Abstract:The first example of C alkylation of 1,2-dihydropyridines with alkyl triflates and Michael acceptors was developed to introduce quaternary carbon centers with high regio- and diastereoselectivity. Hydride or carbon nucleophile addition to the resultant iminium ion also proceeded with high diastereoselectivity. Carbon nucleophile addition results in an unprecedented level of substitution to provide piperidine rings with adjacent tetrasubstituted carbon atoms.

Abstract:Piperidines are prevalent in natural products and pharmaceutical agents and are important synthetic targets for drug discovery and development. We report on a methodology that provides highly substituted piperidine derivatives with regiochemistry selectively tunable by varying the strength of acid used in the reaction. Readily available starting materials are first converted to dihydropyridines via a cascade reaction initiated by rhodium-catalyzed carbon-hydrogen bond activation. Subsequent divergent regio- and diastereoselective protonation of the dihydropyridines under either kinetic or thermodynamic control provides two distinct iminium ion intermediates that then undergo highly diastereoselective nucleophilic additions. X-ray structural characterization of both the kinetically and thermodynamically favored iminium ions along with density functional theory calculations provide a theoretical underpinning for the high selectivities achieved for the reaction sequences.

Abstract:A new catalyst, poly( p -phenylenediamine) functionalized with copper nanoparticles stabilized on layered double hydroxide (LDH@poly( p -phenylenediamine)@CuI), was prepared. The structure of the new catalyst was identified and confirmed using different analyzes such as FESEM, EDX, XRD, TGA and elemental mapping. The efficiency of the mentioned catalyst was confirmed in the three-component one-pot synthesis of various derivatives of dihydropyrano[3,2-c]chromene in aqueous ethanol solvent. The structure of the synthesized products was confirmed using infrared spectrometry (FT-IR), nuclear magnetic resonance ( 1 H NMR and 13 C NMR) and Mass techniques. High yield of products, short reaction time, easy purification, reuse of catalyst, and the use of a soft reaction method can be considered as the main advantages of this method. Additionally, this catalyst was recycled four times without a significant loss of catalytic power.

Abstract:A nickel ferrite nanocatalyst was synthesized in the presence of simple and cost-effective polysorbates assisted sol gel method as an effective catalyst has been successfully used for one pot multicomponent Hantzsch condensation reaction of different aromatic aldehydes, dimedone and ammonium acetate under solvent-free condition at 110 °C-120 °C. The highlights feature for the synthesis of acridinediones are the recyclability of catalyst, solvent-free reaction condition, use of inexpensive catalyst, highly efficient, facile product isolation, clean reaction, nontoxic substances and great yields in short reaction time. The recovered catalyst can run four times without loss its activity.

Abstract:Piperidine and pyrrolidine derivatives are important nitrogen heterocyclic structures with a wide range of biological activities. However, reported methods for their construction often face problems of requiring the use of expensive metal catalysts, highly toxic reaction reagents or hazardous reaction conditions. Herein, an efficient route from halogenated amides to piperidines and pyrrolidines was disclosed. In this method, amide activation, reduction of nitrile ions, and intramolecular nucleophilic substitution were integrated in a one-pot reaction. The reaction conditions were mild and no metal catalysts were used. The synthesis of a variety of N-substituted and some C-substituted piperidines and pyrrolidines became convenient, and good yields were obtained.

Abstract:Piperidines are widely present in small molecule drugs and natural products. Despite many methods have been developed for their synthesis, new approaches to polysubstituted piperidines are highly desirable. This work presents a radical (4+2) cycloaddition reaction for synthesis of piperidines featuring dense substituents at 3,4,5-positions that are not readily accessible by known methods. Using commercially available diboron(4) compounds and 4-phenylpyridine as the catalyst precursors, the boronyl radical-catalyzed cycloaddition between 3-aroyl azetidines and various alkenes, including previously unreactive 1,2-di-, tri-, and tetrasubstituted alkenes, has delivered the polysubstituted piperidines in generally high yield and diastereoselectivity. The reaction also features high modularity, atom economy, broad substrate scope, metal-free conditions, simple catalysts and operation. The utilization of the products has been demonstrated by selective transformations. A plausible mechanism, with the ring-opening of azetidine as the rate-limiting step, has been proposed based on the experimental and computational results.

Abstract:In this study, a series of benzimidazolium salts were synthesized as asymmetric N-heterocyclic carbene (NHC) precursors. Nine novel palladium complexes with the general formula [PdX2(NHC)(pyridine)] were synthesized using benzimidazolium salts in the PEPPSI (Pyridine Enhanced Precatalyst Preparation, Stabilization and Initiation) theme. All synthesized Pd(ii) complexes are stable. The synthesized compounds were thoroughly characterized by respective spectroscopic techniques, such as 1HNMR, 13C NMR, FTIR spectroscopy, X-ray crystallography and elemental analysis. The geometric structure of the palladium N-heterocyclic carbene has been optimized in the framework of density functional theory (DFT) using the B3LYP-D3 dispersion functional with LANL2DZ as a basis set. The on/off mechanism of pyridine assisted Pd-NHC complexes made them the best C-H functionalized catalysts for regioselective C-5 arylated products. Five membered heterocyclic compounds such as 2-acetyl furan, furfuryl acetate 2-acetylthiophene and N-methylpyrrole-2-carboxaldehyde were treated with numerous aryl bromides and arylchlorides under optimal catalytic reaction conditions. Interestingly, all the prepared catalysts possessed essential structural features that facilitated the formation of desired coupling products in quantitative yield with excellent selectivity. The arylation reaction of bromoacetophenone was highly catalytically active with only 1 mol% catalyst loading at 150 °C for 2 hours. To check the efficiency of the synthesized complexes, three different five member heterocyclic substrates (2-acetylfuran, 2-acetylthiophen, 2-propylthaizole) were tested with a number of aryl bromides bearing both electron-donating and electron-withdrawing groups on para position. The data in Tables 2-4. Indicated that electron-donating groups on the para position of aryl halide decreased the catalytic conversion while electron-withdrawing groups increased the catalytic conversion this was due to the high nucleophilicity of the electron-donating substituents.

