Abstract:A new Schiff-base fluorescence probe CS containing benzimidazole and coumarin fluorophores was synthesized and characterized by NMR, IR and ESI-MS. Fluorescence and absorption spectra show the unique changes in the presence of Hg2+ and Cu2+. Hg2+ induces a remarkable fluorescence enhancement and results in mercury-promoted hydrolysis of a mercury-CS complex to convert the imine group to the aldehyde group. Cu2+ leads to a colorimetric change from colorless to pale yellow and causes formation of a copper-CS complex. Thus, CS can act as a fluorescent chemodosimeter for Hg2+ and a naked-eye chemosensor Cu2+. The cell imaging experiments for Hg2+ show a slow enhancement of a bright blue fluorescence with increasing incubation time, while Cu2+ instantly quenches the fluorescence and then the fluorescence intensity has no further change by extending the incubation time. Therefore, the probe CS shows its potential application for simultaneous differential detection of Hg2+ and Cu2+ in living cells based on the fluorescence response time. (C) 2016 Elsevier B.V. Abstract:Mercury (Hg) is a heavy metal with high toxicity and easy migration; it can be enriched through the food chain, and cause serious threats to the natural environment and human health. So, the development of a method that can be used to detect mercury ions (Hg2+) in the environment, in cells, and in organisms is very important. Here, a new 7-hydroxycoumarin-derived carbonothioate-based probe (CC-Hg) was designed and synthesized for detection of Hg2+. After addition of Hg2+, a large fluorescence enhancement was observed due to the formation of 7-hydroxyl, which reinforced the intramolecular charge transfer process. The CC-Hg probe had good water solubility and selectivity. Moreover, the probe was able to detect Hg2+ quantitatively over the concentration range 0-2 mu M and with a detection limit of 7.9 nM. Importantly, we successfully applied the probe to detect Hg2+ in water samples, in living cells, and in zebrafish. The experimental results demonstrated its potential value in practical applications.

Abstract:Mercury ions are highly toxic and can accumulate along food chains in water, soil, crops and animals. Effective detection of mercury ions in various media is of great significance for maintaining the ecological environment and protecting people's health. In this work, a mercury ions specific fluorescent probe was developed by a simple one-step reaction of commercial substrates of 4-chloro-7-nitro-2,1,3-benzoxadiazole and 1-(2-aminoethyl)-4-methylpiperazine. Investigation on sensing behavior showed that this probe had high sensitivity and selectivity towards mercury ions. Furthermore, this probe could be used as a tool to track the level of mercury ions in living system. In living cells, the probe with green emission emitted a bright red fluorescence when it was bound to mercury ions. In Arabidopsis thaliana, similar red emission could be detected from the root tip and stalk when A. thaliana was grown in culture medium containing mercury ions. The imaging in zebrafish showed that mercury ions were mainly concentrated in the stomach and head of zebrafish. Especially, this probe could be applied in quantitative analysis of mercury ions in tap water, green tea, sea shrimp and soil. This work provided a practical tool for the detection of mercury ions in living systems and quantitative analysis in real samples.

Abstract:In this work, a simple dichlororesorufin-based colorimetric and fluorescent probe R-Hg with the recognition receptor of carbon- othioate was designed, synthesized, and exploited to determine mercury ions in aqueous solution and living cells as well as in zebrafish. After a series of analytical tests, probe R-Hg showed some excellent characteristics, such as ultrasensitivity, high selectivity, and good water solubility. The probe solution exhibited a very large fluorescence enhancement (about 150-fold) upon addition of He2+. Additionally, probe R-Hg exhibited excellent sensitivity, which could quantificationally detect Hg2+ at the nanomolar level, and the detection limit was 0.49 nM. Moreover, probe R-Hg could be used as a "naked-eye" probe for monitoring Hg2+, because the color of its solution transformed remarkably from yellow into pink with the participation of Hg2+. Importantly, probe R-Hg has potential application value and has been successfully applied to the determination of He2+ in practical water samples and in living RAW 264.7 cells as well as in zebrafish as a bioimaging reagent.

Abstract:Mercury, as a highly toxic heavy metal, can cause very serious harm to human health and even death in severe cases. Therefore, we synthesized a novel ratiometric fluorescent probe for detecting mercury ions, with mercaptoethanol as the recognition receptor. Probe CMER could determine mercury ions in 0-1.6 μM and the detection limit is 7.6 nM. Moreover, CMER manifested a fast response for Hg2+ (within 5 s) and simultaneously observed that the color changed from light yellow to orange by naked eye. In addition to these preeminent spectral properties, the probe also had satisfactory bioimaging results in RAW 264.7 macrophage cells and zebrafish.

