Abstract:The reconstruction task in photoacoustic tomography can vary a lot depending on measured targets, geometry, and especially the quantity we want to recover. Specifically, as the signal is generated due to the coupling of light and sound by the photoacoustic effect, we have the possibility to recover acoustic as well as optical tissue parameters. This is referred to as quantitative imaging, i.e, correct recovery of physical parameters and not just a qualitative image. In this chapter, we aim to give an overview on established reconstruction techniques in photoacoustic tomography. We start with modelling of the optical and acoustic phenomena, necessary for a reliable recovery of quantitative values. Furthermore, we give an overview of approaches for the tomographic reconstruction problem with an emphasis on the recovery of quantitative values, from direct and fast analytic approaches to computationally involved optimisation based techniques and recent data-driven approaches.

Abstract:We report on experimental demonstration of photoacoustic tomography for simultaneous recovery of the optical and acoustic properties of heterogeneous media. Photoacoustic images are reconstructed from a series of tissue-like phantom experiments using a finite element-based reconstruction algorithm coupled with a scanning photoacoustic imaging system. The results show that the images obtained are quantitative in terms of the shape, size, location and optical and acoustic properties of the heterogeneities examined.

Abstract:A framework for reconstruction of optical diffusion and absorption coefficients in quantitative photoacoustic tomography is presented. This framework is based on a Tikhonov-type functional with a regularization term promoting sparsity of the absorption coefficient and a prior involving a Kubelka-Munk absorption-diffusion relation that allows to obtain superior reconstructions. The reconstruction problem is formulated as the minimization of this functional subject to the differential constraint given by a photon-propagation model. The solution of this problem is obtained by a fast and robust sequential quadratic hamiltonian algorithm based on the Pontryagin maximum principle. Results of several numerical experiments demonstrate that the proposed computational strategy is able to obtain reconstructions of the optical coefficients with high contrast and resolution for a wide variety of objects.

Abstract:This paper proposes a multigrid inversion framework for quantitative photoacoustic tomography reconstruction. The forward model of optical fluence distribution and the inverse problem are solved at multiple resolutions. A fixed-point iteration scheme is formulated for each resolution and used as a cost function. The simulated and experimental results for quantitative photoacoustic tomography reconstruction show that the proposed multigrid inversion can dramatically reduce the required number of iterations for the optimization process without loss of reliability in the results.

Abstract:Photo-acoustic tomography is a newly developed hybrid imaging modality that combines a high-resolution modality with a high-contrast modality. We analyze the reconstruction of diffusion and absorption parameters in an elliptic equation and improve an earlier result of Bal and Uhlmann to the partial date case. We show that the reconstruction can be uniquely determined by the knowledge of 4 internal data based on well-chosen partial boundary conditions. Stability of this reconstruction is ensured if a convexity condition is satisfied. Similar stability result is obtained without this geometric constraint if 4n well-chosen partial boundary conditions are available, where $n$ is the spatial dimension. The set of well-chosen boundary measurements is characterized by some complex geometric optics (CGO) solutions vanishing on a part of the boundary.

Abstract:Photoacoustic (PA) tomography is a rapidly developing imaging modality which can provide high contrast and spatial-resolution images of light absorption distribution in tissue. However, the quantitative reconstruction of absorption distribution is still a challenge. In this study, we propose an adaptive and quantitative reconstruction algorithm for reducing amplification of noises and artifacts in deep position due to light fluence compensation. In this method, the quantitative processing is integrated into the iterative reconstruction, and absorption coefficient distribution is iteratively updated. At each iteration step, the residual is calculated from detected PA signals and the signals calculated from a forward model by using the initial pressure which is calculated from the production of voxel value and the light fluence. By minimizing the residual, the reconstructed values are converged to the true absorption coefficient distributions. Since this method uses a global optimized compensation, better CNR can be obtained. The results of simulation and phantom experiment indicate that the proposed method provide better CNR at deep region. We expect that the capability of increasing imaging depth will broaden clinical applications.

