Abstract:Ru(II) complexes which can undergo photo-induced ligand dissociation display potentials for biological applications in some aspects. However, their short excitation wavelengths limit tissue penetration and therefore applications. Two-photon technic is a general way to shift the excitation wavelength to the near-infrared region (NIR), but few Ru(II) complexes with photolabile ligands having large two-photon absorption cross sections (δ2) have been reported so far. In this work, by modification with a pyrene group on the tpy ligand, the resultant complex [Ru(tpy-py)(CH3CN)3]2+ (1) displayed enhanced two-photon induced ligand dissociation compared with the parent complex [Ru(tpy)(CH3CN)3]2+ (2), probably through an intramolecular energy transfer mechanism. The ligand dissociation process was investigated in detail by absorption, 1H NMR, ESI MS spectra and theoretical calculations.

Abstract:Dinuclear complexes bearing Ru(II) photoactive centers are of interest for the development of efficient dual catalysts for many photocatalyzed reactions. Ditopic polypyridine ligands, bis(pyridine-2-yl)amino-1,10-phenanthrolines, containing an additional coordination site (bis(pyridine-2-yl)amine, dpa) at positions 3, 4 or 5 of the 1,10-phenanthroline core (Phen-3NPy2, Phen-4NPy2 and Phen-5NPy2) were synthesized. They were used as bridging ligands to obtain dinuclear complexes [(bpy)2Ru(Phen-NPy2)PdCl2](PF6)2 (Ru(Phen-NPy2)Pd) in good yields via stepwise complexation. In these complexes Ru(II) is coordinated to 1,10-phenanthroline, while Pd(II) is bound to the dpa chelating moiety, as established by NMR spectroscopy and X-ray single crystal analysis. The influence of the position of dpa in the phenanthroline ring on the structural, optical and electrochemical properties of Ru(Phen-NPy2)Pd complexes was studied. The complexes exhibit photoluminescence in argon-saturated MeCN solution with maxima in the range of 615–625 nm, with emission quantum yields ranging from 0.11 to 0.15 for Ru(Phen-NPy2) complexes and from 0.018 to 0.026 for dinucear Ru(Phen-NPy2)Pd complexes. All the complexes absorb visible light in the range of 370–470 nm with high extinction coefficients and can be considered useful as photocatalysts. The Ru2+/3+ potential in Ru(Phen-NPy2)Pd complexes showed no significant dependence on the dpa position, while the Pd2+/0 reduction potential was significantly lower for Ru(Phen-3NPy2)Pd and Ru(Phen-NPy2)Pd, than for Ru(Phen-NPy2)Pd (–0.57 V and –0.72 V vs. Ag/AgCl, KCl(sat.), respectively). The complexes were used as photoactivated precatalysts in Сu-free Sonogashira coupling under blue LEDs (12 W) irradiation. The reaction proceeded roughly three times faster when Ru(Phen-4NPy2)Pd and Ru(Phen-3NPy2)Pd were used as catalyst precursors compared to the mixed catalytic system Ru(bpy)3(PF6)2/(RNPy2)PdCl2.

The Reasons for Ligand‐Dependent Quantum Yields and Absorption Spectrum of Four Polypyridylruthenium(II) Complexes with a Tetrazolate‐Based Ligand: TDDFT Study

Abstract:The quantum yield, lifetime, and absorption spectrum of four [Ru(bpy)(2)L](+) [where bpy is 2,2'-bipyridyl; L is represented by the deprotonated form of 2-(1H-tetrazol-5-yl)pyridine (L1) or 2-(1H-tetrazol-5-yl)pyrazine (L2)], as well as their methylated complexes [Ru(bpy)(2)LMe](2+) (RuL1Me and RuL2Me) are closely ligand dependent. In this paper, density functional theory (DFT) and time-dependent DFT (TDDFT) were performed to compare the above properties among these complexes. The calculated results reveal that the replacement of pyridine by pyrazine or the attachment of a CH3 group to the tetrazolate ring greatly increases the pi-accepting ability of the ancillary ligands. The stronger pi-accepting ability of the ancillary ligands in RuL2 results in stronger interaction with polar solvents and a relaxed geometrical structure; these are the main reasons for the lowest quantum yields and the shortest lifetime. The electron-donating CH3 group buffers the interaction between RuL2Me and the solvent, and together with the enhanced MLCT participation, these account for the highest quantum yields and the longest lifetime. This theoretical contribution allows the factors determining the efficiency of radiative and nonradiative decay pathways in this series of complexes to be rationalized and also allows design of new ruthenium(II) complexes with higher phosphorescence efficiency. Pyrene-Functionalized Polyacetylenes: Synthesis and Photoluminescence Property

