Abstract:Flowering Chinese cabbage is one of the most economically important stalk vegetables. However, the molecular mechanisms underlying bolting, which is directly related to stalk quality and yield, in this species remain unknown. Previously, we examined five key stem development stages in flowering Chinese cabbage. Here, we identified a gene, BcSOC1 (SUPPRESSOR OF OVEREXPRESSION OF CONSTANS1), in flowering Chinese cabbage using transcriptome analysis, whose expression was positively correlated with bolting. Exogenous gibberellin (GA3) and low-temperature treatments significantly upregulated BcSOC1 and promoted early bolting and flowering. Additionally, BcSOC1 overexpression accelerated early flowering and stem elongation in both Arabidopsis and flowering Chinese cabbage, whereas its knockdown dramatically delayed bolting and flowering and inhibited stem elongation in the latter; the inhibition of stem elongation was more notable than delayed flowering. BcSOC1 overexpression also induced cell expansion by upregulating genes encoding cell wall structural proteins, such as BcEXPA11 (cell wall structural proteins and enzymes) and BcXTH3 (xyloglucan endotransglycosidase/hydrolase), upon exogenous GA3 and low-temperature treatments. Moreover, the length of pith cells was correlated with stem height, and BcSOC1 interacted with BcAGL6 (AGAMOUS-LIKE 6) and BcAGL24 (AGAMOUS-LIKE 24). Thus, BcSOC1 plays a vital role in bolting and stem elongation of flowering Chinese cabbage and may play a novel role in regulating stalk development, apart from the conserved function of Arabidopsis SOC1 in flowering alone.

Abstract:Precise flowering timing is critical for the plant life cycle. Here, we examined the molecular mechanisms and regulatory network associated with flowering in Chinese cabbage (Brassica rapa L.) by comparative transcriptome profiling of two Chinese cabbage inbred lines, “4004” (early bolting) and “50” (late bolting). RNA-Seq and quantitative reverse transcription PCR (qPCR) analyses showed that two positive nitric oxide (NO) signaling regulator genes, nitrite reductase (BrNIR) and nitrate reductase (BrNIA), were up-regulated in line “50” with or without vernalization. In agreement with the transcription analysis, the shoots in line “50” had substantially higher nitrogen levels than those in “4004”. Upon vernalization, the flowering repressor gene Circadian 1 (BrCIR1) was significantly up-regulated in line “50”, whereas the flowering enhancer genes named SUPPRESSOR OF OVEREXPRESSION OF CONSTANCE 1 homologs (BrSOC1s) were substantially up-regulated in line “4004”. CRISPR/Cas9-mediated mutagenesis in Chinese cabbage demonstrated that the BrSOC1-1/1-2/1-3 genes were involved in late flowering, and their expression was mutually exclusive with that of the nitrogen signaling genes. Thus, we identified two flowering mechanisms in Chinese cabbage: a reciprocal negative feedback loop between nitrogen signaling genes (BrNIA1 and BrNIR1) and BrSOC1s to control flowering time and positive feedback control of the expression of BrSOC1s.

Analysis of Interactions Between AGL18 Family Members and Flowering Time Integrator Factor SOC1 in Brassica juncea

Abstract:AGAMOUS-LIKE 18 (AGL18) is a key regulatory factor in flowering time control of Brassicajuncea.In order to clarify the regulatory mechanism of AGL18 protein interact with SOC1 in flowering pathways,AGL18-1 gene from reproductive shoots as well as AGL18-2 and AGL18-3 from vegetative shoots in ‘QJ’ germplasm of Brassicajuncea,were cloned respcetively.AGL18-1,AGL18-2 and A GL18-3 were members of AGL 18 family and respectively encoded 257,257 and 258 amino acids.Sequences analysis showed that they were more than 90％ similar to Brassica rapa and Brassica napus.Both Yeast two hybrid and BiFC showed that SOC1 protein could not interact with AGL18-1,AGL18-2 or AGL18-3.Yeast one hybrid and Dual-Glo(R) Luciferase assays indicated that SOC1 promoter could interact with AGL 18-1 but not AGL 18-2 and AGL 18-3.To screen the interaction regions of AGL 18-1/SOC1,the M-domain and IKC-domain truncations of AGL18-1 protein were separated and found that only the M-domain truncation could interact with SOC1 promoter.It suggested that M-domain was the key region that mediated the interaction between AGL18-1 and SOC1 promoter.The work provided valuable information for in-depth studies on the molecular mechanisms of AGL18 protein with SOC1 and the flowering time in Brassicajuncea.
Orphan gene BR2 positively regulates bolting resistance through the vernalization pathway in Chinese cabbage

