Abstract:Chilling injury (CI) is a physiological disorder induced by cold, which heavily limit crop production and postharvest preservation worldwide. Methyl jasmonate (MeJA) can alleviate CI in various fruit species, including peach; however, the underlying molecular mechanism is poorly understood. Here, changes in contents of phenolics, lipids, and jasmonic acid (JA) and gene expressions are compared between MeJA and control fruit. Exogenous MeJA inhibited expressions of PpPAL1, PpPPO1, and PpPOD1/2 but did not affect the phenolic content. Furthermore, MeJA fruit showed lower relative electrolyte leakage, indicating less membrane damage. Meanwhile, the enrichment of linoleic acid in the potential lipid biomarkers, especially phosphatidylcholine, phosphatidylethanolamine, and phosphatidylglycerol, coincided with lower expressions of PpFAD8.1 but higher PpLOX3.1 and JA content. In the JA signaling pathway, MeJA significantly upregulated expressions of PpMYC2.2 and PpCBF3 but downregulated PpMYC2.1. In conclusion, adjustments of fatty acids in phospholipids contribute to MeJA-induced alleviation of CI in peach fruit via induction of the JA-mediated C-repeat-binding factor pathway.

Abstract:Chilling stress is an important constraint of global production of maize. This study was undertaken to compare the chilling responses of different maize seedling tissues and to analyze changes in polyamines as a result of chilling stress. Reponses to chilling were characterized in two maize ( Zea mays L.) inbred lines, ‘HuangC’ and ‘Mo17’, that putatively differ in chilling sensitivity. Seedlings were exposed to low temperature (5°C) and chilling injury was estimated by electrical conductivity (EC), malonaldehyde (MDA) concentration, and by changes in putrescine (Put), spermidine (Spd) and spermine (Spm) concentrations in root, mesocotyl, and coleoptile tissues. Membrane permeability (as measured by EC), MDA concentrations and Put concentrations in the three tissue of maize seedlings increased after chilling stress, except for the Put concentration in roots. Spd and Spm concentrations in the three tissues of seedlings decreased after chilling stress. The EC for cold stressed tissues were lower in HuangC than Mo17. Also, the EC of coleoptile tissues were lower than for mesocotyl in both inbred lines. We suggest that mesocotyl tissue can be used to evaluate cold tolerance in maize. Stepwise regression analyses showed that chilling injury in roots was generally correlated with Spd concentration while in the mesocotyl injury was mainly correlated with Put and Spd concentrations. Spermidine and Spm concentrations in the coleoptile were correlated with chilling injury. Characteristics changes of polyamines in chill-tolerant maize seedling combined with regression analysis are a reliable method for evaluating chill tolerance in maize lines.
Spermidine Suppressed the Inhibitory Effects of Polyamines Inhibitors Combination in Maize (Zea mays L.) Seedlings under Chilling Stress

