Abstract:Mitochondrial one-carbon metabolism provides carbon units to several pathways, including nucleic acid synthesis, mitochondrial metabolism, amino acid metabolism, and methylation reactions. Late-onset Alzheimer's disease is the most common age-related neurodegenerative disease, characterised by impaired energy metabolism, and is potentially linked to mitochondrial bioenergetics. Here, we discuss the intersection between the molecular pathways linked to both mitochondrial one-carbon metabolism and Alzheimer's disease. We propose that enhancing one-carbon metabolism could promote the metabolic processes that help brain cells cope with Alzheimer's disease-related injuries. We also highlight potential therapeutic avenues to leverage one-carbon metabolism to delay Alzheimer's disease pathology.

What problem does this paper attempt to address?
The paper primarily explores the relationship between mitochondrial one-carbon metabolism and Alzheimer's disease (AD), and proposes that enhancing one-carbon metabolism may help slow down or prevent the pathological progression of Alzheimer's disease. Specifically, the core issues addressed in the paper are: 1. **The importance of mitochondrial one-carbon metabolism**: Mitochondrial one-carbon metabolism provides carbon units required for several important biochemical pathways, including nucleotide synthesis, mitochondrial metabolism, amino acid metabolism, and methylation reactions. 2. **Characteristics of Alzheimer's disease**: Alzheimer's disease is a common age-related neurodegenerative disease, one of whose features is impaired energy metabolism, which may be related to mitochondrial bioenergetics. 3. **The link between one-carbon metabolism and Alzheimer's disease**: The authors discuss the intersections between one-carbon metabolism and molecular pathways of Alzheimer's disease, and hypothesize that enhancing one-carbon metabolism may promote metabolic processes that help brain cells cope with Alzheimer's disease damage. 4. **Therapeutic possibilities**: The paper also emphasizes the potential of utilizing one-carbon metabolism as a therapeutic approach to delay the pathological changes of Alzheimer's disease. In summary, the main aim of this study is to explore how mitochondrial one-carbon metabolism affects the development of Alzheimer's disease and to identify potential methods to intervene in the disease by regulating this metabolic pathway.
