Abstract:The toxic metal cadmium (Cd) poses a serious threat to plant growth and human health. Populus euphratica calcium-dependent protein kinase 21 (CPK21) has previously been shown to attenuate Cd toxicity by reducing Cd accumulation, enhancing antioxidant defense and improving water balance in transgenic Arabidopsis. Here, we confirmed a protein-protein interaction between PeCPK21 and Arabidopsis nuclear transcription factor YC3 (AtNF-YC3) by yeast two-hybrid and bimolecular fluorescence complementation assays. AtNF-YC3 was induced by Cd and strongly expressed in PeCPK21-overexpressed plants. Overexpression of AtNF-YC3 in Arabidopsis reduced the Cd inhibition of root length, fresh weight and membrane stability under Cd stress conditions (100 µM, 7 d), suggesting that AtNF-YC3 appears to contribute to the improvement of Cd stress tolerance. AtNF-YC3 improved Cd tolerance by limiting Cd uptake and accumulation, activating antioxidant enzymes and reducing hydrogen peroxide (H2O2) production under Cd stress. We conclude that PeCPK21 interacts with AtNF-YC3 to limit Cd accumulation and enhance the reactive oxygen species (ROS) scavenging system and thereby positively regulate plant adaptation to Cd environments. This study highlights the interaction between PeCPK21 and AtNF-YC3 under Cd stress conditions, which can be utilized to improve Cd tolerance in higher plants.

Abstract:Cadmium is a toxic heavy metal that has serious impacts on human and ecosystem health. Poplars play an active role in the phytoremediation of heavy metals in the environment; whereas only a few studies have investigated the molecular mechanisms by which trees absorb heavy metals from the environment. This study identified an abscisic acid-responsive element binding protein, PtrAREB3, in Populus trichocarpa . PtrAREB3 was principally expressed in leaves and markedly induced following the ABA and Cd treatments. Moreover, the poplar PtrAREB3 acts as direct regulator of target genes. Under Cd stress, PtrAREB3 further activates target genes associated with stress response and promotes cadmium tolerance of poplar by reducing the production of oxides, reducing chlorophyll degradation, increasing the synthesis of proline, and activating the antioxidant system. Furthermore, PtrAREB3 might indirectly regulate gene expression relating to Cd uptake, transport, and detoxification; ultimately, this increases Cd accumulation in plants. After 28 days of Cd treatment, the PtrAREB3 over-expression transgenic lines showed 18.2%− 25.7% higher dry biomass accumulation with a net photosynthesis rate than WT. In addition, overexpression of PtrAREB3 increased Cd accumulation in plants by 40.4%− 52.1%. Overall, the overexpression of PtrAREB3 enhanced Cd accumulation and tolerance, which making it an ideal molecular target for genetic improvement in Populus plants grown in metal contaminated areas in the future.

Enhanced cadmium phytoremediation capacity of poplar is associated with increased biomass and Cd accumulation under nitrogen deposition conditions

Abstract:Nitrogen (N) deposition plays a significant role in soil cadmium (Cd) phytoremediation, and poplar has been considered for the remediation of contaminated soil because of its enormous biomass and strong Cd resistance. To reveal the underlying physiological and root phenotypic mechanisms of N deposition affecting Cd phytoextraction in poplar, we assessed root phenotypic characteristics, Cd absorption and translocation, chlorophyll fluorescence performance, and antioxidant enzyme activities of a clone of Populus deltoides × P. nigra through combined greenhouse Cd and N experiments. Our results showed that Cd significantly changed the root phenotype by reducing root length, tip number, and diameter. Cd also caused the peroxidation of lipids, damaged the photosystem II (PSII) reaction centre, and reduced photosynthetic capacity, resulting in a decrease in biomass accumulation in poplar. The N60 (60 kg N·ha-1·yr-1) and N90 (90 kg N·ha-1·yr-1) treatments promoted the net photosynthetic rate of poplar by increasing the activity of antioxidant enzymes and proline content and repairing the PSII reaction centre, thus increasing the biomass accumulation of poplar exposed to Cd stress. Simultaneously, the N60 and N90 treatments might have increased Cd uptake from the soil by upregulating total root length, root tips, and fine root length. Cd mainly accumulated in roots and stems but not in leaves. The N30 (30 kg N·ha-1·yr-1) treatment had no obvious effects on these parameters compared with the single Cd treatment. Consequently, our study suggested that adequate N can improve biomass and Cd accumulation to enhance the phytoremediation capacity of poplar for Cd, which might be related to the improvement of leaf physiological defence and the change in root phenotypic characteristics.