Abstract:Chilling stress greatly inhibited the seed germination, plant growth, development and productivity in this study. The current research aimed to study the effects of different polyamine (PA) inhibitor combinations (Co), e.g., D-arginine (D-Arg), difluoromethylormithine (DFMO), aminoguanidine (Ag) and methylglyoxyl-bis-(guanyhydrazone) (MGBG) at different doses, i.e., 10 mu M Co, 100 mu M Co, 500 mu M Co, 1000 mu M Co and 1000 mu M Co + 1 mM Spd (Spermidine) in two inbred lines of maize (Zea mays L.), i.e., Mo17 and Huang C, a sensitive and tolerant chilling stress, respectively. The combination treatments of PA inhibitors reduced the biosynthesis of putrescine (Put) in the tissues of both studied inbred lines. Application with 500 mu M Co and 1000 mu M Co did not result in a significant difference in Put concentrations, except in the coleoptile of Mo17. However, combining Spd to 1000 mu M of PA inhibitors enhanced the Put, Spd, spermine (Spm) and total PAs in the roots, coleoptile and mesocotyls. Put and total PAs were increased by 39.7% and 30.54%, respectively, when Spd + 1000 mu M Co were applied relative to their controls. Chilling stress and PA inhibitors treatments affected both inbred lines and resulted in differences in the PA contents. Results showed that enzymes involved in the biosynthesis of PAs (ornithine decarboxylase as ODC and S-adenosylmethionine decarboxylase as SAMDC) were significantly downregulated by 1000 mu M Co in the tissues of both inbred lines. In contrast, the activity of PAO, a Pas degradation enzyme, was significantly improved by 1000 mu M Co under chilling stress. However, Spd + 1000 mu M Co significantly improved the activities of ODC and SAMDC and their transcript levels (ODC and SAMDC2). While it significantly downregulated the PAO activity and their relative genes (PAO1, PAO2 and PAO3) under chilling stress. Overall, this study elucidates the specific roles of Spd on the pathway of PA inhibitors and PA biosynthesis metabolism in maize seed development in response to chilling stress. Moreover, the Huang C inbred line was more tolerant than Mo17, which was reflected by higher activities of PA biosynthesis-related enzymes and lower activities of PAs' degradative-related enzymes in Huang C.
Polyamine Biosynthetic Pathways and Their Relation with the Cold Tolerance of Maize (zea Mays L.) Seedlings

Abstract:ABSTRACT Background The present study was designed to investigate the inhibition role of two polyamine biosynthesis inhibitors, i.e., D-arginine (D-Arg) and DL-α-difluoromethylornithine (DFMO), in polyamine biosynthesis under chilling stress in different tissues of two maize inbred lines – Huang C (chilling-tolerance) and Mo17 (chilling-sensitive). Results The results showed that exposure to the lower concentration of polyamine biosynthesis inhibitors improved seedlings growth, such as the root length, root and shoot fresh weight, chlorophyll a (chl a). The effectiveness of 10 µM D-Arg treatments was more prominent than those of 10 µM DFMO. However, the higher concentration of inhibitors suppressed seedlings growth, and the exposure to 100 µM DFMO caused stronger decreases in the photosynthetic pigments, such as chlorophyll a (chl a), chlorophyll b (chl b), total chlorophyll and carotenoids, than the other treatments. Meanwhile, the inhibitor treatments caused the lower content of putrescine (Put) in roots, mesocotyls and coleoptiles in both maize inbred lines as compared with untreated plants. However, the lower concentration (10 µM) of polyamine biosynthetic inhibitors improved the Spd content, except 10 µM D-Arg in root of Huang C, and 10 µM DFMO in coleoptiles of both Mo17 and Huang C. The correlation analysis found that Spd was positively significantly correlated with root length and shoot fresh weight of seedling. Conclusion It was showed that the Spd played an important role in seedling growth improvement. At the same concentration of polyamine biosynthetic inhibitors, the Put contents in different tissues of the seedlings treated with DFMO were generally lower than those treated with D-Arg, except for Put contents in root of Mo17 with 10 µM treatment. Moreover, the treatments of 100 µM were more prominent than those of 10 µM treatments. Exposure to 100 µM D-Arg and 100 µM DFMO could each decrease the activities of Arginine decarboxylase (ADC), Ornithine decarboxylase (ODC) and S-adenosylmethionine decarboxylase (SAMDC) in all maize tissues. However, the decrease of the ADC activity was more prominent in 100 µM D-Arg-treated seedlings, while the decrease of SAMDC and ODC activities was prominent in 100 µM DFMO-treated seedlings. Genes involved in polyamine biosynthesis, such as ADC, ODC, SAMDC, and PAO, showed different expression patterns in response to chilling stress and polyamine biosynthesis inhibitors. This study suggested that Put was synthesized via both the ADC and ODC pathways after chilling stress, with the ODC pathway being the major one.
SlMYC2 targeted regulation of polyamines biosynthesis contributes to methyl jasmonate-induced chilling tolerance in tomato fruit