Abstract:Mercury ions (Hg2+) cause great harm to the environment and living organisms owing to their strong migration and transformation ability. Therefore, the detection of Hg2+ in the environment and living systems becomes significant. In this study, we constructed a novel amino coordination type-specific Hg2+ fluorescent probe TD-Hg. The probe provides diaminomaleonitrile as the coordination group for Hg2+ and could respond to Hg2+ in a very short time (< 5 s). Moreover, probe TD-Hg can detect Hg2+ in environmental water samples with a high sensitivity, which proves that the probe has good environmental application potential. In addition, the probe possesses good biocompatibility verified by the experiments on cytotoxicity, behavioral toxicology, hepatotoxicity, cardiotoxicity, the toxicity of the blood vessels, and immunotoxicity using cultured cell model or zebrafish larvae. Note that the probe can monitor the Hg2+ content in cultured cells and zebrafish larvae, which makes it possess a high ability for biological application in monitoring Hg2+. In conclusion, the construction of probe TD-Hg provides an effective tool for the specific detection of Hg2+.

Abstract:The detection of ionic mercury (Hg2+) is very important because it is a highly toxic environmental pollutant that could cause serious diseases and threaten human health. Herein, we designed a new carbonothioate-based far-red fluorescent probe, CBRB, with a seminaphthorhodafluor dye as the fluorophore for the detection of Hg2+. The CBRB probe by itself exhibited very weak fluorescence due to the enhanced photo-induced electron transfer (PET) effect and inhibited the intramolecular charge transfer (ICT) process caused by the carbonothioate moiety. Upon addition of Hg2+, a tremendous fluorescence enhancement was achieved, attributed to the removal of the carbonothioate group via a specific mercury-promoted desulfurization reaction. Moreover, the probe displayed a large Stokes shift (about 105 nm) and was used to quantitatively measure the concentration of Hg2+ for concentrations ranging from 0 to 1 M (DL = 3.6 nM). In addition, CBRB in our experiments responded exclusively to Hg2+, even in the presence of high concentrations other ions. Gratifyingly, this probe was successfully used to monitor Hg2+ in environmental water samples and to image Hg2+ in living cells as well as in zebrafish.

Abstract:A highly selectivity and senstivity fluorescent probe for Hg2+. A simple naphthalimide‐based fluorescent probe was designed and synthesized for the determination of mercury ion (Hg2+). The probe showed a noticeable fluorescence quenching response for Hg2+. When added with Hg2+, the fluorescence intensity of the probe at 560 nm was remarkably decreased with the color changed from yellow to colorless under ultraviolet (UV) light. The probe had a notable selectivity and sensitivity for Hg2+ and displayed an excellent sensing performance when detecting Hg2+ at low concentration (19.5 nM). The binding phenomenon between the probe and Hg2+ was identified by Job's method and high‐resolution mass spectrometry (HRMS). Moreover, the probe was not only utilized to identify Hg2+ in real samples with satisfactory results (92.00%–110.00%) but also was successfully used for bioimaging in cells and zebrafish. The recognition mechanism has been verified by transmission electron microscopy (TEM) for the first time. All the results showed that the probe could be used as a potent useful tool for detection of Hg2+.

Abstract:The accumulation of mercury pollution in plants can induce severe injury to human beings. It is a great challenge to monitor ultra-low concentrations of mercury in complicated matrixes. In this study, we successfully developed a strategy via Hg2+-triggered naphthalene-based fluorescent probe 1, which formed excimer that subsequently emitted fluorescence for the successful detection of ultra-low concentrations of Hg2+. The coordination of N and S atoms with Hg2+ facilitated the formation of excimer from the naphthalene-conjugated planes that were in sufficiently close proximity. Suppression of CN bond rotation also induced the chelation-enhanced fluorescence (CHEF) effect, and the cumulative result of these effects was obvious fluorescent enhancement. Compared with probe 2, the other key factor for detection of Hg2+ is that the electrons of the hydroxyl group can easily transfer to a naphthalene moiety, resulting in an augmented π-electron density that enhanced the π-π stacking of the naphthalene-conjugated excimer. After detailed spectral studies and mechanism discussions, it was realized that probe 1 was able to detect ultra-low concentrations of Hg2+ in PBS buffer solution. The detection limit was calculated to be 1.98 nM. On account of the excellent performances, the probe was successfully applied in monitoring Hg2+ in water and pea sprouts with the potential for application as an advanced warning of contamination.

Furthermore, the confocal fluorescence images demonstrated that XL-1 could be used for monitoring Hg2+ in living cells.

Abstract:Mercury ions (Hg 2+ ) are a class of heavy metal ions that can cause serious harm to the human body due to their toxicity, fluidity, and bioaccumulation. Direct contact with Hg 2+ can cause adverse effects on the human body. In addition, long-term exposure to Hg 2+ can lead to extreme toxicity of enzymes and proteins in the body, disrupting the cell cycle and thus endangering human life. It is becoming increasingly important to develop a method that is reliable, stable, and sensitive for detecting Hg 2+ . In this study, a near-infrared "turn-on" fluorescent probe based on dicyanomethylene-4 H -pyran, DCM-Hg, was designed and synthesized to determine Hg 2+ over other metal ions. Probe DCM-Hg had a detection limit (LOD) of 3.35 μM when used to detect Hg 2+ . The Hg 2+ concentrations and the fluorescence intensities at 695 nm showed a good linear correlation. Additionally, Hg 2+ in HepG2 cells was successfully visualized using the current probe.

