Abstract:The physiological titer of molting hormones in insects depends on relative activities of synthesis and degradation pathways. Ecdysone oxidase (EO) is a key enzyme in the inactivation of ecdysteroid. However, there are only a few reports on ecdysteroid inactivation and its enzymes in silkworm. In this study, we cloned and characterized the Bombyx mori EO (BmEO). The BmEO cDNA contains an ORF of 1,695 bp and the deduced protein sequence contains 564 amino acid residues. The deduced protein sequence contains two functional domains of glucose-methanol-choline oxidoreductase in N-terminal and C-terminal. Comparing the expression levels of BmEO in different tissues, high transcription was mainly present in hemocytes. Reduced expression of this enzyme is expected to lead to pathological accumulation of ecdysone in the hemolymph of silkworm larvae or pupae. Our data show that RNA inference of BmEO transcripts resulted in the accumulation of ecdysteroid and death of larvae or pupae. We infer that EO is a crucial element in the physiology of insect development. (C) 2011 Wiley Periodicals, Inc.

Abstract:Insect molting is an important developmental process of metamorphosis, which is initiated by molting hormone. Molting includes the activation of dermal cells, epidermal cells separation, molting fluid secretion, the formation of new epidermis and old epidermis shed and other series of continuous processes. Polyphenol oxidases, dopa decarboxylase and acetyltransferase are necessary enzymes for this process. Traditionally, the dopa decarboxylase ( Bm Ddc) was considered as an enzyme for epidermal layer’s tanning and melanization. This work suggested that dopa decarboxylase is one set of the key enzymes in molting, which closely related with the regulation of ecdysone at the time of biological molting processes. The data showed that the expression peak of dopa decarboxylase in silkworm is higher during molting stage, and decreases after molting. The significant increase in the ecdysone levels of haemolymph was also observed in the artificially fed silkworm larvae with ecdysone hormone. Consistently, the dopa decarboxylase expression was significantly elevated compared to the control. Bm Ddc RNAi induced dopa decarboxylase expression obviously declined in the silkworm larvae, and caused the pupae appeared no pupation or incomplete pupation. Bm Ddc was mainly expressed and stored in the peripheral plasma area near the nucleus in Bm N cells. In larval, Bm Ddc was mainly located in the brain and epidermis, which is consisted with its function in sclerotization and melanization. Overall, the results described that the dopa decarboxylase expression is regulated by the molting hormone, and is a necessary enzyme for the silkworm molting.
Identification of a Novel Prothoracicostatic Hormone and Its Receptor in the Silkworm Bombyx Mori

Abstract:The insect brain regulates the activity of the prothoracic glands to secrete ecdysteroids, which affect growth, molting, and metamorphosis. Here we report the identification of a novel prothoracicostatic factor and its receptor in the silkworm Bombyx mori. The prothoracicostatic factor purified from pupal brains of B. mori is a decapeptide with the conserved structure of an insect myosuppressin and thus named Bommo-myosuppressin. Bommo-myosuppressin dose dependently suppressed the cAMP level and inhibited ecdysteroidogenesis in the larval prothoracic glands at much lower concentrations than the prothoracicostatic peptide, the other prothoracicostatic factor reported previously. In vitro analyses using a prothoracic gland incubation method revealed that Bommo-myosuppressin and prothoracicostatic peptide regulate the prothoracic gland activity via different receptors. In situ hybridization and immunohistochemistry revealed the existence of Bommo-myosuppressin in the brain neurosecretory cells projecting to neurohemal organs in which it is stored. We also identified and functionally characterized a specific receptor for Bommo-myosuppressin and showed its high expression in the prothoracic glands. All these results suggest that Bommo-myosuppressin functions as a prothoracicostatic hormone and plays an important role in controlling insect development.

