Abstract:The mechanisms and chemo- and regioselectivities of Ru(II)-catalyzed decarboxylative C-H alkenylation of aryl carboxylic acids with alkynes were investigated with density functional theory (DFT) calculations. The catalytic cycle involves sequential carboxylate-directed C-H activation, alkyne insertion, decarboxylation and protonation. The facile tether-assisted decarboxylation step directs the intermediate toward the desired decarboxylative alkenylation, instead of typical annulation and double alkenylation pathways. The decarboxylation barrier is very sensitive to the tether length, and only the seven-membered ring intermediate can selectively undergo the designed decarboxylation, suggesting a tether-dependent chemoselectivity. This tether-dependent chemoselectivity also applies to the alkyl tethers. In addition, the polarity of solvent is found to control the chemoselectivity between the decarboxylative alkenylation and [4 + 2] annulation. Solvent with low polarity (toluene) favors the decarboxylation pathway, leading to the decarboxylative alkenylation. Solvent with high polarity (methanol) favors the ionic stepwise C-O reductive elimination pathway, leading to the [4 + 2] annulation. To understand the origins of regioselectivity with asymmetric alkynes, the distortion/interaction analysis was applied to the alkyne insertion transition states, and led to a predictive frontier molecular orbital model. The asymmetric alkynes selectively use the terminal with the larger HOMO orbital coefficient to form the C-C bond in the insertion step.

Abstract:Density functional theory (DFT) calculations were performed to investigate the detailed mechanisms of metal-catalyzed reactions of alkynyl thioethers with isoxazoles/anthranils. The origins of the product selectivities were also explored. The results show that when [IPrAu(CH3CN)]+ was used as the catalyst, β-selective addition gave a sulfenylated pyrrole/indole as the primary product. However, the major product was a sulfur-substituted β-keto enamide, which was formed via α-addition, when [Zn(OTf)]+ was used. The product selectivity was determined by the first nucleophilic attack step. We found that the catalyst and substrate both played crucial roles in guiding the product selectivity. The electronic effects of ligands on the catalyst and substituents on the substrate were the dominant factor. For the anthranil substrate, the preference for C1–C6 bond coupling rather than C1–C4 bond coupling can be attributed to effective conjugation between the carbonyl group and the phenyl moiety during C1–C6 bond coupling. These insights into the mechanisms of metal-catalyzed reactions of alkynyl thioethers with isoxazoles/anthranils will facilitate the design of new catalytic reactions of isoxazoles with electron-deficient alkynes.

Rhodium(III)-Catalyzed Asymmetric Borylative Cyclization of Cyclohexadienone-Containing 1,6-Dienes: An Experimental and DFT Study.

Abstract:Because of the inherent difficulty in differentiating two olefins, the development of metal-catalyzed asymmetric cyclization of 1,6-dienes remains challenging. Herein, we describe the first rhodium(III)-catalyzed asymmetric borylative cyclization of cyclohexadienone-tethered mono-, 1,1di-, and (E)-1,2-disubstituted alkenes (1,6-dienes), affording optically pure cis-bicyclic skeletons bearing three or four contiguous stereocenters with high yields (25-93%), and excellent diastereoselectivities (>20:1 dr) and enantioselectivities (90-99% ee). This mild catalytic approach is generally compatible with a wide range of functional groups, which allows several facile conversions of the cyclization products. Furthermore, on the basis of our SAESI-MS experiment and computational study, a Rh(I)/(III) catalytic cycle is proposed in this tandem reaction, and the Rh(I) active species catalyzes the overall transformation via sequential oxidative addition of B(2)pin(2), olefin insertion, cyclizing conjugate addition, and reductive elimination. The irreversible conjugate addition determines the overall regioselectivity of borylative cyclization, and the ring strain favors the formation of 5,6-bicyclic structure. This highlights the control of ring strain in diene cyclizations, which provides a useful basis for future reaction designs.

Abstract:The Rh(I)-catalyzed reaction of siloxyvinylcyclopropanes and diazoesters leads to the formation of siloxyvinylcyclobutane and 1,4-diene derivatives. With [Rh(cod)Cl]2 as the catalyst, the formation of 1,4-diene was favored over the formation of siloxyvinylcyclobutane. By changing the catalyst to [Rh(cod)2OTf], siloxyvinylcyclobutane derivatives are formed with excellent chemoselectivities and in moderate to good yields. The alkene products are also obtained as single E configured isomers. A detailed mechanism for this transformation is proposed on the basis of mechanistic experiments and DFT calculations. The effect of catalysts on the chemoselectivity of these reactions is also examined computationally.