Abstract:Orphan genes (OGs) are unique to the specific species or lineage, and whose homologous sequences cannot be found in other species or lineages. Furthermore, these genes lack recognizable domains or functional motifs, which make their characterization difficult. Here, we identified a Brassica rapa OG named BOLTING RESISTANCE 2 (BR2) that could positively modulate bolting resistance. The expression of BR2 was developmentally regulated and the BR2 protein was localized to the cell membrane. BR2 overexpression not only markedly delayed flowering time in Arabidopsis transgenic plants, but substantially affected the development of leaves and flower organs. Flowering repressor AtFLC gene was significantly up-regulated transcribed in Arabidopsis BR2 overexpression lines, while AtFT and AtSOC1 expression was decreased. In addition, the BR2 expression was enhanced in bolting-resistant type Chinese cabbage and was reduced in non-resistant type. Moreover, chilling stress inhibited the BR2 expression levels. Overexpression of BR2 also delayed flowering time in Chinese cabbage. In vernalized Chinese cabbage BR2 overexpression plants, BrVIN3.b and BrFRI were significantly down-regulated, while BrFLC5 was substantially up-regulated. Key floral factors, including three BrSOC1s, two BrLFYs, and four BrFTs were down-regulated. The expression changes of these key genes were consistent with the delayed flowering phenotype of Chinese cabbage BR2 overexpressing plants. Thus, we predicted that BR2 may predominantly function via the vernalization pathway. Our findings propose that the OG BR2 acts as a novel modulator of flowering time in Chinese cabbage, which provides a new insight on the breeding of varieties that are resistant to bolting.
Protein interactions of SOC1 with SVP are regulated by a few crucial amino acids in flowering pathways of Brassica juncea

Abstract:SUPPRESSOR OF OVEREXPRESSION OF CONSTANS 1 (SOC1) is a vital flowering signal integrator to promote flowering, which is inhibited by a MADS-box transcription factor, SHORT VEGETATIVE PHASE (SVP). However, it remains elusive about how SOC1 interacts with SVP in flowering pathways of Brassica juncea. Here, B. juncea SOC1 (BjuSOC1) gene was cloned and it expressed differently between stem apexes and leaves during the low-temperature vernalization and long-day photoperiod pathways. Yeast two-hybrid and BiFC assays indicated that BjuSOC1 directly interacted with BjuSVP in vitro and in vivo. Interestingly, further studies indicated that mutants of BjuSOC1K108V, BjuSOC1R109L, BjuSOC1C137K could no more interact with BjuSVP, and BjuSVPR137L also led to loss of the protein interaction. It suggested that the 108th, 109th, and 137th of BjuSOC1 and 137th of BjuSVP regulated the protein interactions between BjuSOC1 and BjuSVP. The results provided valuable information for further study on the control of flowering time in B. juncea.

Abstract:Glucosinolates (GS) are a group of sulfur- and nitrogen-rich plant secondary metabolites that originate from amino acids and exist mainly in plants in the order Brassicales, such as Arabidopsis thaliana (Arabidopsis) and Chinese cabbage (Brassica rapa ssp. pekinensis). To date, several regulatory components responsible for GS biosynthesis have been identified in Arabidopsis. However, the functions of GS biosynthesis regulators in Chinese cabbage have not been clarified. In our current study, a putative ATR1/MYB34 orthologous gene, BrMYB34.2, was isolated from Chinese cabbage leaves. To investigate the function of this gene, we engineered Arabidopsis plants that overexpress BrMYB34.2 ectopically and phenotypic analysis was performed. Moreover, we assayed the accumulation levels of indolic GS (IGS) and aliphatic glucosinolates in transgenic plants and test the expression of key genes of IGS biosynthesis and tryptophan synthesis by Real-time quantitative PCR. And further analysed the resistance of transgenic plants in 5MT stress treatment. The results indicate that ectopic expression of the BrMYB34.2 gene in Arabidopsis was able to up-regulate the accumulation level of IGS due to the increased expression of IGS and Trp biosynthetic genes. Moreover, overexpression of BrMYB34.2 conferred Arabidopsis 5MT resistance. These results suggest that the BrMYB34.2 gene may function as one of the regulators of IGS and Trp biosynthesis in Chinese cabbage.
Isolation and Characteristic Analysis of a Novel Bhlh Gene,BcBHLHogu, in Chinese Cabbage