Abstract:Four pyrene-functionalized polyacetylenes were designed and prepared through a typical post-polymerization modification route, which is the highly efficient reaction between activated ester and primary anime groups. The chemical structures of the resultant polymers were characterized with multiple spectroscopic techniques and the data indicated the successful functionalization of the polyacetylenes. The introduction of the pyrene moieties into the polymer structure allowed us to investigate the interactions between the polymer backbone and side chains. For the mono-substituted polyacetylenes, both the monomer and excimer emission features of the pyrene groups could be recorded, while for the di-substituted polyacetylenes, the fluorescence from the pyrene excimer vanished and the fluorescence intensity from the pyrene monomer decreased, the fluorescence from the polymer chain predominated the emission features. The concomitant energy transfer from the pyrene monomer and excimer to poly(diphenylacetylene) backbone was associated with the underlying mechanism. In addition to the substitution modes, the linkage between the poly(diphenylacetylene) backbone and the pyrene moiety also played a significant role in the determination of the emission species. A long alkyl spacer was beneficial to the pyrene monomer emission while a short one may be helpful to the formation of the excimer and intramolecular energy transfer.

Abstract:The easy tunable photophysical properties of Ru(II) polypyridyl complexes, together with their water solubility, photochemical stability and high biocompatibility make them suitable as agents for photodynamic therapy (PDT). The development of complexes containing new ligands is a step forward in the improvement and application of these compounds as photosensitizer and photocytotoxic agents. We report the synthesis of a set of Ru(II)-polypyridyl complexes with different electronic properties and delocalized π systems: one homoleptic complex [Ru II (dpbpy) 3 ](PF 6 ) 2 ( 1 ), and three heteroleptic complexes [Ru II (dpbpy) 2 (phen)](PF 6 ) 2 ( 2 ), [Ru II (dpbpy)(phen) 2 ](PF 6 ) 2 ( 3 ) and [Ru II (dpbpy) 2 (CN–Me)](PF 6 ) 2 ( 4 ). All of them contain 4,4′-diphenyl-2,2′-bipyridine ( dpbpy ) and 1,10-phenantroline ( phen ) or N-methyl-N′-2-pyridilimidazolium (CN–Me) ligands. The complexes have been characterized by spectroscopic, structural and electrochemical methods. UV–vis absorption in solution show the red shift for the metal to ligand charge transfer (MLCT) transitions after changing phen by dpbpy and carbene ligands. The photoluminescence emission spectra of 1 – 4 supported by theoretical calculations using time-dependent density-functional theory (TDDFT) suggest that the lowest energy excited state is mainly 3 MLCT. Complex 2 exhibits the highest phosphorescence emission with a quantum yield (Φ P ) in CH 2 Cl 2 of 44.1 %, followed by complex 1 (Φ P = 40.0 %), whereas complex 4 shows the lowest quantum efficiency (Φ P = 16.0 %), what suggest that the introduction of the CN–Me carbene ligand produces a significant quenching of the phosphorescence. Phosphorescence triplet state lifetimes between 0.96 and 1.74 μs were shown for 1 – 4 in degassed CH 2 Cl 2 . All complexes are proven to be efficient singlet oxygen photosensitizers. The trend in the redox potentials becomes more positive by increasing the number of phen ligands in the complexes, in full agreement with the HOMO's energy level and the blue shift of the MLCT transitions in the absorption spectra. Complexes internalization was analysed in tumorigenic mammary epithelial SKBR-3 cells, being complexes 1 and 2 the most well internalized. The effect of the photodynamic treatment using light-activated complexes 1 and 2 for 10 min demonstrated to increase cell death, being the homoleptic complex 1 an outstanding candidate as potential theranostic agent for bioimaging and PDT.

