Abstract:Peroxynitrite (ONOO-) has been proven to participate in various physiological and pathological processes, and may also be a contributing factor in many diseases. In view of this, there is a need to develop detection tools for unambiguously tracking a small amount of endogenous ONOO- to reveal its exact mechanisms. In this paper, a colorimetric and red-emitting fluorescent probe Red-PN, based on a rhodamine-type fluorophore and hydrazide reactive site is described. The probe Red-PN possesses the advantages of rapid response (within 5 s), visual color change (from colorless to pink), preeminent sensitivity (detection limit = 4.3 nM) and selectivity. Because of these outstanding performances, it was possible to accurately detect endogenous ONOO-. It was encouraging that the probe Red-PN could be used effectively for tracking the relatively low levels of endogenous and exogenous ONOO-in living cells and zebrafish. Thus, it is envisioned that the probe Red-PN would have promising prospects in applications for imaging ONOO- in a variety of biological settings.

Abstract:In this work, taking full advantage of the intramolecular charge transfer (ICT) mechanism, a hydroxynaphthalimide-based ratiometric two-photon fluorescent probe RTP-PN was synthesized to detect ONOO-. Probe RTP-PN could accurately detect ONOO- in the range of 1.4 nM-1.4 mM with the detection limit of 1.4 nM by a ratiometric fluorescence spectroscopy method. Additionally, probe RTP-PN exhibited an ultrafast response for ONOO- than other various species including H2O2 and ClO-. Abstract:One kind of novel dicyanisophorone-based colorimetric fluorescent probe 3 has been prepared for recognizing peroxynitrite in aqueous condition. Large bathochromic shift of the absorption band is observed upon titration with ONOO–, inducing a clear solution color change from yellow to pale pink which enabling "naked-eye" detection possible. Moreover, probe 3 can react instantly with ONOO–accompanied by significant fluorescence enhancement at 621 nm while the detection limit is as low as 37 nM. Most importantly, probe 3 exhibits high selectivity and sensitivity towards ONOO– over other competitive ions in the aqueous solutions. Probe 3 has also been successfully applied in living MCF-7 cells and the result suggests that probe 3, could be applied as a potential candidate for the detection of ONOO–.

Abstract:Peroxynitrite is a highly reactive molecule involved in cell signaling and pathological processes. We hereby report a novel genetically encoded probe, pnGFP, which can selectively sense peroxynitrite. A boronic acid moiety was site-specifically introduced into circularly permuted fluorescent proteins. By examining different protein templates followed with site-targeted random mutagenesis, we identified a selective peroxynitrite sensor, which is essentially unresponsive to other common cellular redox signaling molecules. The new probe has been genetically introduced into mammalian cells to image peroxynitrite at physiologically relevant concentrations.

Abstract:Based on ESIPT, we designed and synthesized a naphthalimide chemosensor N-CBT for selectively visualizing endo/exogenous peroxynitrite (ONOO-) in living cells. The incorporation of 2-benzothiazoleacetonitrile offers N-CBT a rare pre-existing eight-membered ring hydrogen bonding configuration, which is able to generate two types of emission of naphthalimide. Confirmed by calculation results, fast proton transfer from the hydroxyl group to the carbonyl group occurs along with excited-state energy transfer via the intramolecular H-bond, leading to a tautomeric transformation from the excited enol form to the excited keto form. In aqueous solution, the formation of intermolecular hydrogen bonding with water perturbs ESIPT and destroys the stable planar construction. By breaking the cyano carbon-carbon double bond in the presence of ONOO-, green fluorescence can be regenerated efficiently. As a result, 34-fold fluorescence enhancement at 518 nm was observed in response, and it showed a good linear relationship in the range of 1 to 14 mu M with a detection limit of 37 nM. Subsequently, N-CBT was applied in visualizing cellular ONOO-, and it demonstrated great potential in selectively visualizing endo/exogenous peroxynitrite (ONOO-) in living cells.

