Abstract:In this pedagogical paper a coherent explanation of the resting potential of nerve cells is given in terms of its determining factors. These are the currents of active transport of the ions to which the membrane is permeable, their membrane permeabilities and their concentrations in the extracellular fluid. They play the role of the independent variables in the problem and simultaneously also determine the concentrations of the permeating ions inside the cell. The resting state is assumed to be a stationary state, and the resting potential is understood to be the membrane potential in this state. The explanation presented here differs from the conventional one found in textbooks with respect to the handling of the intracellular concentrations of the permeant ions. In the conventional description the causal relations between these and the independent variables are not taken into account, and the intracellular concentrations are treated like additional independent variables. The "causal" theory presented here leads to a simple formula for the resting potential in terms of the currents of active transport (pumping by the Na,K-pump and carrier transport) and the membrane permeabilities for sodium and potassium ions. In addition it leads to two results not obtained in the conventional treatment: An upper bound exists for the magnitude of the resting potential, and the difference between the resting potential and the diffusion potential, which can be measured after poisoning of the ion pumps, is small.

Abstract:In this paper, analysis of symbol error probability (SEP) is carried over frequency non-selective slowly Rician fading channel with N branch receiver diversity using maximal ratio combining (MRC) technique. We assume that channel side information is known to the receiver. Symbol error rate expression of coherent M-ary phase shift keying (MPSK) and non coherent M-ary frequency shift keying (MFSK) are obtained through numerical computation based on moment generating function (MGF) approach in order to avoid complex numerical calculation. Error performance plots of these two modulation techniques has been drawn and compared for different values of Rician factor K, diversity order N and modulation order M.
Defined extracellular ionic solutions to study and manipulate the cellular resting membrane potential

Abstract:All cells possess an electric potential across their plasma membranes and can generate and receive bioelectric signals. The cellular resting membrane potential (RMP) can regulate cell proliferation, differentiation and apoptosis. Current approaches to measure the RMP rely on patch clamping, which is technically challenging, low-throughput and not widely available. It is therefore critical to develop simple strategies to measure, manipulate and characterize the RMP. Here, we present a simple methodology to study the RMP of non-excitable cells and characterize the contribution of individual ions to the RMP using a voltage-sensitive dye. We define protocols using extracellular solutions in which permeable ions (Na+, Cl- and K+) are substituted with non-permeable ions [N-Methyl-D-glucamine (NMDG), gluconate, choline, SO42-]. The resulting RMP modifications were assessed with both patch clamp and a voltage sensitive dye. Using an epithelial and cancer cell line, we demonstrate that the proposed ionic solutions can selectively modify the RMP and help determine the relative contribution of ionic species in setting the RMP. The proposed method is simple and reproducible and will make the study of bioelectricity more readily available to the cell biology community.This article has an associated First Person interview with the first author of the paper.

Abstract:Neuronal activity is associated with transmembrane ionic redistribution, which can lead to an osmotic imbalance. Accordingly, activity-dependent changes of the membrane potential are sometimes accompanied by changes in intracellular and/or extracellular volume. Experimental data that include distributions of ions and volume during neuronal activity are rare and rather inconsistent partly due to the technical difficulty of performing such measurements. However, progress in understanding the interrelations among ions, voltage and volume has been achieved recently by computational modelling, particularly "charge-difference" modelling. In this work a charge-difference computational model was used for further understanding of the specific roles for cations and anions. Our simulations show that without anion conductances the transmembrane movements of cations are always osmotically balanced, regardless of the stoichiometry of the pump or the ratio of Na+ and K+ conductances. Yet any changes in cation conductance or pump activity are associated with changes of the membrane potential, even when a hypothetically electroneutral pump is used in calculations and K+ and Na+ conductances are equal. On the other hand, when a Cl- conductance is present, the only way to keep the Cl-equilibrium potential in accordance with the changed membrane potential is to adjust cell volume. Importantly, this voltage-evoked Cl--dependent volume change does not affect intracellular cation concentrations or the amount of energy that is necessary to support the system. Taking other factors into consideration (i.e. the presence of internal impermeant poly-anions, the activity of cation-Cl- cotransporters, and the buildup of intra- and extracellular osmolytes, both charged and electroneutral) adds complexity, but does not change the main principles.
Resting membrane potential and intracellular [Na+] at rest, during fatigue and during recovery in rat soleus muscle fibres in situ

