Amyloid plaques: hotspots for microglial turnover

Abstract:Microglial division and death rates are highest in the vicinity of amyloid plaques, according to new research in a mouse model of Alzheimer disease (AD). The researchers injected a mixture of three lentiviral constructs, each encoding a different fluorophore to achieve multi-colour labelling of microglia, into wild-type and AD mice. This technique enabled tracking of individual cells, which were imaged in vivo through a cranial window. The basic migration properties of microglia, including speed and fraction of migrating cells, were similar between AD and wild-type mice. However, in AD mice, a substantial fraction (33.0%) of migration was directed towards amyloid plaques. Furthermore, 69.0% of cell division and 54.4% of cell death occurred within the plaque vicinity, implicating amyloid plaques as a major site for microglia turnover in AD.
