Abstract:Recently, various efforts have been put forward on the development of technologies for the synthesis of methane from CO2 and H2, since it can offer a solution for renewable H2 storage and transportation. In parallel, this reaction is considered to be a critical step in reclaiming oxygen within a closed cycle. Over the years, extensive fundamental research works on CO2 methanation have been investigated and reported in the literatures. In this updated review, we present a comprehensive overview of recent publications during the last 3 years. Various aspects on this reaction system are described in detail, such as thermodynamic considerations, catalyst innovations, the influence of reaction conditions, overall catalytic performance, and reaction mechanism. Finally, the future development of CO2 methanation is discussed.

Abstract:Methane bio-oxidation plays an important role in the global methane balance and greenhouse gases mitigation. Oxygen (O2) is one of the significant factors in methane bio-oxidation. The O2 concentration in the environments not only can affect community structure and activity of aerobic methanotrophs and the distribution of methane-derived carbon, but also aerobic methanotrophs have various ways of metabolism at different O2 concentrations. It is meaningful for carbon cycle and biodiversity of methane-driven ecosystem to understand the hypoxia-tolerance mechanisms of aerobic methanotrophs and methane bio-oxidation in O2-limited environments. In this paper, the activity and community structure of aerobic methanotrophs in O2-limited environments were summarized. The hypoxia-tolerance mechanism of aerobic methanotrophs and the relationship between methanotrophs and non-methanotrophs in O2-limited environments were reviewed. Future studies about aerobic methanotrophs were also discussed.

Abstract:Methane bio-oxidation plays an important role in the global methane balance and warming mitigation, while copper has a crucial function in methane bio-oxidation. On one side, copper is known to be a key factor in regulating the expression of the genes encoding the two forms of methane monooxygenases (MMOs) and is the essential metal element of the particulate methane monooxygenase (pMMO). On the other side, the content and fractionation of copper in the environment have great effects on the distribution of methanotrophs and their metabolic capability of methane and non-methane organic compounds, as well as on the copper-specific uptake systems in methanotrophs. Thus, it is meaningful to know the role of copper in methane bio-oxidation for comprehensive understanding of this process and is valuable for guiding the application of methanotrophs in greenhouse gas removal and pollution remediation. In this paper, the roles of copper in methane oxidation were reviewed, including the effect of copper on methanotrophic community structure and activity, the expression and activity of MMOs as well as the copper uptake systems in methanotrophs. The future studies of copper and methane oxidation were also discussed.

Abstract:Particulate methane monooxygenase (pMMO) is a three-subunit integral membrane enzyme that catalyzes the oxidation of methane to methanol. Although pMMO is the predominant methane oxidation catalyst in nature, it has proved difficult to isolate, and most questions regarding its molecular structure, active site composition, chemical mechanism, and genetic regulation remain unanswered. Copper ions are believed to play a key role in both pMMO regulation and catalysis, and there is some evidence that the enzyme contains iron as well. A number of research groups have solubilized and purified or partially purified pMMO. These preparations have been characterized by biochemical and biophysical methods. In addition, aspects of methane monooxygenase gene regulation and copper accumulation in methanotrophs have been studied. This review summarizes for the first time the often controversial pMMO literature, focusing on recent progress and highlighting unresolved issues.

Abstract:Methane monooxygenases (MMOs) catalyze the conversion of methane to methanol as the first committed step in the assimilation of this hydrocarbon into biomass and energy by methanotrophs, thus playing a significant role in the biogeochemistry of this potent greenhouse gas. Two distinct enzymes, a copper-dependent membrane protein, particulate methane monooxygenase (pMMO), and an iron-dependent cytosolic protein, soluble methane monooxygenase (sMMO), carry out this transformation using large protein scaffolds that help to facilitate the timely transport of hydrocarbon, O2, proton, and electron substrates to buried dimetallic active sites. For both enzymes, reaction of the reduced metal centers with O2 leads to intermediates that activate the relatively inert C–H bonds of hydrocarbons to yield oxidized products. Among synthetic and biological catalysts, MMOs are unique because they are the only ones known to hydroxylate methane at ambient temperatures. As a need for new industrial catalysts and green chemical transformations increases, understanding how the different MMO metal centers efficiently accomplish this challenging chemistry has become the focus of intense study. This chapter examines current understanding of the sMMO and pMMO protein structures, their methods for substrate channeling, and mechanisms for the dimetallic activation of O2 and C–H bonds.