Abstract:Cadmium (Cd) is one of the most toxic heavy metals for plant growth in soil. ATP-binding cassette (ABC) transporters play important roles in biotic and abiotic stresses. However, few ABC transporters have been characterized in poplar. In this study, we isolated an ABC transporter gene PtoABCG36 from Populus tomentosa. The PtoABCG36 transcript can be detected in leaves, stems and roots, and the expression in the root was 3.8 and 2 times that in stems and leaves, respectively. The PtoABCG36 expression was induced and peaked at 12 h after exposure to Cd stress. Transient expression of PtoABCG36 in tobacco showed that PtoABCG36 is localized at the plasma membrane. When overexpressed in yeast and Arabidopsis, PtoABCG36 could decrease Cd accumulation and confer higher Cd tolerance in transgenic lines than in wild-type (WT) lines. Net Cd2+ efflux measurements showed a decreasing Cd uptake in transgenic Arabidopsis roots than WT. These results demonstrated that PtoABCG36 functions as a cadmium extrusion pump participating in enhancing tolerance to Cd through decreasing Cd content in plants, which provides a promising way for making heavy metal tolerant poplar by manipulating ABC transporters in cadmium polluted areas.

Abstract:A cadmium (Cd) tolerance protein ( SpCTP3 ) involved in the Sedum plumbizincicola response to Cd stress was identified. However, the mechanism underlying the Cd detoxification and accumulation mediated by SpCTP3 in plants remains unclear. We compared wild-type (WT) and SpCTP3 -overexpressing transgenic poplars in terms of Cd accumulation, physiological indices, and the expression profiles of transporter genes following with 100 μmol/L CdCl 2 . Compared with the WT, significantly more Cd accumulated in the above-ground and below-ground parts of the SpCTP3 -overexpressing lines after 100 μmol/L CdCl 2 treatment. The Cd flow rate was significantly higher in the transgenic roots than in the WT roots. The overexpression of SpCTP3 resulted in the subcellular redistribution of Cd, with decreased and increased Cd proportions in the cell wall and the soluble fraction, respectively, in the roots and leaves. Additionally, the accumulation of Cd increased the reactive oxygen species (ROS) content. The activities of three antioxidant enzymes (peroxidase, catalase, and superoxide dismutase) increased significantly in response to Cd stress. The observed increase in the titratable acid content in the cytoplasm might lead to the enhanced chelation of Cd. The genes encoding several transporters related to Cd 2+ transport and detoxification were expressed at higher levels in the transgenic poplars than in the WT plants. Our results suggest that overexpressing SpCTP3 in transgenic poplar plants promotes Cd accumulation, modulates Cd distribution and ROS homeostasis, and decreases Cd toxicity via organic acids. In conclusion, genetically modifying plants to overexpress SpCTP3 may be a viable strategy for improving the phytoremediation of Cd-polluted soil.