Abstract:Developing some methods that can simply and effectively detect mercury ions (Hg^2+) in the environment and biological systems are very important due to the problems of high toxicity and biological accumulation. Herein, we report a simple rhodol-derived colorimetric and fluorescent probe rhodol-Hg with a recognition receptor of carbonothioate for the specific determination of Hg^2+. The color of probe rhodol-Hg solution changed remarkably from colorless to pink in the presence of Hg^2+, thus rhodol-Hg could act as a “naked-eye” probe for Hg^2+. Additionally, this probe exhibited high selectivity and ultrasensitivity in aqueous solution with the limit of detection (LOD) of 1.4 nM toward Hg^2+, and the linear range was 0–0.8 μM determined by turn-on fluorescence spectrometry. Importantly, this probe has been successfully used for the detection of Hg^2+ in environmental waters and living cells.

Abstract:As a kind of heavy metal ions, mercury ions (Hg2+) have important effects on ecological environment and human health. Since Hg2+ interact with mercaptan molecules in the living body, they gather in the living body and endanger health or even life, so the detection of Hg2+ is indispensable. In this research, taking advantage of thymine moiety as a specific ligand for chelating with Hg2+, a new type of "T-Hg-T" fluorescent probe BC-Hg was designed and synthesized to specially detect Hg2+ in real water samples and living systems. The barbituric acid unit in the probe can react with Hg2+ and then deprotonate to form polymer and precipitate. Probe BC-Hg has a rapid response to the detection of Hg2+ and strong anti-interference ability. It has a good linear detection of Hg2+ in the range of 0-1.5 mu M, so it can quantitatively detect Hg2+ within a certain concentration range. Probe BC-Hg has obvious "naked eye" colorimetric effect and can be used to determine the generation of Hg2+ polymer by the Tyndall effect. In addition, probe BC-Hg can also be used to detect Hg2+ in real water samples, cells, and zebrafish. Therefore, the probe can serve as an effective tool for detecting Hg2+ in environment and organisms.

Abstract:Environmental pollution problem, especially mercury ions (Hg2+) pollution, seriously threatens the organism health. Numerous researches have revealed that even extremely low concentration of Hg2+ can severely damage the organisms. Therefore, it is essential for real-time detection of Hg2+. Fluorescent probe technology with great advantages has become the preferred technique for environmental detection and in vivo analysis of Hg2+. In recent years, lots of contributions have been committed to designing and synthesizing novel fluorescent probes for environmental pollutant Hg2+ analysis. In this feature article, we review the recent advances in fluorescent probes for Hg2+ analysis, focusing on probe response modes, including signal-decreased fluorescent probes, signal-enhanced fluorescent probes, ratiometric fluorescent probes as well as fluorescent probes for Hg2+ and other species analysis. Moreover, the design strategies, performance and applications of fluorescent probes are summarized in detail. Challenges and prospects for fluorescent probe are proposed, orienting new design strategies and applications expanding.

Abstract:Since mercury could accumulate along with the food chains and undermine environment and human health, development of efficient mercury detection tools in water samples, plants and living systems is of great significance. In this work, we designed a hemicyanine-based fluorescent probe HCYS for mercury detection. The probe exhibited distinct near-infrared fluorescence intensity. Based on the spin orbit coupling effect and heavy atom effect of Hg 2+ , a significant fluorescent change was exhibited since Hg 2+ chelated with the probe by inhibition of the intramolecular charge transfer process in HCYS . The limit of detection (LOD) was calculated to be 1.41 μM. Exploiting the benefits of fast response time (less than 1 min), high selectivity and noninvasively track capacity, the probe has been applied in biological systems. The solid results proved that the probe HCYS is a powerful tool for Hg 2+ imaging in living cells, zebrafish, pea spouts and water samples.

Abstract:A water-soluble fluorescent probe for Hg(2+) based on a rhodamine B derivative was designed and synthesized. The new probe showed reversible colorimetric and fluorescent response to Hg(2+) in a fully aqueous solution. The probe exhibited real-time detection of Hg(2+) with high selectivity in media containing less than 1% organic cosolvent. Furthermore, bioimaging studies indicated that the new probe was cell permeable and suitable for the real-time imaging of Hg(2+) in living cells by confocal microscopy.

Abstract:Mercury ions (Hg2+), as one of heavy transition metals (HTM), is a highly toxic metal that is hazardous to human health. Here an aggregation-induced emission (AIE) fluorescent probe is designed for the highly sensitive and selective detection of Hg2+. The probe is engineered with a tetraphenylethene (TPE) derivative as the fluorophore and thiopropionic acid as the site of recognition for Hg2+. Due to the different solubilities of the probe AIE-COOH and its corresponding product after reaction with Hg2+. The probe demonstrates a maximum detection limit of 22 nM and a fast response time of ∼100 s. Simultaneously, AIE-COOH exhibits outstanding detectivity and hypersensitivity for the detection of Hg2+ in aqueous solutions. These characteristics demonstrate that AIE-COOH hold a great potential in environmental, food and biological systems. Moreover, we have also successfully applied it to Hg2+ fluorescence imaging in in living cells.
