Abstract:Real time control is one of the possibilities to minimise the impact of the integrated urban wastewater system (sewer system and treatment plant) on the receiving water quality. Integrated control uses information about the river state to act in the sewer system or in treatment plant. In order to test and tune these integrated controllers, a simplified integrated model is needed. Even with these simplified models, the simulation times may be too long and further model reduction is needed. In this paper, dependency-structure based model reduction is proposed as a technique to further reduce model complexity. Three steps are proposed: relocation of the upstream system boundaries to just upstream of the first control point, relocation of the downstream boundaries to just downstream of the last measurement point, and third, a further model simplification based on an analysis of the sensitivity of the control actions on submodel elimination. The effect of applying the different reduction approaches on the control strategy and on the resulting river water quality is discussed on the basis of a case study of the catchment of Tielt.
Blocking of alpha 5 integrin stimulates production of TGF-beta and PAI-1 by human mesangial cells

Abstract:Expression of integrin, which mediates cell-matrix interaction, is affected by several cytokines, in particular by transforming growth factor-beta (TGF-beta). However, it is unknown whether, in an opposite way, a specific integrin is involved in cytokine synthesis. We tested this hypothesis. Function-blocking anti-alpha 5 integrin (fibronectin receptor) antibody increased TGF-beta secretion in growth-arrested human mesangial cells (2.3-fold) compared with control IgG or anti-alpha v beta 3 integrin (receptor for several matrix proteins) antibody. It also increased the secretion of plasminogen activator inhibitor-1 (PAI-1), a protein associated with matrix increase, by 3.2-fold. The increase in PAI-1 secretion induced by anti-alpha 5 integrin antibody was not abrogated by anti-TGF-beta neutralizing antibody. These results indicate that function-blocking of anti-alpha 5 integrin stimulates TGF-beta as well as PAI-1 production, suggesting that alpha 5 integrin is involved in fibrotic process. Function-modulation of a specific integrin thus appears to play a role in glomerular remodeling.
Intercellular Adhesion Molecule-1 And Tumour Necrosis Factor-Alpha Expression In Human Glomerulonephritis

Abstract:The relationship between renal expression of intercellular adhesion molecule-1 (ICAM-1), glomerular hypercellularity, renal function and renal tumour necrosis factor-alpha (TNF-alpha) expression was examined by immunohistochemistry staining in 64 cases of human glomerulonephritis. Glomerular anti-ICAM-1 antibody staining was increased in most cases of IgA nephropathy and lupus nephritis, but was unchanged compared to normal in membranous nephropathy and minimal change disease, and reduced in glomerular sclerosis. However, when taken together, patients with mild or no glomerular hypercellularity (group A) showed normal ICAM-1 expression, those with moderate to severe hypercellularity (group B) had increased glomerular ICAM-1 expression (P < 0.001), while those with glomerular sclerosis (group C) had reduced glomerular ICAM-1 expression. Patients in groups B and C also showed a significant increase in tubular ICAM-1 expression (P < 0.01) and interstitial infiltration of ICAM-1(+) cells (P < 0.001). Indeed, tubular ICAM-1 expression correlated with decreased creatinine clearance (r = -0.352; P < 0.05). In situ hybridization demonstrated that increase in tubular ICAM-1 staining was due to de novo gene expression, rather than absorption of soluble ICAM-1 from the lumen. Focal expression of tumour necrosis factor-a was seen in areas of leucocyte infiltration and strong ICAM-1 expression. Indeed, TNF-alpha staining correlated with increased renal ICAM-1 expression in both glomerular and tubulointerstitial compartments (r = 0.81; P<0.01). To confirm that TNF-alpha can directly stimulate renal ICAM-1 expression, TNF-alpha was shown to transiently increase ICAM-1 mRNA synthesis for 4-8 h and cause a progressive increase in ICAM-1 protein on the surface of cultured human mesangial cells. In summary; (i) increased glomerular ICAM-1 expression was restricted to cases of moderate to severe hypercellularity; (ii) tubular ICAM-1 expression correlated with both creatinine clearance and interstitial infiltration of ICAM-1(+) cells; and (iii) TNF-alpha expression was shown to correlate with the degree of renal ICAM-1 expression, suggesting that local TNF-alpha plays an important role in the up-regulation of ICAM-1 in human glomerulonephritis.
Down-regulation of Integrin Β1 and Focal Adhesion Kinase in Renal Glomeruli under Various Hemodynamic Conditions.