Abstract:Methanotrophs are a group of bacteria capable of utilizing methane as the sole carbon and energy source for their anabolism and catabolism. Since methanotrophs contain the unique enzymes of methane monooxygenases (MMOs), which can catalyze the oxidation of methane and short-chain alkanes and alkenes, they have potential applications in carbon recycle of nature and industrial biotechnology. Therefore, methanotrophs have been paid much more attention by the researchers in recent 20 years. In this paper, the latest progresses in studies of methanotrophs and MMOs were reviewed, including taxonomy, function and distribution of methanotrophs, and structure, function and genetic engineering of MMOs. The future research directions of methanotrophs and MMOs as well as their applications were also discussed.

Abstract:Methane monooxygenases (MMOs) mediate the facile conversion of methane into methanol in methanotrophic bacteria with high efficiency under ambient conditions. Because the selective oxidation of methane is extremely challenging, there is considerable interest in understanding how these enzymes carry out this difficult chemistry. The impetus of these efforts is to learn from the microbes to develop a biomimetic catalyst to accomplish the same chemical transformation. Here, we review the progress made over the past two to three decades toward delineating the structures and functions of the catalytic sites in two MMOs: soluble methane monooxygenase (sMMO) and particulate methane monooxygenase (pMMO). sMMO is a water-soluble three-component protein complex consisting of a hydroxylase with a nonheme diiron catalytic site; pMMO is a membrane-bound metalloenzyme with a unique tricopper cluster as the site of hydroxylation. The metal cluster in each of these MMOs harnesses O2 to functionalize the C—H bond using different chemistry. We highlight some of the common basic principles that they share. Finally, the development of functional models of the catalytic sites of MMOs is described. These efforts have culminated in the first successful biomimetic catalyst capable of efficient methane oxidation without overoxidation at room temperature.

Abstract:Climate warming is a hot environmental issue of global concern. As one of the major methane sinks, the process of methane oxidation coupled with denitrification (MOD) reduces the environmental impact brought by the greenhouse effect and water eutrophication. In addition, as an energy substance, methane can also improve its economic value by transforming into other liquid chemicals. Previous studies on the mechanism of the process have mainly focused on the extracellular electron transfer between species. However, in recent years, the production of intermediates influenced by different factors, and the existence of a large number of acid-producing bacteria and methanogens under anaerobic conditions, has led some researchers to pursue research into a new mechanism of the process. Moreover, the discovery of CO2 as a potential electron acceptor in products is certainly exciting, being a big opportunity under the ‘carbon neutral’ policy. This review looks back at the development of the MOD process and describes its functional microorganism and mechanism in detail when summarizing the types of microorganisms and intermediates at different oxygen levels, and introduces some traditional and novel biotechnologies, such as metagenomics, meta-transcriptomics, and meta-proteomics, etc., to help explore the novel mechanism of the process of MOD mediated by intermediates.

Abstract:Particulate methane monooxygenase (pMMO) is an integral membrane metalloenzyme that oxidizes methane to methanol in methanotrophic bacteria, organisms that live on methane gas as their sole carbon source. Understanding pMMO function has important implications for bioremediation applications and for the development of new, environmentally friendly catalysts for the direct conversion of methane to methanol. Crystal structures of pMMOs from three different methanotrophs reveal a trimeric architecture, consisting of three copies each of the pmoB, pmoA, and pmoC subunits. There are three distinct metal centers in each protomer of the trimer, mononuclear and dinuclear copper sites in the periplasmic regions of pmoB and a mononuclear site within the membrane that can be occupied by copper or zinc. Various models for the pMMO active site have been proposed within these structural constraints, including dicopper, tricopper, and diiron centers. Biochemical and spectroscopic data on pMMO and recombinant soluble fragments, denoted spmoB proteins, indicate that the active site involves copper and is located at the site of the dicopper center in the pmoB subunit. Initial spectroscopic evidence for O(2) binding at this site has been obtained. Despite these findings, questions remain about the active site identity and nuclearity and will be the focus of future studies.