Abstract:Chilling injury (CI) is a major limiting factor in the retention of the postharvest quality of chilling-sensitive vegetables and fruit stored at low temperatures. The enhanced chilling tolerance induced by methyl jasmonate (MeJA) treatment might be related to the polyamines biosynthesis. However, the molecular mechanism of polyamines biosynthesis induced by MeJA is far from clear. Here, we found that the application of 0.05 mmol L−1 MeJA enhanced the activities of arginase, arginine decarboxylase and ornithine decarboxylase, as well as the transcripts of SlARG1, SlARG2, SlADC and SlODC, promoted the accumulations of polyamines and further inhibited CI development. In addition, polyamines induced by MeJA were strongly positively correlated with the SlMYC2 expression level. Moreover, MeJA-induced polyamines biosynthesis was largely inhibited due to the silencing of SlMYC2. The (SlMYC2-silenced + MeJA)-treated fruit possessed higher incidence and index of CI than the MeJA-treated fruit. Combining these findings with results of the principal component analysis, we concluded that SlMYC2 is involved in MeJA-induced chilling tolerance in postharvest tomato fruit by regulating polyamines biosynthesis. Furthermore, the electrophoretic mobility shift and dual-luciferase reporter assays indicated that SlMYC2 could activate the transcription of SlARG1, SlARG2, SlADC and SlODC by binding directly to G/E-box elements in their promoters. From the findings, it was revealed that the targeted up-regulation of SlARG1, SlARG2, SlADC and SlODC by SlMYC2 is involved in MeJA-induced polyamines biosynthesis, which enhances chilling tolerance in tomato fruit.

Abstract:Chilling injury (CI) is a common phenomenon during low temperature storage for most fruits, reducing the effectiveness of refrigeration methods. Methyl jasmonate (MeJA) has been extensively employed to extend the shelf life of fruits during cold storage. However, the mechanism of MeJA inducing fruit resistance to CI requires further investigation. The study found that treating tomatoes with 0.05 mM MeJA inhibited CI by regulating the reactive oxygen species (ROS) metabolism, polyamines, and jasmonic acid (JA) signaling pathway, as well as cold response pathway. Interestingly, the expression of SlMsrB5 in tomato was up-regulated under MeJA treatment, showing an increase of 36.44% to 161.75% compared to the control fruit. Notably, silencing the expression of SlMsrB5 led to a continuously increased in CI incidence by 52.17% and CI index by 36.58% compared to the control fruit at the end of storage. Additionally, the silencing of SlMsrB5 weakened the inhibitory impact of MeJA on CI and reduced its induction effects on the aforementioned indicators. In addition, correlation analysis showed that the transcription of SlMsrB5 has a negative correlation with ROS related indicators under MeJA treatment, while was highly correlated with the aforementioned induction effects controlled by MeJA. Therefore, this study demonstrates a new basis for revealing the underlying mechanisms in the MeJA-regulated chilling resistance in postharvest fruits.

Transcriptome and DNA Methylome Analysis Reveal New Insights into Methyl Jasmonate-Alleviated Chilling Injury of Peach Fruit after Cold Storage