Abstract:Ecdysteroid is one of the two most important insect hormones, which controls growth and developmental activities of insect. In silkworm, Bombyx mori L., basically ecdysteroid induces molting and metamorphosis in larvae and exogenous administration hastens the silk synthetic activity and cocoon spinning process but it strictly depends on the time of administration. In the present study, we administered phytogenous ecdysteroid to the 48-h-old 5th instar silkworm, B. mori at a single dose of 2 mug per larva to study its effects on the larval growth, cocoon and silk variables. The possible role of ecdysteroid in altering the developmental sequence of silkworm and thereby its effect on cocoon and silk production at molecular level have been tried to get elucidated. The genomic DNA was isolated from the posterior silk gland on day 5 and 7 of the 5th instar from the ecdysteroid treated and the control larvae and was randomly amplified with arbitrary primers. The result presented notable variation in the amplified product suggesting the participation of ecdysteroid in regulating the silk gene. The feeding period of treated larvae was unaffected while the cocoon characters exhibited considerable improvement. The filament traits also were improved notably in the treated larvae. The participation of ecdysteroid in the silk biosynthetic process with its physiological and molecular implications was discussed.
Bombyx Prothoracicostatic Peptides Activate the Sex Peptide Receptor to Regulate Ecdysteroid Biosynthesis.

Abstract:Insect molting and metamorphosis are induced by steroid hormones named ecdysteroids, whose production is regulated by various neuropeptides. We cloned the gene and analyzed the expression of the prothoracicostatic peptide, a unique neuropeptide shown to suppress the production of ecdysteroids in the prothoracic gland of the silkworm, Bombyx mori. We also characterized a Bombyx G protein-coupled receptor, which has previously been identified as an ortholog of the Drosophila sex peptide receptor, as a functional prothoracicostatic peptide receptor. This receptor responded specifically to the prothoracicostatic peptides when examined using a heterologous expression system. The receptor was highly expressed in the prothoracic gland on the day before each larval and pupal ecdysis, when prothoracicostatic peptides are synthesized at a high level in the epiproctodeal glands. These results suggest that the sex peptide receptor functions as a prothoracicostatic peptide receptor in Bombyx and that the peripheral neurosecretory cells as well as the central neuroendocrine system play stage-specific roles in regulating ecdysteroidogenesis.

Abstract:For mass production of human epidermal growth factor ( hEGF), silkworm baculovirus expression vector system (BEVS) was adopted in this study. hEGF gene was in-frame fused with polyhedrin (Ph) gene under the control of Ph promoter and was used to co-transfect BmN cell with the modified Bombyx mori baculovirus DNA to obtained recombinant virus. The ELISA showed a maximum expression on day 4 in larvae and pupae. Both cellular extracts and haemolymph of silkworm larvae infected with rBacPh-EGF could all support the proliferation of Balb/c3T3 cell. The corresponding materials from BmN cell and silkworm larvae infected with wild virus also indicated a weak effect of upregulation on 3T3 cells proliferation. The animal study showed that both pupae infected with rBacPh-EGF virus and wild virus protected the gastric mucosa against ethanol-induced damage in rats, although the protection from pupae infected with wild virus was slightly weak. The mechanism is under investigating.

Abstract:According to the large-scale sequencing of cDNA library from silkworm pupae, the cDNA of a novel gene with blank research background was identified and temporarily named Bm-X. The length of this cDNA is 778 bp. We obtained its ORF for further study by bioinformatics analysis. It is 444 bp and encodes 147 amino acid residues, with a predicted molecular weight (MW) of 16.4 kD and an isoelectric point (pI) of 3.69. In this study, we successfully constructed a recombinant plasmid pET-28a(+)-Bm-X and expressed it in Escherichia coli. We used the fusion protein rBm-X which purified by Niaffinity chromatography to produce polyclonal antibodies against Bm-X for Western blot analysis. The analysis revealed that Bm-X was expressed in the larval midgut, the epidermis and the silk gland. In addition, the subcellular localization analysis of silkworm ovary epithelial cells (BmN cells) showed that Bm-X protein was located both in cytoplasm and nucleus, and the signal was stronger in cytoplasm than in nucleus. Our findings indicate that Bm-X gene is a novel species-specificity gene and its expression product can be detected in tissues of the fifth silkworm instar larvae and BmN cells.