Abstract:Dedicated to Professor H. Ila on her 80 th birthday N-Arylated piperidones are present as pharmacophores in many pharmaceuticals and serve as useful precursors for the construction of important new molecules. We have developed a transition-metal-free, cost-effective, and mild approach for the synthesis of N-(hetero)arylated piperidones and their ketals by using ketals of piperidones and 2-oxo-5,6-dihydro-2 H -benzo[ h ]chromene-3-carbonitriles as precursors. The desired products were obtained in two steps: amination of the 2-oxo-5,6-dihydro-2 H -benzo[ h ]chromene-3-carbonitrile from piperidone, followed by ring transformation using a suitable nucleophile source. We have successfully tethered functionalized dihydrophenanthrenes, hydrobenzo[ c ]phenanthrenes, and benzoquinolines to piperidinone moieties under transition-metal-free conditions. Abstract:In this project, poly(4-vinylpyridinum) trinitromethanide [P4VPH]C(NO 2 ) 3 as a newly reported heterogeneous catalyst was utilized for the promotion of the synthesis of benzo[ c ]acridine derivatives via the three-component condensation of various aromatic aldehydes, dimedone, and 1-naphthylamine. All the obtained products were identified with the comparison of their physical properties and spectral data with the authentic samples. By using this catalyst, the relevant products were obtained during reasonable times with high yields. Also, this heterogeneous catalyst can be reused up to three times without significant loss of its catalytic activity.

Abstract:AIM AND OBJECTIVE:The synthesis of bipyridines, especially 2, 2'-bipyridines, remains challenging because the catalytic cycle can be inhibited due to coordination of bipyridine to transition metal. Thus, the development of efficient methods for the synthesis of bipyridines is highly desirable. In the present work, we presented a promising approach for preparation of bipyridines via a Pd-catalyzed reductive homocoupling reaction with simple piperazine as a ligand. MATERIALS AND METHODS:Simple and inexpensive piperazine was used as a ligand for Pd-catalyzed homocoupling reaction. The combination of Pd(OAc)2 and piperazine in dimethylformamide (DMF) was observed to form an excellent catalyst and efficiently catalyzed the homocoupling of azaarenyl halides, in which DMF was used as the solvent without excess reductants although stoichiometric reductant was generally required to generate the low-oxidation-state active metal species in the catalytic cycles. RESULTS:In this case, good to excellent yields of bipyridines and their (hetero) aromatic analogues were obtained in the presence of 2.5 mol% of Pd(OAc)2 and 5 mol% of piperazine, using K3PO4 as a base in DMF at 140°C. CONCLUSION:According to the results, piperazine as an inexpensive and efficient ligand was used in the Pd(OAc)2-catalyzed homocoupling reaction of heteroaryl and aryl halides. The coupling reaction was operationally simple and displayed good substrate compatibility.

Abstract:A base-induced synthesis of 2-(4-(2-(phenylthio)ethyl)piperazinyl) acetonitriles by reaction of disulfides, 1-(chloromethyl)-4-aza-1-azonia bicyclo[2.2.2]octane chloride and trimethylsilyl cyanide is reported. The scope of the method is demonstrated with 30 examples. The reaction mechanism research indicates that the three-component reaction would be a SN2 reaction. The products exhibit good activities towards advanced synthesis of aqueous soluble acyl-CoA: cholesterol O-acyltransferase-1 (ACAT-1) inhibitors. Our work is superior as it uses less-odor disulfides as carbon sources and EtOH as solvent in a water and dioxygen insensitive reaction system, followed by a simple purification process.

Abstract:Herein, we describe a novel protocol for visible-light-induced α,γ-C(sp3)–H difunctionalization of piperidines. This redox-neutral, atom-economical protocol, which exhibits a broad substrate scope and good functional group compatibility, constitutes a concise, practical method for constructing piperidine-containing bridged-ring molecules. Abstract:In recent times, the research on the synthesis of highly functionalized piperidines has become a challenge and an attractive research field among synthetic chemists because these compounds have many biological, pharmaceutical and industrial applications. In this regard, in this paper we would like to report a green, rapid and efficient method for the synthesis of highly functionalized piperidines based on the use of zinc (II) chloride supported on Fe3O4 NPs modified with Dopamine and 5H-Cyclopenta[2,1-b:3,4-b′]dipyridin-5-one [Fe3O4@Dop/CP-diPy-ZnCl2 as a novel magnetically reusable catalyst. The structure of Fe3O4@Dop/CP-diPy-ZnCl2 nanocatalyst was well identified by several spectroscopic techniques including: FT-IR, TEM, SEM, EDX, ICP-OES, TGA, XRD, VSM and Elemental mapping technique. One-pot three-component reactions of aromatic amines with different aryl aldehydes and β-ketoesters were performed in water and all desired highly functionalized piperidines were synthesized with high to excellent yields, which indicates the high efficiency of this catalytic system. The reusability tests shown that that Fe3O4@Dop/CP-diPy-ZnCl2 catalyst is a reusable and stable catalyst.Graphical Abstract
