Abstract:Temperature stress affect a stress response of aquatic animals including physiological metabolism, immune defense and other metabolic processes, damaged their immune response even cause dead. In this study, the physiological and histological changes of Procambarus clarkii under the high temperature (30 degrees C) and the low temperature (10 degrees C) stress were investigated. With the increase of temperature, the pyruvate kinase activity in the hepatopancreas and gill tissues of crayfish was gradually increased, and the fatty acid synthase activity at 10 degrees C and 30 degrees C were significantly lower than the control group of 25 degrees C. Furthermore, lysozyme, catalase and superoxide dismutase in 10 degrees C and 30 degrees C were significantly lower than that of 25 degrees C. ELISA revealed that HSP90 activities was enhanced with the increasing temperature in hepatopancreas after 36h of treatment. HE sections showed that high temperature stress or hypothermia caused deformation of R cells (storage cells), rupture of B cells (secretory cells), and partial rupture of the basement membrane (BM) in hepatopancreas, as well as the deformation of epithelial cells and respiratory epithelial cells of BM in gill tissues. While the temperature stress (30 degrees C and 10 degrees C treatment) caused the deformation of epithelial cells and respiratory epithelial cells, making rupture of the BM, comparing with the group at 25 degrees C. The study provides the data about the response of crayfish to temperature stress at the molecular level, and benefit to develop the strategy to defend the damage in the crayfish aquaculture caused by the temperature neither in the different regions nor in the different season.

Abstract:1. Adaptation of muscarinic receptors (MR)--muscarinic stimulation--induced intracellular Ca2+ mobilization during short-heat exposure (33 degrees C). 2. Heat-exposure for 48 hr decreased the carbachol (CCh)-stimulated cytosolic Ca2+ concentration increase. 3. The number of MR on cell surface increased transiently at 24 hr with a subsequent decrease at 48 hr. 4. CCh-stimulated inositol triphosphate (IP3) formation decreased at 48 hr. 5. In saponin-permeabilized cells, 1,4,5-IP3-induced 45Ca2+ release decreased at 24 hr. 6. The data suggest that the adaptation for increased muscarinic stimulation occurs at IP3 generating sites as well as at intracellular IP3 receptor sites during heat exposure.

Abstract:The heat shock response has been found in many strains of bacteria to human beings. Besides heat stimuli, many kinds of factors could also induce the synthesis of heat shock proteins (hsp). It is still unknown whether neurotransmitter could induce the increase of hsp expression in mammalian cells. In the present study, the effects of glutamic acid and acetylcholine (ACh) on the induction of hsp70 mRNA in PC12 cells were studied by Northern blot method. The probe used is specific for inducible hsp70 mRNA. Our results showed that the glutamic acid under limited conditions (such as at 50-500 mumol/L and action time 5-30 min) could induce the expression of hsp70 mRNA, which was partly mediated by NMDA receptors. On the other hand, ACh (0.1-1,000 mumol/L) could not induce the expression of hsp 70 mRNA.

Abstract:Historical data, epidemiological surveys, laboratory and animal experiments, clinical studies and observations in ordinary dental practice all illustrate that starchy fibrous foods have a low cariogenic potential. The recommended substitution of sucrose in Western diets by starchy foods would lead to a reduction of caries provided that these other carbohydrates replace the between meal consumption of sugar-rich products. The low cariogenic potential of a diet containing carbohydrates with a fibrous character is probably not solely due to its low sucrose content. Such a diet promotes mastication and saliva secretion which tend to reduce the caries risk. Furthermore, unrefined foods may contain caries-protective substances like phytates, trace elements and antibacterial factors. Lectins, which are found in many plants could be of importance by influencing the attachment of micro-organisms to the teeth in a favourable manner. The tradition to end a meal with fruit may thus have other oral effects than hitherto considered. In addition, a diet containing carbohydrates with a fibrous character and a low amount of sucrose probably has a low pathogenic potential for the periodontal tissues. However, no epidemiological data are available to support this assumption.

