Abstract:Objective To develop and understand the therapy of electrical stimulation as well as to offer the physiological evidence, the present study focused on the activation in human brain during the electrical stimulation of different frequency. Methods Functional magnetic resonance imaging (fMRI) covering the whole brain of healthy volunteer was performed to observe the response of human brain to frequency-specific electrical stimulation in the wrist and elbow. Results There were different activation areas observed in brain evoked by electrical stimulation of different frequency. And the areas, especially the motor cortex and somatosensory cortex, showed significant activation. Conclusion The work suggested that frequency-specific electrical stimulation acted through different neuro-pathways served for different nerve function in the central nervous system,and the present study made a light for choosing the better frequency of electrical stimulation during electrical stimulation therapy.
Cortical Oscillatory Dynamics of Tactile Selective Sensation - for a Novel Type of Somatosensory Brain-computer Interface

Abstract:In this study, cortical oscillatory dynamics with respect to tactile selective sensation tasks were investigated. Subjects were required to perform three tactile attention tasks, prompted by cues presented in random order and while both wrists were simultaneously stimulated: 1) selective sensation of the left hand (SS-L), 2) selective sensation of the right hand (SS-R), 3) bilateral selective sensation (SS-B). Even-related (de)synchronization (ERD/ERS) analysis revealed a clear contralateral activation of somatosensory cortex during the SS-L and SS-R tasks, and a bilateral activation during SS-B tasks. Additionally, we found a clear ERS in occipital region of the brain during all the SS tasks. Diverse activation pattern among SS-L, SS-R and SS-B offers novel brain signals for somatosensory Brain-computer Interfaces.
Neuromodulation with Submillimeter Spatial Precision by Optoacoustic Fiber Emitter

Abstract:New techniques to non-invasively modulate neural activities with high precision are constantly being sought in neuroscience. Ultrasound is a promising neuromodulation modality due to its non-invasive potential. However, conventional piezo-based ultrasound neuromodulation using a transducer usually have a spatial resolution of more than a few millimeters, limiting research on specific brain area or neural circuits. Here, a spatially confined optoacoustic emitter is developed for high-precision neuromodulation, allowing neurons to be activated at submillimeter spatial precision. By designing a candle soot-PDMS layered optoacoustic fiber tip (CPOF) with a diameter of 350 mu m, we demonstrate the generation of local ultrasound waves with a peak frequency of 3.5 MHz. The ultrasound generated by CPOF can directly activate cortical neurons in culture within a radius of 450 mu m around the fiber tip, generating intracellular calcium transients. The CPOF promises to be a novel technique for high-precision neural stimulation and for investigating the mechanism of neurostimulation.
A Vibro-tactile Stimulation and Vibro-signature Synchronization Device for SSSEP-Based Study.

Abstract:A vibro-tactile stimulation and vibro-signature synchronization device was proposed in this paper. The device was used to elicit steady-state somatosensory evoked potential (SSSEP) and synchronize the actual vibraion information with the corresponding electroencephalography (EEG) signals in temporal domain. The device provided five independent stimulation channels and could generate vibro-tactile stimulation with arbitrary waveform, amplitude and frequency. Each channel used the intended stimulation waveform to drive a linear resonant actuator (LRA). The vibro-signature of each channel could be detected by a force sensing resistor (FSR) and fed back into the EEG recording system. Four stimulation patterns with a random sequence were applied to the index finger of three healthy subjects. Results showed that SSSEP features could be evoked with different stimulation patterns and the actual vibro-signature could be synchronized with the EEG signals. Comparing the actual stimulation with the intended stimulation, the errors of carrier frequency and modulation frequency were quite small. Results indicated that this novel device stood a good chance of serving in SSSEP-based studies.