Abstract:Phytoremediation offers a great potential for affordable remediation of heavy metal (HM)-polluted soil and water. Screening and identifying candidate genes related to HM uptake and transport is prerequisite for improvement of phytoremediation by genetic engineering. Using the cadmium (Cd)-hypersensitive Populus euphratica, an annexin encoding gene facilitating Cd enrichment was identified in this study. With a 12 h exposure to CdCl2 (50-100 μM), P. euphratica cells down-regulated transcripts of annexin1 (PeANN1). PeANN1 was homologue to Arabidopsis annexin1 (AtANN1) and localized mainly to the plasma membrane (PM) and cytosol. Compared with wild type and Atann1 mutant, PeANN1 overexpression in Arabidopsis resulted in a more pronounced decline in survival rate and root length after a long-term Cd stress (10 d, 50 μM), due to a higher cadmium accumulation in roots. PeANN1-transgenic roots exhibited enhanced influx conductance of Cd2+ under cadmium shock (30 min, 50 μM) and short-term stress (12 h, 50 μM). Noteworthy, the PeANN1-facilitated Cd2+ influx was significantly inhibited by a calcium-permeable channel (CaPC) inhibitor (GdCl3) but was promoted by 1 mM H2O2, indicating that Cd2+ entered root cells via radical-activated CaPCs in the PM. Therefore, PeANN1 can serve as a candidate gene for improvement of phytoremediation by genetic engineering.

Abstract:Cadmium, a toxic heavy metal, seriously affects human health and ecological security. The cation/H+ exchanger (CAX) family is a unique metal transporter that plays a crucial role in Cd acquisition, transfer, and remission in plants. Although there are many studies related to the genome-wide analysis of Populus trichocarpa, little research has been done on the CAX family genes, especially concerning Cd stress. In this study, genome-wide analysis of the Populus CAX family identified seven stress-related CAX genes. The evolutionary tree indicated that the CaCA family genes were grouped into four clusters. Moreover, seven pairs of genes were derived by segmental duplication in poplars. Cis-acting element analysis identified numerous stress-related elements in the promoters of diverse PtrCAXs. Furthermore, some PtrCAXs were up-regulated by drought, beetle, and mechanical damage, indicating their possible function in regulating stress response. Under cadmium stress, all CAX genes in the roots were up-regulated. Our findings suggest that plants may regulate their response to Cd stress through the TF-CAXs module. Comprehensively investigating the CAX family provides a scientific basis for the phytoremediation of heavy metal pollution by Populus.

Abstract:Background and Methods: Cadmium (Cd), ranked the 7th most hazardous substance, is one of the most widespread pollutants of soil and water in industrialized nations. Its increased movement in soil-plant systems is posing a serious threat to human health. Cd, without any known functions in plants, was found to be toxic even at minute concentrations, leading to the development of symptoms such as leaf roll, chlorosis and root and shoot growth reduction. Phytoremediation is an emerging cost-effective and environment friendly technology that utilizes high biomass producing plants including Populus plants to remove, transform or stabilize contaminants in soils. The objectives of our study were to record phenotypic variation in a Populus pedigree to Cd exposure, to identify Cd tolerant and susceptible genotypes of Populus, to map QTLs (Quantitative Trait Loci - genomic regions responsible) for Cd tolerance and accumulation in Populus and to predict a mechanism for Cd toxicity and tolerance in susceptible and tolerant Populus genotypes.QTL mapping was accomplished by conducting a greenhouse hydroponic study in which 252 genotypes of a Populus pseudo-backcross progeny were grown for 40 days and treated with 25 µM Cd. Phenotypic variation in total dry weight was recorded on these genotypes and was used for identifying QTL for Cd tolerance. We identified genotypes with contrasting responses to the Cd treatment and conducted a microarray study to identify potential Cd tolerance mechanisms based on gene expression patterns. Results/Conclusions: Significant variation was observed among genotypes in response to Cd treatment based on changes in total dry weight. Cd tolerant and susceptible genotypes were identified based on the least square mean differences (Control-Cd treated for each genotype) among all the genotypes. Three QTLs were identified for Cd tolerance and they accounted for approximately 25% of the phenotypic variation in Cd tolerance measured as total dry weights.In the microarray study, the Cd-susceptible genotype had higher expression of Fe-transporters compared to the tolerant genotypes, and Cd and Fe levels were significantly different in foliage. Even the susceptible genotype controls had higher, though not significant, Fe levels than tolerant genotype controls. We therefore hypothesize that part of the mechanism for Cd tolerance is determined by the differences in the activity of Fe transporters in genotypes with differential Fe homeostasis.
A WRKY transcription factor, PyWRKY71, increased the activities of antioxidant enzymes and promoted the accumulation of cadmium in poplar