Abstract:figure legend With repeated muscle contractions a rundown of the trans‐sarcolemmal sodium (Na+) and potassium (K+) gradients (hence depolarisation of resting membrane potential (EM)) are likely to manifest concomitantly and contribute to fatigue. Therefore, we examined how resting EM and intracellular sodium concentration ([Na+]i) change with brief or fatiguing stimulation of rat soleus fibres in situ. Using ion‐sensitive double‐barrelled microelectrodes, we observed that brief stimulation evoked a large elevation of [Na+]i and hyperpolarisation, whereas fatiguing simulation evoked considerable depolarisation and even higher [Na+]i. [Na+]i values at fatigue were determined by curve‐fitting the recovery data and back‐extrapolating to the end of stimulation. Na+ influx occurs via voltage‐activated (NaV1.4) and other Na+ channels whereas K+ efflux involves delayed rectifier K+ channels (Kv). Raised [Na+]i stimulates Na+–K+ pump activity. Recovery [Na+]i is likely influenced by at least the Na+–K+ pump and sodium–hydrogen exchanger (NHE) activity. The findings show, in single fibres, a severe depolarisation and massive increase of [Na+]i with fatigue in situ, then rapid recovery post‐stimulation. Large trans‐sarcolemmal ionic shifts occur with fatiguing exercise or stimulation of isolated muscles. However, it is unknown how resting membrane potential (EM) and intracellular sodium concentration ([Na+]i) change with repeated contractions in living mammals. We investigated (i) whether [Na+]i (peak, kinetics) can reveal changes of Na+–K+ pump activity during brief or fatiguing stimulation and (ii) how resting EM and [Na+]i change during fatigue and recovery of rat soleus muscle in situ. Muscles of anaesthetised rats were stimulated with brief (10 s) or repeated tetani (60 Hz for 200 ms, every 2 s, for 30 s or 300 s) with isometric force measured. Double‐barrelled ion‐sensitive microelectrodes were used to quantify resting EM and [Na+]i. Post‐stimulation data were fitted using polynomials and back‐extrapolated to time zero recovery. Mean pre‐stimulation resting EM (layer 2–7 fibres) was −71 mV (surface fibres were more depolarised), and [Na+]i was 14 mM. With deeper fibres, 10 s stimulation (2–150 Hz) increased [Na+]i to 38–46 mM whilst simultaneously causing hyperpolarisations (7.3 mV for 2–90 Hz). Fatiguing stimulation for 30 s or 300 s led to end‐stimulation resting EM of −61 to −53 mV, which recovered rapidly (T1/2, 8–22 s). Mean end‐stimulation [Na+]i increased to 86–101 mM with both fatigue protocols and the [Na+]i recovery time‐course (T1/2, 21–35 s) showed no difference between protocols. These combined findings suggest that brief stimulation hyperpolarises the resting EM, likely via maximum Na+‐induced stimulation of the Na+–K+ pump. Repeated tetani caused massive depolarisation and elevations of [Na+]i that together lower force, although they likely interact with other factors to cause fatigue. [Na+]i recovery kinetics provided no evidence of impaired Na+–K+ pump activity with fatigue. Key points It is uncertain how resting membrane potential, intracellular sodium concentration ([Na+]i), and sodium–potassium (Na+–K+) pump activity change during repeated muscle contractions in living mammals. For rat soleus muscle fibres in situ, brief tetanic stimulation for 10 s led to raised [Na+]i, anticipated to evoke maximal Na+‐induced stimulation of the Na+–K+ pump causing an immediate hyperpolarisation of the sarcolemma. More prolonged stimulation with repeated tetanic contractions causes massive elevations of [Na+]i, which together with large depolarisations (via K+ disturbances) likely reduce force production. These effects occurred without impairment of Na+–K+ pump function. Together these findings suggest that rapid activation of the Na+–K+ pump occurs with brief stimulation to maintain excitability, whereas more prolonged stimulation causes rundown of the trans‐sarcolemmal K+ gradient (hence depolarisation) and Na+ gradient, which in combination can impair contraction to contribute to fatigue in living mammals.

