Mycoplasma bovis induced inhibition of bovine PBMC proliferation is ameliorated after 1 blocking the immune-inhibitory programmed death-1 (PD-1) receptor

Abstract:21 Mycoplasma bovis ( M. bovis ) induced-immune suppression is a major obstacle faced by 22 the host for controlling infections. M. bovis impairment of antigen-specific T-cells responses is 23 achieved through inhibiting the proliferation of peripheral-blood mononuclear cells (PBMC). 24 This impairment may contribute to the persistence of M. bovis infection in various sites including 25 lungs and its systemic spread to various organs like joints, with the underlying mechanisms 26 remaining elusive. Here, we elucidated the role of the immune-inhibitory receptor programmed 27 death-1 (PD-1) and its ligand (PD-L1) in M. bovis infection. Flow cytometric (FCM) analyses 28 revealed an upregulation of PD-L1 ligand expression on tracheal and lung epithelial cell lines 29 after M. bovis infection. In addition, we found an increased PD-L1 expression on purified lung 30 lavage macrophages following M. bovis infection by FCM and its localization by 31 immunofluorescence comparing infected and control lung tissue sections. Moreover, M. bovis 32 infection increased expression of the PD-1 receptor on the total PBMC and in gated CD4 + and 33 CD8 + T cells sub-populations. We demonstrated that M. bovis infection induced a significant 34 decrease in CD4 + PD1 INT and CD8 + PD1 INT subsets with intermediate PD1 expression and 35 functioning as progenitor pools giving rise to CD4 + PD1 HIGH and CD8 + PD1 HIGH subsets with high 36 PD1 expression. We blocked PD-1 receptors on PBMC using anti-PD1 antibody at the beginning 37 of infection leading to significant restoration of PBMCs’ proliferation. Taken together, our data 38 indicates a significant involvement of the PD-1/PD-L1 inhibitory pathway during M. bovis 39 infection and its associated immune exhaustion culminating in impaired host immune responses.
