar/ethanol generation during CrCl 3 -catalyzed organosolv pretreatment, this work also provided theoretical support for biofuel and biochemical production from poplar.

Abstract:Pretreatment using high concentration (>60 %, v/v) of hydrogen peroxide-acetic acid (HPAA) exhibits highly selective and strong delignification capacity. However, the utilization of high concentrations of HPAA is costly and potentially explosive. Although low HPAA concentration is safer, it limits the improvement of the hydrolyzability of poplar. Herein, after the production of xylo-oligosaccharides from poplar by acetic acid (AC) hydrolysis, the obtained lignin-rich (35.6 %) poplar was pretreated with low concentration (10–20 %) of HPAA followed by sodium hydroxide-urea (SU) to enhance the hydrolyzability of AC-hydrolyzed poplar. Firstly, 37.0 % lignin and 93.2 % acetyl groups were removed from AC-hydrolyzed poplar via pretreatment using 10 % HPAA at 120 °C for 10 min. Then, SU pretreatment removed 35.2 % lignin of HPAA-pretreated poplar, which enhanced the glucose yield of the poplar from 21.4 % to 95.5 %. This HPAA-SU process saved 90 % HPAA loading and largely improved the hydrolyzability of lignin-rich poplar in a relatively short time.

Abstract:Abstract Lignocellulosic biomass is an attractive renewable resource to produce biofuel or platform chemicals. Efficient and cost-effective conversion systems of lignocellulosic biomass depend on their appropriate pretreatment processes. Alkali or dilute acid pretreatment of biomass requires a high temperature (> 150 °C) to remove xylan (hemicellulosic sugar) and lignin partially. In this study, peracetic acid was used to pretreat biomass feedstocks, including hardwood and softwood species. It was found that the thermally-assisted dilute acid pretreatment of biomass conducted under the mild temperature of 90 °C up to 5 h resulted in the effective removal of lignin from the biomass with a negligible loss of carbohydrates. This thermally-assisted pretreatment achieved 90% of delignification, and this result was compared with the microwave-assisted pretreatment method. In addition, the crystallinity index ( CrI ), surface morphology, and chemical structure were significantly changed after the acid pretreatment. The biomass digestibility increased significantly with increased reaction time, by 32% and 23% for hardwood and softwood, respectively. From this study, it is clear that peracetic acid pretreatment is an effective method to enrich glucan content in biomass by delignification.

Abstract:Most of the current commercial production of glacial acetic acid (GAA) is by petrochemical routes, primarily methanol carbonylation. GAA is an intermediate in the production of plastics, textiles, dyes, and paints. GAA production from biomass might be an economically viable and sustainable alternative to petroleum-derived routes. Separation of acetic acid from water is a major expense and requires considerable energy. This study evaluates and compares the technical and economic feasibility of GAA production via bioconversion using either ethyl acetate or alamine in diisobutylkerosene (DIBK) as organic solvents for purification. Models of a GAA biorefinery with a production of 120,650 tons/year were simulated in Aspen software. This biorefinery follows the path of pretreatment, enzymatic hydrolysis, acetogen fermentation, and acid purification. Estimated capital costs for different scenarios ranged from USD 186 to 245 million. Recovery of GGA using alamine/DIBK was a more economical process and consumed 64% less energy, due to lower steam demand in the recovery distillation columns. The estimated average minimum selling prices of GGA were USD 756 and 877/ton for alamine/DIBK and ethyl acetate scenarios, respectively. This work establishes a feasible and sustainable approach to produce GGA from poplar biomass via fermentation.