Characterization of the electrogenic sodium pump in cardiac Purkinje fibres

Abstract:1. The Na pump is examined in sheep cardiac Purkinje fibres using a two micro-electrode voltage clamp technique.2. After reducing the external K concentration, [K](o), to zero for 2 min or more, subsequent addition of an ;activator cation' (known to activate the Na pump in other preparations) produces a transient increase of outward current. This outward current transient is abolished by 10(-5)M-strophanthidin (cf. Gadsby & Cranefield, 1979a).3. It is concluded that this transient increase of outward current is a result of a transient stimulation of the sodium pump by the raised [Na](i) following exposure to 0-K(o). Although this current transient may reflect the activity of an electrogenic Na pump, it is difficult to use K as the activator cation to establish this point. This is due to the extracellular K depletion that occurs during Na pump reactivation and the subsequent change that this K depletion produces in the current-voltage relationship of the Purkinje fibre.4. Rb(o) or Cs(o) have been used instead of K(o) to reactivate the Na pump when examining the transient increase of outward current. On adding either of these cations after exposing a preparation to a solution without such ;activator cations', the outward current transient is relatively voltage independent over a wide range of potentials (-90 to +10 mV). It is concluded that, following the addition of Rb(o) or Cs(o), the transient increase of outward current is a direct measure of the transient increase of the electrogenic Na pump current.5. Increasing [Rb](o) or [Cs](o) over the range of 0-40 mM increases the rate of decay of the electrogenic Na pump current transient. Using a simple model (cf. Rang & Ritchie, 1968), it is shown that the decay rate constant of the electrogenic Na pump current transient is a good measure of the degree of activation of the external site of the Na pump. At a given concentration of activator cation, Rb(o) produces a greater activation of the Na pump than does Cs(o). The K(0.5) for Rb(o) is 6.3 mM and for Cs(o) is 14.2 mM. Li(o) activates the Na pump more weakly than Rb(o) and Cs(o).6. The coupling ratio of the Na pump is shown to be independent of Rb(o) or Cs(o) over the range 2-40 mM. Furthermore, consistent with the results of Gadsby & Cranefield (1979a), the coupling ratio is independent of Na(i) over the range considered.7. The Q(10) for the electrogenic Na pump current transient varies between 1.6 and 2.3 over the range of temperature 26-46 degrees C.8. A maximum Na pump current of about 0.78 muA cm(-2) is obtained. Assuming a coupling ratio of 3Na/2K, the rate of Na ion transport into the cell is estimated to be about 23 p-mole cm(-2) sec(-1). Assuming a Na pump turnover of 150 sec(-1), we estimate that there are about 1000 Na pump sites per mum(2) of cell surface.9. We conclude that the electrogenic Na pump current transient provides a good measure of the activity of the Na pump when Rb or Cs are used as ;activator cations'. This measure can be used in the intact preparation to investigate the relationship between Na pump rate and other cellular events such as the regulation of tension (Eisner & Lederer, 1980).
Resting trabecular meshwork cells experience constitutive cation influx

Abstract:A quintessential sentinel of cell health, the membrane potential in nonexcitable cells integrates biochemical and biomechanical inputs, determines the driving force for ionic currents activated by input signals and plays critical functions in cellular differentiation, signaling, and pathology. The identity and properties of ion channels that subserve the resting potential in trabecular meshwork (TM) cells is poorly understood, which impairs our understanding of intraocular pressure regulation in healthy and diseased eyes. Here, we identified a powerful cationic conductance that subserves the TM resting potential. It disappears following Na+ removal or substitution with choline or NMDG+, is insensitive to TTX, verapamil, phenamil methanesulfonate, amiloride and GsMTx4, is substituted by Li+ and Cs+, and inhibited by Gd3+ and Ruthenium Red. Constitutive cation influx is thus not mediated by voltage-operated Na+, Ca2+, epithelial Na+ (ENaC) channels, Piezo channels or Na+/H+ exchange but may involve TRP-like channels. Transcriptional analysis detected expression of many TRP genes, with the transcriptome pool dominated by TRPC1 followed by expression of TRPV1, TRPC3, TRPV4 and TRPC5. Pyr3 and Pico1,4,5 did not affect the standing current whereas SKF96365 promoted rather than suppressed, Na+ influx. SEA-0400 induced a modest hyperpolarization, indicating residual contribution from Na+/Ca2+ exchange. The resting membrane potential in human TM cells is thus maintained by a constitutive monovalent cation leak current with properties not unlike those of TRP channels. This conductance is likely to influence conventional outflow by setting the homeostatic steady-state and by regulating the magnitude of pressure-induced currents in normotensive and hypertensive eyes.

