The Effects of Hypoxia and Macromolecular Crowders on Extracellular Matrix Production by Human Endometrial Mesenchymal Stromal Cells

Abstract:Recent decades have been marked by intense research on extracellular matrix (ECM) biology, which controls major cellular functions—from proliferation and differentiation to migration and apoptosis The bioactive properties of ECM open up wide prospects for its use in bioengineering and regenerative medicine. In this context, the key technology is ECM production by decellularization of organs, tissues, or cell cultures. The problem of rapid production of large quantities of bioactive ECM of cultured cells for medical purposes seems to be very relevant; at the same time, the question remains open regarding human endometrial mesenchymal stromal cells (eMSCs). In order to optimize the conditions for ECM production by cultivated eMSCs, we studied the impact of macromolecular compounds (crowders)—Ficoll and polyethylene glycol—on ECM protein deposition depending on the time, concentration, and molecular weight of the crowders in normoxic and hypoxic conditions. As is shown by immunofluorescence, Ficoll 400 is most effective for the production of key matrix components—fibronectin, type IV collagen and, to a lesser extent, type III collagen. Comparison of normoxic (20% O 2 ) and hypoxic (3% O 2 ) cultivation conditions revealed that cells produced ECM with a more developed structure at reduced oxygen concentration. It is significant that, under these conditions, Ficoll 400 promotes ECM deposition only at low serum content in the growth medium. To summarize, we can conclude that the combination of hypoxia, Ficoll 400 and low serum content in the growth medium provides an optimal way to produce an ECM. We have demonstrated for the first time the phenomenon of macromolecular crowding in the context of improving the deposition and organization of the ECM structure in eMSCs.