Abstract:The mechanism, solvent effects, and origins of selectivities in Ru(II)-catalyzed intramolecular (5+2) cycloaddition and ene reaction of vinylcyclopropanes (VCPs) and alkynes have been studied using density functional theory. B3LYP/6-31G(d)/LANL2DZ optimized structures were further evaluated with the M06 functional, 6-311+G(2d,p) and LANL2DZ basis sets, and the SMD solvent model. The favored mechanism involves an initial ene-yne oxidative cyclization to form a ruthenacyclopentene intermediate. This mechanism is different from that found earlier with rhodium catalysts. The subsequent β-hydride elimination and cyclopropane cleavage are competitive, determining the experimental selectivity. In trans-VCP, the cyclopropane cleavage is intrinsically favored and leads to the (5+2) cycloaddition product. Although the same intrinsic preferences occur with the cis-VCP, an unfavorable rotation is required in order to generate the cis-double bond in seven-membered ring product, which reverses the selectivity. Acetone solvent is found to facilitate the acetonitrile dissociation from the precatalyst, destabilizing the resting state of the catalyst and leading to a lower overall reaction barrier. In addition, the origins of diastereoselectivities when the allylic hydroxyl group is trans to the bridgehead hydrogen are found to be the electrostatic interactions. In the pathway that generates the favored diastereomer, the oxygen lone pairs from the substituent are closer to the cationic catalyst center and provide stabilizing electrostatic interactions. Similar pathways also determine the regioselectivities, that is, whether the more or less substituted C-C bond of cyclopropane is cleaved. In the trans-1,2-disubstitued cyclopropane substrate, the substituent from the cyclopropane is away from the reaction center in both pathways, and low regioselectivity is found. In contrast, the cleavage of the more substituted C-C bond of the cis-1,2-disubstituted cyclopropane has steric repulsions from the substituent, and thus higher regioselectivity is found.

Abstract:A rhodium-catalyzed dehydrogenative cycloisomerization of dienylcyclopropane compounds is reported, which provides a straightforward approach to a variety of highly substituted toluene derivatives in 67-85% yields. The dienylcyclopropaneimides are produced by a single-step formal three-component olefination procedure. Preliminary mechanistic studies indicated that an electron-withdrawing group as R plays a critical role in completing this transformation.

Abstract:The mechanism and origins of selectivities in [Ni(NHC)]-catalyzed intramolecular (5 + 2) cycloadditions and homo-ene reactions of vinylcyclopropanes (VCPs) and alkynes have been studied using density functional theory. The preferred mechanism involves oxidative alkyne-alkene cyclization to form a metallacyclopentene intermediate, in contrast to a cyclopropane cleavage pathway in the reaction with Rh(I) catalysts. The selectivity between the (5 + 2) and homo-ene products is determined in the subsequent competing reductive elimination and β-hydride elimination steps. Two similar-sized N-heterocyclic carbene (NHC) ligands, SIPr and ItBu, yielded reversed product selectivity, favoring the (5 + 2) and homo-ene products respectively. This is attributed to the anisotropic steric environment of these NHC ligands, which positions the bulky substituents on the ligand toward different directions and leads to distinct steric control in the reductive elimination and β-hydride elimination transition states.

The whole process of the current reaction involves the nucleophilic cyclic oxyrhodation/ stereodefined carborhodation/ beta-H elimination/ reductive elimination. Calculated results reveal that the carborhodation step is not only the rate-limiting step but also determines the stereo-selectivity.

Abstract:We report a cobalt-catalyzed desymmetrizing isomerization of exo-cyclic alkenes to generate chiral 1-methylcyclohexene derivatives with good yields and enantioselectivities. A novel chiral thiazolinyl iminoquinoline ligand and its cobalt complex were designed and synthesized to control the establishment of tertiary or quaternary carbon centers at a remote position. This protocol is operationally simple, and a model for the stereochemical outcome has been proposed.

Abstract:Combined computational and experimental studies elucidated the distinctive mechanistic features of electrochemical cobalt-catalyzed C-H oxygenation. A sequential electrochemical-chemical (EC) process was identified for the formation of an amidylcobalt(iii) intermediate. The synthesis, characterization, cyclic voltammetry studies, and stoichiometric reactions of the related amidylcobalt(iii) intermediate suggested that a second on-cycle electro-oxidation occurs on the amidylcobalt(iii) species, which leads to a formal Co(iv) intermediate. This amidylcobalt(iv) intermediate is essentially a cobalt(iii) complex with one additional single electron distributed on the coordinating heteroatoms. The radical nature of the coordinating pivalate allows the formal Co(iv) intermediate to undergo a novel carboxylate-assisted HAT mechanism to cleave the arene C-H bond, and a CMD mechanism could be excluded for a Co(iii/i) catalytic scenario. The mechanistic understanding of electrochemical cobalt-catalyzed C-H bond activation highlights the multi-tasking electro-oxidation and the underexplored reaction channels in electrochemical transition metal catalysis.