Abstract:Significance: Peroxynitrite is a short-lived intracellular redox signaling species that is closely related to a number of diseases. Current detection methods for this anion lack the selectivity over other redox-active species present in the cell. In this report the authors demonstrate the selective detection of peroxynitrite by using an amphiphilic block copolymer through an in situ dioxirane oxidation–fragmentation strategy. Comment: The reaction mechanism involves: 1) in situ dioxirane formation by reaction between a trifluoromethyl ketone and peroxynitrite; 2) intramolecular oxygen atom transfer by oxidizing the electron-rich arene; 3) the resulting peroxide undergoes fragmentation, releasing a hydrophilic carboxylic acid group that causes the self-assembled structures to dissolve. This chemistry enables selective detection of peroxynitrite over a number of other redox-active interferents such as hydrogen peroxide. Abstract:Peroxynitrite is a short-lived endogenous reactive species and plays important roles in many physiological and pathological processes. In this work, we synthesized a near-infrared probe based on the structure of benzothiazole derived cyanine for determination of peroxynitrite (ONOO-). The designed probe specifically reacted with ONOO- through oxidative cleavage of conjugated C˭C double bonds and generating the non-fluorescent product. Meanwhile, the characteristic absorption of the probe at 630 nm greatly decreased after reaction with ONOO-, accompanied by drastic color change from bright blue to green yellow, which exhibited a distinct visual feature. It was demonstrated that the probe could be used to measure ONOO- in a dose-response manner and had a detection limit lower as 26 nM. Furthermore, the probe Cy-SN was applied for the imaging of endogenous ONOO- in living cells by confocal microscopy, which showed good cell permeability and low cytotoxicity. Successful application of probe for exogenous colorimetric detection and endogenous fluorescence imaging of ONOO- is suggesting its great potential applications in biological analysis.

Abstract:Peroxynitrite (ONOO-), a highly reactive oxygen species generated by the reaction of nitric oxide and superoxide radical anion, is involved in numerous physiological and pathological processes in the human body. To identify important pathogenic mechanisms, it is crucial to develop a reliable tool for detecting peroxynitrite in living systems. In the present study, a new difluoroboron β-diketonate-based fluorescent probe for detecting exogenous and endogenous peroxynitrite in living systems was designed. The red emitting fluorophore can be synthesized in a simple three-step procedure. This probe reacts quickly and selectively with peroxynitrite and its detection limit is determined to be as low as 19.8 nM. It allows for clear imaging of peroxynitrite in RAW 264.7 cells and was successfully applied to visualize changes of intracellular peroxynitrite induced by reactive oxygen species inhibitors. This designed probe is an effective tool for investigating the physiological and pathological role of peroxynitrite in living cells.

Abstract:Peroxynitrite is widely present in organisms and closely related to many pathophysiological functions. Therefore, it is of great physiological significance to develop capable probes for detecting ONOO − . In this work, a novel fluorescent probe B-Ch was designed based on the intramolecular charge transfer (ICT) effect. By means of molecular engineering, the replacement from diethylamine group to hydroxyl group has improved the detection sensitivity of the probe. After the addition of ONOO − , the solution color and fluorescence showed noticeable changes, which were visible to the naked eye. The probe showed excellent advantages: visualization, good selectivity, low sensitivity (22.4 nM), good stability and biocompatibility, exogenous and endogenous imaging of ONOO − in HeLa cells.

Abstract:Herein we developed a fast-responsive fluorescent probe 2-(benzo[d]thiazol-2-yl)-4-methylphenol (HMBT)-Py for ONOO- detection. HMBT-Py can react with ONOO- by an activated C=C oxidation cleavage reaction, causing a ratiometric change in the fluorescence spectra. In addition, HMBT-Py showed high selectivity to ONOO- over other reactive species.