Abstract:【Objective】This study was carried out in order to better understand the mechanism of nuclear-cytoplasmic interactions in OguCMS Chinese cabbage pak-choi (Brassica campestris L. ssp. chinensis Makino var. communis Tsen et Lee cv. Aijiaohuang).【Method】Expression profiling was performed using cDNA-AFLP technology on floral buds of OguCMS lines and maintainers in Chinese cabbage pak-choi. A differentially expressed cDNA fragment was obtained,which was unexpressed in early-stage buds of maintainers, but expressed steably in OguCMS floral buds. The full cDNA, designated as BcBHLHogu (GenBank accession numbers: EF127860), was obtained by RACE. 【Result】 It was found that BcBHLHogu coded 122 amino acids, contained a bHLH domain, and shared high homology with AtBHLH060. Phylogenetic analysis indicated that BcBHLHogu and AtBHLHs060/048 were orthologs genes. Its function is similar to BEEs1/2/3, which are three redundant brassinostreriod early response genes.【Conclusion】 It is speculated that BcBHLHogu is a novel bHLH transcription factor in Chinese cabbage, and it appears to be involved in developmental signaling events for floral processes and has a function related to brassinosteriods signaling response.
Cloning and functional analysis of the flowering gene GmSOC1-like, a putative SUPPRESSOR OF OVEREXPRESSION CO1 / AGAMOUS - LIKE 20 ( SOC1 / AGL20 ) ortholog in soybean

Abstract:Key message The major insight in this manuscript is that we identified a new flowering regulator, GmSOC1-like, which may participate in the initiation and maintenance of flowering in soybean. Abstract Flowering is pivotal for the reproductive behavior of plants, and it is regulated by complex and coordinated genetic networks that are fine-tuned by endogenous cues and environmental signals. To better understand the molecular basis of flowering regulation in soybean, we isolated GmSOC1 and GmSOC1 - like , two putative soybean orthologs for the Arabidopsis SUPPRESSOR OF OVEREXPRESSION OF CO1/AGAMOUS - LIKE 20 ( SOC1 / AGL20 ). The expression pattern of GmSOC1 - like was analyzed by qRT-PCR in Zigongdongdou , a photoperiod-sensitive soybean cultivar. GmSOC1 - like was widely expressed at different levels in most organs of the soybean, with the highest expression in the shoot apex during the early stage of floral transition. In addition, its expression showed a circadian rhythm pattern, with the highest expression at midnight under short-day (SD) condition. Intriguingly, GmSOC1 - like was induced 4 days earlier than GmSOC1 during flowering transition in SD, suggesting that GmSOC1 and GmSOC1 - like expression might be differentially regulated. However, under long-day (LD) condition, the expression of GmSOC1 and GmSOC1 - like decreased gradually in the shoot apex of Zigongdongdou , which is in accordance with the fact that Zigongdongdou maintains vegetative growth in LD. In addition, overexpression of GmSOC1 - like stimulated the flowering of Lotus corniculatus cv. supperroot plants. In conclusion, the results of this study indicate that GmSOC1 - like may act as a flowering inducer in soybean.