Abstract:The dirhodium(II) complexes with bridging phosphine and OAc ligands showed high reactivity and selectivities in olefin dehydrosilylation. In order to determine the structure of the actural catalyst which cannot be determined experimentally, the geometries of the dirhodium catalyst, the detailed catalytic mechanism, as well as the stereo- and chemoselectivities of the title reaction were studied by DFT calculations. The results showed that one OAc group is monodentate and the other is bidentate in the dirhodium catalyst C'. The determined catalytic cycle consists of four processes: Rh-H bond activation in C', Si-H bond activation in alkoxysilane, alkylene insertion into Rh-Si bond, followed by β-H elimination or σ-metathesis reaction. Among them, the alkylene insertion process is the rate-determining state. The stereoselectivity of the title reaction is controlled by the steric effect and orbital interactions between the alkyene and dirhodium catalysts in the process of β-H elimination process. The chemoselectivity is regulated by the presence of the axial ligand in the dirhodium catalyst, when there is an axial ligand coordinated to the Rh atom, E-alkene is the main product, whereas alkane would be obtained in the absence of axial ligand. Our work determines the structure of the actural catalyst, and provides explanations and predictions for the activity, chemo- and stereoselectivity control of olefin dehydrosilylation.

Abstract:Olefin isomerization is a significant transformation in organic synthesis, which provides a convenient synthetic route for internal olefins and remote functionalization processes. The selectivity of an olefin isomerization process is often thermodynamically controlled. Thus, to achieve selectivity under kinetic control is very challenging. Herein, we report a novel cobalt-catalyzed regioselective olefin isomerization reaction. By taking the advantage of fine-tunable NNP-pincer ligand structures, this catalytic system features high kinetic control of regioselectivity. This mild catalytic system enables the isomerization of 1,1-disubstituted olefins bearing a wide range of functional groups in excellent yields and regioselectivity. The synthetic utility of this transformation was highlighted by the highly selective preparation of a key intermediate for the total synthesis of minfiensine. Moreover, a new strategy was developed to realize the selective monoisomerization of 1-alkenes to 2-alkenes dictated by installing substituents on the gamma-position of the double bonds. Mechanistic studies supported that the in situ generated Co-H species underwent migratory insertion of double bond/beta-H elimination sequence to afford the isomerization product. The less hindered olefin products were always preferred in this cobalt-catalyzed olefin isomerization due to an effective ligand control of the regioselectivity for the beta-H elimination step.

Abstract:A highly enantioselective cobalt-catalyzed desymmetrizing reductive cyclization of alkynyl cyclodiketones has been developed. Under mild reaction conditions by employing HBpin as reducing agent and ferrocene-based PHOX as chiral ligand, a series of polycyclic tertiary allylic alcohols bearing contiguous quaternary stereocenters are achieved in moderate to excellent yields with excellent enantioselectivities (up to 99%). Broad substrate scope and high functional group compatibility are observed in this reaction. A CoH-catalyzed pathway involving alkyne hydrocobaltation followed by nucleophilic addition to C=O bond is proposed. Synthetic transformations of the product are conducted to demonstrate the practical utilities of this reaction.

Abstract:DielsAlder reaction between electronically neutral dienes and dienophiles is usually sluggish under thermal conditions and has to be catalyzed by transition metal catalysts. We report here our DFT study of the mechanism and stereochemistry of the Rh-catalyzed DielsAlder reaction between electronically neutral dienes and dienophiles (alkenes and alkynes), finding that this reaction includes a reaction sequence of oxidative cyclization between diene and alkene/alkyne and a reductive elimination step. The alkynes oxidative cyclization is much faster than alkenes due to the additional coordination of alkyne to the Rh center in the oxidative cyclization transition state. For both intermolecular and intramolecular reactions, the reductive elimination step in the catalytic cycle is rate-determining. The different reactivity of ene-diene and yne-diene substrates can be rationalized by the model that reductive elimination to form a C(sp(2))C(sp(3)) bond is easier than that for the formation of a C(sp(3))C(sp(3)) bond, due to the additional coordination of the double bond to the Rh center in the transition state in the former. We also uncovered the reasons for the high para-selectivity of the intermolecular DielsAlder reaction of dienes and alkynes. In addition, DFT calculations aiming to understand the high diastereoselectivity of an intramolecular [4 + 2] reaction of ene-dienes with substituents adjacent to the diene and ene moieties of the substrates found that the substituents in the substrates favor staying away from the Rh center in the oxidative cyclization transition states. This preference leads to the generation of the final [4 + 2] products with the substituents and the bridgehead hydrogen atoms in a cis-configuration.