Abstract:1. The mechanism by which the fluorescent, cationic dye diS‐C3‐(5) responds to the membrane potential of red blood cells has been investigated. 2. The dye in aqueous solution absorbs most strongly at 650 nm. Addition of white, haemoglobin‐free membranes red shifts the absorption maximum ca. 20 nm, while addition of membrane‐free cell lysate results in the appearance of a new dye absorption peak at 590 nm. Thus the dye binds both to cell membranes and to cell contents. The component of the cytoplasm which binds the dye is non‐dialysable, presumably haemoglobin. 3. Dye added to a suspension of intact cells shows a strong absorption at 590 nm indicating that the dye has bound to the cell contents and that the membrane is permeable to the dye. 4. The amount of dye which partitions into (and on to) the cells can be determined, as reported by Sims, Waggoner, Wang & Hoffman (1974), from the fluorescence of the dye remaining in the supernatant after the cells are centrifuged to the bottom of the suspension. In most conditions the proportion of the cell associated dye which is either free inside the cell or bound to the outside face of the membrane is negligible compared to the proportion bound to the cell contents. 5. On the assumption that the dye is not actively transported, the ratio of the equilibrium dye activities inside and outside the cell, ai/ao, is determined by the membrane potential according to the Nernst relation. Driving the membrane potenial negative then increases the cell associated dye by increasing the activity ratio and hence ai and the amount of dye bound to cell contents. 6. At the known Donnan equilibrium potential the internal dye activity can be calculated from the external activity. An empirical relation between cell associated dye and internal activity has been determined by measuring the dye partition between cells and medium at different external activities. 7. Using this empirial relation, and providing that any changes in cell composition do not affect the dye binding, the internal activity at any potential can be calculated from the measured amount of cell associated dye. The external activity can be estimated fluorimetrically. The membrane potential is then calculated from the activity ratio. 8. The membrane potenial of cells has been altered by adding valinomycin in the presence of different K gradients. Under the conditions used, the 'constant field' permeability for K‐Val is 15‐20 times that of Cl. 9. Dye binding to haemoglobin is influenced by pH and thus dye partitioning into cells changes with intracellular pH. Increasing intracellular pH increases the amount of dye partitioned, while decreasing pH decreases this amount. 10. When large potentials are produced with valinomycin there is no change in intracellular pH. This result indicates that in red blood cells intracellular pH is determined by the external pH and the Cl concentration ratio and not by the membrane potentials. 11. The resting membrane potential of Drosophila melanogaster larval muscle depends strongly on external calcium concentration

Abstract:The resting membrane potential (RMP) of most cells is not greatly influenced by the transmembrane calcium gradient because at rest, the membrane has very low permeability to calcium. We have observed, however, that the resting membrane potential of muscle cells in the larval bodywall of Drosophila melanogaster varies widely as the external calcium concentration is modified. The RMP depolarized as much as 21.8 mV/mM calcium at low concentrations, and on average, about 10 mV/mM across a range typical of neurophysiological investigations. The extent to which muscle RMP varies has important implications for the measurement of synaptic potentials as well. Two parameters of excitatory junctional potential (EJP) voltage were compared across a range of RMPs. EJP amplitude (DeltaV) and peak voltage (maxima) change as a function of RMP; on average, a 10 mV change in RMP elicits a 4-5 mV change in EJP amplitude and peak voltage. The influence of the calcium gradient on resting and synaptic membrane potentials led us to investigate the endogenous ion concentrations of larval hemolymph. In addition to the major monovalent ions and calcium, we report the first voltammetric analysis of magnesium concentration in larval fruit fly hemolymph.
Emerging Roles of the Membrane Potential: Action Beyond the Action Potential

Abstract:Whilst the phenomenon of an electrical resting membrane potential (RMP) is a central tenet of biology, it is nearly always discussed as a phenomenon that facilitates the propagation of action potentials in excitable tissue, muscle, and nerve. However, as ion channel research shifts beyond these tissues, it became clear that the RMP is a feature of virtually all cells studied. The RMP is maintained by the cell's compliment of ion channels. Transcriptome sequencing is increasingly revealing that equally rich compliments of ion channels exist in both excitable and non-excitable tissue. In this review, we discuss a range of critical roles that the RMP has in a variety of cell types beyond the action potential. Whereas most biologists would perceive that the RMP is primarily about excitability, the data show that in fact excitability is only a small part of it. Emerging evidence show that a dynamic membrane potential is critical for many other processes including cell cycle, cell-volume control, proliferation, muscle contraction (even in the absence of an action potential), and wound healing. Modulation of the RMP is therefore a potential target for many new drugs targeting a range of diseases and biological functions from cancer through to wound healing and is likely to be key to the development of successful stem cell therapies.