Abstract:Peroxynitrite (ONOO(-)) is a highly reactive species implicated in the pathology of numerous diseases and there is currently great interest in developing fluorescent probes that can selectively detect ONOO(-) in living cells. Herein, a polymeric micelle-based and cell-penetrating peptide-coated fluorescent nanoprobe that incorporates ONOO(-) indicator dye and reference dye for the ratiometric detection and imaging of ONOO(-) has been developed. The nanoprobe effectively avoids the influences from enzymatic reaction and high-concentration ·OH and ClO(-). The improved ONOO(-) selectivity of the nanoprobe is achieved by a delicate complementarity of properties between the nanomatrix and the embedded molecular probe (BzSe-Cy). This nanoprobe also has other attractive properties, such as good water solubility, photostability, biocompatibility, and near-infrared excitation and emission. Fluorescence imaging experiments by confocal microscopy show that this nanoprobe is capable of visualizing ONOO(-) produced in living cells and it exhibits very low toxicity and good membrane permeability. We anticipate that this technique will be a potential tool for the precise pathological understanding and diagnosis of ONOO(-)-related human diseases.

Abstract:Peroxynitrite (ONOO−), as one of the important reactive oxygen/nitrogen species, is associated with many physiological and pathological activities. Therefore, effective detection methods with high sensitivity and rapid response time are essential to reveal its functions. A novel Rhodol-based fluorescent probe RN is constructed and synthesized by introducing of 1,1-dimethylhydrazone as a recognition group. Due to the oxidative breakage of the CN bond by ONOO−, the probe has significant fluorescence enhancement change with excellent detection properties such as rapid response time, high sensitivity and selectivity. Moreover, probe RN show low cytotoxicity and is successfully capable of imaging endogenous and endogenous ONOO− in living cells. These properties indicate that RN has great application potential for imaging of ONOO− in biological systems.

Abstract:A water-soluble boronate-based fluorescent probe was evaluated for the detection of peroxynitrite (ONOO-) in the presence of a monosaccharide. The enhanced fluorescence of the probe when bound with o-fructose was switched off in the presence of peroxynitrite. In contrast, other reactive oxygen/nitrogen species (ROS/RNS) led to only slight fluorescence decreases due to protection by an internal N-B interaction. The interaction of the probe with o-fructose not only strengthens the fluorescence signal, but also protects the boronic acid from oxidation by other ROS/RNS. Therefore, under conditions generating various ROS/RNS, the boronate-based saccharide complex preferentially reacts with peroxynitrite. The probe was used in cell imaging experiments for the detection of endogenous and exogenous peroxynitrite. The sensor displays good "on-off" responses towards peroxynitrite, both in RAW 264.7 cells and HeLa cells.

Abstract:The peroxynitrite ion (ONOO-) has important roles in many biological processes. We have developed a multicolor ONOO--sensing probe (SiONNOH) that undergoes deprotonation and desilylation processes, which result in several changes in the emission wavelengths. In response to different concentrations of ONOO-, the probe exhibits fluorescence changes from pink (595 nm at 2 eq. ONOO-) to green (540 nm at 6 eq. ONOO-) via orange (3 eq. ONOO-) and yellow (4 eq. ONOO-) under physiological conditions until no fluorescence signal is observed after ONOO- is completely eliminated by lipoic acid. The probe shows the high selectivity for ONOO- and the limit of detection is calculated to be 1.27 mu M. Moreover, the probe shows the capacity to monitor the concentration ranges of ONOO- through multicolor fluorescence in living cells, which will greatly facilitate the rapid detection of ONOO- concentration ranges by the naked eye under a UV light without any precision instrumentation.

Abstract:Peroxynitrite (ONOO-) plays a crucial role in the regulation of diverse pathophysiological processes, and high level of ONOO- is profound association with numerous diseases. Herein, we developed an anthraquinone-based fluorescent probe L for ONOO- determination by a new recognition mechanism: amido oxidized nitroso-group by ONOO-. Probe L with amine-based recognition receptor is more selective to ONOO- than other reactive oxygen species, including H2O2 and ClO-. Furthermore, ONOO- could be rapidly detected by probe L with a Limit of Detection of 13 nM. More importantly, L could be used to monitor intracellular ONOO- in SMMC-7721 cells. (C) 2020 Elsevier B.V.