Abstract:The Bregman method is utilized to quantitatively improve the simultaneous reconstruction of absorption and scattering coefficients for both Jacobian-based and gradient-based methods in quantitative photoacoustic tomography with multiple optical excitations. It is the synergistic combination of Bregman method and the total-variation regularization in achieving the desired quantitative improvement. In particular, the Bregman method is directly applied to the minimization of the nonlinear and nonconvex functional in the gradient-based method, which is efficiently solved via split Bregman method with limited-memory BFGS as inner loops. We show in simulations the feasibility of 3D simultaneous reconstruction.

Abstract:Photoacoustic (PA) tomography (PAT) is a cutting-edge imaging modality for visualizing the internal structure and light-absorption distribution in tissue. However, reconstruction of the absorption distribution has been limited by nonuniform light fluence. This paper introduces a novel method for quantitative reconstruction of the distribution of optical absorption coefficients in tissue. In this method, we implement an iterative algorithm for recovering absorption coefficients from optical absorbed energy maps based on a 3D Monte Carlo simulation of light transport and integrated with fluence compensation to obtain the initialization parameters. In the iteration algorithm, we calculate the deviation between the detected and the computed absorbed energy distribution at each iteration. By minimizing the deviation in the absorbed energy, the recovered values converge to the true absorption distribution. The results of numerical simulation and phantom experiment theoretically and experimentally demonstrate that the proposed method performs an accurately quantitative estimate of the distribution of optical absorption coefficients. This work expects to provide accurate quantitative information for absorbers within tissues or organs, and thereby broaden the clinical applications of PAT.

Abstract:The development of efficient and accurate image reconstruction algorithms is one of the cornerstones of computed tomography. Existing algorithms for quantitative photoacoustic tomography currently operate in a two-stage procedure: First an inverse source problem for the acoustic wave propagation is solved, whereas in a second step the optical parameters are estimated from the result of the first step. Such an approach has several drawbacks. In this paper we therefore propose the use of single-stage reconstruction algorithms for quantitative photoacoustic tomography, where the optical parameters are directly reconstructed from the observed acoustical data. In that context we formulate the image reconstruction problem of quantitative photoacoustic tomography as a single nonlinear inverse problem by coupling the radiative transfer equation with the acoustic wave equation. The inverse problem is approached by Tikhonov regularization with a convex penalty in combination with the proximal gradient iteration for minimizing the Tikhonov functional. We present numerical results, where the proposed single-stage algorithm shows an improved reconstruction quality at a similar computational cost.

Abstract:Two-photon absorption photoacoustic tomography (TP-PAT) is a recent hybrid imaging modality that aims at reconstructing two-photon absorption properties of heterogeneous media from measured ultrasound signals generated by the photoacoustic effect. While there have been extensive experimental studies in recent years to show the great promises of TP-PAT, very little has been done on developing computational methods for quantitative image reconstruction in this imaging modality. In this work, we present a mathematical model for quantitative TP-PAT in diffusive media. We implemented a computational strategy for the reconstruction of the optical absorption coefficients, and provide numerical simulations based on synthetic acoustic data to demonstrate the feasibility of quantitative reconstructions in TP-PAT.

Abstract:Photon scattering imposes a fundamental restriction on optical imaging in turbid media. In this work, we propose a compressive-sensing-based photoacoustic imaging modality that allows single-shot tomography with a single detector in a scattering medium. The nonuniform optical speckle grains created in the diffusive regime, which act as a conventional drawback of optical modality, are used to generate ultrasound locally. The photoacoustic signals from spatial positions can be well extracted from a superimposed signal via introduction of locally modulated time delays. Taking advantage of the compressed measurement assisted by the acoustic mask, we demonstrate a theoretical compressed framework of three-dimensional photoacoustic surface tomography with a broad field of view after one-time optical illumination. This approach can increase the photoacoustic imaging efficiency greatly and reveals the potential for dynamic imaging in optically diffusive media.