Abstract:The work and the professional relationships of clinicians who use hypnosis and their research counterparts always contain a certain degree of tension. This is especially true when one group or the other claims to have a purchase on truth. A review of the strengths and limitations of each subspecialty is provided along with an understanding of some of the differences between the clinical and experimental perspectives. The author raises a number of points with the hope that this article will promote discussion among those who are engaged in clinical and experimental work and those few who are active in both. Several suggestions for integration and collaboration are offered for consideration by both groups.
Dependence of cell's membrane potential on extracellular voltage observed in Chara globularis

Abstract:The membrane potential (V m ) of a cell results from the selective movement of ions across the cell membrane. Recent studies have revealed the presence of a gradient of voltage within a few nanometers adjacent to erythrocytes. Very notably this voltage is modified in response to changes in cell's membrane potential thus effectively extending the potential beyond the membrane and into the solution. In this study, using the microelectrode technique, we provide experimental evidence for the existence of a gradient of negative extracellular voltage (V z ) in a wide zone close to the cell wall of algal cells, extending over several micrometers. Modulating the ionic concentration of the extracellular solution with CO 2 alters the extracellular voltage and causes an immediate change in V m . Elevated extracellular CO 2 levels depolarize the cell and hyperpolarize the zone of extracellular voltage (ZEV) by the same magnitude. This observation strongly suggests a coupling effect between V z and V m . An increase in the level of intracellular CO 2 (dark respiration) leads to hyperpolarization of the cell without any immediate effect on the extracellular voltage. Therefore, the metabolic activity of a cell can proceed without inducing changes in V z . Conversely, V z can be modified by external stimulation without metabolic input from the cell. The evolution of the ZEV, particularly around spines and wounded cells, where ion exchange is enhanced, suggests that the formation of the ZEV may be attributed to the exchange of ions across the cell wall and cell membrane. By comparing the changes in V m in response to external stimuli, as measured by electrodes and observed using a potential-sensitive dye, we provide experimental evidence demonstrating the significance of extracellular voltage in determining the cell's membrane potential. This may have implications for our understanding of cell membrane potential generation beyond the activities of ion channels.

Abstract:Abstract The standard model of the cell membrane potential Vm describes it as arising from diffusion currents across a membrane with a constant electric field, with zero electric field outside the cell membrane. However, the influence of Vm has been shown to extend into the extracellular space where it alters the cell’s ζ-potential, the electrical potential measured a few nm from the cell surface which defines how the cell interacts with charged entities in its environment, including ions, molecules, and other cells. The paradigm arising from surface science is that the ζ-potential arises only from fixed membrane surface charge, and has consequently received little interest. However, if the ζ-potential can mechanistically and dynamically change by alteration of Vm, it allows the cell to dynamically alter cell–cell and cell-molecule interactions and may explain previously unexplained electrophysiological behaviours. Whilst the two potentials Vm and ζ are rarely reported together, they are occasionally described in different studies for the same cell type. By considering published data on these parameters across multiple cell types, as well as incidences of unexplained but seemingly functional Vm changes correlating with changes in cell behaviour, evidence is presented that this may play a functional role in the physiology of red blood cells, macrophages, platelets, sperm, ova, bacteria and cancer. Understanding how these properties will improve understanding of the role of electrical potentials and charges in the regulation of cell function and in the way in which cells interact with their environment. Insight The zeta (ζ) potential is the electrical potential a few nm beyond the surface of any suspensoid in water. Whilst typically assumed to arise only from fixed charges on the cell surface, recent and historical evidence shows a strong link to the cell’s membrane potential Vm, which the cell can alter mechanistically through the use of ion channels. Whilst these two potentials have rarely been studied simultaneously, this review collates data across multiple studies reporting Vm, ζ-potential, electrical properties of changes in cell behaviour. Collectively, this points to Vm-mediated ζ-potential playing a significant role in the physiology and activity of blood cells, immune response, developmental biology and egg fertilization, and cancer among others.