Abstract:This study aimed to investigate whether acute passive heat stress 1) decreases muscle Maximal Voluntary Contraction (MVC); 2) increases peripheral muscle fatigue; 3) increases spinal cord excitability, and 4) increases key skeletal muscle gene signaling pathways in skeletal muscle. Examining the biological and physiological markers underlying passive heat stress will assist us in understanding the potential therapeutic benefits. MVCs, muscle fatigue, spinal cord excitability, and gene signaling were examined after control or whole body heat stress in an environmental chamber (heat; 82 °C, 10% humidity for 30 min). Heart Rate (HR), an indicator of stress response, was correlated to muscle fatigue in the heat group (R = 0.59; p < 0.05) but was not correlated to MVC, twitch potentiation, and H reflex suppression. Sixty-one genes were differentially expressed after heat (41 genes >1.5-fold induced; 20 < 0.667 fold repressed). A strong correlation emerged between the session type (control or heat) and principal components (PC1) (R = 0.82; p < 0.005). Cell Signal Transduction, Metabolism, Gene Expression and Transcription, Immune System, DNA Repair, and Metabolism of Proteins were pathway domains with the largest number of genes regulated after acute whole body heat stress. Acute whole-body heat stress may offer a physiological stimulus for people with a limited capacity to exercise.

Abstract:The intracellular build-up of thermally damaged proteins following exposure to heat stress results in the synthesis of a family of evolutionarily conserved proteins called heat shock proteins (Hsps) that act as molecular chaperones, protecting the cell against the aggregation of denatured proteins. The transcriptional regulation of heat shock genes by heat shock factor 1 (HSF1) has been extensively studied in model systems, but little research has focused on the role HSF1 plays in Hsp gene expression in eurythermal organisms from broadly fluctuating thermal environments. The threshold temperature for Hsp induction in these organisms shifts with the recent thermal history of the individual but the mechanism by which this plasticity in Hsp induction temperature is achieved is unknown. We examined the effect of thermal acclimation on the heat-activation of HSF1 in the eurythermal teleost Gillichthys mirabilis. After a 5-week acclimation period (at 13, 21 or 28 degrees C) the temperature of HSF1 activation was positively correlated with acclimation temperature. HSF1 activation peaked at 27 degrees C in fish acclimated to 13 degrees C, at 33 degrees C in the 21 degrees C group, and at 36 degrees C in the 28 degrees C group. Concentrations of both HSF1 and Hsp70 in the 28 degrees C group were significantly higher than in the colder acclimated fish. Plasticity in HSF1 activation may be important to the adjustable nature of the heat shock response in eurythermal organisms and the environmental control of Hsp gene expression.

Quantitative Proteomics Provided Insights into the Protective Effects of Heat Acclimation on the Rat Hypothalamus after Exertional Heatstroke

Abstract:Background: The effects of heat acclimation (HA) on the hypothalamus after exertional heatstroke (EHS) and the specific mechanism have not been fully elucidated, and this study aimed to address these questions. Methods: In the present study, rats were randomly assigned to the control, EHS, HA, or HA + EHS groups (n = 9). Hematoxylin and eosin (H&E) staining was used to examine pathology. Tandem mass tag (TMT)-based proteomic analysis was utilized to explore the impact of HA on the protein expression profile of the hypothalamus after EHS. Bioinformatics analysis was used to predict the functions of the differentially expressed proteins. The differential proteins were validated by western blotting. An enzyme-linked immunosorbent assay was used to measure the expression levels of inflammatory cytokines in the serum. Results: The H&E staining (n = 5) results revealed that there were less structural changes in hypothalamus in the HA + EHS group compared with the EHS group. Proteomic analysis (n = 4) revealed that proinflammatory proteins such as argininosuccinate synthetase (ASS1), high mobility group protein B2 (HMGB2) and vimentin were evidently downregulated in the HA + EHS group. The levels of interleukin (IL)-1β, IL-1, and IL-8 were decreased in the serum samples (n = 3) from HA + EHS rats. Conclusions: HA may alleviate hypothalamic damage caused by heat attack by inhibiting inflammatory activities, and ASS1, HMGB2 and vimentin could be candidate factors involved in the exact mechanism.
Investigation into the signal transduction pathway via which heat stress impairs intestinal epithelial barrier function

Abstract:Background and aims: Intact protein absorption is thought to be a causative factor in several intestinal diseases, such as food allergy, celiac disease and inflammatory bowel disease. However, the mechanism remains unclear. The aim of this study was to characterize a novel signal transduction pathway via which heat stress compromises intestinal epithelial barrier function. Methods: Heat stress was carried out by exposing confluent human intestinal epithelial cell line T84 cell monolayers to designated temperatures (37-43 degrees C) for 1 h. Transepithelial electric resistance (TER) and permeability to horseradish peroxidase (HRP, molecular weight = 44 000) were used as indicators to assess the intestinal epithelial barrier function. Phosphorylated myosin light chain (MLC), MLC kinase (MLCK) and protein kinase C (PKC) protein of the T84 cells were evaluated in order to identify the signal transduction pathway in the course of heat stress-induced intestinal epithelial barrier dysfunctions. Results: The results showed that exposure to heat stress significantly increased intact protein transport across the intestinal epithelial monolayer; the amount of phospho-PKC, phospho-MLCK and phospho-MLC proteins in T84 cells decreased significantly at 41 degrees C and 43 degrees C although they increased at 39 degrees C. The heat stress-induced T84 monolayer barrier dysfunction was inhibited by pretreatment with PKC inhibitor, MLCK inhibitor, or HSP70. Conclusion: Heat stress can induce intestinal epithelial barrier dysfunction via the PKC and MLC signal transduction pathway.

