Synthesis and Antitubercular Evaluation of Diverse Glycosylated Ureas from D-Glucose

Abstract:N, N-disubstituted glycosylated ureas have been synthesized in good-to-excellent yields by 1,4-conjugate addition of amines to glycosylated olefinic esters followed by reaction of resulting glycosyl β-amino esters with diisocyanates. The developed sugar-based molecules were well characterized by extensive standard spectroscopic analysis including NMR (1H, 13C, and DEPT), IR, MS, and elemental analysis and were screened for their biological activity against Mycobacterium tuberculosis (M. Tb.), where some of them displayed potent antitubercular activity. The molecules have been designed keeping in view the well-known inhibitors of the enzymes involved in the biosynthesis of mycobacterial cell wall polymers and may be further explored as lead molecules for the successful development of a potential antitubercular drug candidate.Dedicated to Dr. Rama P. Therefore, it is crucial to design new classes of anti - tuberculosis drugs to reduce the treatment time and the factors for the development of drug resistance. ### Research methods The authors synthesized N,N - disubstituted glycosylureas through the following steps: 1. **Synthesis of glycosyl β - amino esters**: Starting from D - glucose, through multiple standard synthesis steps, including 1,2; 5,6 - di - O - isopropyl protection, 3 - O - methylation, selective 5,6 - isopropyl deprotection, NaIO₄ oxidation and HEW enylation reaction, and finally the glycosyl β - amino esters were prepared through 1,4 - conjugate addition reaction. 2. **Synthesis of glycosylureas**: The above - mentioned glycosyl β - amino esters were reacted with diisocyanates to generate the required glycosylurea derivatives. 3. **Structural characterization**: The synthesized compounds were characterized in detail by means of nuclear magnetic resonance (NMR), infrared spectroscopy (IR), mass spectrometry (MS) and elemental analysis. 4. **Biological activity evaluation**: The anti - tuberculosis activities of the compounds against Mycobacterium tuberculosis were evaluated using the microplate blue assay (MABA) and the agar microdilution technique. ### Main findings - Most of the compounds did not show significant activity against Mycobacterium tuberculosis, and their minimum inhibitory concentration (MIC) values were greater than 25 or 50 μg/mL. - However, compounds 3a, 4b and 4c showed significant antimicrobial activities, and their MIC values were 12.5 μg/mL, indicating that these compounds have the potential to be developed into new anti - tuberculosis drugs. - Structure - activity relationship studies showed that the optimal hydrophilic and hydrophobic balance is crucial for improving the inhibitory effect. For example, compounds 4b and 4c have a hydrophilic hydroxyethyl substituent at the C - 5 position and a hydrophobic 3 - O - benzyl substituent, showing good anti - tuberculosis activities. ### Conclusion By synthesizing and characterizing a series of glucose - based glycosylurea derivatives, the authors found some lead molecules with potential anti - tuberculosis activities. Further optimization of these compounds is expected to develop new anti - tuberculosis drugs.
