Biocompatible chlorocoumarins from harmful chlorophenols, their synthesis and biomedicinal evaluation

Abstract:In this work, two chlorophenols with particular toxicities were transformed into their 14 corresponding coumarins (CMs). Their chemical structures were confirmed by examining the spectra generated by various spectroscopic instruments. The created CMs were evaluated as influential, multifaceted, and biocompatible therapeutic applicants. This was done through three evaluational studies, which are bioactivity, biosafety, and computer-based druggable analysis. In the first study type, the possibilities of the created CMs to function as antitumor applicants were inspected against six tumor-acquired cellular populations through a MTT-based methodology. Also, the function as an antioxidant was studied using two free radical types, while the antidiabetic function was assessed employing two glucose-regulating enzymes. Moreover, their function as broad-spectrum antimicrobial applicants was evaluated against six infectious aerobic bacteriomers, four anaerobic bacteriomers, and two fungiomers. Concerning the second evaluational study, the biosafety of the created CMs was investigated by following their impact on the regular growth of one normal cellular population and one normal bacterium. In the last study type, the capacity of the created CMs to consider drug applicants was examined using two online platforms. The results of these three study types indicated that the created CMs have good antitumor, antioxidant, and antidiabetic activities with broad-spectrum antimicrobial potential in relation to the applied test settings and references. As antitumor applicants, the created CMs showed an intriguing antitumor impact, with IC 50 values ranging from 13.08 μg/ml to 103.20 μg/ml. With IC 50 values ranging between 62.45 μg/ml and 112.01 μg/ml, these CMs showed considerable neutralizing action towards the free structures under investigation. Given the standard results, the created CMs had a good-to-moderate antidiabetic effect with IC 50 values ranging between 324.46–424.02 μg/ml. In the antimicrobial investigations, the created CMs showed broad-spectrum microbiocidal activity in general and particularly against the test fungiomers. Also, these multifunctional attributes were coupled with outstanding biocompatibility and drug-like properties. The author comes to the conclusion that the transformation of toxic chlorophenols into their CMs modified their effects in a way that served better in therapeutics. Also, this work may open the door to repurposing toxic phenols into biocompatible medicinal applications.
