Abstract:We describe a new method for determining receptor-ligand association/dissociation events across the interface of two surfaces (two-dimensional binding) by monitoring abrupt decrease/resumption in thermal fluctuations of a biomembrane force probe. Our method has been validated by rigorous control experiments and kinetic experiments. We show that cellular on-rate of association can be measured by analysis of intervals from a dissociation event to the next association event (waiting times). Similarly, off-rate of molecular dissociation can be measured by analysis of intervals from an association event to the next dissociation event (bond lifetimes). Different types of molecular bonds could be distinguished by different levels of reduction in thermal fluctuations. This novel method provides a powerful tool to study cell adhesion and signaling mediated by single or multiple receptor-ligand species.

Abstract:Biological signaling is imagined as a combination of activation and transport. The former is triggered by local molecular interactions and the latter is the result of molecular diffusion. However, other fundamental physical principles of communication have yet to be addressed. We have recently shown, that lipid interfaces allow for the excitation and propagation of sound pulses. Here we demonstrate, that these reversible perturbations can control the activity of membrane embedded enzymes without the necessity of molecular transport. They therefore allow for the rapid communication between distant biological entities (e.g. receptor and enzyme) at the speed of sound, which is here in the order of 1 m/s within the membrane. The mechanism reported provides a new physical framework for biological signaling.

Abstract:An approach to optical transduction and amplification of amphiphile-triggered orientational responses of liquid crystals (LCs) based on the interference effect was developed. The sensitive substrate was obtained by lading 4'-pentyl-4-cyanobiphenyl (5CB) into three-dimensionally ordered silica colloidal crystal (SCC) films. Changes in the optical thickness (Delta OT) of the substrates, which are inverted by their Fabry-Perot fringes, depend on the changes of the refractive index caused by the differences in the orientations of LCs. The orientation changes of LCs loading into SCC films have the effect of amplifying signals. These are based on the interactions between surfactants (alkyl trimethylammonium halides (CnTABs, n = 8, 10, 12, 14, and 16) and sodium lauryl sulfonate (SLS)) and LCs, which induce a particular orientation of the LCs molecules. In this flowing system, the reversibility of the signal response for the adsorption of amphiphile was related to the length of the surfactant chain and its critical micelle concentration (CMC). A new method capable of real-time sensing adsorbate-triggered anchoring transitions based on LC-infiltrated SCC films was accomplished. These results provide basics and principles for online, label-free, and real-time analysis of molecules and their interactions in a flowing environment based on the interference effect.
Evidence for 2D Solitary Sound Waves in a Lipid Controlled Interface and its Biological Implications

Abstract:Biological membranes by virtue of their elastic properties should be capable of propagating localized perturbations analogous to sound waves. However, the existence and the possible role of such waves in communication in biology remains unexplored. Here we report the first observations of 2D solitary elastic pulses in lipid interfaces, excited mechanically and detected by FRET. We demonstrate that the nonlinearity near a maximum in the susceptibility of the lipid monolayer results in solitary pulses that also have a threshold for excitation. These experiments clearly demonstrate that the state of the interface regulates the propagation of pulses both qualitatively and quantitatively. We elaborate on the striking similarity of the observed phenomenon to nerve pulse propagation and a thermodynamic basis of cell signaling in general.

Abstract:The existence of acoustic pulse propagation in lipid monolayers at the air-water interface is well known. These pulses are controlled by the thermodynamic state of the lipid membrane. Nevertheless, the role of acoustic pulses for intra- and intercellular communication are still a matter of debate. Herein, we used the dye di 4- -ANEPPDHQ, which is known to be sensitive to the physical state and transmembrane potential of membranes, in order to gain insight into compression waves in lipid-based membrane interfaces. The dye was incorporated into lipid monolayers made of phosphatidylserine or phosphatidylcholine at the air-water-interface. A significant blue shift of the emission spectrum was detected when the state of the monolayer was changed from the liquid expanded (LE) to the liquid condensed (LC) phase. This transition-sensitivity of di-4-ANEPPDHQ was generalized in experiments with the bulk solvent dimethyl sulfoxide (DMSO). Upon crystallization of solvent, the emission spectrum also underwent a blue shift. During compression pulses in lipid monolayers, a significant fluorescence response was only observed when in the main transition regime. The optical signature of these waves, in terms of sign and magnitude, was identical to the response of di-4-ANEPPDHQ during action potentials in neurons and excitable plant cells. These findings corroborated the suggestion that action potentials are nonlinear state changes that propagate in the cell membrane.