Abstract:We propose electrocorticographic temporal alteration mapping (ETAM) for motor cortex mapping by utilizing movement-related cortical potentials (MRCPs) within the low-frequency band [0.05-3] Hz. This MRCP waveform-based temporal domain approach was compared with the state-of-the-art electrocorticographic frequency alteration mapping (EFAM), which is based on frequency spectrum dynamics. Five patients (two epilepsy cases and three tumor cases) were enrolled in the study. Each patient underwent intraoperative direct electrocortical stimulation (DECS) procedure for motor cortex localization. Moreover, the patients were required to perform simple brisk wrist extension task during awake craniotomy surgery. Cross-validation results showed that the proposed ETAM method had high sensitivity (81.8%) and specificity (94.3%) in identifying sites which exhibited positive DECS motor responses. Moreover, although the sensitivity of the ETAM and EFAM approaches was not significantly different, ETAM had greater specificity compared with EFAM (94.3 vs. 86.1%). These results indicate that for the intraoperative functional brain mapping, ETAM is a promising novel approach for motor cortex localization with the potential to reduce the need for cortical electrical stimulation.

Abstract:Foot sole somatosensory feedback is critical to motor control and declines with aging and disease. To enable study of cortical networks underlying foot sole somatosensation, we developed a pneumatic tactile stimulator capable of producing one degree-of-freedom (DOF) oscillations with preset waveform, frequency (≤10 Hz), force magnitude (5-500 N), duty cycle (20-100%), and contacted surface area over which pressures are applied to the foot sole. Image tests (anatomical/functional/field map) of a phantom demonstrated that the device is compatible with 3 T MRI. Gradient-recalled echo-planar images of seven healthy young adults using a typical block-designed 1 Hz sinusoidal stimulation protocol revealed significant activation contralaterally within the primary somatosensory cortex and paracentral gyrus, and bilaterally within the secondary somatosensory cortex. The stimulation system may therefore serve as a research tool to study functional brain networks involved in the perception and modulation of foot sole somatosensation and its relationship to motor control.

Abstract:Intracortical electrical micro-stimulation has been applied widely for the attempts on reconstruction of sensory functions. More recently, thalamic electrical stimulation has been proposed as a promising target for somatosensory stimulation. However, the cortical activations and mechanisms evoked by VPM stimulation remained unclear. In this report, the cortical neural responses to electrical stimulations were recorded by optical imaging of intrinsic signals. The impact of stimulation parameters was characterized to illustrate how the VPM stimulation alter cortical activities. Significant increases were found in cortical responses with increased stimulation amplitude or pulse width. However, frequency modulation exhibited significant inhibition with higher frequency stimulation. Our results suggest that optical imaging of intrinsic signals is sensitive and reliable to deep brain stimulations. These results may not only help to understand the modulation effects through thalamocortical pathway, but also show the possibility to use VPM stimulation to evoke frequency-tuned tactile sensations in rats.

Abstract:Electrocutaneous stimulation (ES) relies on the application of an electrical current flowing through the surface of the skin, eliciting a tactile percept. It can be applied for somatosensory mapping approaches at functional magnetic resonance imaging (fMRI) to obtain somatotopic maps illustrating the spatial patterns reflecting the functional organization of the primary somatosensory cortex (S1). However, its accessibility remains constrained, particularly in applications requiring multiple stimulation channels. Furthermore, the magnetic resonance (MR) environment poses several limitations in this regard. This study presents a prototype of a multichannel electrocutaneous stimulation device designed for somatosensory stimulation of the upper limbs of human participants in an MR environment in an inexpensive, safe, customizable, controlled, reproducible, and automated way. Our current-controlled, voltage-limited, stimulation device comprises 20 stimulation channels that can be individually configured to deliver various non-simultaneous combinations of personalized electrical pulses, depending on the subject, stimulation site, and stimulation paradigm. It can deliver a predefined electrical stimulus during fMRI acquisition, synchronized with the stimulation task design and triggered upon initiation of the acquisition sequence. Regarding device assessment, we conducted tests using an electrical circuit equivalent to the impedance of the human body and the electrode-skin interface to validate its feasibility. Then, we evaluated user acceptability by testing the device in human participants. Considering the stringent conditions of the MR environment, we performed a comprehensive set of safety and compatibility evaluations using a phantom. Lastly, we acquired structural and functional MR data from a participant during a somatosensory stimulation experiment to validate brain activity elicited by electric stimulation with our device. These assessments confirmed the device's safety in fMRI studies and its ability to elicit brain activity in the expected brain areas. The scope of application of our device includes fMRI studies focused on somatosensory mapping and brain-computer interfaces related to somatosensory feedback.