Abstract:Cadmium (Cd) pollution poses significant threats to the ecological environment and human health. Currently, phytoremediation is recognized as an environmentally friendly approach for mitigating Cd pollution, with increasing attention on the utilization of transgenic plants in Cd-contaminated soil remediation. In this study, we isolated and cloned PyWRKY71 from Populus yunnanensis and conducted a pot experiment to validate its enhanced functionality in conferring Cd tolerance to woody plants (poplar). During the experiment, the increase in plant height of the OE-87 line (overexpression poplar) was 1.46 times than that of the wild type (WT). Moreover, PyWRKY71 significantly promoted the accumulation of Cd in poplar, especially in the roots, where the Cd content in the OE-45 and OE-87 lines was 1.42 times than that in the WT. The chlorophyll content of transgenic poplar leaves was higher than that of the WT, reflecting a protective mechanism of PyWRKY71. Additionally, the activities of other antioxidants, including POD, SOD, CAT, and MDA, were elevated in transgenic poplars, bolstering their tolerance to Cd stress. In summary, PyWRKY71 exhibits substantial potential in regulating plant tolerance to Cd stress. This study not only provides a solid scientific foundation but also introduces a novel modified poplar variety for the remediation of Cd pollution.

Populus euphratica R2R3-MYB transcription factor RAX2 binds ANN1 promoter to increase cadmium enrichment in Arabidopsis

Abstract:The expression of R2R3-MYB transcription factor PeRAX2 increased transiently upon CdCl 2 exposure (100 μM, 48 h) in leaves and roots of Populus euphratica. We observed that overexpression of PeRAX2 increased Cd 2+ concentration in Arabidopsis root cells and Cd 2+ amount in whole plant, which was due to the increased Cd 2+ influx into root tips. However, the Cd 2+ influx facilitated by PeRAX2 overexpression was substantially reduced by LaCl 3 (an inhibitor of Ca 2+ -channels), suggesting that PeRAX2 could promote the Cd 2+ entering through PM Ca 2+ -permeable channels (CaPCs) in the roots. It is noting that the expression of annexin1 (AtANN1), which mediates the influx of divalent cations through the PM calcium channels, was upregulated by Cd 2+ in PeRAX2-transgenic Arabidopsis. Bioinformatic analysis revealed that the AtANN1 promoter (AtANN1-pro) contains four cis-elements for MYB binding. The PeRAX2 interaction with AtANN1-pro was validated by LUC reporter assay, EMSA, and Y1H assay. Our data showed that PeRAX2 binds to the AtANN1 promoter region to regulate gene transcription and that AtANN1 mediates the Cd 2+ entry through CaPCs in the PM, leading to a Cd 2+ enrichment in transgenic plants. The PeRAX2-stimulated Cd 2+ enrichment consequently resulted in high H 2 O 2 production in root cells of transgenic plants. The expression of AtSOD and AtPOD and activities of CAT, SOD, POD increased in the transgenic lines under Cd 2+ stress. However, the Cd 2+ -upregulated expression and activity of antioxidative enzymes were less pronounced in the PeRAX2-overexpressed lines, compared to the wildtype and vector controls. As a result, root length and plant growth were more suppressed by Cd 2+ in the transgenic lines. Our data suggest that transcriptional regulation of AtANN1 by PeRAX2 can be utilized to improve Cd 2+ enrichment and phytoremediation, although the enriched Cd 2+ affected antioxidant defense system and plant growth in the model species.

