Synthesis of Vitamin B 12 -Antibiotic Conjugates with Greatly Improved Activity against Gram-Negative Bacteria

Abstract:There is an urgent need to discover new antibiotics and improve the efficacy of known antibiotics against Gram-negative bacteria. "Trojan horse" conjugates are novel and promising antibiotics. Herein we report the design and synthesis of vitamin-B<sub>12</sub>–ampicillin conjugates, which exhibited more than 500 times improved activity against <i>Escherichia coli</i> compared with ampicillin itself. What problem does this paper attempt to address?
This paper aims to solve the problem of the insufficient efficacy of antibiotics against Gram - negative bacteria. Specifically, the researchers designed and synthesized Vitamin B12 - ampicillin conjugates, hoping to improve the activity of ampicillin against Escherichia coli through the specific transport pathway of Vitamin B12. The experimental results showed that the antibacterial activity of these conjugates was more than 500 times higher than that of ampicillin alone. In addition, the study also explored whether this strategy could be applied to other antibiotics, especially those whose targets are located in the bacterial cytoplasm. Through these studies, the authors demonstrated that the use of the transport mechanism of Vitamin B12 can significantly enhance the effectiveness of antibiotics, providing new ideas and methods for the development of new antibiotics.
