Abstract:Small interfering RNA (siRNA) technology, which could be able to silence specific targeted mRNAs rapidly and efficiently, has been regarded as a novel and potent gene therapeutic approach for diseases, such as cancers, infections, inflammations and neurological disorders. However, some conspicuous intrinsic defects of siRNA including the poor cellular uptake, low biological stability, unfavorable pharmacokinetics and bio-distribution properties, have hindered further applications in clinic. Therefore, it is prerequisite to design safe and effective siRNA delivery systems for the successful development of siRNA-based various diseases therapeutics. This review aims to analyze and summarize these supramolecular assembly models in the process of siRNA delivery, namely recent nanotechnological strategies for carrying siRNA by using diverse non-viral vehicles such as cationic liposomes, stable nucleic acid lipid particles (SNALPs), cationic polymers, cationic dendrimers, inorganic nanoparticles and others. Eventually, it is concluded with future perspectives of siRNA therapeutics in the field of non-viral carriers.

Abstract:In the past decade, nucleic acid therapeutics have experienced rapid development, with RNA interference (RNAi) based technology emerging as a versatile tool widely used in the treatment of various diseases. Small interfering RNA (siRNA), as a sequence-specific gene silencing method, has provided an effective and specific means for studying gene function and developing new therapeutic strategies. Consequently, there has been significant interest in utilizing siRNA as a method to target specific gene functions in therapy. However, in order to make RNAi technology valuable and effective, it is crucial to chemically modify siRNA and develop efficient siRNA delivery strategies. These measures aim to improve the stability and cellular uptake capability of siRNA, enhance sequence specificity, and reduce non-specific gene silencing and biotoxicity. This review provides a concise introduction to the chemical modifications of siRNA aimed at enhancing its resistance to nucleases and stability. Additionally, the development of siRNA delivery systems, including lipid-based and polymer-based carrier systems, represents a significant research direction. These systems aim to improve the pharmacokinetics of siRNA, enhance intracellular delivery, and achieve tumor targeting. Finally, this review summarizes and discusses the technological bottlenecks and future trends in siRNA drug delivery, providing guidance for the further application of siRNA as a therapeutic strategy.

Abstract:Surface chemistry is essential for the biomedical applications of functional nanomaterials. Here, a supramolecular container-based surface engineering approach is designed to impart excellent water dispersibility and precisely control the orientation of surface targeting ligands of the nanoparticles. An acyclic cucurbituril (aCB) molecular container is used as a chemical bridge to incorporate nanoparticles and targeting ligands via a bilateral host-guest complexation, enabling the bioactive moieties of targeting ligands to be fully exposed and faced outward to facilitate biological targeting. The enhanced biological targeting effect as well as targeted imaging performance of aCB-engineered nanoparticles are demonstrated in vitro and in vivo. Molecular dynamic simulations illustrate a tight binding of targeting ligand to the relevant receptor with the assistance of the aCB molecular container for the enhanced targeting efficiency, representing an attractive extension of supramolecular chemistry-based technology for nanoparticle surface engineering and supramolecularly regulated biological targeting.

Abstract:A synthetic carrier of small interfering RNA (siRNA) with customizable size, pH‐responsive degradability and optimized surface population of cell‐targeting agents is constructed precisely through a series of interlocked thermodynamically self‐regulated processes. This system consists of a unimolecular polyplex core formed from each individual molecule of networked cationic polymer and a multi‐functional shell assembled from a rationally designed triblock copolymer. The core‐forming polymer of defined size is synthesized via Zeta potential‐regulated condensation of branched and linear multi‐amine bearing oligomers through pH‐responding imidazole conjugated imine linkages. The shell‐forming copolymer consists of a multi‐carboxyl saccharide end block to guide to the surface assembly, a hydrophobic central block to form an encapsulating layer, and a poly(ethylene glycol) end block equipped with a highly selective active ester for “hooking” various cell‐targeting agents. Animal assays confirm the flexibility and convenience of this system to equip with selected functional components for systemic delivery of siRNA.
Delivery Pathway Regulation of 3′,3″-Bis-peptide-sirna Conjugate Via Nanocarrier Architecture Engineering

Abstract:Small interfering RNA (siRNA) has been continuously explored for clinical applications. However, neither nanocarriers nor conjugates have been able to remove the obstacles. In this study, we employed a combined nanochemistry strategy to optimize its delivery dilemma, where different interactions and assembly modes were cooperatively introduced into the forming process of siRNA/lipids nanoplexes. In the nanoplexes, the 3′,3″-bis-peptide-siRNA conjugate (pp-siRNA) and gemini-like cationic lipids (CLDs) were employed as dual regulators to improve their bio-behavior. We demonstrated that the “cicada pupa”-shaped nanoplexes of MT-pp-siRNA/CLDs (MT represented the mixed two-phase method) exhibited more compact multi-sandwich structure (∼25 layers), controllable size (∼150 nm), and lower zeta potential (∼22 mV) than other comparable nanoplexes and presented an increased siRNA protection and stability. Significantly, the nanoplex was internalized into melanoma cells by almost caveolae-mediated endocytosis and macropinocytosis (∼99.46%), and later reduced/avoided lysosomal degradation. Finally, the nanoplex facilitated the silencing of mRNA of the mutant B-Raf protein (down by ∼60%). In addition, pp-siRNA had a high intracellular sustainability, a significantly prolonged circulating time, and accumulation in tumor tissues in vivo. Our results have demonstrated that the combined approach can improve the intracellular fate of siRNA, which opens up novel avenues for efficient siRNA delivery.