Abstract:Quantitative photoacoustic tomography aims at estimating optical parameters from photoacoustic images that are formed utilizing the photoacoustic effect caused by the absorption of an externally introduced light pulse. This optical parameter estimation is an ill-posed inverse problem, and thus it is sensitive to measurement and modeling errors. In this work, we propose a novel way to solve the inverse problem of quantitative photoacoustic tomography based on the perturbation Monte Carlo method. Monte Carlo method for light propagation is a stochastic approach for simulating photon trajectories in a medium with scattering particles. It is widely accepted as an accurate method to simulate light propagation in tissues. Furthermore, it is numerically robust and easy to implement. Perturbation Monte Carlo maintains this robustness and enables forming gradients for the solution of the inverse problem. We validate the method and apply it in the framework of Bayesian inverse problems. The simulations show that the perturbation Monte Carlo method can be used to estimate spatial distributions of both absorption and scattering parameters simultaneously. These estimates are qualitatively good and quantitatively accurate also in parameter scales that are realistic for biological tissues.

Abstract:The field of photoacoustic tomography has experienced considerable growth in the past few years. Although several commercially available pure optical imaging modalities, including confocal microscopy, two-photon microscopy, and optical coherence tomography, have been highly successful, none of these technologies can provide penetration beyond $\sim$1 mm into scattering biological tissues, because they are based on ballistic and quasi-ballistic photons. Heretofore, there has been a void in high-resolution optical imaging beyond this penetration limit. Photoacoustic tomography, which combines high ultrasonic resolution and strong optical contrast in a single modality, has broken through this limitation and filled this void. In this paper, the fundamentals of photoacoustics are first introduced. Then, scanning photoacoustic microscopy and reconstruction-based photoacoustic tomography (or photoacoustic computed tomography) are covered.

Abstract:Photoacoustic tomography is a promising and rapidly developed methodology of biomedical imaging. It confronts an increasing urgent problem to reconstruct the image from weak and noisy photoacoustic signals, owing to its high benefit in extending the imaging depth and decreasing the dose of laser exposure. Based on the time-domain characteristics of photoacoustic signals, a pulse decomposition algorithm is proposed to reconstruct a photoacoustic image from signals with low signal-to-noise ratio. In this method, a photoacoustic signal is decomposed as the weighted summation of a set of pulses in the time-domain. Images are reconstructed from the weight factors, which are directly related to the optical absorption coefficient. Both simulation and experiment are conducted to test the performance of the method. Numerical simulations show that when the signal-to-noise ratio is -4 dB, the proposed method decreases the reconstruction error to about 17%, in comparison with the conventional back-projection method. Moreover, it can produce acceptable images even when the signal-to-noise ratio is decreased to -10 dB. Experiments show that, when the laser influence level is low, the proposed method achieves a relatively clean image of a hair phantom with some well preserved pattern details. The proposed method demonstrates imaging potential of photoacoustic tomography in expanding applications.

Abstract:Quantitative photo-acoustic tomography (QPAT) seeks to reconstruct a distribution of optical attenuation coefficients inside a sample from a set of time series of pressure data that is measured outside the sample. The associated inverse problems involve two steps, namely acoustic and optical, which can be solved separately or as a direct composite problem. We adopt the latter approach for realistic acoustic media that possess heterogeneous and often not accurately known distributions for sound speed and ambient density, as well as an attenuation following a frequency power law that is evident in tissue media. We use a Diffusion Approximation (DA) model for the optical portion of the problem. We solve the corresponding composite inverse problem using three total variation (TV) regularised optimisation approaches. Accordingly, we develop two Krylov-subspace inexact-Newton algorithms that utilise the Jacobian matrix in a matrix-free manner in order to handle the computational cost. Additionally, we use a gradient-based algorithm that computes a search direction using the L-BFGS method, and applies a TV regularisation based on the Alternating Direction Method of Multipliers (ADMM) as a benchmark, because this method is popular for QPAT and direct QPAT. The results indicate the superiority of the developed inexact Newton algorithms over gradient-based Quasi-Newton approaches for a comparable computational complexity.