Abstract:Methyl jasmonate (MeJA) has been used to alleviate the chilling injury (CI) caused by postharvest cold storage of horticultural crops involving peach fruit. However, the molecular mechanism remains largely unknown. Here, we treated melting peach fruit (Prunus persica L. Batsch., cv. Hujingmilu) with different concentrations of MeJA, followed by storage at 0 degrees C up to 28 d. Peach fruit treated with 1 mM MeJA delayed the development of CI during 3 d shelf-life followed by 28 d cold after storage (C28dS3). The alleviated CI was associated with significant higher content of JA-Ile in peach fruit after MeJA treatment than controls. RNA-Seq revealed significant alternations in expression of genes related to ethylene synthesis, fruit softening, flesh browning and aroma-related volatile synthesis. Transcription factors associated with fruit ripening such as PpNAC1 and JA signaling such as PpMYC2.2 showed higher transcript levels in MeJA treated peach fruit than controls. Peach fruit with CI symptom had higher genome methylation level than MeJA treated fruit after cold storage at C28dS3. Reverse patterns between transcript abundance and DNA methylation in promoter were observed for transcription factor PpNAC1 and downstream PpACS1 for ethylene synthesis, PpExp1 for softening, and PpAAT1 for volatile ester and lactone production. Overall, in addition to providing new insights into the regulatory mechanism for MeJA in alleviating fruit chilling injury through integration of transcriptome and DNA methylome, our study also establishes a foundation for fruit quality maintaining after cold storage of peach fruit.

Abstract:BACKGROUND Energy status is associated with ripening, senescence and physiological disorder of various types of fruit after harvest, including peaches. Methyl jasmonate (MeJA) was effective in enhancing chilling tolerance in cold-stored peach fruit. The effect of MeJA on energy metabolism in peach fruit was investigated. RESULTS MeJA treatment increased the contents of adenosine triphosphate and adenosine diphosphate, but decreased the content of adenosine monophosphate, which resulted in a higher level of energy charge in peach fruit. Enzyme activities including H+-adenosine triphosphatase, Ca2+-adenosine triphosphatase, succinic dehydrogenase and cytochrome c oxidase were also higher in MeJA-treated fruit than those in control fruit. Meanwhile, MeJA treatment markedly inhibited the increase of ion leakage and the accumulation of malondialdehyde, super-oxidant radical and hydrogen peroxide. CONCLUSION These results indicated that MeJA could enhance chilling tolerance of peach fruit by inducing enzyme activities related to energy metabolism and maintaining high levels of ATP and energy charge.
CsMYB Transcription Factors Participate in Jasmonic Acid Signal Transduction in Response to Cold Stress in Tea Plant (Camellia sinensis)

Abstract:Low-temperature stress is an increasing problem for the cultivation of tea (Camellia sinensis), with adverse effects on plant growth and development and subsequent negative impacts on the tea industry. Methyl jasmonate (MeJA), as a plant inducer, can improve the cold-stress tolerance in tea plants. R2R3-MYB transcription factors (TFs) are considered potentially important regulators in the resistance to cold stress in plants. However, the molecular mechanisms, by which MYB TFs via the jasmonic acid pathway respond to cold stress in the tea plant, remain unknown. In this study, physiological and biochemical assays showed that exogenous MeJA application could effectively promote ROS scavenging in the tea plant under cold stress, maintaining the stability of the cell membrane. Sixteen R2R3-MYB TFs genes were identified from the tea plant genome database. Quantitative RT-PCR analysis showed that three CsMYB genes were strongly induced under a combination of MeJA and cold-stress treatment. Subcellular localization assays suggest CsMYB45, CsMYB46, and CsMYB105 localized in the nucleus. Exogenous MeJA treatment enhanced the overexpression of CsMYB45, CsMYB46, and CsMYB105 in E. coli and improved the growth and survival rates of recombinant cells compared to an empty vector under cold stress. Yeast two-hybrid and bimolecular fluorescence complementation experiments confirmed that CsMYB46 and CsMYB105 interacted with CsJAZ3, CsJAZ10, and CsJAZ11 in the nucleus. Taken together, these results highlight that CsMYB45, CsMYB46, and CsMYB105 are not only key components in the cold-stress signal response pathway but also may serve as points of confluence for cold stress and JA signaling pathways. Furthermore, our findings provide new insight into how MYB TFs influence cold tolerance via the jasmonic acid pathway in tea and provide candidate genes for future functional studies and breeding.
MaJAZ1 Attenuates the MaLBD5-Mediated Transcriptional Activation of Jasmonate Biosynthesis Gene MaAOC2 in Regulating Cold Tolerance of Banana Fruit.

