Volatile Oil from Magnolia sieboldii Improve Neurotransmitter Disturbance and Enhance the Sleep-Promoting Effect in p-Chlorophenylalanine-Induced Sleep-Deprived Mice

Abstract:Sleep-promoting effects of Magnolia sieboldii K.Koch, Magnoliaceae, volatile oils and elucidate their potential mechanisms in Kunming mice with p-chlorophenylalanine-induced insomnia were explored. In addition, the components of M. sieboldii oil were analyzed by gas chromatography-mass spectrometry. A pentobarbital-induced sleep test was performed to examine the effect of different volatile oil concentrations on sleep. Immunohistochemical staining was performed to determine the protein expression levels of 5-hydroxytryptamine (5-HT1A) and gamma-aminobutyric acid type A subunit alpha 1 (GABAARα1) receptors in brain tissues. Nissl staining was used to detect the neuronal numbers. Western blotting and reverse transcription quantitative PCR were used to determine the relative number of cDNA and the expression of 5-hydroxytryptamine and GABAARα1. The result shows that β-elemene (22.11%), α-pinene (9.83%), and germacrene D (6.76%) were the major components. Nissl staining revealed that Magnolia oil increased the number of normal neurons in the cerebral cortex to different degrees. Based on immunohistochemical and western blot results, low and medium concentrations (50 and 100 mg/kg), significantly increased the expression levels of 5-hydroxytryptamine and GABAARα1 (p < 0.01) when compared with control group. Different oil concentrations could improve the mental state in insomnia-induced mice to different degrees. Magnolia oil facilitated sleep by increasing the number of normal neurons in the brain and protein levels of 5-hydroxytryptamine and GABAARα1 in brain regions.Graphical Abstract