Abstract:Photoacoustic imaging, an acoustic imaging modality with potentially optical resolution in an optical turbid medium, has attracted great attention. However, the convergence of wavefront optimization and raster scanning in computational photoacoustic imaging leads to the challenge of fast mapping, especially for a spatial resolution approaching the acoustic deep-subwavelength regime. As a sparse sampling paradigm, compressive sensing has been applied in numerous fields to accelerate data acquisition without significant quality losses. In this work, we propose a dual-compressed approach for photoacoustic surface tomography that enables high-efficiency imaging with 3D spatial resolution unlimited by the acoustics in a turbid environment. The dual-compressed photoacoustic imaging with single-pixel detection, enabled by spatially optical modulation with synchronized temporally photoacoustic coding, allows decoding of the fine optical information from the modulated acoustic signal even when the variance of original photoacoustic signals is weak. We perform a proof-of-principle numerical demonstration of dual-compressed photoacoustic imaging, that resolves acoustic sub-acoustic-wavelength details with a significantly reduced number of measurements, revealing the potential for dynamic imaging. The dual-compressed concept, which transforms unobtrusive spatial difference into spatio-temporal detectable information, can be generalized to other imaging modalities to realize efficient, high-spatial-resolution imaging.

Abstract:In this paper, we present a numerical reconstruction method for quantitative photoacoustic tomography (QPAT), based on the radiative transfer equation (RTE), which models light propagation more accurately than diffusion approximation (DA). We investigate the reconstruction of absorption coefficient and scattering coefficient of biological tissues. An improved fixed-point iterative method to retrieve the absorption coefficient, given the scattering coefficient, is proposed for its cheap computational cost; the convergence of this method is also proved. The Barzilai–Borwein (BB) method is applied to retrieve two coefficients simultaneously. Since the reconstruction of optical coefficients involves the solutions of original and adjoint RTEs in the framework of optimization, an efficient solver with high accuracy is developed from Gao and Zhao (2009 Transp. Theory Stat. Phys. 38 149–92). Simulation experiments illustrate that the improved fixed-point iterative method and the BB method are competitive methods for QPAT in the relevant cases.

Abstract:Quantitative photoacoustic tomography (qPAT) is an imaging technique aimed at estimating chromophore concentrations inside tissues from photoacoustic images, which are formed by combining optical information and ultrasonic propagation. The application of qPAT as a transcranial imaging modality is complicated by shear waves that can be produced when ultrasound waves travel from soft tissue to bone. Because of this, the estimation of chromophores distributions near the skull can be problematic. In this paper, we take steps towards compensating for aberrations of the recorded photoacoustic signals caused by elastic wave propagation. With photoacoustic data simulated in a coupled elastic-acoustic domain, we conduct inversions in a purely acoustic domain. Estimation of the posterior density of the initial pressure is achieved by inversion under the Bayesian framework. We utilize the Bayesian approximation error approach to compensate for the modelling errors arising from approximating a coupled elastic-acoustic domain with a purely fluid domain. The resulting reconstructions and corresponding uncertainty estimates are then used to evaluate the posterior density of the optical absorption parameter. In the sense of the posterior uncertainty, the results show that the Bayesian approximation error approach yields a more feasible estimate for the posterior model of the initial pressure which, in turn, yields a more feasible estimate for the posterior model of the absorption coefficient.

Abstract:Quantitative photoacoustic tomography (PAT) reconstructs optical maps using ultrasonic measurements, with improved resolution from conventional optical imaging due to significantly smaller acoustic scattering than optical scattering for detecting signals in depth. In this work, formulating quantitative PAT as a nonlinear least-squares problem with l(1)-norm sparsity regularization, we develop an efficient gradient-based reconstruction algorithm using a forward-backward splitting method, and prove its convergence for such a nonconvex problem.