Abstract:Previous studies indicated that methyl jasmonate (MeJA) treatment could effectively reduce the chilling injury of many fruits, including banana, but the underlying mechanism is poorly understood. In this study, one lateral organ boundaries (LOB) domain (LBD) gene, designated as MaLBD5, was isolated and characterized from banana fruit. Expression analysis revealed that accumulation of MaLBD5 was induced by cold temperature and MeJA treatment. Subcellular localization and transactivation assays showed that MaLBD5 was localized to the nucleus and possessed transcriptional activation activity. Protein-protein interaction analysis demonstrated that MaLBD5 physically interacted with MaJAZ1, a potential repressor of jasmonate signaling. Furthermore, transient expression assays indicated that MaLBD5 transactivated a jasmonate biosynthesis gene, termed MaAOC2, which was also induced by cold and MeJA. More interestingly, MaJAZ1 attenuated the MaLBD5-mediated transactivation of MaAOC2. These results suggest that MaLBD5 and MaJAZ1 might act antagonistically in relation to MeJA-induced cold tolerance of banana fruit, at least partially via affecting jasmonate biosynthesis. Collectively, our findings expand the knowledge of the transcriptional regulatory network of MeJA-mediated cold tolerance of banana fruit.

Abstract:Bud dormancy, which enables damage from cold temperatures to be avoided during winter and early spring, is an important adaptive mechanism of deciduous fruit trees to cope with seasonal environmental changes and temperate climates. Understanding the regulatory mechanism of bud break in fruit trees is highly important for the artificial control of bud break and the prevention of spring frost damage. However, the molecular mechanism underlying the involvement of MYB TFs during the bud break of peach is still unclear. In this study, we isolated and identified the PpMYB52 (Prupe.5G240000.1) gene from peach; this gene is downregulated in the process of bud break, upregulated in response to ABA and downregulated in response to GA. Overexpression of PpMYB52 suppresses the germination of transgenic tomato seeds. In addition, Y2H, Bimolecular fluorescence complementation (BiFC) assays verified that PpMYB52 interacts with a RING-type E3 ubiquitin ligase, PpMIEL1 , which is upregulated during bud break may positively regulate peach bud break by ubiquitination-mediated degradation of PpMYB52 . Our findings are the first to characterize the molecular mechanisms underlying the involvement of MYB TFs in peach bud break, increasing awareness of dormancy-related molecules to avoid bud damage in perennial deciduous fruit trees.

Novel Insights into Modified Atmosphere Mediated Cold Tolerance in Peach Fruit During Postharvest Storage

Abstract:Internal browning is a common symptom of chilling injury (CI) of peach fruit during postharvest cold storage, leading to a significant loss in the marketplace. In this study, modified atmosphere (MA) storage was applied to mitigate the occurrence of CI in both 'Jinxiu' and 'Zhonghuashoutao' peach and the overall transcriptomic changes were profiled. In two cultivars, MA treatment elicited multiple chilling responses affecting production of reactive oxygen species (ROS) and lipid metabolisms, as well as jasmonic acid (JA), gamma-aminobutyric acid (GABA) and proline synthesis, accompanied by accumulation of JAs (JA and JA-Ile), GABA and proline. Correlation analysis demonstrated that JAs and GABA served as significant regulators involved in cold tolerance under MA treatment. Weighted correlation network analysis (WGCNA) suggested that ethylene responsive factors (ERFs), WRKYs and NACs might serve as central transcription factors in the chilling-response pathway. Notably, PpERF61 was confirmed to activate the expression of JA biosynthetic genes Pp13S-LOX, PpAOS and PpOPR3 as well as GABA synthesis gene PpGAD. Taken together, this study unveils novel insights into the underlying mechanism of enhanced cold resistance in MA-treated peach fruit.
MaMYB13 is involved in response to chilling stress via activating expression of VLCFAs and phenylpropanoids biosynthesis-related genes in postharvest banana fruit

