Abstract:A broad range of N-sulfonyformamidines, widely used intermediates for drugs, were synthesized in moderate to excellent yields from 2-Pyridinemethanamine as N-source via Copper-catalyzed C-N cleavage. Firstly, N-(2-pyridinylmethyl)benzenesulfonamides were smoothly synthesized via 2-pyridinemethanamine and sulfonyl chlorides, then reacted with N,N-dimethylformamide dimethyl acetal to obtain the corresponding N-Sulfonylformamidines analogs, during which pyridin-2-ylmethyl and sulfonyl groups were essential for the C-N bond cleavage. The current work presents a valuable complementarity to the synthesis of N-sulfonyformamidines as 2-pyridinemethanamine can provide the N source and sulfonyl chloride,s which could be original materials. Background: N-sulfonylamidines have gained considerable attention from schools and industries because of their unique bioactivity. Since Pinner's strategy, expanding the synthesis methods of N-sulfonylamidines has been the goal of many organic chemists over the past decades. Besides the crash reaction conditions and the participation of undesirable reagents, the production of N-sulfonylamidines commonly required unstable ammonia and azides as the source of nitrogen that hindered the further development and application of N-sulfonylamidine derivatives. Objective: The study aims to find a stable N source to replace NaN3 or NH3 to synthesize N-sulfonylamidines from sulfonyl chlorides. Methods: Firstly, N-(2-pyridinylmethyl)benzenesulfonamides were smoothly synthesized via 2-pyridinemethanamine and sulfonyl chlorides. Then the reaction conditions of N-(2-pyridinylmethyl) benzenesulfonamides and N,N-dimethylformamide dimethyl acetal (DMF-DMA) were screened and optimized. The reaction was processed in glycol at 80 degrees C for 8 hours with the addition of 5 mol% Cu(OAc)(2)center dot H2O as a catalyst. Results: Taking advantage of pyridin-2-ylmethyl, a scope of N-Sulfonylformamidines were synthesized from those N-(2-pyridinylmethyl)benzenesulfonamides under copper-catalyzed C-N bond cleavage. Conclusion: This ready synthetic method will be more of a promising inspiration for bioactive compound synthesis and drug development than for an innovative approach to synthesizing N-sulfonylformamidines.

Abstract:A general method for one‐pot synthesis of thioamides is developed through a three‐component reaction involving chlorohydrocarbon, amide and elemental sulfur. Such a strategy does not only avoid residual transition metal in the product but also prevent the generation of C−N coupling by‐product. The latter is prone to be generated when alkane halide and amine are present. With the protocol proposed in this work, both alkyl and aryl thioamides can be obtained in moderate to excellent yields with a high tolerance of various functional groups. External oxidants are not required in the reaction. In addition, the reaction mechanisms are addressed using a combination of controlling experiments and quantum chemical calculations.

Abstract:A facile method for the synthesis of tetrahydropyrimidines is described. Diethylamine was treated with methyl propiolate to provide Michael adduct (beta-amino acrylate). In the presence of organic acid, the cyclization between imines with beta-aminoacrylateoccurred at room temperature and completed within several minutes to give tetrahydropyrimidines with liberation of ammonium salt. Formaldimines show good toleration to the reaction.

Abstract:A novel synthesis of 2-iminoimidazoles and 2-aminoimidazoles, via nucleophilic addition and a subsequent sodium hydroxide mediated intramolecular alkyne hydroamination from propargylamines and carbodiimides, is developed. This transition-metal-free cascade process represents an atom-economic and step-economic procedure for the construction of imidazole derivatives with high regioselectivity and moderate to good yields.

Abstract:A highly regioselective iodosulfonylation of allenes in the presence of CuI and 1,10-phenanthroline has been developed for the synthesis of various useful ( E)-α-iodomethyl vinylsulfones in moderate to excellent yields. This practical reaction is fast, operationally simple, and in particular, proceeds under very mild conditions to afford the target products with high regio- and stereoselectivity. The selectivity was illustrated by a conceptual DFT analysis.

Abstract:A new strategy for the C(sp2)–H imidation and 1,2-imidofluorination of vinylsulfides has been established through simple treatment with N-fluorobis(benzenesulfonyl)imide, which acts as both oxidant and nitrogen source. For alkyl and electron-rich aryl vinylsulfides, alkyl/arylthioenamines are produced in up to 92% total yields. However, for steric and electron-deficient aryl vinylsulfides, 1,2-imidofluorination products are prepared in up to 73% yields. The chemoselectivity is controlled by the steric and electronic effects of aryl substituents. Thionium ions are proposed as key intermediates in the two reactions.

Abstract:The palladium-catalyzed [3+2] decarboxylative cycloaddition of 4-vinyl-4-butyrolactones with sulfamate-derived cyclic imines has been developed, providing the sulfamate-fused pyrrolidine derivatives in high yields with good diastereoselectivities. The scale-up reaction and further derivation of the product worked well, demonstrating the potential application of the current reaction in organic synthesis. A plausible reaction mechanism was also proposed.

Abstract:Herein we report the facile synthesis of a series of novel N-O reagents and identify N ′-di-Boc- N ′′-[(mesitylsulfonyl)oxy]-guanidine as an efficient imidating reagent. This reagent is synthetically scalable and bench-stable, yet it shows unprecedented reactivity and chemoselectivity towards S(II) in buffer/hexafluoroisopropanol (HFIP) system, to generate a new class of N -guanyl sulfilimines. Further transformation of N -guanyl sulfilimines to N -guanyl sulfoximines provides us a chance to investigate these rarely reached compounds, which are shown to be versatile precursors to a variety of N -heterocyclic and NH- sulfoximines. Our work provides an addition to the tool box of both sulfur imidation and guanidine synthesis, especially for the need of rapid and selective modifications of sulfur(II) center under biocompatible conditions.

Abstract:The utility of sulfilimine is increasingly recognized in pharmaceuticals, agrochemicals, and other complex molecular structures. The synthesis of such compounds has been historically hindered by laborious procedures, harsh conditions, and limited substrate scope for wide applications. Here, we report a highly efficient Cu-catalyzed S-vinylation of sulfenamides with alkenyl iodonium salts to access α,β-unsaturated sulfilimines. The reaction proceeds under mild conditions, providing high yields of the desired products while demonstrating a broad substrate scope. Notably, the introduction of a 2,3-dimethylpropyl group as the S-substituent enables the efficient synthesis of novel S-divinylsulfilimines.

Abstract:Regiospecific and direct imidation of the methyl C(sp3)–H bond of thioanisoles is realized under mild and metal-free conditions with N-fluorobis(benzenesulfonyl)imide as an oxidant and nitrogen source. Proposed mechanism suggests that thionium ion intermediates and a Pummerer-type reaction are involved. Abstract:Sulfenyl imidinium salts are a virtually unexplored class of intermediates in organic chemistry. Herein, we demonstrate how sulfonium rearrangements can be deployed to access these versatile synthetic intermediates, bearing three contiguous (and congested) stereogenic centers, with high levels of selectivity. The synthetic value of the scaffold was unraveled by selective transformations into a range of building blocks, including 1,4-dicarbonyl derivatives and sulfonolactones.