Abstract:Abstract Long non-coding RNAs (lncRNAs) play essential roles in plant abiotic stress responses, but the response of lncRNA-mediated genetic networks to cadmium (Cd) treatment remain elusive in trees, the promising candidates for phytoremediation of Cd contamination. We identified 172 Cd-responsive lncRNAs and 295 differentially expressed target genes in the leaves of Cd-treated Populus tomentosa. Functional annotation revealed that these lncRNAs were involved in various processes, including photosynthesis, hormone regulation, and phenylalanine metabolism. Association studies identified 78 significant associations, representing 14 Cd-responsive lncRNAs and 28 target genes for photosynthetic and leaf physiological traits. Epistasis uncovered 83 pairwise interactions among these traits, revealing Cd-responsive lncRNA-mediated genetic networks for photosynthesis and leaf physiology in P. tomentosa. We focused on the roles of two Cd-responsive lncRNA–gene pairs, MSTRG.22608.1–PtoMYB73 and MSTRG.5634.1–PtoMYB27, in Cd tolerance of Populus, and detected insertions/deletions within lncRNAs as polymorphisms driving target gene expression. Genotype analysis of lncRNAs and heterologous overexpression of PtoMYB73 and PtoMYB27 in Arabidopsis indicated their effects on enhancing Cd tolerance, photosynthetic rate, and leaf growth, and the potential interaction mechanisms of PtoMYB73 with abiotic stresses. Our study identifies the genetic basis for the response of Populus to Cd treatment, facilitating genetic improvement of Cd tolerance in trees.

Cadmium-induced protein AS8: A protein to improve Cd accumulation and transport via Cd uptake in poplar

Abstract:The pollution of soil with heavy metals (HMs) has become an environmental problem of global concern. Phytoremediation, whereby plants extract HMs from soil, can efficiently and substantially reduce HM pollution in soil in an environmentally friendly manner. Cadmium-induced protein AS8 (CIPAS8) is present in many plants and its expression is induced by HMs. In this study, PeCIPAS8 and SlCIPAS8 were transformed into 84K poplar to study their effects on tolerance to, and translocation of, cadmium (Cd) in woody plants. Localization analyses showed that two CIPAS8 proteins were localized at the plasma membrane when transiently expressed in tobacco leaf epidermal cells. Compared with wild-type 84K poplar seedlings, transgenic poplar lines overexpressing PeCIPAS8 or SlCIPAS8 showed increased Cd contents and decreased Cd tolerance. Transgenic poplar lines overexpressing PeCIPAS8 or SlCIPAS8 accumulated more Cd in the roots, stems, and leaves, but the plant height did not differ significantly, compared with wild-type 84K poplar under Cd stress during the vegetative stage. CIPAS8 increased the Cd influx rate of transgenic poplar roots compared with that of the wild type, and affected the transcription levels of other metal transporters. These findings show that CIPAS8 increases Cd flux into plant tissues and demonstrate moderate Cd sensitivity of the plant. Therefore, CIPAS8 is an influx transporter with the potential to increase the uptake of toxic HMs by woody plants growing in HM-contaminated soils.