Supramolecular affinity can act as a main driving force either in the self-assembly of carriers or in the loading of drugs. It is also possible to employ strong recognitions to achieve self-delivery of drugs. Due to dynamic controllable drug-release properties, the supramolecular nanoscale drug-delivery system provides a promising platform for precision medicine.

Abstract:Small interfering RNA (siRNA) can be used to suppress gene expression, thereby providing a new avenue for the treatment of various diseases. However, the successful implementation of siRNA therapy requires the use of delivery platforms that can overcome the major challenges of siRNA delivery, such as enzymatic degradation, low intracellular uptake and lysosomal entrapment. Here, a protocol for the preparation and use of a biocompatible and effective siRNA delivery system is presented. This platform consists of polyethylenimine (PEI) and arginine (Arg)-grafted porous silicon microparticles, which can be loaded with siRNA by performing a simple mixing step. The silicon particles are gradually degraded over time, thereby triggering the formation of Arg-PEI/siRNA nanoparticles. This delivery vehicle provides a means for protecting and internalizing siRNA, without causing cytotoxicity. The major steps of polycation functionalization, particle characterization, and siRNA loading are outlined in detail. In addition, the procedures for determining particle uptake, cytotoxicity, and transfection efficacy are also described.

Abstract:Gene therapy using small interfering RNA (siRNA) is emerging as a novel therapeutic approach to treat various diseases. However, safe and efficient siRNA delivery still constitutes the major obstacle for clinical implementation of siRNA therapeutics. Here we report an ionizable supramolecular dendrimer vector, formed via self-assembly of a small amphiphilic dendrimer, as an effective siRNA delivery system with a favorable safety profile. By virtue of the ionizable tertiary amine terminals, the supramolecular dendrimer has a low positively charged surface potential and no notable cytotoxicity at physiological pH. Nonetheless, this ionizable feature imparted sufficient surface charge to the supramolecular dendrimer to enable formation of a stable complex with siRNA via electrostatic interactions. The resulting siRNA/dendrimer delivery system had a surface charge that was neither neutral, thus avoiding aggregation, nor too high, thus avoiding cytotoxicity, but was sufficient for favorable cellular uptake and endosomal release of the siRNA. When tested in different cancer cell lines and patient-derived cancer organoids, this dendrimer-mediated siRNA delivery system effectively silenced the oncogenes Myc and Akt2 with a potent antiproliferative effect, outperforming the gold standard vector, Lipofectamine 2000. Therefore, this ionizable supramolecular dendrimer represents a promising vector for siRNA delivery. The concept of supramolecular dendrimer nanovectors via self-assembly is new, yet easy to implement in practice, offering a new perspective for supramolecular chemistry in biomedical applications.

Abstract:Cytoplasmic transportation of therapeutic nucleic acids is deemed as an onerous task with aim of precise knockdown towards the targeted genes. Pertaining to the programed functionalities of natural virus in circumventing the biological barriers, we tailored multifaceted chemistries into manufacture of synthetic siRNA delivery vehicles in resembling the functionalities of viral vectors to dynamically tackle with a sequential of biological obstacles encountered in the journey of systemic anti-tumor RNAi therapy. Once harnessing ligands with RGD motif for specific internalization into subcellular endosomal compartments of the tumor cells, the architecture of the proposed delivery vehicles was subjected to facile transformation responsive to pH stimuli in acidic endosomal compartments. The external biocompatible PEGylation palisade was consequently detached, unveiling the cytomembrane-lytic cationic components to commit disruptive potencies to the anionic endosomal membranes for translocation of siRNA conjugates into cytosol. Eventually, liberation of active siRNA could be accomplished due to its responsiveness to the strikingly high level of glutathione in cytosol, thereby contributing to potent RNAi. Hence, our elaborated virus-mimicking platform has demonstrated significant anti-tumor efficacy through systemic administration of anti-angiogenic RNAi payloads, which inspired prosperous potentials in a variety of therapeutic applications.

Abstract:Acombinatorial strategy based on nucleic acid assembly and electrostatic complexation is developed for efficient small interfering ribonucleic acid(siRNA) delivery. In this approach, siRNAs are first loaded into a well-defined nanotube through programmable nucleic acid self-assembly. Compared to small rigid siRNA duplex, the obtained siRNA-bearing nanotube with large architecture is more readily to complex with cationic and ionizable poly(β-amino ester), resulting in the formation of a novel platform for efficient siRNA delivery.