Abstract:Eight metal complexes of the type [M(tpy)(2)](2+) (tpy = 2,2':6',2 ''-terpyridine) featuring four pendant pyridine rings are reported and characterised by NMR, MS, absorption spectroscopy and electrochemical methods. Palladium-mediated Suzuki and Sonogashira cross-coupling reactions were performed on both free 4'-(3,5-dibromophenyl)-tpy and its Ru(II) complex in good yields. The ready N-alkylation of the pendant pyridyl units has significant influence on the absorption and electrochemical reduction of the complexes, processes which are localised on the periphery and leaves the [Ru(tpy)(2)](2+) core essentially unaffected. The binding of metal ions by the free pyridines is also demonstrated as means of assembling larger ordered non-covalent structures.
Near-IR/Visible-Emitting Thiophenyl-Based Ru(II) Complexes: Efficient Photodynamic Therapy, Cellular Uptake, and DNA Binding.

Abstract:Near-IR-emitting and/or efficiently photodynamic water-soluble Ru(II) complexes that hold great application potentials as photodynamic therapy and/or photodetection agents for cancers have been poorly explored. In this paper, the solvatochromism, calf thymus DNA binding, and singlet oxygen generation properties of a known ruthenium(II) complex of visible-emitting [Ru(bpy)2(dtdpq)](ClO4)2 (Ru1) and a new homoleptic complex of near-IR-emitting [Ru(dtdpq)3](ClO4)2 (Ru2) (bpy = 2,2'-bipyridine, dtdpq = 2,3-bis(thiophen-2-yl)pyrazino[2,3-f][1,10]phenanothroline) in water are reported. Moreover, DNA photocleavage, singlet oxygen generation in HeLa cells, cellular uptake/localization, and in vitro photodynamic therapy for cancer cells of water-soluble Ru1 are described in detail. The results show that Ru1 acted as potent photodynamic cancer therapy and mitochondrial imaging agents. Ru2 exhibited very strong solvatochromism from a visible emission maximum at 588 nm in CH2Cl2 to the near-IR region at 700 nm in water and singlet oxygen generation yield in water (23%) and DNA binding properties (intercalative DNA binding constant on the order of 106 M-1) comparable to those of Ru1, which should make Ru2 attractive for the aforementioned applications of Ru1 if the water solubility of Ru2 can be improved enough for the studies above.

Abstract:Ruthenium(ii) terpyridyl species typically suffer from a distortion away from the ideal octahedral geometry due to the rigidity of the terpyridine ligand. In this study we report a series of new sulfur-bridged terpyridyl ligands (1a-c) which offer enhanced flexibility due to sulfur spacers linking the pyridine rings, in addition to the ability to tune electronic properties with electron-donating (Me) and electron-withdrawing (CF3) substituents in the 4 and 4 ''-positions of the sulfur-bridged ligands. The resulting Ru(ii) complexes are detailed herein as both homoleptic (2a-c) and heteroleptic (3a-c) species, with the synthesis, structural characterization, photophysical and redox properties discussed, and the sulfur-bridged ligands allowing geometries closer to that of a "perfect" octahedron. All six species were found to be non-emissive at room temperature (with emissivity increasing on cooling to 77 K), and the electrochemical properties were greatly altered due to the increased bite angle of the sulfur-bridged ligands. The electrochemical and photophysical HOMO-LUMO gap can be easily tuned by altering the substituents in the 4 and 4 ''-positions.
Substituted Pyridazines As Ligands in Homoleptic (fac and Mer) and Heteroleptic Ru(II) Complexes.