Abstract:High temperature has become a global concern because it seriously affects the growth and reproduction of plants. Exposure of plant cells to high temperatures result in cellular damage and can even lead to cell death. Part of the damage can be ascribed to the action of reactive oxygen species (ROS), which accumulate during abiotic stresses such as heat stress. ROS are toxic and can modify other biomacromolecules including membrane lipids, DNA, and proteins. In order to protect the cells, ROS scavenging is essential. In contrast with their inherent harms, ROS also function as signaling molecules, inducing stress tolerance mechanisms. This review examines the evidence for crosstalk between the classical heat stress response, which consists of heat shock factors (HSFs) and heat shock proteins (HSPs), with the ROS network at multiple levels in the heat response process. Heat stimulates HSF activity directly, but also indirectly via ROS. HSFs in turn stimulate the expression of HSP chaperones and also affect ROS scavenger gene expression. In the short term, HSFs repress expression of superoxide dismutase scavenger genes via induction of miRNA398, while they also activate scavenger gene expression and stabilize scavenger protein activity via HSP induction. We propose that these contrasting effects allow for the boosting of the heat stress response at the very onset of the stress, while preventing subsequent oxidative damage. The described model on HSFs, HSPs, ROS, and ROS scavenger interactions seems applicable to responses to stresses other than heat and may explain the phenomenon of crossacclimation.

Abstract:Abstract The adrenal gland responds to heat stress by epinephrine and glucocorticoid release to alleviate the adverse effects. This study investigated the effect of acute heat stress on the protein profile and histone modification in the adrenal gland of layer-type country chickens. A total of 192 roosters were subject to acute heat stress and thereafter classified into a resistant or susceptible group according to body temperature change. The iTRAQ analysis identified 80 differentially expressed proteins, in which the resistant group had a higher level of somatostatin and hydroxy-δ-5-steroid dehydrogenase but a lower parathymosin expression in accordance with the change of serum glucocorticoid levels. Histone modification analysis identified 115 histone markers. The susceptible group had a higher level of tri-methylation of histone H3 lysine 27 (H3K27me3) and showed a positive crosstalk with K36me and K37me in the H3 tails. The differential changes of body temperature projected in physiological regulation at the hypothalamus–pituitary–adrenal axis suggest the genetic heterogeneity in basic metabolic rate and efficiency for heat dissipation to acclimate to thermal stress and maintain body temperature homeostasis. The alteration of adrenal H3K27me3 level was associated with the endocrine function of adrenal gland and may contribute to the thermotolerance of chickens.

[Effect of nifedipine on sinus node and atrioventricular node function following autonomic blockade in the human].

Abstract:The effect of intravenously administered nifedipine (7.5 micrograms/kg) on sinus node and atrioventricular (AV) node function was evaluated in 17 patients, 9 of whom had sinus node disease after autonomic blockade (propranolol 0.2 mg/kg and atropine 0.04 mg/kg i.v.). The patients' ages ranged from 18 to 77 years. After nifedipine there was a nonsignificant increase in the median values for sinus cycle length from 750 ms to 760 ms, for sinus node recovery time from 1150 ms to 1240 ms, for corrected sinus node recovery time from 440 ms to 460 ms, and for sinoatrial conduction time from 63 ms to 75 ms. Nevertheless, nifedipine produced a significant increase in the AV node refractory periods (median values): the effective refractory period lengthened from 300 ms to 305 ms (rate 100/min) and from 290 ms to 300 ms (rate 120/min) respectively. The functional refractory period increased from 380 ms to 400 ms (rate 100/min) and from 385 ms to 410 ms (rate 120/min) respectively. The Wenckebach period was significantly prolonged from 370 ms to 390 ms. We conclude, therefore, that in the absence of autonomic control intravenous administration of nifedipine exerts a mild depressive effect on AV node but not on sinus node function.
Monitoring the biochemical and cellular responses of marine bivalves during thermal stress by using biomarkers