Abstract:Given that integrin β1 is an important component of the connection to maintain glomerular structural integrity, by binding with multiple extracellular matrix proteins and mediating intracellular signaling. Focal adhesion kinase (FAK) is the most essential intracellular integrator in the integrin β1-FAK signalling pathway. Here, we investigated the changes of the two molecules and visualized the possible interaction between them under various hemodynamic conditions in podocytes. Mice kidney tissues were prepared using in vivo cryotechnique (IVCT) and then were stained and observed using light microscopy, confocal laser scanning microscopy and immunoelectron microscopy. The expression of these molecules were examined by western blot. Under the normal condition, integrin β1 stained continually and evenly at the membrane, and FAK was located in the cytoplasm and nuclei of the podocytes. There were significant colocalized plaques of two molecules. But under acute hypertensive and cardiac arrest conditions, integrin β1 decreased and stained intermittently. Similarly, FAK decreased and appeared uneven. Additionally, FAK translocated to the nuclei of the podocytes. As a result, the colocalization of integrin β1 and FAK reduced obviously under these conditions. Western blot assay showed a consistent result with the immunostaining. Collectively, the abnormal redistribution and decreased expressions of integrin β1 and FAK are important molecular events in regulating the functions of podocytes under abnormal hemodynamic conditions. IVCT could offer considerable advantages for morphological analysis when researching renal diseases.
[Expression of Transforming Growth Factor-Beta1 in Renal Fibrosis of Human Mesengial Proliferative Glomerulonephritis].

Abstract:OBJECTIVE:To explore the possible effect of transforming growth factor-beta(1) (TGF -beta(1)) on the development of renal fibrosis in human mesengial proliferative glomerulonephritis (MsPGN).METHODS:Immunohistochemistry method, sirius red staining polarization microscopy and the computer imaging analysis system were used to detect the expression of TGF-beta(1), the distribution of collagen I, collagen III and collagen IV.RESULT:In MsPGN with renal fibrosis, collagen IV was increased markedly,and collagen I and collagen III appeared in the expanded mesengial matrix abnormally. Collagen III and collagen IV were increased markedly in tubulointerstitium. TGF-beta(1) expression was positively correlated with the expression of collagen I, collagen III and collagen IV in tubulointerstitium (r=0.82 0.92,P<0.01), and negatively correlated with I/III, I/IV and III/IV (r=-0.83,-0.92, P<0.001).CONCLUSION:Abnormal increase of TGF-beta(1) may be one of the important factors associated with glomerular sclerosis and tubulointerstitial fibrosis through the increment and abnormal distribution of collagen I, collagen III and collagen IV.
The characterization of alpha5-integrin expression on tubular epithelium during renal injury

Abstract:The hallmark of progressive chronic kidney disease is the deposition of extracellular matrix proteins and tubulointerstitial fibrosis. Integrins mediate cell-extracellular matrix interaction and may play a role tubular epithelial injury. Murine primary tubular epithelial cells (TECs) express alpha(5)-integrin, a fibroblast marker and the natural receptor for fibronectin. Microscopy localized alpha(5)-integrin on E-cadherin-positive cells, confirming epithelial expression. The expression of alpha(5)-integrin increased in TECs grown on fibronectin and occurred in parallel with an upregulation of alpha-smooth muscle actin (alphaSMA), a marker of epithelial-mesenchymal transition (EMT). Exposure of TECs to transforming growth factor (TGF)-beta also increased TEC alpha(5)-integrin expression in association with alphaSMA and EMT. Knock-down of alpha5-integrin expression with short interfering RNA attenuated the TGF-beta induction of alphaSMA but did not alter morphologic EMT. Rather, alpha5-integrin was necessary for epithelial cell migration on fibronectin but not type IV collagen during cell spreading and epithelial wound healing in vitro. Immunohistochemistry revealed basolateral tubular epithelial alpha(5)-integrin expression in mouse kidneys after unilateral ureteric obstruction but not in contralateral control kidneys. In patient biopsies of nondiabetic kidney disease, alpha(5)-integrin expression was increased significantly in the renal interstitium. Focal basolateral staining was also detected in injured, but not in normal, tubular epithelium. In summary, these data show that TECs are induced to express alpha(5)-integrin during EMT and tubular epithelial injury in vitro and in vivo. These results increase our understanding of the biology of integrins during EMT and tubular injury in chronic kidney disease.