Abstract:Methane, a potent greenhouse gas, has gained significant attention due to its environmental impact and economic potential. Chemical industries have focused on specialized catalytic systems, like zeolites, to convert methane into methanol. However, inherent limitations in selectivity, irreversibility, and pore blockages result in high costs and energy requirements, thus hindering their commercial viability and profitability. In contrast, biological methane conversion using methanotrophs has emerged as a promising alternative, offering higher conversion rates, self-renewability, improved selectivity, and economically feasible upstream processes. Nevertheless, biological methane oxidation encounters challenges including the difficulty in cultivating methanotrophs and their slow growth rates, which hinder large-scale bioprocessing. Another highlighted limitation is the limited mass transfer of methane into liquid in bioreactors. Practical strategies to enhance methane oxidation in biological systems, including optimizing reactor design to improve mass transfer, altering metal concentrations, genetic engineering of methane monooxygenases, enzyme encapsulation, and utilizing microbial consortia are discussed. By addressing the limitations of chemical approaches and highlighting the potential of biological methods, the review concluded that the utilization of genetically engineered methanotrophic biofilms on beads within a biotrickling reactor, along with enhanced aeration rates, will likely enhance methane oxidation and subsequent methane conversion rates.

Abstract:Methane is an important greenhouse gas which is produced from many natural and anthropogenic sources. It plays an important role in overall global warming. A significant amount of methane is removed through microbiological oxidation by methanotrophic bacteria, which are widespread in the environment, including many extreme environments. The key enzyme of these microorganisms, methane monooxygenase (MMO), especially the soluble MMO, is remarkable in its broad substrate specificity. This unique capability, i.e. catalyzing reactions of environmental importance, has attracted great attention for applied microbiologists and biochemical engineers. In this review, recent advances in the application of methanotrophs to environmental bioengineering are summarized, including biodiversity, catalytic properties, and cultivation, etc. We have focused on two aspects of the application and potential value of methanotrophs in environmental bioengineering, namely methane removal and biodegradation of toxic compounds. The removal of methane produced from landfills has been widely studied, and much of this work can be used as a source of reference for coal mine gas removal. Many bioreactors using methanotrophs in bioremediation have been developed in recent years. These reactors have two forms of configuration, single-stage and multi-stage. Current limitations which may affect the engineering applications of methanotrophs are discussed, such as the lack of suitable methanotrophic isolate, gas transfer limitation, competitive inhibition of MMO, regeneration of reducing equivalents for MMO and product toxicity.

Abstract:Microbial methane oxidation is the major biological methane (CH4) sink in the carbon cycle. Methanotrophs can use various electron acceptors in addition to oxygen; understanding the role and contribution of methanotrophs is thus an important topic. However, anaerobic oxidation of methane (AOM) mediated by methanotrophs is poorly explored and understood. This article summarizes the role aerobic methanotrophic bacteria play in AOM. Though AOM was originally considered to be mediated by anaerobic methanotrophic archaea, intra-aerobic methane-oxidizing bacteria (Candidatus Methylomirabilis oxyfera) appear to be involved in nitrite-dependent AOM. In addition, aerobic methanotrophs of the Methylomonadaceae and Methylocystaceae, are more versatile than previously assumed and can also be involved in nitrate/nitrite- or mineral oxide-dependent AOM under oxygen-limitation. Furthermore, the simultaneous reduction of nitrous oxide and oxidation of CH4 may be another new metabolic trait of aerobic methanotrophs. We discuss the potential metabolic pathways of CH4 oxidation under hypoxic conditions. It is of great ecological importance not only for the quantification of CH4 oxidation and emissions, but also for the definition of a new function of aerobic methanotrophs in anaerobic/hypoxic environments.