Abstract:Many MCM1-AGAMOUS-DEFICIENS-SRF (MADS) genes have been proved to play an important role in the flowering time regulation of plants. The flowering-inhibiting factor AGAMOUS-LIKE 18 (AGL18) integrates into the two flowering-activating factors SUPPRESSOR OF OVEREXPRESSION OF CONSTANS 1 (SOC1) and AGAMOUS-LIKE 24 (AGL24), which play an important role during the plant developmental stages of the flowering pathway. However, it remains unknown whether and how the AGL18 protein directly interacts with SOC1 and/or AGL24 genes to regulate flowering time in Brassica juncea. In this study, three members (AGL18-1 in florescence, AGL18-2 and AGL18-3 in young seedlings) of the AGL18 family, and SOC1 and AGL24 in florescence were cloned in Brassica juncea. Yeast One-Hybrid assays and Dual-Glo® Luciferase assays showed that the SOC1 and AGL24 promoters interacted only with AGL18-1 protein, not AGL18-2 and AGL18-3. The typical conserved structure of the M-domain of AGL18-1 was the key region that mediated the interaction between the AGL18-1 protein and SOC1 promoter, and the I-domain, K-domain and C-domain did not regulate the interaction of AGL18-1/SOC1. In contrast, the K-domain and M-domain in AGL18-1 could mediate the interaction between the AGL18-1 protein and AGL24 promoter. This indicated that the AGL18-1 protein must have its unique functions that differed from AGL18-2 and AGL18-3. This work provides valuable information for in-depth studies into the molecular mechanisms of the AGL18 protein with SOC1 and AGL24 for flowering time control of Brassica juncea.
Genome-Wide Identification of BrassinaZole Resistant ( BZR ) Gene Family in Brassica rapa ssp. pekinensis and Its Expression Pattern During Flowering Process

Abstract:The BrassinaZole Resistant (BZR) gene family mediates the signal transduction of brassinolide, which is vital in regulating vegetative and reproductive growth of plants. In this research, a comprehensive analysis of all candidate BrBZR genes in Chinese cabbage ( Brassica rapa ssp. pekinensis ) was conducted using bioinformatics methods, including protein physicochemical properties, evolutionary relationship, gene structure, and conserved domains. Additionally, these candidate genes’ expression patterns during flowering development were analyzed utilizing transcriptome data, and key genes that regulate flowering of Chinese cabbage were screened. In the results, 14 BrBZR genes were identified, with coding sequence (CDS) lengths of 460–1162 bp, and encoding proteins of 152–386 amino acids. The majority of BrBZRs had a highly conserved motif 1 as well as numerous regulatory elements associated with hormone and abiotic stress responses. Transcriptome sequencing showed that 12 BrBZRs were expressed during the flowering development, among which BrBES1.1 and BrBES1.2 were highly expressed, with FPKM ranges of 61.78–99.91 and 43.71–76.69, respectively. Their expression was upregulated during vernalization and flower bud differentiation, suggesting that they may play a key role in regulating the flowering process of Chinese cabbage. Overall, our study provides comprehensive insights into the BrBZR gene family, and has important implications for the further elucidation of the mechanisms that regulate flower development in Chinese cabbage and potentially other related Brassica species.

Abstract:SOC1, a MADS-box type II transcription factor, integrates environmental and endogenous cues to promote flowering in angiosperms. Recent reports implicating SOC1 in roles beyond floral transition prompted functional characterization of SOC1 in polyploid rapeseed mustard genomes. Gene characterization in Brassicas necessitates analysis of composite homeolog function. While insertional mutagenesis is untenable in Brassicas owing to gene redundancy, gain-of-function approach entails serial characterization of individual homeologs. Herein, we demonstrate modulated floral promotive effects in natural variants of Brassica SOC1 and provide lateral branching as a probable outcome of polyploidy-induced gene diversification. Ectopic expression of two B genome specific SOC1 variants in Arabidopsis thaliana resulted in differential floral acceleration and manifestation of multiple vegetative rosettes. Characterization of composite homeolog function in B. juncea via introgression of Brassica SOC1 specific artificial miRNA, designed to target homeologs, also exhibited modifications in floral transition and lateral branching. Comprehensive analysis of field performance of B. juncea transgenics displayed altered fitness across 11 agronomic traits. Crucially, reduced SOC1 levels directly impacted two developmental traits, namely, flowering time and number of lateral branches which in turn influenced several dependent agronomic traits. While delayed flowering and crop maturity resulted in altered fatty acid composition with higher SFA and lower PUFA in transgenics relative to controls, reduction in overall count of lateral branches caused a concomitant decrease in silique count which ultimately impacted total seed yield in transgenics. Statistical analysis revealed number of secondary branches as the most critical trait influencing seed yield. Based on our findings, we propose enhancing levels Brassica SOC1, a key target, for achieving earliness in flowering, improved seed yield and oil quality, and studying trait trade-offs.
Genome-Wide Identification and Analysis of SRO Gene Family in Chinese Cabbage (Brassica rapa L.)