Abstract:Synopsis Although the ecdysteroid of the silkworm had been studied for decades, the proteome of the prothoracic gland, the primary source of ecdysteroid hormones, has not been studied previously. In the present paper, we utilized a proteomic approach to investigate the fifth instar prothoracic gland during the growth and development of the silkworm, Bombyx mori L. The two-dimensional electrophoresis results showed that the majority of proteins were acidic proteins, especially concentrated in the area of 25-65 kDa, with pl values of between 4 and 7, and the difference was not distinct. When compared with Qiufeng (Japanese strain), the interspecific distinction was larger than the intraspecific distinction, and 19 particular spots, excized from the third, fifth and ninth days of p5O (Chinese strain) and Qiufeng were subjected to MALDI-TOF-MS (matrix-assisted laser-desorption ionization-time-of-flight MS) analysis. We sorted them into seven catagories: energetics and/or metabolism, storage proteins, protection, lipid metabolism, signal transduction, cell function and unknown function proteins. Of these proteins, arginine methyltransferase is discussed as playing an important role in regulating the activation of ecdysteroidogenesis via transcription or translation.

Abstract:Molting in insects is regulated by molting hormones (ecdysteroids), which are also crucial to insect growth, development, and reproduction etc. The decreased ecdysteroid in titre results from enhanced ecdysteroid inactivation reactions including the formation of 3-epiecdyson under ecdysone oxidase and 3-dehydroecdysone 3α-reductase (3DE 3α-reductase). In this paper, we cloned and characterized 3-dehydroecdysone 3α-reductase (3DE 3α-reductase) in different tissues and developing stage of the silkworm, Bombyx mori L. The B. mori 3DE 3α-reductase cDNA contains an ORF 783 bp and the deduced protein sequence containing 260 amino acid residues. Analysis showed the deduced 3DE 3α-reductase belongs to SDR family, which has the NAD(P)-binding domain. Using the Escherichia coli, a high level expression of a fusion polypeptide band of approx. 33 kDa was observed. High transcription of 3DE 3α-reductase was mainly presented in the midgut and hemolymph in the third day of fifth instar larvae in silkworm. The expression of 3DE 3α-reductase at different stages of larval showed that the activity in the early instar was high, and then reduced in late instar. This is parallel to the changes of molting hormone titer in larval. 3DE 3α-reductase is key enzyme in inactivation path of ecdysteroid. The data elucidate the regulation of 3DE 3α-reductase in ecdyteroid titer of its targeting organs and the relationship between the enzyme and metamorphosis.
[High Expression of Human EPO Gene in the Larvae and Pupae of the Silkworm, Bombyx Mori].

Abstract:Erythropoietin (EPO) is a glycoprotein hormone produced primarily by the kidney, and is principal factor regulating red blood cell poduction. The synthesized EPO cDNA was inserted into the transfer vector pBlueBac III to generate the recombinant transfer plasmid pBlueBacEPO. Cotransfection of BmN cells with pBlueBacEPO DNA and Wild BmNPV DNA generated the recombinant virus rBmNPVEPO carrying EPO gene driven by the strong promoter of AcNPV polyhedrin gene. The results of Southern blot and PCR reaction confirmed that EPO gene had been correctly inserted in the target position in the BmNPV genome. ELISA assay showed that the EPO gene was expressed with high level in the larvae and pupae of the silkworm. The larvae and pupae produced as high as 62,800 u and 74,000 u in 1 mL hemolymph on the 4th day (larve) and the 5th day(pupae) after infection with the recombinant virus rBmNPVEPO, respectively. Western blot analysis showed the molecular weight of rhEPO produced in the lavae or pupae was about 26 kD. Biologic assay showed the rhEPO had high activity (about 63,000 u in per milliliter of hemolymph) in vitro.
Structural Basis for the Interaction of Diapause Hormone with Its Receptor in the Silkworm, Bombyx Mori