Abstract:Methane is the final product of the anaerobic decomposition of organic matter. The conversion of organic matter to methane (methanogenesis) as a mechanism for energy conservation is exclusively attributed to the archaeal domain. Methane is oxidized by methanotrophic microorganisms using oxygen or alternative terminal electron acceptors. Aerobic methanotrophic bacteria belong to the phyla Proteobacteria and Verrucomicrobia, while anaerobic methane oxidation is also mediated by more recently discovered anaerobic methanotrophs with representatives in both the bacteria and the archaea domains. The anaerobic oxidation of methane is coupled to the reduction of nitrate, nitrite, iron, manganese, sulfate, and organic electron acceptors (e.g., humic substances) as terminal electron acceptors. This review highlights the relevance of methanotrophy in natural and anthropogenically influenced ecosystems, emphasizing the environmental conditions, distribution, function, co-existence, interactions, and the availability of electron acceptors that likely play a key role in regulating their function. A systematic overview of key aspects of ecology, physiology, metabolism, and genomics is crucial to understand the contribution of methanotrophs in the mitigation of methane efflux to the atmosphere. We give significance to the processes under microaerophilic and anaerobic conditions for both aerobic and anaerobic methane oxidizers. In the context of anthropogenically influenced ecosystems, we emphasize the current and potential future applications of methanotrophs from two different angles, namely methane mitigation in wastewater treatment through the application of anaerobic methanotrophs, and the biotechnological applications of aerobic methanotrophs in resource recovery from methane waste streams. Finally, we identify knowledge gaps that may lead to opportunities to harness further the biotechnological benefits of methanotrophs in methane mitigation and for the production of valuable bioproducts enabling a bio-based and circular economy.

Abstract:Methane is a major greenhouse gas,which contributes estimatedly 20% to global warming.Microbially mediated anaerobic oxidation of methane(AOM) is an important way to reduce methane emission in nature.According to different coupling reactions,AOM can be divided into two types,Sulphate-dependent anaerobic methane oxidation(SAMO)and Denitrification-dependent anaerobic methane oxidation(DAMO).SO2-4 and NO-2/NO-3 function as their terminal electron acceptors,separately.This review summarizes types of AOM and microorganisms involved,elaborates mechanisms of the AOMs,and discusses orientation of the future research and prospects of the application of AOM.
Microbial mechanisms of methane production and oxidation in terrestrial ecosystems

Abstract:Terrestrial ecosystems consist of oxic and anoxic environments. Anoxic environments can be permanent or temporary,and they include natural wetlands,paddy field soils,lake and river sediments, rumens, landfills, and anaerobic bioreactors. Large amounts of organic matter are released into these environments annually. Under anoxic conditions,organic matter is decomposed anaerobically,with methane( CH4) as the final product. CH4 can diffuse through the anoxic-oxic interface and enter the atmosphere,and can become a potent greenhouse gas. Methanogenesis appears to be the core process of organic matter decomposition in anoxic environments. Methane oxidation takes place actively in the anoxic-oxic interface through which CH4 diffuses. Microbial production and oxidation of CH4 in terrestrial ecosystems together regulate the atmospheric concentration of CH4 and contribute to a crucial part of global carbon cycling. In this review,we report the recent advances in the mechanistic understanding of microbial CH4 production and oxidation in terrestrial ecosystems. The major topics include: discovery of novel methanogens,particularly of the psychrophilic and acidophilic methanogens,and the 6thand 7thorders of methanogens; syntrophic interaction and direct interspecies electron transfer in the methanogenic oxidation of short-chain fatty acids; the discovery of novel methanotrophs,including anaerobic and aerobic verrucomicrobial methanotrophs; and the novel ecophysiological properties of methanotrophs. These advanceshave greatly increased our understanding of microbial CH4 production and oxidation in terrestrial ecosystems,and with the development of new methods and technologies,more breakthroughs are expected. We also propose a few perspectives for future studies.