Abstract:OBJECTIVE:To study the regulative effects of rhIL-1 beta or rhTNF-alpha on the expression of intercellular adhesion molecule-1 (ICAM-1) and vascular cell adhesion molecule-1 (VCAM-1) on human mesangial cells (HMC).METHODS:At the 4th, 8th, 16th and 32nd hours after stimulation by rhIL-1 beta (25 ng/ml) or rhTNF alpha (100 ng/ml), the mRNA expression of ICAM-1 and VCAM-1 on HMC was determined with Northern blot, and their protein expression was also tested with cell ELISA analysis.RESULTS:mRNA and protein of both ICAM-1 and VCAM-1 were only basically expressed on HMC at very low levels in the control groups without stimulation. After stimulation by rhIL-1 beta or rhTNF alpha, however, their expression was markedly upregulated. The maximal mRNA expression level of ICAM-1 stimulated by rhTNF alpha was present at the 8th hour, but in the remaining groups the maximal mRNA expression levels of ICAM-1 or VCAM-1 were all present at the 4th hour. Compared with control groups, ICAM-1 protein expression was significantly increased from the 4th hour after rhIL-1 beta stimulation (P < 0.05), but in the remaining groups ICAM-1 or VCAM-1 protein expression was significantly raised from the 8th hour after stimulation (P < 0.001).CONCLUSION:These results suggest that upregulated expression of ICAM-1 and VCAM-1 by inflammatory cytokines IL-1 beta and TNF alpha may play a pathogenic role in glomerulonephritis.

Abstract:Objective: To investigate that integrine platelete membrane glycoprotein IIb-IIIa(GPIIb-IIIa) was changed in human mesangial proliferative glomerulonephritis (MsPGN) and also to evaluate integrine GPIIb-IIIa expressed in mesangial pe oliferation glomerulonephritis. Methods: Renal biopsy tissues of 18 cases were stained by HE, PAS, PASM * Masson, and studied by ligh-microscopy, electron microscopy and immunofluorescence. GPIIb, GPIIIa, P-selestin(P-140), fibronetin (Fn), Laminin(LN) expression were detected by immunohistochemistry with SABC method, and the computer imaging analysis system. Results: 1PAS and PASM positive stained materials, FN, LN were significantly increased; 2GPIIb, GPIIIa, P-140 expressed upregulate; 3The number of cell in glumerulus(n), average volume of glumerlus (AVG) and glonerulosclersis index (SI) were significantly higher than that in nomal; Rend tubules and interstitial tissue were damaged. Conclusion: Integrine GPIIb-IIIa expressed upregulate in tubular epithelium, glomerlus and intertitium strong correlation to matrix proliferation, inflammatry cells infiluration and glomerular selerosis. Different actively of GPIIb, GPIIIa in renal diseased with different function are closely related to coagulation reaction, may play arole of platelete in glomerlar disease.
[Regulative Effect of Nuclear Factor Kappa B on the Expression of Intercellular Adhesion Molecular-1 in Human Mesangial Cells].

Abstract:OBJECTIVETo study the regulative effect of nuclear factor kappa B (NF-kappa B) on the expression of intercellular adhesion molecular-1(ICAM-1) in human mesangial cells (HMC) by interleukin I-1 beta (IL-1 beta).METHODSActivation of NF-kappa B was measured by electrophoresis mobility shift assay (EMSA). ICAM-1 expression was detected by Northern Blot and Cell ELISA.RESULTSrhIL-1 beta (10 ng/ml) could rapidly stimulate activation and translocation of NF-kappa B, and also could enhance the expression of ICAM-1 in mRNA (0.14 vs 0.35) and protein (0.92 +/- 0.10 vs 1.35 +/- 0.11, P < 0.01) level. After pretreatment with 100 mumol/L L-1-chlor-3-(4-tosylamido)-4-phenyl-2 butanon (TPCK), an inhibitor of NF-kappa B, both ICAM-1 levels of mRNA and protein stimulated by rhIL-1 beta were blocked by about 50% in these cells (0.46 +/- 0.05 vs 1.29 +/- 0.12, P < 0.01) compared with the rhIL-1 beta-stimulated group.CONCLUSIONThese results suggest that NF-kappa B is one of the signaling factors for IL-1-stimulated ICAM-1 expression in HMC. It may modulate the immune-inflammatory process in glomerular diseases.