Abstract:We have developed a novel tactile pattern presentation device to assess tactile cognitive function by active, passive and dual touch. The device is magnetic resonance imaging (MRI) compatible and can serve to investigate the underlying neural mechanisms of tactile pattern discrimination. The primary components of the system are one disk for pattern presentation (DPP) driven by an ultrasonic motor, one set of fingers position control unit and one reaction key pad. In order to evaluate the performance of the presentation device, two functional MRI tasks were executed. First, we used MRI-Phantom to test the practicability of the tactile pattern presentation device. Second, we designed a tactile raised line cognitive task with five young subjects to investigate the brain activity areas concerning tactile shape cognition by dual touch. The results suggested that this device to be unaffected by the magnetic field, and the brain imaging results were consistent with previous neuroimaging study. The most significantly of present study is that the device provides a powerful means of investigating neural mechanisms of perception and cognitive function for touch discrimination.

Abstract:Abstract This study presents an open-transmit and 24-channel flexible receiver head coil assembly tailored for high-resolution ultra-high field fMRI of the human somatosensory and motor cortex. Leveraging the increased signal-to-noise ratio (SNR) and spatial resolution of ultra-high field MRI, we address the technical challenges inherent in fMRI coil design. The open birdcage transmit coil enhances patient comfort and enables visual task implementation, demonstrating superior performance in transmit efficiency and SAR distribution compared to conventional coils. Furthermore, the 24-channel flexible receiver head coil offers enhanced SNR and image quality, facilitating sub-millimeter VASO imaging for precise functional mapping. These advancements provide valuable tools for unraveling the intricacies of somatosensory and motor cortex function. By enriching human brain functional studies, they contribute significantly to our understanding of the mechanisms underlying somatosensory and motor cortex function, with potential implications for clinical applications in neurology and neuroscience research.

Abstract:Movement constraints in stroke survivors are often accompanied by additional impairments in related somatosensory perception. A complex interplay between the primary somatosensory and motor cortices is essential for adequate and precise movements. This necessitates investigating the role of the primary somatosensory cortex in movement deficits of stroke survivors. The first step towards this goal could be a fast and reliable functional Magnetic Resonance Imaging (fMRI)-based mapping of the somatosensory cortex applicable for clinical settings. Here, we compare two 3 T fMRI-based somatosensory digit mapping techniques adapted for clinical usage in seven neurotypical volunteers and two sessions, to assess their validity and retest-reliability. Both, the traveling wave and the blocked design approach resulted in complete digit maps in both sessions of all participants, showing the expected layout. Similarly, no evidence for differences in the volume of activation, nor the activation overlap between neighboring activations could be detected, indicating the general feasibility of the clinical adaptation and their validity. Retest-reliability, indicated by the Dice coefficient, exhibited reasonable values for the spatial correspondence of single digit activations across sessions, but low values for the spatial correspondence of the area of overlap between neighboring digits across sessions. Parameters describing the location of the single digit activations exhibited very high correlations across sessions, while activation volume and overlap only exhibited medium to low correlations. The feasibility and high retest-reliabilities for the parameters describing the location of the single digit activations are promising concerning the implementation into a clinical context to supplement diagnosis and treatment stratification in upper limb stroke patients.

Abstract:In this paper we developed an integrated device comprised of a multi-electrode array coupled with optical fiber for deep-brain optical stimulation and electrical recording. We characterized the array device both electrically and optically, and conducted in vivo experiments on free moving rats for validation. This design of array device provides a viable tool for neuromodulation and neural signal acquisition in optogenetics and in other fields of neuroscience studies perspectively.