Abstract:WRKY transcription factors (TFs) play an important role in regulating plant growth and responses to environmental stress. However, the molecular mechanism of WRKY to cadmium (Cd) stress is unclear, which prevents phytoremediation of Cd-contaminated soil from widely application. Results showed that the Cd content in the aboveground part of the two transgenic poplar lines were 1.57 and 1.99 times higher than that of wild type (WT), and lateral roots, GSH, PCs content and GST activity increased significantly. RNA-seq. data about transgenic and WT poplars revealed that 2074 differentially expressed genes (DEGs) in roots, 4325 in leaves, and 499 in both tissues. And these DEGs were mainly concentrated in ABC transport protein (PaABC), heavy-metal binding protein (PaHIPP), and transportation and loading of xylem (PaNPF, PaBSP) proteins, and they enhanced Cd accumulation. Meanwhile, PyWRKY48 increased the Cd tolerance of transgenic poplars by up-regulating the expression of PaGRP, PaPER and PaPHOS, which encode cell wall proteins, antioxidant enzyme, and heavy metal-associated proteins, respectively. In addition, overexpression PyWRKY48 promoted poplar growth by increasing the chlorophyll and carotenoid content. ENVIRONMENTAL IMPLICATION: This study generated PyWRKY48-overexpressing poplars and functionally verified them in Cd-contaminated soil, to analyze the effects of the gene on poplar growth, Cd tolerance and Cd accumulation. RNA seq. data revealed that several genes are involved in Cd exposure. This may provide a strong molecular basis and new ideas for improving the phytoremediation efficiency of Cd-contaminated soils. Importantly, the transgenic poplars grew better and accumulated more Cd than the wild-type. Therefore, PyWRKY48-overexpressing poplars could be considered useful for mitigating environmental pollution.
Ectomycorrhizal Fungal Strains Facilitate Cd2+ Enrichment in a Woody Hyperaccumulator under Co-Existing Stress of Cadmium and Salt

Abstract:Cadmium (Cd2+) pollution occurring in salt-affected soils has become an increasing environmental concern in the world. Fast-growing poplars have been widely utilized for phytoremediation of soil contaminating heavy metals (HMs). However, the woody Cd2+-hyperaccumulator, Populus × canescens, is relatively salt-sensitive and therefore cannot be directly used to remediate HMs from salt-affected soils. The aim of the present study was to testify whether colonization of P. × canescens with ectomycorrhizal (EM) fungi, a strategy known to enhance salt tolerance, provides an opportunity for affordable remediation of Cd2+-polluted saline soils. Ectomycorrhization with Paxillus involutus strains facilitated Cd2+ enrichment in P. × canescens upon CdCl2 exposures (50 μM, 30 min to 24 h). The fungus-stimulated Cd2+ in roots was significantly restricted by inhibitors of plasmalemma H+-ATPases and Ca2+-permeable channels (CaPCs), but stimulated by an activator of plasmalemma H+-ATPases. NaCl (100 mM) lowered the transient and steady-state Cd2+ influx in roots and fungal mycelia. Noteworthy, P. involutus colonization partly reverted the salt suppression of Cd2+ uptake in poplar roots. EM fungus colonization upregulated transcription of plasmalemma H+-ATPases (PcHA4, 8, 11) and annexins (PcANN1, 2, 4), which might mediate Cd2+ conductance through CaPCs. EM roots retained relatively highly expressed PcHAs and PcANNs, thus facilitating Cd2+ enrichment under co-occurring stress of cadmium and salinity. We conclude that ectomycorrhization of woody hyperaccumulator species such as poplar could improve phytoremediation of Cd2+ in salt-affected areas.

Abstract:The basic helix-loop-helix (bHLH) gene family is integral to various aspects of plant development and the orchestration of stress response. This study focuses on the bHLH genes within Populus × canescens, a poplar species noted for its significant tolerance to cadmium (Cd) stress. Through our comprehensive genomic analysis, we have identified and characterized 170 bHLH genes within the P. canescens genome. These genes have been systematically classified into 22 distant subfamilies based on their evolutionary relationships. A notable conservation in gene structure and motif compositions were conserved across these subfamilies. Further analysis of the promoter regions of these genes revealed an abundance of essential cis-acting element, which are associated with plant hormonal regulation, development processes, and stress response pathway. Utilizing quantitative PCR (qPCR), we have documented the differential regulation of PcbHLHs in response to elevated Cd concentrations, with distinct expression patterns observed across various tissues. This study is poised to unravel the molecular mechanism underpinning Cd tolerance in P. canescens, offering valuable insights for the development of new cultivars with enhanced Cd accumulation capacity and tolerance. Such advancements are crucial for implementing effective phytoremediation strategies to mitigate soil pollution caused by Cd.
Identification of Quantitative Trait Loci and Candidate Genes for Cadmium Tolerance in Populus.