Abstract:OBJECTIVE:To explore the expressions of transforming growth factor-beta1 (TGF-beta1) and its signaling transduction molecule Smad 2 and their significance in the development of glomerulosclerosis.METHODS:Using in situ hybridization and immunohistochemistry to detect Smad 2 mRNA expression and TGF-beta1, collagen IV, fibronectin expression in renal biopsies from 61 cases with a spectrum of glomerulonephritis including IgA nephropathy (40 cases), membranous glomerulonephritis (10 cases) and sclerosing glomerulonephritis (11 cases), compared with 11 cases of glomerular mild lesion with image analysis system.RESULTS:With the exception of Smad 2 mRNA expression in mild type IgA nephropathy, all other types of diseased glomeruli showed increased expression of both TGF-beta1 and Smad 2 mRNA when compared with the 11 cases of mild glomerular lesions. The expressions of glomerular TGF-beta1 and Smad 2 mRNA positively correlated with collagen IV and fibronectin deposition in the glomeruli.CONCLUSIONS:TGF-beta1 and Smad 2 may be involved in the excessive deposition of glomerular extracellular matrix and play an important role in the development of glomerulosclerosis.
The Expression of Α-Smooth Muscle Actin and Collagen in Renal Tissue of Patients with IgA Nephropathy

Abstract:Objective To quantitatively evaluate the expression of α-smooth muscle actin (α-SMA) and collagens in the renal tubulointerstitium of patients with IgA nephropathy and to explore its relationship to clinical pathology. Methods The levels of α-SMA and collagenⅠ, Ⅲ, Ⅳ were measured by immunohistochemistry in 65 cases with IgA nephropathy and the results were correlated with the clinicopathological data of the patients. Results The α-SMA levels in the renal tubulointerstitium were significantly increased with advancing of histopathological grade. The expression of tubulointerstitial α-SMA was significantly correlated with that of collagenⅠ, Ⅲ, Ⅳ (r=0.855,P0.001;r=0.744,P0.001; r=0.864,P0.001 respectively). Conclusion α-SMA expression in renal interstitial cell may play an important role in mediating the tubulointerstitial fibrosis of IgA nephropathy.
CTGF in kidney fibrosis and glomerulonephritis

Abstract:BackgroundGlomerulonephritis, which causes inflammation in glomeruli, is a common cause of end-stage renal failure. Severe and prolonged inflammation can damage glomeruli and lead to kidney fibrosis. Connective tissue growth factor (CTGF) is a member of the CCN matricellular protein family, consisting of four domains, that regulates the signaling of other growth factors and promotes kidney fibrosis.Main body of the abstractCTGF can simultaneously interact with several factors with its four domains. The microenvironment differs depending on the types of cells and tissues and differentiation stages of these cells. The diverse biological actions of CTGF on various types of cells and tissues depend on this difference in microenvironment. In the kidney, CTGF is expressed at low levels in normal condition and its expression is upregulated by kidney fibrosis. CTGF expression is known to be upregulated in the extra-capillary and mesangial lesions of glomerulonephritis in human kidney biopsy samples. In addition to involvement in fibrosis, CTGF modulates the expression of inflammatory mediators, including cytokines and chemokines, through distinct signaling pathways, in various cell systems. In anti-glomerular basement membrane (GBM) glomerulonephritis, systemic CTGF knockout (Rosa-CTGF cKO) mice exhibit 50% reduction of proteinuria and decreased crescent formation and mesangial expansion compared with control mice. In addition to fibrotic markers, the glomerular mRNA expression of Ccl2 is increased in the control mice with anti-GBM glomerulonephritis, and this increase is reduced in Rosa-CTGF cKO mice with nephritis. Accumulation of MAC2-positive cells in glomeruli is also reduced in Rosa-CTGF cKO mice. These results suggest that CTGF may be required for the upregulation of Ccl2 expression not only in anti-GBM glomerulonephritis but also in other types of glomerulonephritis, such as IgA nephropathy; CTGF expression and accumulation of macrophages in the mesangial area have been documented in these glomerular diseases. CTGF induces the expression of inflammatory mediators and promotes cell adhesion.Short conclusionCTGF plays an important role in the development of glomerulonephritis by inducing the inflammatory process. CTGF is a potentiate target for the treatment of glomerulonephritis.

Expression of Immunoglobulin A in Human Mesangial Cells and Its Effects on Cell Apoptosis and Adhesion.

