Abstract:
Ischemic stroke is a syndrome of severe neurological responses that cause neuronal death, damage to the neurovascular unit and inflammation. Notoginsenoside R1 (NG-R1) is a neuroprotective drug that is commonly used to treat neurodegenerative and cerebrovascular diseases. However, its potential mechanisms on the regulation of small molecule metabolism in ischemic stroke are largely unknown. The aim of this study was to explore the potential mechanisms of NG-R1 on the regulation of small molecule metabolism after ischemic stroke. Here, we found that NG-R1 reduced infarct size and improved neurological deficits by ameliorating neuronal damage and inhibiting glial activation in MCAO/R rats. Furthermore, using matrix-assisted laser desorption/ionization mass spectrometry imaging (MALDI-MSI), we clarified that NG-R1 regulated ATP metabolism, the tricarboxylic acid (TCA) cycle, the malate-aspartate shuttle, antioxidant activity, and the homeostasis of iron and phospholipids in the striatum and hippocampus of middle cerebral artery occlusion/reperfusion (MCAO/R) rats. In general, NG-R1 is a promising compound for brain protection from ischemic/reperfusion injury, possibly through the regulation of brain small molecule metabolism.

What problem does this paper attempt to address?
### Problems the Paper Attempts to Solve This paper aims to explore the effects of the newly extracted phytoestrogen Notoginsenoside R1 (NG-R1) from Panax notoginseng on small molecule metabolism after cerebral ischemia/reperfusion injury and its potential mechanisms. #### Main Research Content: 1. **Background and Issues**: - Ischemic stroke is a severe neurovascular syndrome that leads to neuronal death, neurovascular unit damage, and inflammation. - The specific mechanisms by which NG-R1 regulates small molecule metabolism in ischemic stroke are currently unclear. 2. **Research Objectives**: - To investigate the potential mechanisms by which NG-R1 regulates small molecule metabolism after ischemic stroke. - To determine whether NG-R1 alleviates ischemia/reperfusion injury by regulating small molecule metabolism in the brain. #### Specific Experimental Methods: - **Animal Model**: The effects of NG-R1 were evaluated using the MCAO/R (middle cerebral artery occlusion/reperfusion) model. - **Drug Treatment**: NG-R1 was injected into rats with the MCAO/R model, followed by a series of tests including neurological function scoring, histological staining, and immunofluorescence staining. - **Mass Spectrometry Imaging Technology**: Matrix-assisted laser desorption/ionization mass spectrometry imaging (MALDI-MSI) was used to analyze the distribution of small molecule metabolites in the brain. #### Main Findings: - **Reduction of Infarct Volume and Improvement of Neurological Deficits**: NG-R1 significantly reduced the infarct volume and improved neurological deficits in MCAO/R rats. - **Inhibition of Neuronal Apoptosis and Astrocyte Activation**: NG-R1 reduced neuronal apoptosis and inhibited astrocyte activation. - **Regulation of Metabolic Pathways**: NG-R1 regulated ATP metabolism, the citric acid cycle, the glutamate-aspartate shuttle system, antioxidant activity, and the homeostasis of iron and phospholipids. In summary, this paper aims to reveal the neuroprotective effects of NG-R1 in ischemic stroke and its specific mechanisms through the regulation of small molecule metabolism.