Abstract:Currently, biological signaling is envisaged as a combination of activation and movement, triggered by local molecular interactions and molecular diffusion, respectively. Abstract:ABSTRACT Over recent decades, lipid membranes have become standard models for examining the biophysics and biochemistry of cell membranes. Interrogation of lipid domains within biomembranes is generally done with fluorescence microscopy via exogenous chemical probes. However, most fluorophores have limited partitioning tunability, with the majority segregating in the least biologically relevant domains ( i.e. , low-density liquid domains). Therefore, a molecular-level picture of the majority of non-labeled lipids forming the membrane is still elusive. Here, we present simple, label-free imaging of domain formation in lipid monolayers, with chemical selectivity in unraveling lipid and cholesterol composition in all domain types. Exploiting conventional vibrational contrast in spontaneous Raman imaging, combined with chemometrics analysis, allows for examination of ternary systems containing saturated lipids, unsaturated lipids, and cholesterol. We confirm features commonly observed by fluorescence microscopy, and provide an unprecedented analysis of cholesterol distribution at the single-membrane level.

Abstract:Local changes in pH are known to significantly alter the state and activity of proteins and in particular enzymes. pH variations induced by pulses propagating along soft interfaces (e.g. the lipid bilayer) would therefore constitute an important pillar towards a new physical mechanism of biochemical regulation and biological signaling. Here we investigate the pH-induced physical perturbation of a lipid interface and the physiochemical nature of the subsequent acoustic propagation. Pulses are stimulated by local acidification of a lipid monolayer and propagate, in analogy to sound, at velocities controlled by the two-dimensional compressibility of the interface. With transient local pH changes of 0.6 units directly observed at the interface and velocities up to 1.4 m/s this represents hitherto the fastest protonic communication observed. Furthermore simultaneously propagating mechanical and electrical changes in the lipid interface up to 8 mN/m and 100 mV are detected, exposing the thermodynamic nature of these pulses. Finally, these pulses are excitable only beyond a threshold for protonation, determined by the pKa of the lipid head groups. When combined with an enzymatic pH-optimum, the proposed communication can be very specific, thus providing a new physical basis for intra- and intercellular signaling via two-dimensional sound waves at interfaces.
Quantitative Assessment of the Dynamic Modification of Lipid-DNA Probes on Live Cell Membranes

Abstract:Synthetic lipid-DNA probes have recently attracted much attention for cell membrane analysis, transmembrane signal transduction, and regulating intercellular networks. These lipid-DNA probes can spontaneously insert onto plasma membranes simply after incubation. The highly precise and controllable DNA interactions have further allowed the programmable manipulation of these membrane-anchored functional probes. However, we still have quite limited understanding of how these lipid-DNA probes interact with cell membranes and also what parameters determine this process. In this study, we have systematically studied the dynamic process of cell membrane modification with a group of lipid-DNA probes. Our results indicated that the hydrophobicity of the lipid-DNA probes is strongly correlated with their membrane insertion and departure rates. Most cell membrane insertion stems from the monomeric form of probes, rather than the aggregates. Lipid-DNA probes can be removed from cell membranes through either endocytosis or direct outflow into the solution. As a result, long-term probe modifications on cell membranes can be realized in the presence of excess probes in the solution and/or endocytosis inhibitors. For the first time, we have successfully improved the membrane persistence of lipid-DNA probes to more than 24 h. Our quantitative data have dramatically improved our understanding of how lipid-DNA probes dynamically interact with cell membranes. These results can be further used to allow a broad range of applications of lipid-DNA probes for cell membrane analysis and regulation.