Abstract:The present work aimed to study the cellular, biochemical and molecular biomarkers in the digestive glands and hemocytes of Modiolus barbatus and whether there is a hierarchy in their response to thermal stress. We determined a) the neutral red retention assay (NRR) in heamotocytes and b) the lysosomal membrane stability (LMS), the levels of second messenger cAMP, the activity of acetylcholinesterase (AChE) in the digestive glands of Modiolus barbatus after acclimation to 18 °C, 24 °C, 28 °C or 30 °C for 30 days. Moreover, in order to estimate the threshold of temperature inducing expression of stress proteins we determined the levels of Hsp70 and Hsp90 in the digestive glands. Hsps are expressed at lower temperature than those causing reduction in the LMS and NNR times. The reduction in the LMS and NNR times at high temperatures of acclimation might be related to inability of Modiolus barbatus to gain energy from the ingested food.
Physiological Responses of Corythucha Ciliata Adults to High Temperatures under Laboratory and Field Conditions.

Abstract:Under high temperature conditions, insects can tolerate to survive through various physiological mechanisms, which have been well documented in laboratory studies. However, it is still unclear as to whether these laboratory data can scale up to those in the field. Here we studied dynamics of heat-induced metabolites in Corythucha ciliata adults under both laboratory and field conditions to examine their significance in thermal tolerance of the species. We compared the effects of controlled thermal treatments (2h at 33-43 °C at 2 °C intervals in the laboratory) and naturally increasing thermal conditions (10:00-14:00 at 2-h intervals (33.5-37.2 °C) on a hot summer day in a field in Shanghai, China) on water content and levels of water-soluble protein, triglycerides, mannitol, and sorbitol in the adult bodies. The results showed that water content significantly decreased and all other metabolic parameters significantly increased in response to temperature stresses with similar patterns in both the laboratory and field, although the respective threshold temperatures were different under the two conditions. The close linkage observed in the two conditions suggests that a short period of heat stress induces water loss and accumulation of thermal metabolites in C. ciliata adults. This heat-resistance provides a defense mechanism counteracting thermal damage in C. ciliata.

Abstract:BackgroundChanges in ambient temperature seriously affect physiological regulation and biochemical reactions in ectotherms. However, transient elevation in oceanic temperature occurs naturally during the day. Short-term elevation in the ambient temperature affects different physiological responses in marine fish, including cellular protein stability and osmotic balance of the internal environment. Since fish gills are vital osmoregulatory organ which directly contacts external environment, activation of cytoprotective responses to maintain gill cell viability and biological function is essential for fish survival under challenging environmental conditions. The purpose of this study was to investigate the short-term effects of elevated temperature on physiological regulation in the gills of a marine teleost, blue green damselfish (Chromis viridis).ResultsAs part of the stress response, plasma glucose levels were induced by short-term hyperthermic exposure (12 h). Furthermore, upregulation of the levels of gill heat shock proteins (HSPs) and ubiquitinated proteins was essential for preventing the accumulation of protein aggregations in branchial cells of C. viridis under hyperthermic stress. The specific activity of branchial Na+/K+-ATPase (NKA), however, significantly reduced while the amount of protein was similar between normal and high-temperature groups.ConclusionsThe present study provided the evidence to illustrate that activation of the branchial protein quality control mechanism to carry out cytoprotective response was involved in coping with thermal stress. However, plasma osmolality and muscle water content, respectively, that slightly but evidently increased and decreased might result from impaired osmoregulatory ability due to hyperthermia-decreased gill NKA activity.

The effect of 15 consecutive days of heat-exercise acclimation on heat shock protein 70

Abstract:The purpose of this study was to investigate the alterations in serum heat shock protein (Hsp) 70 levels during a 15-consecutive-day intermittent heat-exercise protocol in a 29-year-old male ultra marathon runner. Heat acclimation, for the purpose of physical activities in elevated ambient temperatures, has numerous physiological benefits including mechanisms such as improved cardiac output, increased plasma volume and a decreased core temperature (T (c)). In addition to the central adaptations, the role of Hsp during heat acclimation has received an increasing amount of attention. The acclimation protocol applied was designed to correspond with the athlete's tapering period for the 2007 Marathon Des Sables. The subject (VO(2)max = 50.7 ml.kg(-1).min(-1), peak power output [PPO] = 376 W) cycled daily for 90 min at a workload corresponding to 50% of VO(2)max in a temperature-controlled room (average WBGT = 31.9 +/- 0.9 degrees C). Venous blood was sampled before and after each session for measurement of serum osmolality and serum Hsp70. In addition, T (c), heart rate (HR) and power output (PO) was measured throughout the 90 min to ensure that heat acclimation was achieved during the 15-day period. The results show that the subject was successfully heat acclimated as seen by the lowered HR at rest and during exercise, decreased resting and exercising T (c) and an increased PO. The heat exercise resulted in an initial increase in Hsp70 concentrations, known as thermotolerance, and the increase in Hsp70 after exercise was inversely correlated to the resting values of Hsp70 (Spearman's rank correlation = -0.81, p < 0.01). Furthermore, the 15-day heat-exercise protocol also increased the basal levels of Hsp70, a response different from that of thermotolerance. This is, as far as we are aware, the first report showing Hsp70 levels during consecutive days of intermittent heat exposure giving rise to heat acclimation. In conclusion, a relatively longer heat acclimation protocol is suggested to obtain maximum benefit of heat acclimation inclusive of both cellular and systemic adaptations.
Effects of Chronic Stress on Behavior and Heat Shock Protein 70 Expression in Immune Cells