Abstract:Diapause hormone (DH) is a 24-aa amidated neuropeptide that elicits the embryonic diapause of the silkworm, Bombyx mori (Bommo), via sensitive and selective interaction with its receptor, Bommo DH receptor (Bommo-DHR). Previous studies of the structure-activity relationship of Bommo-DH were all based on an in vivo diapause-induction bioassay, which has provided little information on the structure of Bommo-DHR or its iteration with DH. Here, to unveil the interaction of Bommo-DH with its receptor, N-terminally truncated analogs and alanine-scanning mutants of Bommo-DH were chemically synthesized and functionally evaluated by using a Cy5.5-labeled Bommo-DH competitive binding assay and Bommo-DHR-based functional assays, including cAMP assay and Ca2+ mobilization assay. Our study demonstrates that the C-terminal residues of Arg23 and Leu24 of Bommo-DH are essential for the binding and activation of Bommo-DHR, and that Trp19 and Phe20 also contribute to the functional activity of Bommo-DH. In contrast, when Gly21 or Pro22 were replaced with alanine, both mutants exhibited binding and signaling activities that were indistinguishable from the wild-type peptide. Furthermore, our homology modeling and molecular dynamics simulations, together with experimental validations, have identified the residues of Glu89, Phe172, Phe194, and Tyr299 in Bommo-DHR that are critically involved in the interaction with Bommo-DH. Abstract:Prothoracicostatic peptide is one of insect brain neuropeptides, which was observed to inhibit the prothoracic glands of the silkworms, Bombyx mori, at active stage of ecdysone biosynthesis, in the experiments in vitro. In the present paper, prothoracicostatic peptide was injected into the silkworms in different dosages with different methods, single or strengthened injection, and hemolyms were taken thereafter in different intervals. By using 3H-ecdysteriod-radioimmunoassay, a remarkable in vivo activity of prothoracicostatic peptide and developmental changes of ecdysteroid titer in the silkworm in vivo, caused by prothoracicostatic peptide treatment were gained. This is the first evidence that prothoracicostatic peptide significantly inhibits ecdysteroidogenesis in an insect in vivo.

Abstract:Rombyx prothoracicostatic peptide (Bom-PTSP) is a brain neuropeptide that has recently been reported to have in vitro inhibitory activity to prothoracicotropic hormone (PTFH)-stimulated ecdysteroid biosynthesis in the prothoracic gland of the silkworm, Bombyx mori. In the present report, gom-PTSP has been shown to significantly decrease hemolymph ecdysteroid titer in the fifth instar larvae when gom-PTSP was injected into the fifth instar day 8 silkworm larvae, resulting in significant delay in spinning behavior. Abstract:In insects, steroid hormones named ecdysteroids elicit molting and metamorphosis. The prothoracic gland (PG) is a predominant source of ecdysteroids, where their biosynthesis (ecdysteroidogenesis) is regulated by several neuropeptides. Here, we report that FMRFamide-related peptides (FaRPs) regulate ecdysteroidogenesis through direct innervation of the PG in the silkworm Bombyx mori. We purified a previously uncharacterized Bombyx FaRP, DPSFIRFamide, and identified the corresponding Bombyx FMRFamide gene (Bommo-FMRFamide, BRFa), which encodes three additional FaRPs. All BRFa peptides suppressed ecdysteroidogenesis in the PG by reducing cAMP production by means of the receptor for Bommo-myosuppressin, another FaRP we have previously shown to act as a prothoracicostatic factor. BRFa is predominantly expressed in neurosecretory cells of thoracic ganglia, and the neurons in the prothoracic ganglion innervate the PG to supply all four peptides to the gland surface. Electrophysiological recordings during development confirmed the increased firing activity of BRFa neurons in stages with low PG activity and decreased ecdysteroid levels in the hemolymph. To our knowledge, this study provides the first report of peptides controlling ecdysteroidogenesis by direct innervation.
Larva-relevant Genes Expression in Embryonic Development Stages of Domesticated Silkworm (bombyx Mori L.)

Abstract:Gene expression in embryonic stages of silkworm was investigated through analysis of protein expression profile. Proteins from embryonic stages were separated by using two-dimensional electrophoresis (2-DE). The 2-DE images were analyzed using software ImageMaster Elite. Some spots with high abundance would express throughout the following specific stages once they were detected. These proteins may play an important role in embryonic development and metabolism. 5 spots of them were excised from the gels and analyzed their sequences and function. These sequences were identified by using matrix-assisted laser desorption ionization- time of flight mass spectrometry (MALDI-TOF-MS). It is found that five larval functional proteins were expressed in four embryonic stages after homology searching based on peptide mass fingerprinting MS results against related database, that is,juvenile hormone binding protein presented in shortening stage, epidermal growth factor receptor presented in head thorax differentiation stage, larval cuticle protein and accessory gland specific peptide presented in tubercle stage, amylase presented in body pigmentation stage. These functional proteins are seen to express in certain stages for preparing to meet the certain physiological demands, which suggest the larvae relevant genes have been expressed orderly and timely in embryonic stages.