Abstract:Abstract Although the inner potential plays a fundamental role in electrochemistry, its precise definition is not clear. In fact, there are two different concepts: a physical quantity which can be measured by high energy electron scattering and calculated by density functional theory. For water, it is of the order of 4 V, and it is irrelevant for electrochemistry. For our purpose, the relevant quantity is the electrostatic potential experienced by an ion, which enters into the electrochemical potential. This is of the order of 0 to $$-$$ - 0.5 V, and may be slightly different for different ions.

Abstract:A two-parameter model describes the resistance of a membrane challenged with different salt concentrations on either side of the membrane. This model treats the membrane as a circuit consisting of a resistor located between two switches. One switch is on the diluate side of the membrane. The second switch is on the concentrated side. The probability of the diluate side switch being active (loaded with a counter-ion) is β (dimensionless) and the probability of the switch being active (loaded with a counter-ion) on the concentrated side is γ (dimensionless). The probability of the circuit being active is the product of the probabilities βγ in which case the circuit is actively transporting a counter-ion across the membrane. The probability functions are calculated from the conductivity of the diluate λ D (mS/cm), the concentrate λ C (mS/cm) and a fitted parameter { K sat } (mS/cm). The functions are β = λ D /(λ D +{K sat }) and γ = λ D /(λ D + { K sat }). These functions derived from enzyme kinetics where { K sat } is analogous to the Michaelis-Menton saturation coefficient. The second fitted parameter is the minimum resistance r min (Ωˑcm 2 ) representing the case where the product βγ approaches unity (λ D and λ C ≫ { K sat }). The product of the minimum resistance r min and the applied voltage is analogous to the reciprocal of the maximum rate of an enzyme reaction. The resistance of the membrane r mem (Ωˑcm 2 ) is r mem = r min /(βγ). Measurements made with equal salt solutions on both sides of the membrane are used to calculate the resistance under any salt conditions. The probability functions β and γ represent the relative activity of the counter-ions on the surfaces of the membrane. This suggests that the Nernst potential across the membrane v mem (Volts) is expressed as v mem = (ɌT/Ƒ)ˑln (γ/β) where Ɍ (8.314 J/°K⋅mol) is the gas law constant, T (°K) is the temperature, and Ƒ (96,485 C/mol) is the Faraday constant. The current density (A/cm 2 ) generated across the membrane under reverse electrodialysis conditions is i = v mem /r mem and the power density P (W/cm 2 ) is P = i 2 r mem . It is, therefore, possible to estimate the performance of a membrane from a limited data set of resistance data such as may be found in the literature.

Abstract:The mitochondrial adenosine 5′-diphosphate (ADP)/adenosine 5′-triphosphate (ATP) carrier imports ADP into the mitochondrion and exports ATP to the cell. Here, we demonstrate that 3.3 positive charges are translocated with the negatively charged substrate in each transport step. They can be assigned to three positively charged residues of the central substrate-binding site and two asparagine/arginine pairs. In this way, the membrane potential stimulates not only the ATP 4− export step, as a net −0.7 charge is transported, but also the ADP 3− import step, as a net +0.3 charge is transported with the electric field. These positive charge movements also inhibit the import of ATP and export of ADP in the presence of a membrane potential, allowing these nucleotides to be maintained at high concentrations in the cytosol and mitochondrial matrix to drive the hydrolysis and synthesis of ATP, respectively. Thus, this is the mechanism by which the membrane potential drives adenine nucleotide exchange with high directional fluxes to fuel the cellular processes.