Abstract:Excellent and unique diastereoselectivity is observed in the iridium-catalyzed deoxygenation of tertiary cyclohexanols and cyclopentanols. The substituent effect on the diastereoselectivity and detailed control models are analyzed case by case, using tertiary monocyclic and polycyclic cyclohexanols, bicyclic bridged cycloalkanols, and cyclopentanols as the model substrates. The selectivity is decided by the steric environment of the carbocation intermediates and is independent of the catalyst loading. Stereoelectronically, the iridium hydride approaches the carbocation in directions perpendicular to the carbocation plane. The sterically large iridium hydride delivers its hydride in the sterically least hindered direction to the carbocation. The deoxygenation has found important applications in the stereospecific arylations of sterically complex compounds. Our deoxygenation is stereochemically very different from the coupling reactions and can be used to specifically synthesize stereoisomers that are not available via cross-couplings.

Abstract:The recent success of Krische et al. ( Angew. Chem., Int. Ed. 2017 , 56 , 14667 -14671 ) in achieving a ruthenium(0)-catalyzed transfer hydrogenative cycloaddition of 1,2-diols with cyclohexadiene and norbornadiene in excellent yield with exo- and diastereoselectivity represents an exciting development in the synthesis of bridged carbocycles. In the present work, the possible catalytic mechanisms and origin of the exo- and diastereoselectivity for cyclohexadiene and norbornadiene were studied in detail by density functional theory calculations. The theoretical results indicate that the exoselective pathway for the cyclohexadiene substrate proceeds by a novel two-step successive C-C coupling, while the endoselective pathway undergoes the C-C coupling reaction in a conventional concerted manner. The origin of the preferential chemoselectivity of dione-cyclohexadiene C-C coupling over aromatization to benzene was investigated. Aromatization to benzene is unfavorable because of the large distortion energy of the three-membered ring in the transition state of hydrogen migration. From distortion/interaction analysis, for norbornadiene, the distortion energy plays the main role in determining the exoselectivity.

Abstract:Crystallographic characterization of RuX(CO)(η3-C3H5)(JOSIPHOS), where X = Cl, Br, or I, reveals a halide-dependent diastereomeric preference that defines metal-centered stereogenicity and, therefrom, the enantioselectivity of C−C coupling in ruthenium-catalyzed anti-diastereo- and enantioselective C−C couplings of primary alcohols with 1-aryl-1-propynes to form products of carbonyl anti-(α-aryl)allylation. Computational studies reveal that a non-classical hydrogen bond between iodide and the aldehyde formyl CH bond stabilizes the favored transition state for carbonyl addition. Abstract:The mechanism and origin of stereoselectivity of rhodium(II)-catalyzed cyclopropanation reactions with diazooxindole and styrene has been studied using density functional theory calculations. The catalyzed reactions by achiral Rh-2(OAc)(4) and chiral Rh-2(S-PTTL)(4) as well as the uncatalyzed model were comparatively studied. The computational results indicate that the cyclopropanation step in both Rh-2(OAc)(4) and Rh-2(S-PTTL)(4) models is a single concerted but asynchronous process. The nitrogen extrusion step is found to be the rate-limiting step of the catalytic cycle, whereas the cyclopropanation step is the stereoselectivity-determining step. The diastereomeric ratios (dr) and the enantiomeric excess (ee) values are successfully predicted, which are in good agreement with the experimental values. The high trans-diastereoselectivity might be governed by the pi-pi interactions between the syn indole ring in carbenoid ligand and the phenyl group in styrene, whereas the good enantioselectivity can be ascribed to the steric interaction between the phenyl ring in styrene and the phthalimido group in the catalyst as well as the aromatic interactions (pi-pi and CH-pi) in the transition states. Additionally, the methodological study using different functionals demonstrated the importance of considering the dispersion interactions in the current reaction systems. This theoretical study will help in understanding the mechanism of the asymmetric cyclopropanations of olefins through carbene-transfer reactions.