Abstract:Fruit chilling injury is the result of physiological dysfunction due to membrane lipid phase change, oxidative damage of biomacromolecules and respiratory metabolism abnormality. However, the involvement of transcription factors in response to fruits chilling tolerance remains largely unclear. Here, MaMYB13 was identified to participate in banana fruit response to chilling stress. MaMYB13 has transcriptional activation activity. When exposed to low temperature, expression of MaMYB13 was enormously induced. Moreover, MaMYB13 promoter was activated by chilling stress. MaMYB13 bound to the promoters of several important very-long-chain fatty acids (VLCFAs) and phenylpropanoids biosynthesis-related genes, including MaKCS11, Ma4CL6 and MaAAE1, and activated their transcription. Furthermore, MaKIN10 X1/3 interacted with MaMYB13 and enhanced MaMYB13-mediated transcriptional activation possibly via phosphorylation. Altogether, our results unravel the mechanism of MaMYB13-MaKIN10 X1/3 interaction regulating banana fruit chilling tolerance through activating the expression of MaKCS11, Ma4CL6 and MaAAE1, providing new insights into the regulatory network of MYB transcription factor.
PpERF-CRF3 selected by transcriptomic analysis plays key roles in the regulation of ABA alleviating chilling injury in peach fruit

Abstract:Abscisic acid (ABA) is widely utilized to mitigate chilling injury (CI) of fruit. However, the molecular mechanism of ABA alleviates CI in peach fruit remain unclear. Herein, 10-4 M ABA treatment significantly mitigated the CI of peach fruit by reducing relative conductivity and malondialdehyde content, while increasing proline and endogenous ABA content. Transcriptomic analysis indicated that an abundant number of differentially expressed genes were altered by ABA treatment, which primarily enriched pathways such as plant hormone signal transduction, glycerophospholipid metabolism and phenylpropanoid biosynthesis. RNA-Seq results indicate that ABA modulates the transcription of genes involved in auxin, ABA and ethylene signal transduction, as well as in cell wall degradation, antioxidant, fatty acid desaturation and proline metabolism. RT-qPCR confirmed the RNA-Seq results, ABA treatment induced the transcription of proline metabolism related genes (PpP5CR2, PpP5CS, PpP5CS1) and PpERF-CRF3. Particularly noteworthy, as a nuclear protein, PpERF-CRF3 activated the expression of PpP5CR2 and PpP5CS by directly binding to their promoters and over-expression PpERF-CRF3 increased proline content and enhanced PpP5CR2 and PpP5CS expression. Overall, these findings suggest that ABA mitigates CI in peach fruit may be by mediating these pathways, and PpERF-CRF3 potentially involves this process by stimulating the expression of genes related to proline synthesis.
MaC2H2-like regulates chilling stress response of 'Fenjiao' banana by modulating flavonoid synthesis and fatty acid desaturation

Abstract:Chilling injury (CI) is a major problem that affects fruit quality and ripening. Herein, chilling stress severely inhibited the expression of transcription factor MaC2H2-like. MaC2H2-like activates the expression of genes associated with flavonoid synthesis (MaC4H-like1, Ma4CL-like1, MaFLS, and MaFLS3) and fatty acid desaturation (MaFAD6-2 and MaFAD6-3), the leading indicators of chilling tolerance. MaC2H2-like interacts with MaEBF1 and boosts the transcriptional activity of MaFAD6-2, MaFAD6-3, Ma4CL-like1, and MaFLS. The overexpression of MaC2H2-like reduced fruit CI, induced the expression of these genes and increased the content of flavonoid and unsaturated fatty acid. Meanwhile, the silencing of MaC2H2-like increased fruit CI and downregulated the expression of those genes and reduced the content of flavonoid and unsaturated fatty acid. These results indicate that MaC2H2-like function as new player in modulating fruit CI by regulating flavonoid synthesis and fatty acid desaturation. MaC2H2-like could be a useful candidate gene for improving cold tolerance in 'Fenjiao' banana.

