Abstract:The four types of CD3 signaling chains of TCR have functional diversityCD3ε recruits Csk and p85 via its mono-phosphorylated ITAM and BRS motif respectivelyIncorporation of CD3ε into 28Z CAR alters signaling to promote antitumor functionE28Z CAR-T cells have reduced cytokine production but enhanced persistenceA T cell receptor (TCR) mediates antigen-induced signaling through its associated CD3ε, δ, γ, and ζ, but the contributions of different CD3 chains remain elusive. Using quantitative mass spectrometry, we simultaneously quantitated the phosphorylation of the immunoreceptor tyrosine-based activation motif (ITAM) of all CD3 chains upon TCR stimulation. A subpopulation of CD3ε ITAMs was mono-phosphorylated, owing to Lck kinase selectivity, and specifically recruited the inhibitory Csk kinase to attenuate TCR signaling, suggesting that TCR is a self-restrained signaling machinery containing both activating and inhibitory motifs. Moreover, we found that incorporation of the CD3ε cytoplasmic domain into a second-generation chimeric antigen receptor (CAR) improved antitumor activity of CAR-T cells. Mechanistically, the Csk-recruiting ITAM of CD3ε reduced CAR-T cytokine production whereas the basic residue rich sequence (BRS) of CD3ε promoted CAR-T persistence via p85 recruitment. Collectively, CD3ε is a built-in multifunctional signal tuner, and increasing CD3 diversity represents a strategy to design next-generation CAR.

Download : Download high-res image (132KB)

Download : Download full-size image

What problem does this paper attempt to address?
The paper attempts to address two main issues: 1. **Understanding the multiple roles of CD3 ε in TCR signaling**: - The paper investigates the phosphorylation dynamics of different CD3 chains in the TCR receptor complex, particularly the phosphorylation pattern of CD3 ε, using quantitative mass spectrometry. - The study found that the ITAM (Immunoreceptor Tyrosine-based Activation Motif) of CD3 ε exhibits monophosphorylation characteristics under TCR stimulation, which is related to the selectivity of the Lck kinase. - Further research indicates that monophosphorylated CD3 ε ITAM specifically recruits the inhibitory kinase Csk, thereby attenuating TCR signaling, suggesting that TCR is a self-limiting signaling mechanism that includes both activation and inhibitory motifs. 2. **Exploring the application of CD3 ε in CAR-T cell therapy**: - Current CAR-T cell designs typically include only the activating CD3 z chain, neglecting the potential functions of other CD3 chains. - Researchers integrated the intracellular segment of CD3 ε into the second-generation CAR (28Z CAR) to construct new CAR-T cells (E28Z CAR). - Experimental results show that E28Z CAR-T cells produce fewer cytokines under antigen stimulation but exhibit greater persistence, thereby improving the anti-tumor function of CAR-T cells. In summary, this paper aims to explore the potential application of CD3 ε in CAR-T cell therapy by deeply studying the signaling mechanism of CD3 ε, with the goal of enhancing the therapeutic efficacy and safety of CAR-T cells.