Abstract:Cell membranes are incredibly complex environments containing hundreds of components. Despite substantial advances in the past decade, fundamental questions related to lipid-lipid interactions and heterogeneity persist. This review explores the complexity of lipid membranes, showcasing recent advances in vibrational spectroscopy to characterize the structure, dynamics, and interactions at the membrane interface. We include an overview of modern techniques such as surface-enhanced infrared spectroscopy as a steady-state technique with single-bilayer sensitivity, two-dimensional sum-frequency generation spectroscopy, and two-dimensional infrared spectroscopy to measure time-evolving structures and dynamics with femtosecond time resolution. Furthermore, we discuss the potential of multiscale molecular dynamics (MD) simulations, focusing on recently developed simulation algorithms, which have emerged as a powerful approach to interpret complex spectra. We highlight the ongoing challenges in studying heterogeneous environments in multicomponent membranes via current vibrational spectroscopic techniques and MD simulations. Overall, this review provides an up-to-date comprehensive overview of the powerful combination of vibrational spectroscopy and simulations, which has great potential to illuminate lipid-lipid, lipid-protein, and lipid-water interactions in the intricate conformational landscape of cell membranes. Abstract:Membrane tension plays a significant role in many cellular processes including cell adhesion, migration and spreading. Despite the importance of membrane tension, it remains difficult to measure in vivo. Recently, the development of non-invasive fluorescent probes have made great progress, especially excited-state deplanarization in molecular rotors has been applied to image membrane tension in living cells. Nevertheless, an intrinsic limitation of such kind of probe is that they depend on the lipid packing, and how the lipid packing responds to the membrane tension change remains unclear. Therefore, in this work, we used a polarity-sensitive membrane probe to investigate the possible response mechanism of lipid packing to the change of membrane tension that was regulated by osmotic shocks. The results showed that an increase in membrane tension could stretch the lipids apart with large displacements, and this change was not homogeneous on the whole membrane, instead, increase of membrane tension induced phase separation.

Abstract:Probing the interaction between molecules and protocells is crucial for understanding the passive transport of functional molecules in and out of artificial and real cells. Second-harmonic generation (SHG) has been proven to be a powerful method for analyzing the adsorption and cross-membrane transport of molecules on lipid bilayers. In this study, we used SHG and two-photon fluorescence (TPF) imaging to study the interaction of charged dye molecules (D289) with a lipid vesicle. Unexpectedly, it was observed that the transport of D289 at a relatively high concentration is not as efficient as that at a lower dye concentration. Periodic shrinking of the model protocell and discharging of D289 out from the vesicle were revealed by combined analyses of SHG and TPF images. The response of the vesicle to a surfactant was also analyzed with D289 as a probe. This work demonstrates that the combined SHG and TPF imaging method is a unique approach that can provide detailed information on the interaction of molecules and lipids (both morphology and molecular kinetics). Determining these subtle interfacial kinetics in molecules is important for understanding the mechanism of many biophysical processes occurring on lipids.

Abstract:Membrane receptors, among other membrane proteins that universally exist on the cell surface, are a class of important macromolecules responsible for cell-signaling processes. They are activated or inhibited through selective binding with endogenous ligand molecules. These binding activities are implicated in physiologic as well as pathologic events if errors in signaling interactions occur. The thorough understanding of receptor-ligand recognition is of paramount importance for fundamental biological studies and, in particular, could improve the precision of disease diagnosis and therapy. However, current techniques capable of tracking the dynamic actions of membrane receptors are elusive. This review highlights our recent progress in the development of fluorescent probes and supramolecular materials for receptor-targeting biosensing and bioimaging. Strategies in probe construction and the practical biomedical applications that have been achieved are discussed. Perspectives and challenges with respect to this interdisciplinary field are offered.

Abstract:By reverting to spectroscopy, changes in the biological environment of a fluorescent probe can be monitored and the presence of various phases of the surrounding lipid bilayer membranes can be detected. This study highlights the important differences in orientation and location and therefore in efficiency between the probes when they are used in fluorescence microscopy to screen various lipid bilayer membrane phases. Dependent on the lipid composition, the angle between the transition state dipole moments of both probes and the normal to the membrane are found to deviate clearly from 90{\deg}. It is seen that the DiI-C18(5) probe is located in the headgroup region of the SM:Chol mixture, in close contact with water molecules. A fluorescence anisotropy study indicates also that DiI-C18(5) gives rise to a distinctive behavior in the SM:Chol membrane compared to the other considered membranes. The latter behavior has not been seen for the studied BODIPY probe, which is located deeper in the membrane.
Thermodynamic state of the interface during acoustic cavitation in lipid suspensions

Abstract:The thermodynamic state of lipid interfaces was observed during shock wave induced cavitation in water with submicrosecond resolution, using the emission spectra of hydration-sensitive fluorescent probes colocalized at the interface. The experiments show that the cavitation threshold is lowest near a phase transition of the lipid interface. The cavitation collapse time and the maximum state change during cavitation are found to be a function of both the driving pressure and the initial state of the lipid interface. The experiments show dehydration and crystallization of lipids during the expansion phase of cavitation, suggesting that the heat of vaporization is absorbed from within the interface, which is adiabatically uncoupled from the free water. The study underlines the critical role of the thermodynamic state of the interface in cavitation dynamics, which has mechanistic implications for ultrasound-mediated drug delivery, acoustic nerve stimulation, ultrasound contrast agents, and the nucleation of ice during cavitation.