Abstract:Methyl jasmonate (MeJA) is a crucial cellular regulator involved in various developmental processes and defense mechanisms against environmental stresses. Hydrogen sulfide (H 2 S), a novel gas transmitter, has been shown to play a significant role in stress mitigation in plants. While there is established crosstalk between these two signaling molecules in plants, the evidence of their interaction within fruit remains notably insufficient. In this study, we investigated whether H 2 S serves as a downstream signal for MeJA during the cold storage of peach fruit. Exogenous MeJA application reduced cold damage in peach fruit, as indicated by a notable decrease in the browning index (BI) and malondialdehyde (MDA) content. This mitigation was attributed to the downregulation of polyphenol oxidase (PPO) gene family transcription in peach, like PpPPO1 , PpPPO2 , and PpPPO8 . Furthermore, endogenous H 2 S levels in MeJA-treated peaches increased during storage. We performed a genome-wide analysis on H 2 S biosynthesis genes, PpLCDs and PpDCDs , and assessed their expression levels in peaches. Among them, PpLCD1 , PpLCD3 , PpLCD5 , PpDES1 , PpDES3 , and PpDES5 were identified as contributors to H 2 S biosynthesis in peaches. Additionally, PpLCD3 , PpLCD5 , PpDES1 , PpDES2 , and PpDES4 were observed to transiently respond to MeJA application. Several JA signaling-related cis-elements, including TGACG-motif, CGTCA-motif, G-box, and low-temperature responsive element, were found in the promoters of these LCD/DCD genes. In vitro experiments with exogenous H 2 S applications further confirmed the protective role of H 2 S in cold-stored peaches. Our findings suggest that H 2 S-mediated MeJA signaling alleviates chilling injury in peach fruit during low-temperature storage.

Abstract:Auxin can inhibit or promote fruit ripening, depending on the species. Melting flesh (MF) peach fruit (Prunus persica L. Batsch) cultivars produce high levels of ethylene caused by high concentrations of indole-3-acetic acid (IAA), which leads to rapid fruit softening at the late stage of development. In contrast, due to the low concentrations of IAA, the fruit of stony hard (SH) peach cultivars does not soften and produces little ethylene. Auxin seems necessary to trigger the biosynthesis of ethylene in peach fruit; however, the mechanism is not well understood. In this study, we identified miRNA gene family members ppe-miR393a and ppe-miR393b that are differentially expressed in SH and MF fruits. RNA ligase-mediated 5 ' rapid amplification of cDNA ends and transient transformation of Nicotiana benthamiana revealed TRANSPORT INHIBITOR RESPONSE 1 (PpTIR1), part of the auxin perception and response system, as a target of ppe-miR393a and b. Yeast 2-hybrid assay and bimolecular fluorescence complementation assay revealed that PpTIR1 physically interacts with an Aux/IAA protein PpIAA13. The results of yeast 1-hybrid assay, electrophoretic mobility shift assay, and dual-luciferase assay indicated that PpIAA13 could directly bind to and trans-activate the promoter of 1-aminocyclopropane-1-carboxylic acid synthase 1 (PpACS1), required for ethylene biosynthesis. Transient overexpression and suppression of ppe-miR393a and PpIAA13 in peach fruit induced and repressed the expression of PpACS1, confirming their regulatory role in ethylene synthesis. Gene expression analysis in developing MF and SH fruits, combined with postharvest a-naphthalene acetic acid (NAA) treatment, supports a role for a ppe-miR393-PpTIR1-PpIAA13-PpACS1 module in regulating auxin-related differences in ethylene production and softening extent in different types of peach.
PpBBX32 and PpZAT5 modulate temperature-dependent and tissue-specific anthocyanin accumulation in peach fruit