Abstract:IgA nephropathy (IgAN) is characterized by predominant IgA deposition in the glomerular mesangium. It has been considered that the deposited IgA is synthesized by B cells, although recent reports have suggested the implication of other cell types. Therefore, the present study investigated whether glomerular mesangial cells could produce IgA by themselves. Semi-quantitative reverse transcription-polymerase chain reaction, and immunostaining analysis revealed that the IgA protein and gene transcripts were expressed in primary human renal mesangial cells (HRMCs). Furthermore, the IgA heavy chain (1 and 2) and the light chain ( and ) were localized in the cytoplasm or were located on the cell membranes of human mesangial cells (HMCs). Mass spectrometry results indicated that Ig 1 and Ig 2 were secreted in the culture media of HMCs. The transcripts of Ig , Ig and Ig constant regions were detected. The predominant rearrangement pattern of the variable region of Ig , was V3-20*01/J1*01 in HMCs and V1-12*01/J4*01 in HRMCs. In addition, knockdown of Ig 1 expression by small interfering RNA (siRNA) inhibited cell adhesion and promoted apoptosis. Our findings demonstrate that HMCs can express IgA, and that this expression is associated with cell functions, which may contribute to the deposition of IgA in patients with IgAN.
T Cell Infiltration is Associated with Kidney Injury in Patients with Anti-Glomerular Basement Membrane Disease.

Abstract:Cell-mediated autoimmunity, particularly that involving autoreactive T cells, participates in mediating anti-glomerular basement membrane (GBM) disease. However, direct kidney injury mediated by renal infiltrated T cells has not been clearly elucidated in humans. The T cell profile (CD3, CD4, CD8, IL-17, and foxp3) and macrophage (CD68) were examined by immunohistochemistry on renal biopsy tissues from 13 patients with anti-GBM disease. The correlation between cell infiltration and clinical data was also analyzed. We found that the distribution of T cell infiltration was predominant in the peri-glomerular and interstitial areas. CD3+ T cell infiltratrion around the glomeruli with cellular crescent formations was significantly higher than that around the glomeruli with mild mesangial proliferation. CD8+ T cells significantly accumulated around the glomeruli with cellular crescents without IgG deposits compared to those with IgG deposits. The prevalence of infiltrating CD8+ T cells was correlated with the percentage of ruptured Bowman's capsules. In conclusion, cellular immunity may play a crucial role in the inflammatory kidney injury in anti-GBM patients. The periglomerular infiltration of T cells, especially CD8+ T cells, may participate in the pathogenic mechanism of glomerular damage.
Immunohistological Study on the Mechanism of Mesangial Proliferation

Abstract:Inflammatory cells infiltrated in glomeruli and proliferating glomerular cells were immunohistochemically studied in 87 renal biopsies from patients with glomerulonephritides (GN) of various types, by using monoclonal antibody to Mac387 and PCNA (proliferative cell nuclear antigen) respectively. The results showed that the expressions of Mac387 and PCNA were generally increased in proliferative GN, compared with non-proliferative GN. The expression of Mac 387 was related to the severity of mesangial cell proliferation determined by light microscopy, but the increase of PCNA expression was found in severely proliferative GN. Although certain relationship between Mac387 and PCNA expression on glomerular cells was observed, it was not statistically significant. Finally, the possible mechanism of glomerular mesangial proliferation was discussed.

Abstract:Immunoglobulin A nephropathy (IgA-N) is a primary glomerulonephritis characterized by mesangial cell proliferation and expansion. It still leads to end-stage renal disease over the long course of the disease in many patients. Although glomerular endothelial cells (GE) have been implicated in the pathogenesis of IgA-N, their role remains poorly understood. We conducted single-cell and spatial transcriptomic analysis using human specimens with mild IgA-N compared with normal. We observed the significantly higher expression of several inflammation-associated molecules which have been reported and not reported to be involved in IgA-N pathogenesis, respectively. Furthermore, we show that several important molecules were also highly expressed in GE of IgA-N at protein level by tissue immunostaining. Our analysis suggests that the characteristic pathways identified in GE that involve these molecules are associated with the progression of IgA-N. These findings offer important insights on both the pathogenesis of IgA-N and its treatment at an early stage.