Abstract:Understanding genetic variation for the response of Populus to heavy metals like cadmium (Cd) is an important step in elucidating the underlying mechanisms of tolerance. In this study, a pseudo-backcross pedigree of Populus trichocarpa Torr. & Gray and Populus deltoides Bart. was characterized for growth and performance traits after Cd exposure. A total of 16 quantitative trait loci (QTL) at logarithm of odds (LOD) ratio ≥ 2.5 were detected for total dry weight, its components and root volume. Major QTL for Cd responses were mapped to two different linkage groups and the relative allelic effects were in opposing directions on the two chromosomes, suggesting differential mechanisms at these two loci. The phenotypic variance explained by Cd QTL ranged from 5.9 to 11.6% and averaged 8.2% across all QTL. A whole-genome microarray study led to the identification of nine Cd-responsive genes from these QTL. Promising candidates for Cd tolerance include an NHL repeat membrane-spanning protein, a metal transporter and a putative transcription factor. Additional candidates in the QTL intervals include a putative homolog of a glutamate cysteine ligase, and a glutathione-S-transferase. Functional characterization of these candidate genes should enhance our understanding of Cd metabolism and transport and phytoremediation capabilities of Populus.
Plant Cadmium Resistance 2 (sapcr2) Facilitates Cadmium Efflux in the Roots of Hyperaccumulator Sedum Alfredii Hance

Abstract:Hyperaccumulators are the preferred materials for phytoremediation. Sedum alfredii Hance is a cadmium (Cd) hyperaccumulator plant in China, although its detoxification mechanism remains unresolved. In our study, we cloned a gene belonging to the plant cadmium resistance (PCR) family, named SaPCR2, from the hyperaccumulating ecotype (HE) of S. alfredii. Sequence analysis indicated that SaPCR2 contained a cysteine-rich domain highly conserved in the PCR family and played an important role in Cd detoxification. Based on the relative quantitative results, SaPCR2 was highly expressed in the roots of HE S. alfredii, but not the shoots and Cd exposure did not significantly affect SaPCR2 expression. In contrast, the expression level of SaPCR2 was very low in plants of its non-hyperaccumulating ecotype (NHE). The subcellular localization of SaPCR2 in tobacco leaves and yeasts showed that SaPCR2 was localized on the plasma membrane and the expression of the SaPCR2 protein in a Zn/Cd-sensitive yeast Δzrc1 significantly increased its tolerance to Cd stress by decreasing the Cd content in cells. Heterologous expression of SaPCR2 in plants of both Arabidopsis thaliana and NHE S. alfredii significantly reduced the Cd levels in the roots, but not in the shoots. These results suggest that the overexpression of SaPCR2 in plants provides a route for Cd leak out of the root cells and protects the root cells against phytotoxicity of Cd stress. To the best of our knowledge, this is the first study of transporter-mediated root efflux of Cd in hyperaccumulator S. alfredii.

Abstract:PcWRKY1 binds to the Yellow Stripe‐Like3 (PcYSL3) promoter region and represses PcYSL3 expression to negatively regulate cadmium‐nicotianamine (Cd‐NA) unloading from xylem vessels to adjacent ray parenchyma cells and Cd accumulation in the phloem of Populus × canescens (Aiton) Sm. A considerable amount of cadmium (Cd) can accumulate in the bark of poplar stems, but the Cd transport pathway and its underlying molecular mechanisms remain unknown. Here, a Cd radial transport pathway in poplar stems and a previously unrecognized PcWRKY1‐Yellow Stripe‐Like 3 (PcYSL3) module that regulates Cd transport are identified in Populus × canescens (Aiton) Sm. Cadmiun‐nicotianamine (Cd‐NA) in xylem vessels in poplar stem‐wood is unloaded to adjacent ray parenchyma cells and further radially transported to bark‐phloem. PcYSL3 is putatively identified as involved in Cd radial transport in poplar stems. PcYSL3 is highly expressed in ray parenchyma cells adjacent to xylem vessels and the encoded protein localizes on the plasma membrane. Cd accumulation is greater in the wood and bark of PcYSL3‐overexpressing poplars than the wild type, whereas the opposite is observed in PcYSL3‐knockdown plants. PcWRKY1 can bind to the PcYSL3 promoter sequence and represses its expression. PcWRKY1 inhibits Cd accumulation in the wood and bark of plants. Thus, PcWRKY1 suppresses PcYSL3 transcription to negatively regulate Cd‐NA unloading from xylem vessels to adjacent ray parenchyma cells and its radial transport in poplar stem. The findings have provided new insights into breeding of poplars for more effective remediation of heavy metal‐contaminated soils.