Abstract:High-resolution awake mouse fMRI remains challenging despite extensive efforts to address motion-induced artifacts and stress. This study introduces an implantable radiofrequency (RF) surface coil design that minimizes image distortion caused by the air/tissue interface of mouse brains while simultaneously serving as a headpost for fixation during scanning. Using a 14T scanner, high-resolution fMRI enabled brain-wide functional mapping of visual and vibrissa stimulation at 100x100x200m resolution with a 2s per frame sampling rate. Besides activated ascending visual and vibrissa pathways, robust BOLD responses were detected in the anterior cingulate cortex upon visual stimulation and spread through the ventral retrosplenial area (VRA) with vibrissa air-puff stimulation, demonstrating higher-order sensory processing in association cortices of awake mice. In particular, the rapid hemodynamic responses in VRA upon vibrissa stimulation showed a strong correlation with the hippocampus, thalamus, and prefrontal cortical areas. Cross-correlation analysis with designated VRA responses revealed early positive BOLD signals at the contralateral barrel cortex (BC) occurring 2 seconds prior to the air-puff in awake mice with repetitive stimulation, which was not detectable with the randomized stimulation paradigm. This early BC activation indicated learned anticipation through the vibrissa system and association cortices in awake mice under continuous training of repetitive air-puff stimulation. This work establishes a high-resolution awake mouse fMRI platform, enabling brain-wide functional mapping of sensory signal processing in higher association cortical areas.

Abstract:We have developed a way to map brain-wide networks using focal pulsed infrared neural stimulation in ultrahigh-field magnetic resonance imaging (MRI). The patterns of connections revealed are similar to those of connections previously mapped with anatomical tract tracing methods. These include connections between cortex and subcortical locations and long-range cortico-cortical connections. Studies of local cortical connections reveal columnar-sized laminar activation, consistent with feed-forward and feedback projection signatures. This method is broadly applicable and can be applied to multiple areas of the brain in different species and across different MRI platforms. Systematic point-by-point application of this method may lead to fundamental advances in our understanding of brain connectomes.

Optoacoustic brain stimulation at submillimeter spatial precision

Abstract:Low-intensity ultrasound is an emerging modality for neuromodulation. Yet, transcranial neuromodulation using low-frequency piezo-based transducers offers poor spatial confinement of excitation volume, often bigger than a few millimeters in diameter. In addition, the bulky size limits their implementation in a wearable setting and prevents integration with other experimental modalities. Here, we report spatially confined optoacoustic neural stimulation through a miniaturized Fiber-Optoacoustic Converter (FOC). The FOC has a diameter of 600 μm and generates omnidirectional ultrasound wave locally at the fiber tip through the optoacoustic effect. We show that the acoustic wave generated by FOC can directly activate individual cultured neurons and generate intracellular Ca2+ transients. The FOC activates neurons within a radius of 500 μm around the fiber tip, delivering superior spatial resolution over conventional piezo-based low-frequency transducers. Finally, we demonstrate direct and spatially confined neural stimulation of mouse brain and modulation of motor activity in vivo.
Densely sampled stimulus-response map of human cortex with single pulse TMS-EEG and its relation to whole brain neuroimaging measures

Abstract:Large-scale networks underpin brain functions. How such networks respond to focal stimulation can help decipher complex brain processes and optimize brain stimulation treatments. To map such stimulation-response patterns across the brain non-invasively, we recorded concurrent EEG responses from single-pulse transcranial magnetic stimulation (i.e., TMS-EEG) from over 100 cortical regions with two orthogonal coil orientations from one densely-sampled individual. We also acquired Human Connectome Project (HCP)-styled diffusion imaging scans (six), resting-state functional Magnetic Resonance Imaging (fMRI) scans (120 mins), resting-state EEG scans (108 mins), and structural MR scans (T1- and T2-weighted). Using the TMS-EEG data, we applied network science-based community detection to reveal insights about the brain's causal-functional organization from both a stimulation and recording perspective. We also computed structural and functional maps and the electric field of each TMS stimulation condition. Altogether, we hope the release of this densely sampled (n=1) dataset will be a uniquely valuable resource for both basic and clinical neuroscience research.