Abstract:Cooperative beamforming and jamming are two efficient schemes to improve the physical-layer security of a wireless relay system in the presence of passive eavesdroppers. However, in most works these two techniques are adopted separately. In this letter, we propose a joint cooperative beamforming and jamming scheme to enhance the security of a cooperative relay network, where a part of intermediate nodes adopt distributed beamforming while others jam the eavesdropper, simultaneously. Since the instantaneous channel state information (CSI) of the eavesdropper may not be known, we propose a cooperative artificial noise transmission based secrecy strategy, subjected to the individual power constraint of each node. The beamformer weights and power allocation can be obtained by solving a second-order convex cone programming (SOCP) together with a linear programming problem. Simulations show the joint scheme greatly improves the security.

Abstract:Diapause of the silkworm,Bombyx mori,a kind of egg diapause, happens in the early stages of embryonic development,decided by both inheritance and environment. Diapause hormone (DH),which is secreted by the suboesophageal ganglion of the silkworm in pupae period,plays a key role in the process of the embryo diapause. The expression of DH gene arose the secretion of DH in 12 cells of suboesophageal ganglion,then the peptide goes into the eggs through the DH receptor on the surface of ovarian cells,regulating the material metabolism,ultimately deciding on the start or the end of diapause. The function and structure of DH peptide,the character and expression of DH gene as well as DH receptor gene are discussed here.
Molecular Characterization of A Peritrophic Membrane Protein from the Silkworm, Bombyx Mori

Abstract:The peritrophic membrane lines the gut of most insects at one or more stages of their life cycles. It facilitates the digestive processes in the guts and protects from invasion by pathogens or food particles. In the current study, a novel PM protein, designated as BmMtch, was identified from the silkworm, Bombyx mori. The open reading frame of BmMtch is 888 bp in length, encoding 295 amino acid residues consisting of two domains (Mito_carr domains) and three transmembrane regions. They are localized on the 11th chromosome as single copy with one exon only. Quantitative real time PCR analysis (qRT-PCR) revealed that BmMtch was mainly expressed in larval fat bodies, Malpighian tubules, testis and ovaries, and could be detected through all stages of the life cycle of silkworm. Immuno-fluorescence analysis indicated that BmMtch was localized within the goblet cell of larval midgut. Western blotting analysis showed that BmMtch were detected in total proteins of PM and larval midgut. The characteristics of BmMtch indicated that BmMtch represents a novel member of insect PM proteins, without chitin-binding domains.

Abstract:The silkworm, Bombyx mori, colleterial gland developed very slowly until 2 days before emergence, then markedly enlarged due to the accumulation of a glue-like substances (mainly including 85% water and 11% proteins). However, the No glue (Ng) mutant female moth secreted only very little glue-like substance and laid loose eggs naturally. High-resolution two-dimensional polyacrylamide gel electrophoresis, followed by computer-assisted analysis, was used to screen the secretary region of colleterial gland protein patterns during different development stages to find quantitative and qualitative difference in protein expression during the pupae and moth stages. More than 700 protein spots were resolved in different developmental stages from the secretary region of the glands and most of the proteins were distributed in the mass range from 30 to 70 kD with pH 4-8. Through comparison and analysis, it was found that 3 proteins were only expressed in the later pupae stage (one or two days before emergence) and moth stage. Furthermore, these proteins were not expressed in the Ng mutant especially actin. There was a great variation of some protein expression volume during the development. Protein spots that changed more than 1.5-fold in expression level (relative to day 9), including 6 spots that were down-regulated and 2 spots that were up-regulated in expression were excised for identification by matrix-assisted loser desorption/ionization-time of flight moss spectrometry. Results indicated that actins that participated or regulated the exocytosis of colleterial gland and other differentially expressed proteins might be related to colleterial gland development or the secretion of a glue-like substance.

Abstract:The paper generalizes advances of a new insect brain neuropeptide, prothoracicostatic peptide, which plays an important role in the insect metamorphosis, and was found and identified from silkworm Bombyx mori recent years. The paper introduces its research background, primary structure, function, mutual relationship between it and prothoracicotropic hormone as well as the results of molecular biology research. The evaluation and expectation of this peptide are discussed.