Abstract:Abstract BACKGROUND AND AIMS IgA nephropathy (IgAN) is characterized by the deposition of galactose deficient IgA (gd-IgA1) containing immune complexes in the mesangium leading to activation of the mesangial cells and local inflammation. Hallmarks of the disease are increased by mesangial proliferation and matrix expansion. The exact composition of the mesangial matrix is not known and nor is the full role of the expansion in disease progression. One important group of proteins found in extracellular matrixes is proteoglycans (PGs). PGs are composed of a core protein to which one or several negatively charged glycosaminoglycan (GAG) side chains are attached. Both the core protein and GAGs determine the properties of PGs and changes in the amount and structure of PGs could influence disease progression. In this study, we aimed to elucidate changes in the PG expression in the glomerulus in IgAN as well as how gd-IgA1 affects the mesangial cells’ PG expression. METHOD Using previously published glomerular transcriptomic data from patients with IgAN compared to healthy controls we investigated PG expression as well as changes in expression on a glomerular level [1]. Secondly, we investigated how gd-IgA1 affects the PG production of the cells in vitro using data from a previously published proteomic data set [1] as well as new in vivo experiments using real-time PCR as well as immunohistochemistry. To further explore the effects on PGs in IgAN, we employed a novel glycoproteomic approach that provides the combined characterization of GAG linkage regions and core protein identities in complex sample matrices. RESULTS We found 31 PGs to be expressed in the human glomeruli and of those 6 PGs were found to be significantly regulated in patients with IgAN. SDC1 (syndecan-1), was downregulated, while MXRA5 (matrix-remodeling associated 5), CD44 (CD44), TNC (tenascin C), FMOD (fibromodulin) and PRELP (PRELP) were up-regulated. The proteomic data set on mesangial cells treated with gd-IgA1 revealed that mesangial cells in vitro expressed 19 PGs and of those 10 were significantly regulated. Chondroitin sulfate proteoglycan 4, glypican-1, agrin, aggrecan, versican, lumican, protein AMBP, tenascin, collagen alpha-1(VII) chain and laminin subunit alpha-4. All of the significantly regulated PGs had an increased expression, except aggrecan, which was down-regulated. An interesting finding from the glycoproteomic experiments was the identification of collagen VII as a novel PG carrying chondroitin GAG in mesangial cells. In addition, immunohistochemistry revealed that gd-IgA1 treatment of mesangial cells does not lead to a total increase in GAGs, but rather to a switch from heparan sulfate to chondroitin sulfate GAGs. CONCLUSION In conclusion, this study gives a comprehensive view of the PGs expressed by mesangial cells and their alterations in response to gd-IgA1 as part of the expansion of the mesangium in IgAN. The switch of heparan sulfate GAGs to chondroitin sulfate GAGs is an interesting finding but the role of this switch needs further investigation.

Abstract:IgA deposition in glomerular mesangium and the interaction with mesangial cells may well be the final common pathway to IgA nephropathy (IgAN). Altered hinge-region O-glycosylation of IgA1 from patients with IgAN may predispose to mesangial deposition and activation of the mesangial cell (MC) by IgA1, via a novel IgA1 receptor, and may be a key event in the pathogensis of IgAN. The aim of this study was to investigate the binding capacity and biological effects of IgA1, from both patients with IgAN and healthy controls, on human mesangial cells (HMC). Serum IgA1 was isolated with jacalin affinity chromatography, heated to aggregated form (aIgA1) and labelled with (125)I. Binding capacity of aIgA1 in vitro to cultured primary HMC was evaluated by a radioligand binding assay and the specificity of binding was determined by a competitive inhibition assay. Intracellular calcium release was studied by confocal analysis and phosphorylation of extracellular signal-regulated kinase (ERK) was determined by Western blot analysis. Change of cell cycles was demonstrated by flow cytometry and HMC proliferation was evaluated by direct cell count. Expression of TGF-beta mRNA and production of supernatant fibronectin were tested by RT-PCR and indirect competitive ELISA, respectively. aIgA1 from both the patients with IgAN and normal controls bound to HMC in a dose-dependent, saturable manner, and was saturated at approximately 500 pmoles per 0.5 ml of aIgA1. aIgA1 from patients with IgAN, however, bound to HMC at a higher speed and Scatchard analysis revealed a Kd of (8.89 +/- 2.1) x 10(-8)m versus (4.3 +/- 1.2) x 10(-7)m for aIgA1 from healthy controls (P = 0.026). The binding was specific because it was only inhibited by unlabelled Mono-IgA1 (mIgA1) and not by serum albumin or IgG. aIgA1 from patients with IgAN could induce release of intracellular calcium, phosphorylation of ERK, DNA synthesis, proliferation of HMC, expression of TGF-betamRNA and secretion of fibronectin in HMC in a similar time-dependent manner as aIgA1 from healthy controls, but the effects were much stronger and the durations were much longer (P < 0.05, respectively). We conclude that aIgA1 from patients with IgAN has a higher binding capacity to HMC and stronger biological effects than aIgA1 from healthy controls. This suggests that direct interaction between IgA1 and HMC and subsequential pathophysiological responses may play an important role in the pathogenesis for IgAN.