Abstract:This article reports the preparation of a range of phenyl, pyridyl and pyrazinyl substituted pyridazines via the inverse electron demand [2 + 4] Diels-Alder reaction between 3,6-di(2-pyridyl)-1,2,4,5-tetrazines (bptz) and 3,6-di(2-pyrazinyl)-1,2,4,5-tetrazines (bpztz) and suitable dienophiles including acenaphthalene. The resulting polyaromatic compounds vary systematically in the number of aromatic substituents and the number and position of N-heteroatoms. For four of these compounds, the effect of the molecular changes on the solid-state structures were investigated using single crystal X-ray crystallography. The pyridazines were used as bidentate ligands in {M(II)(bipy)(2)} and tris(homoleptic) complexes (M = Fe, Ru). The optical and electrochemical properties of these complexes reflect the electron accepting character of the new ligands. The facial and meridional isomers of the tris complexes could be separated by column chromatography (on silica), thus allowing a spectral comparison of their absorption and emission properties. The solid-state structures of several of the metal complexes are discussed, including that of the facial isomer of the tris Ru(II) complex of 3,6-bis(2-pyridyl)-4,5-bis(4-pyridyl)pyridazine--a potential preformed geometric motif for the predirected construction of supramolecular assemblies.

Abstract:Synthesis and excellent photocatalytic properties of new ruthenium‐based anthracene and pyrene complexes are presented. Two new monopyridyl‐based Anthracene and Pyrene ligands were synthesized and employed to design Ruthenium‐coordinated organometallic complexes, RuAnthra and RuPyrene, respectively. The ligands and their metal complexes were fully characterized by different analytical techniques, including multinuclear 1D‐ and 2D‐NMR, electrospray ionization‐mass spectrometry, UV–Vis, fluorescence spectroscopy, and elemental analysis. Furthermore, their photophysical properties were studied in different solvents with increasing polarity using UV–Vis and fluorescence spectroscopy, showing less or no significant solvolysis effect. The average emission lifetimes of the complexes were further determined using time‐resolved emission spectroscopy. Additionally, the ligands and their metal complexes showed excellent ability to generate singlet oxygen and can be useful for several important potential applications.

Converting an Almost Noncytotoxic Ru(II) Complex with Photolabile Ligands into a Highly Efficient PACT Agent

Abstract:Ru(II) complexes with weak ligand fields may undergo light‐induced ligand dissociation, and the resulted Ru(II) aqua complexes may bind with biomolecules such as DNA, showing potential as photoactivated chemotherapy (PACT) agents. However, Ru(II) complexes with efficient PACT activity are still rare. Some Ru(II) complexes exhibit efficient photoinduced ligand dissociation but poor cytotoxicity. It is speculated that the low nuclear accumulation levels may account for their low PACT efficacy. In order to confirm this hypothesis, the almost noncytotoxic [Ru(7‐OCH3‐dppz)(4‐OCH3‐py)4](PF6)2 (Ru1) is loaded on nucleus‐targeted C5N2 nanoparticles (NPs). Compared with the free Ru1, Ru1–C5N2 NPs exhibit significantly increased cellular uptake and nuclear accumulation. Therefore, Ru1–C5N2 NPs show efficient PACT activity toward various cancer cell lines (including cisplatin‐resistant one) with half maximal inhibitory concentration (IC50) values of 0.18 × 10−6–0.29 × 10−6 m and phototoxicity index (IC50dark/IC50light) values above 137 under both normoxic and hypoxic conditions. Moreover, Ru1–C5N2 NPs also exhibit efficient PACT activity toward cisplatin‐resistant 3D multicellular tumor spheroids upon two‐photon irradiation (800 nm). The same strategy is also feasible to greatly improve the PACT activity of [Ru(7‐OCH3‐dppz)(py)4]2+, which itself only has a medium effect. The results may provide new sights for developing efficient Ru(II) PACT agents. The almost noncytotoxic [Ru(7‐OCH3‐dppz)(4‐OCH3‐py)4]2+ can be converted into a highly efficient photoactivated chemotherapy (PACT) agent by loading on the nucleus‐targeted C5N2 nanoparticles. Increasing the nuclear accumulation levels of Ru(II) complexes with photolabile ligands greatly improves the PACT activity. The results may provide new sights for developing novel and efficient Ru(II) PACT agents.