Abstract:Regulating membrane potential is key to cellular function. For many animal cells, resting membrane potential is predominantly driven by a family of K2P (two-pore domain) potassium channels. These channels are commonly referred to as leak channels, as their presence results in the membrane being permeable to K+ ions. These channels, along with various pumps and exchangers, keep the cell resting membrane potential (Rp) relatively close to potassium's equilibrium potential (EK); however, in many cells, the resting membrane potential is more depolarized than the EK due to a small Na+ ion leak. Raising [Ca2+]O (extracellular Ca2+ concentration) can result in hyperpolarization of the membrane potential from the resting state. The mechanism for this hyperpolarization likely lies in the blockage of a Na+ leak channel (NALCN) and/or voltage-gated Na+ channels. The effects may also be connected to calcium-activated potassium channels. Using Drosophila melanogaster, we here illustrate that changing [Ca2+]O from 0.5 to 3 mM hyperpolarizes the muscle. Replacing NaCl with LiCl or choline chloride still led to hyperpolarization when increasing [Ca2+]O. Replacing CaCl2 with BaCl2 results in depolarization. K2P channel overexpression in the larval muscle greatly reduces the effects of [Ca2+]O on cell membrane potential, likely because potential is heavily driven by the EK in these muscles. These experiments provide an understanding of the mechanisms behind neuronal hypo-excitability during hypercalcemia, as well as the effects of altered expression of K2P channels on membrane potential.
Nernst-Planck-Gaussian modelling of electrodiffusional recovery from ephaptic excitation between mammalian cardiomyocytes

Abstract:Introduction: In addition to gap junction conduction, recent reports implicate possible ephaptic coupling contributions to action potential (AP) propagation between successive adjacent cardiomyocytes. Here, AP generation in an active cell, withdraws Na + from, creating a negative potential within, ephaptic spaces between the participating membranes, activating the initially quiescent neighbouring cardiomyocyte. However, sustainable ephaptic transmission requires subsequent complete recovery of the ephaptic charge difference. We explore physical contributions of passive electrodiffusive ion exchange with the remaining extracellular space to this recovery for the first time. Materials and Methods: Computational, finite element, analysis examined limiting, temporal and spatial, ephaptic [Na + ], [Cl − ], and the consequent Gaussian charge differences and membrane potential recovery patterns following a Δ V ∼130 mV AP upstroke at physiological (37°C) temperatures. This incorporated Nernst-Planck formalisms into equations for the time-dependent spatial concentration gradient profiles. Results: Mammalian atrial, ventricular and purkinje cardiomyocyte ephaptic junctions were modelled by closely apposed circularly symmetric membranes, specific capacitance 1 μF cm -2 , experimentally reported radii a = 8,000, 12,000 and 40,000 nm respectively and ephaptic axial distance w = 20 nm. This enclosed an ephaptic space containing principal ions initially at normal extracellular [Na + ] = 153.1 mM and [Cl − ] = 145.8 mM, respective diffusion coefficients D Na = 1.3 × 10 9 and D Cl = 2 × 10 9 nm 2 s -1 . Stable, concordant computational solutions were confirmed exploring ≤1,600 nm mesh sizes and Δ t ≤0.08 ms stepsize intervals. The corresponding membrane voltage profile changes across the initially quiescent membrane were obtainable from computed, graphically represented a and w -dependent ionic concentration differences adapting Gauss's flux theorem. Further simulations explored biological variations in ephaptic dimensions, membrane anatomy, and diffusion restrictions within the ephaptic space. Atrial, ventricular and Purkinje cardiomyocytes gave 40, 180 and 2000 ms 99.9% recovery times, with 720 or 360 ms high limits from doubling ventricular radius or halving diffusion coefficient. Varying a , and D Na and D Cl markedly affected recovery time-courses with logarithmic and double-logarithmic relationships, Varying w exerted minimal effects. Conclusion: We thereby characterise the properties of, and through comparing atrial, ventricular and purkinje recovery times with interspecies in vivo background cardiac cycle duration data, (blue whale ∼2000, human∼90, Etruscan shrew, ∼40 ms) can determine physical limits to, electrodiffusive contributions to ephaptic recovery.

Abstract:Although continuous mobile ion passage across the plasma membrane is responsible for the generation of the membrane potential, some experiments suggest that the membrane potential can be generated even without the ion passage across the membrane. This potential could be due to the ion adsorption–desorption phenomenon, and it is even possible to derive a potential formula under such a mechanism. This potential formula is somehow identical to the well-known Nernst equation. We investigated this potential formula in depth and found that the profile of "the membrane surface potential versus the logarithm of ion concentration" exhibits the Nernst slope, and this Nernst slope is the key to the identical potential formula to the Nernst equation even for the system without the passage of ions across the membrane. This work may trigger new information about the membrane potential generation mechanism.