Abstract:Abstract Anthocyanins are important compounds for fruit quality and nutrition. The R2R3 MYB transcription factor PpMYB10.1 is known to be critical for regulating anthocyanin accumulation in peach. However, regulatory factors upstream of PpMYB10.1 which control temperature-dependent, cultivar-contrasted and tissue-specific anthocyanin accumulation remain to be determined. In this study, differential anthocyanin accumulation in the outer flesh near the peel (OF) of peach [Prunus persica (L.) Batsch] was observed between cultivars “Zhonghuashoutao” and “Dongxuemi”, as well as among different storage temperatures and different fruit tissues of “Zhonghuashoutao”. By cross-comparisons of RNA-Seq data of samples with differential anthocyanin accumulation, transcription factor genes PpBBX32 and PpZAT5 were identified. These were functionally characterized as two positive regulators for anthocyanin accumulation via transient expression and genetic transformation. Various interaction assays revealed that both PpBBX32 and PpZAT5 can directly activate the PpMYB10.1 promoter and meanwhile interact at protein level as a PpZAT5-PpBBX32-PpMYB10.1 complex. Furthermore, the results of in silico analysis and exogenous application of methyl jasmonate (MeJA) indicated that MeJA favored anthocyanin accumulation, while it was also found that anthocyanin accumulation as well as PpBBX32 and PpZAT5 expression correlated significantly with endogenous JA and JA-Ile in different fruit tissues. In summary, PpBBX32 and PpZAT5 are upstream activators of PpMYB10.1, allowing JAs to take part in temperature-dependent and tissue-specific anthocyanin accumulation by modulating their expression. This work enriches the knowledge of the transcriptional regulatory mechanisms for differential anthocyanin accumulation under internal and external factors.

Abstract:Pepper fruit (Capsicum annuum L.) is sensitive to chilling stress with chilling injuries occurring below 7 °C; however, chilling injuries occur at different temperatures depending on the genotype. The present study aimed to identify the factors that affect chilling sensitivity in pepper fruits. A total of 112 F2 pepper fruits crossed between chilling-insensitive 'UZB-GJG-1999-51' and chilling-sensitive 'C00562' pepper were grouped according to the seed browning rate, which is a typical chilling symptom of pepper fruit under chilling conditions. Physiological traits, amino acids, fatty acids, as well as ethylene responsive factor (ERF) and jasmonate resistant 1 (JAR1) expression levels were analyzed, and their correlations with the seed browning rate were confirmed. The expression level of JAR1 showed a strong negative correlation with the seed browning rate (r = - 0.7996). The expression level of ERF11 and content of hydrogen peroxide showed strong positive correlation with the seed browning rate (r = 0.7622 and 0.6607, respectively). From these results, we inferred that JAR1 and ERF11 are important factors influencing the chilling sensitivity of pepper fruit.
Phytochrome interacting factor ZmPIF6 simultaneously enhances chilling tolerance and grain size in rice

Abstract:Chilling is a prevalent type of abiotic stress that adversely affects agricultural productivity worldwide. Phytochrome interacting factors (PIFs) are a group of transcription factor that are crucial for plant abiotic stress response. Our research reveals that the maize PIF family gene ZmPIF6 is responsive to chilling stress, which mitigates the negative impacts of chilling through reducing reactive oxygen species content and enhancing cell membrane stability at the physiological and biochemical levels. We also found that the ZmPIF6 overexpression lines showed a significant increase in grain size, encompassing both length and width, which mainly due to the increase in cell size. In addition, digital gene expression results suggested that ZmPIF6 regulates the expression of cold-related and grain size-related genes in rice. In light of these findings, ZmPIF6 has a hopeful prospect as a candidate gene of chilling tolerance and crop productivity in the transgenic breeding.