Cell Wall Polysaccharide-Mediated Cadmium Tolerance Between Two Arabidopsis thaliana Ecotypes

Abstract:Cadmium (Cd) is a toxic metal element and the mechanism(s) underlying Cd tolerance in plants are still unclear. Increasingly more studies have been conducted on Cd binding to plant cell walls (CW) but most of them have focused on Cd fixation by CW pectin, and few studies have examined Cd binding to cellulose and hemicellulose. Here we found that Cd binding to CW pectin, cellulose, and hemicellulose was significantly higher in Tor-1, a Cd tolerant A. thaliana ecotype, than in Ph2-23, a sensitive ecotype, as were the concentrations of pectin, cellulose, and hemicellulose. Transcriptome analysis revealed that the genes regulating CW pectin, cellulose, and hemicellulose polysaccharide concentrations in Tor-1 differed significantly from those in Ph2-23. The expressions of most genes such as pectin methyl esterase inhibitors (PMEIs), pectin lyases, xyloglucan endotransglucosylase/hydrolase, expansins (EXPAs), and cellulose hydrolase were higher in Ph2-23, while the expressions of cellulose synthase-like glycosyltransferase 3 (CSLG3) and pectin ethyl esterase 4 (PAE4) were higher in Tor-1. The candidate genes identified here seem to regulate CW Cd fixation by polysaccharides. In conclusion, an increase in pectin demethylation activity, the higher concentration of cellulose and hemicellulose, regulated by related genes, in Tor-1 than in Ph2-23 are likely involved in enhanced Cd CW retention and reduce Cd toxicity.

Abstract:Trifolium repens (T. repens) can accumulate significant amounts of heavy metal ions, and has strong adaptability to wide environmental conditions, and relatively large biomass, which is considered a potential plant for phytoremediation. However, the molecular mechanisms of T. repens involved in Cd tolerance have not yet been studied in detail. This study was conducted to examine the integrative responses of T. repens exposed to a high-level CdCl2 by investigating the physiological and transcriptomic analyses. The results suggested that T. repens seedlings had a high degree of tolerance to Cd treatment. The roots accumulated higher Cd concentration than leaves and were mainly distributed in the cell wall. The content of MDA, soluble protein, the relative electrolyte leakage, and three antioxidant enzymes (POD, SOD, and APX) was increased with the Cd treatment time increasing, but the CAT enzymes contents were decreased in roots. Furthermore, the transcriptome analysis demonstrated that the differentially expressed genes (DEGs) mainly enriched in the glutathione (GSH) metabolism pathway and the phenylpropanoid biosynthesis in the roots. Overexpressed genes in the lignin biosynthesis in the roots might improve Cd accumulation in cell walls. Moreover, the DEGs were also enriched in photosynthesis in the leaves, transferase activity, oxidoreductase activity, and ABA signal transduction, which might also play roles in reducing Cd toxicity in the plants. All the above, clearly suggest that T. repens employ several different mechanisms to protect itself against Cd stress, while the cell wall biosynthesis and GSH metabolism could be considered the most important specific mechanisms for Cd retention in the roots of T. repens.