Abstract:The hydrogenation of CO to CH4, the CO methanation reaction, has attracted considerable attention due to energy and environment concerns. The reaction is thermodynamically favourable, however, the catalyst should show appreciable activity and durability in this highly exothermic reaction. This chapter focuses on recent advances in methanation catalysis, addressing on the roles of the metals, supports, promoters, the reaction and deactivation mechanisms, and the reactor types, with the aim to provide a foundation for the rational design of CO methanation catalysts and processes that enables the production of synthetic natural gas (SNG) in a more economic and greener manner.

Abstract:Aerobic methane oxidizing bacteria (methanotrophs) can use methane as carbon source and energy source, eliminating 10%-20% of global methane. Methanotrophs can also effectively synthesize valuable methane-derived products. This article introduced the methane oxidizing mechanism of methanotrophs, and summarized the practical application and research hotspots of methanotrophs in the field of methane emission reduction in the landfill, ventilation air methane mitigation in coal mines, valuable chemicals biosynthesis, as well as oil and gas reservoir exploration. Main factors influencing the pollutant removal and the biosynthesis efficiency in various applications were also discussed. Based on the study of large-scale cultivation of methanotrophs, some measures to benefit the application and promotion of aerobic methane oxidizing biotechnology were proposed. This includes investigating the effect of intermediate metabolites on methanotrophs activity and population structure, and exploiting economical and efficient alternative culture media and culture techniques.

Abstract:Methane represents one of the most abundant carbon sources for fuel or chemical production. However, remote geographical locations and high transportation costs result in a substantial proportion being flared at the source. The selective oxidation of methane to methanol remains a grand challenge for catalytic chemistry due to the large energy barrier for the initial C–H activation and prevention of overoxidation to CO2. Indirect methods such as steam reforming produce CO and H2 chemical building blocks, but they consume large amounts of energy over multistage processes. This makes the development of the low-temperature selective oxidation of methane to methanol highly desirable and explains why it has remained an active area of research over the last 50 years.The thermodynamically favorable oxidation of methane to methanol would ideally use only molecular oxygen. Nature effects this transformation with the enzyme methane monooxygenase (MMO) in aqueous solution at ambient temperature with the addition of 2 equiv of a reducing cofactor. MMO active sites are Fe and Cu oxoclusters, and the incorporation of these metals into zeolitic frameworks can result in biomimetic activity. Most approaches to methane oxidation using metal-doped zeolites use high temperature with oxygen or N2O; however, demonstrations of catalytic cycles without catalyst regeneration cycles are limited. Over the last 10 years, we have developed Fe-Cu-ZSM-5 materials for the selective oxidation of methane to methanol under aqueous conditions at 50 °C using H2O2 as an oxidant (effectively O2 + 2 reducing equiv), which compete with MMO in terms of activity. To date, these materials are among the most active and selective catalysts for methane oxidation under this mild condition, but industrially, H2O2 is an expensive oxidant to use in the production of methanol.This observation of activity under mild conditions led to new approaches to utilize O2 as the oxidant. Supported precious metal nanoparticles have been shown to be active for a range of C–H activation reactions using O2 and H2O2, but the rapid decomposition of H2O2 over metal surfaces limits efficiency. We identified that this decomposition could be minimized by removing the support material and carrying out the reaction with colloidal AuPd nanoparticles. The efficiency of methanol production with H2O2 consumption was increased by 4 orders of magnitude, and crucially it was demonstrated for the first time that molecular O2 could be incorporated into the methanol produced with 91% selectivity. The understanding gained from these two approaches provides valuable insight into possible new routes to selective methane oxidation which will be presented here in the context of our own research in this area.This article has not yet been cited by other publications.