Abstract:Chronic stress can suppress organisms' resistance to adverse stimuli in the environment. The stress responses occur at both the system and single-cell levels. Although many studies have investigated the neuroendocrine mechanisms underlying the effects of stress on health, the characteristics of cellular stress response are still poorly understood. One manner in which cells resist damage and/or death induced by stress is to synthesize a highly conserved set of intracellular proteins, termed heat shock protein (HSP). In particular, the 70-kDa HSP (HSP70) is essential for cellular recovery after stress as well as survival and maintenance of normal cellular function. It has been demonstrated that the expression of high levels of HSP70 is associated with an increased resistance of cells to challenges that would otherwise lead to cell injury and/or death. Therefore, the intensity of an HSP70 response to stress is an important biomarker of how the cell tolerates stress damage. This study aimed to determine whether prior chronic stress inhibited the synthesis of HSP70 in immune cells in the peripheral blood and spleen of rats exposed to an acute hot water bath. Twenty-eight rats were randomly divided into 2 groups with 14 rats in each group: the control group (C) and the chronic stress group (CS). CS rats were exposed to chronic unpredictable stress for 4 weeks to induce depressive-like behavior. The controls were stress-free. Thereafter, 8 rats in each group were exposed to acute heat stress (C-heat shock and CS-heat shock) to induce whole-body hyperthermia (maintaining the core temperature at 41℃ for 25 minutes).The other rats remained undisturbed in their home cages (C-no heat shock and CS-no heat shock). All rats were decapitated 6 hours after the completion of the treatments. The blood and spleen were collected and the level of HSP70 expression in the leukocytes was measured using flow cytometry. It was found that CS rats showed a decreased cellular HSP70 expression following the heat treatment, while a significant increase in the synthesis of HSP70 was observed in all types of immune cells from C rats. However, in CS rats, such HSP70 changes were only detected in the monocytes and granulocytes. Further, the increasing degree of heat-induced cellular HSP70 expression in CS rats was less pronounced than that observed in C rats. These results demonstrated that chronic stress decreased heat-induced HSP70 response in immune cells. Further, they suggested that the deficit of HSP70 might be involved in the suppressive modulation of the immune function induced by chronic stress.

Abstract:1. Animal survival and species distribution in the face of global warming and increasing occurrences of heatwave largely depend on how heat tolerance shifts with plastic responses at different spatiotemporal scales, including long‐term acclimation/acclimatization and short‐term heat hardening. However, knowledge about the interaction of these plastic responses is still unclear.2. To understand how plastic responses at different timescales work together to adjust heat tolerance of organisms, we examined the effect of heat hardening on the upper thermal limits of an intertidal mudflat bivalve, the razor clam Sinonovacula constricta, for different seasons by using heart rate as a proxy.3. We observed a stronger heat hardening response of S. constricta in warm seasons, implying that heat hardening worked synchronously with seasonal acclimatization to increase resistance of the clams to high temperatures in warm seasons. In warm seasons, heat hardening increased heat tolerance by 2‐4°C and showed a 24‐h temporal dependence, suggesting an adaptation to the diel fluctuation of thermal regimes in summer.4. Furthermore, thermal stress resembling seasonal maximum environmental temperature induced stronger heat hardening effects, indicating that heat hardening is an essential plastic response to extreme hot weather, complementing seasonal acclimatization.5. Our results suggest that high temperature risk can be alleviated jointly by seasonal acclimatization and heat hardening, and emphasize the importance of considering physiological plasticity on both long‐term and short‐term temporal scales in evaluating and forecasting vulnerability of organisms to climate change.