Abstract:The therapeutic potential of small interfering RNA (siRNA) is monumental, offering a pathway to silence disease-causing genes with precision. However, the delivery of siRNA to target cells in-vivo remains a formidable challenge, owing to degradation by nucleases, poor cellular uptake and immunogenicity. This overview examines recent advancements in the design and application of nucleic acid-based integrated macromolecular complexes for the efficient delivery of siRNA. We dissect the innovative delivery vectors developed in recent years, including lipid-based nanoparticles, polymeric carriers, dendrimer complexes and hybrid systems that incorporate stimuli-responsive elements for targeted and controlled release. Advancements in bioconjugation techniques, active targeting strategies and nanotechnology-enabled delivery platforms are evaluated for their contribution to enhancing siRNA delivery. It also addresses the complex interplay between delivery system design and biological barriers, highlighting the dynamic progress and remaining hurdles in translating siRNA therapies from bench to bedside. By offering a comprehensive overview of current strategies and emerging technologies, we underscore the future directions and potential impact of siRNA delivery systems in personalized medicine.

Abstract:siRNA delivery remains a major challenge in RNAi-based therapy. Here, we report for the first time that an amphiphilic dendrimer is able to self-assemble into adaptive supramolecular assemblies upon interaction with siRNA, and effectively delivers siRNAs to various cell lines, including human primary and stem cells, thereby outperforming the currently available nonviral vectors. In addition, this amphiphilic dendrimer is able to harness the advantageous features of both polymer and lipid vectors and hence promotes effective siRNA delivery. Our study demonstrates for the first time that dendrimer-based adaptive supramolecular assemblies represent novel and versatile means for functional siRNA delivery, heralding a new age of dendrimer-based self-assembled drug delivery in biomedical applications.

Abstract:Small interfering RNA (siRNA) is a powerful tool for modulating gene expression by RNA interference (RNAi). Duplex RNA oligonucleotides induce cleavage of homologous target transcripts, thereby enabling posttranscriptional silencing of potentially any gene. As such, siRNAs may have utility as novel pharmaceuticals for a wide range of diseases. However, a lack of "drug-likeness," physiological barriers, and potential toxicities have meant that systemic delivery of SiRNAs in vivo remains a major challenge. Here we discuss various strategies that have been employed to solve the problem of SiRNA delivery. These include chemical modification of the SiRNA, direct conjugation to bioactive moieties, and nanoparticle formulations.

Abstract:Self-assembly is a fundamental concept and a powerful approach in molecular science. However, creating functional materials with the desired properties through self-assembly remains challenging. In this work, through a combination of experimental and computational approaches, the self-assembly of small amphiphilic dendrons into nanosized supramolecular dendrimer micelles with a degree of structural definition similar to traditional covalent high-generation dendrimers is reported. It is demonstrated that, with the optimal balance of hydrophobicity and hydrophilicity, one of the self-assembled nanomicellar systems, totally devoid of toxic side effects, is able to deliver small interfering RNA and achieve effective gene silencing both in cells - including the highly refractory human hematopoietic CD34(+) stem cells - and in vivo, thus paving the way for future biomedical implementation. This work presents a case study of the concept of generating functional supramolecular dendrimers via self-assembly. The ability of carefully designed and gauged building blocks to assemble into supramolecular structures opens new perspectives on the design of self-assembling nanosystems for complex and functional applications.

Abstract:RNA interference is supposed to be one of the most powerful technologies for suppression of genes and treatment of diverse human diseases while the safe delivery and visualization of siRNA were still challenging. In this text, a novel type of monodispersed conjugated polymer nanoparticles PFNBr with brush-like molecular structure was introduced into siRNA delivery system. The nanoparticles exhibited dual functions conveniently in the delivery system which can not only carry high amount of siRNA to penetrate intracellularly for knocking down targeted mRNA but also act as signal agents for siRNA tracking and cellular imaging. Due to the high density side chains with positive charges and more extended conformation of the spatial structure, PFNBr nanoparticles as nanocarrier for siRNA provided outstanding capture ability (1 mol polymer to more than 32.5 mol siRNA) and enhanced protection capability of siRNA molecules from degradation. Here, it should be noted that the concentration of carrier in the working platform was lowered from the level of mu mol/L to nmol/L compared with other conjugated polymer delivery systems due to the outstanding carrying capacity of PFNBr. And meanwhile, this system acquired high gene silence efficiency and good biocompatibility. The proposed complex nanoparticles efficiently transfected siPlk1 into PANC-1 cells and induced high knockdown efficiency for targeted Plk1 mRNA to 23.9% and no significant cytotoxicity of the PFNBr/siRNA complexes was shown. Therefore, this working platform provides a solution to most of the common problems associated with the siRNA delivery, visualization, and therapeutic applications, and keeps a bright outlook for the development of new nucleic acid-based therapeutics and simultaneously for fluorescent bioimaging.