Abstract:Controlling the interplay between relaxation and charge/energy transfer processes in the excited states of photocatalysts is crucial for the performance of artificial photosynthesis. Metal-to-ligand charge-transfer triplet states (3MLCT*) of ruthenium(II) complexes are broadly implemented for photocatalysis, but an effective means of managing the triplets for enhanced photocatalysis has been lacking. Herein, We proposed a strategy to considerably prolong the triplet excited-state lifetime by decorating a ruthenium(II) phosphine complex (RuP-1) with pendent polyaromatic hydrocarbons (PAHs). Systematic studies demonstrate that in RuP-4 decorated with anthracene, sub-picosecond electron transfer from anthracene to 3MLCT* leads to a charge-separated state that can mediate the formation of the intra-ligand triplet state (3IL) of anthracene, resulting in an exceptionally long excited-state up to several milliseconds. This triplet management strategy enables impressive photocatalytic reduction of CO2 to CO with a turnover number (TON) of 404, an optimized quantum yield of 43% and 100% selectivity, which is the highest reported performance for mononuclear photocatalysts without additional photosensitizers. RuP-4 also catalyzes photochemical hydrogen generation under argon. This work opens up an avenue for regulating the excited-state charge/energy flow for the development of long-lived 3IL multi-functional mononuclear photocatalysts to boost artificial photosynthesis.

Abstract:A new ruthenium (II) complex with N‐functionalized pyrrole derivative is first synthesized. Its optical and electrochemical properties are reported and compared with that of its own each component. This study was carried out to understand what changes the optical and electrochemical properties in the chemical structure will bring when all the pieces of the puzzle are combined step by step to form a chemical structure. In recent years, nitrogen‐based ruthenium complexes have become an important material for dye‐sensitized solar cells and photo‐catalytic H2O and CO2 reduction research and applications, thanks to their charge transfer feature from the metal to the ligand. The electronic and spectral properties of such materials can be easily modified by changing the substituents and ligands. In this study, a new ruthenium (II) complex with N‐functionalized pyrrole derivative ([Ru(bpy)2(bpyCONHArPy)]+2 complex) was successfully synthesized and characterized. Its optical and electrochemical properties were studied by means of fluorescence, UV‐vis spectrometry, FTIR spectroscopy and cyclic voltammetry. These results were compared with that of its components (4‐(1H‐pyrrol‐1‐yl) aniline, N4,N4′‐bis(4‐(1H‐pyrrol‐1‐yl)phenyl)‐[2,2′‐bipyridine]‐4,4′‐dicarboxamide and ligand [Ru(bpy)3]+2). Related theoretical studies were also studied to find energy levels of complex. The present work is carried out to understand what chemical changes bring when combination of all the pieces of the puzzle into a chemical structure step by step.

Abstract:The Ru(II) complexes were obtained by introducing a new ligand containing phenanthrene moiety. Two complexes displayed obvious photocytotoxicity and low dark cytotoxicity towards Hela, A549, and A375 cells. Two complexes might exhibit PDT activities via type II mechanism from TDDFT results. Photodynamic therapy (PDT) has been an effective tumor treatment to reduce the side effects of chemotherapy. Herein, we described the synthesis of two Ru (II) complexes by introducing a phenanthrene unit and their photocytotoxicity. Ru1 and Ru2 displayed high DNA affinities by intercalation. Under light, two complexes produced high singlet oxygen (1O2) quantum yields and cleaved DNA efficiently. Both high DNA affinities and 1O2 quantum yields of two complexes resulted in obvious photocytotoxicity towards Hela, A549, and A375 cells. Furthermore, low dark cytotoxicity was also observed for two complexes, which may promote them to act as potential PDT agents. TDDFT (Time‐dependent density functional theory) calculations also demonstrated two complexes that might display the PDT activities via 1O2 photogeneration.