Abstract:Similar to radical-induced cell death 1 (SROs) is a family of small proteins unique to plants. SRO transcription factors play an important role in plants' response to biotic and abiotic stresses. In this study, we identified 12 BrSRO genes in Chinese cabbage (Brassica rapa L.). Among them, a comprehensive overview of the SRO gene family is presented, including physical and chemical characteristics, chromosome locations, phylogenetic analysis, gene structures, motif analysis, and cis-element analyses. The number of amino acids of BrSRO genes is between 77-779 aa, isoelectric point changed from 6.02 to 9.6. Of the 12 BrSRO genes, 11 were randomly distributed along the 7 chromosomes, while BrSRO12 was located along unassigned scaffolds. Phylogenetic analysis indicated that the SRO proteins from six species, including Arabidopsis, banana, rice, Solanum lycopersicum, Zea mays, and Chinese cabbage were divided into eleven groups. The exon-rich BrSRO6 and BrSRO12 containing 15 exons were clustered to group K. All 12 genes have motif 2, which indicate that motif 2 is a relatively conservative motif. There are many hormone and stress response elements in BrSRO genes. The relative expression levels of 12 BrSRO genes under high temperature, drought, salt, and low temperature conditions were analyzed by real-time fluorescence quantitative PCR. The results indicated the relative expression level of BrSRO8 was significantly up-regulated when plants were exposed to high temperature. The relative expression levels of BrSRO1, 3, 7, 8, and 9 were higher under low temperature treatment. The up-regulated genes response to drought and salt stresses were BrSRO1, 5, 9 and BrSRO1, 8, respectively. These results indicated that these genes have certain responses to different abiotic stresses. This work has provided a foundation for further functional analyses of SRO genes in Chinese cabbage.
[Brassica juncea BjuWRKY71-1 accelerates flowering by regulating the expression of SOC1]

Abstract:Brassica juncea (mustard) is a vegetable crop of Brassica, which is widely planted in China. The yield and quality of stem mustard are greatly influenced by the transition from vegetative growth to reproductive growth, i.e., flowering. The WRKY transcription factor family is ubiquitous in higher plants, and its members are involved in the regulation of many growth and development processes, including biological/abiotic stress responses and flowering regulation. WRKY71 is an important member of the WRKY family. However, its function and mechanism in mustard have not been reported. In this study, the BjuWRKY71-1 gene was cloned from B. juncea. Bioinformatics analysis and phylogenetic tree analysis showed that the protein encoded by BjuWRKY71-1 has a conserved WRKY domain, belonging to class Ⅱ WRKY protein, which is closely related to BraWRKY71-1 in Brassica rapa. The expression abundance of BjuWRKY71-1 in leaves and flowers was significantly higher than that in roots and stems, and the expression level increased gradually along with plant development. The result of subcellular localization showed that BjuWRKY71-1 protein was located in nucleus. The flowering time of overexpressing BjuWRKY71-1 Arabidopsis plants was significantly earlier than that of the wild type. Yeast two-hybrid assay and dual-luciferase reporter assay showed that BjuWRKY71-1 interacted with the promoter of the flowering integrator BjuSOC1 and promoted the expression of its downstream genes. In conclusion, BjuWRKY71-1 protein can directly target BjuSOC1 to promote plant flowering. This discovery may facilitate further clarifying the molecular mechanism of BjuWRKY71-1 in flowering time control, and creating new germplasm with bolting and flowering tolerance in mustard.
The Protein J3 Regulates Flowering Through Directly Interacting with the Promoter of SOC1 in Brassica Juncea

Abstract:DNA J HOMOLOG 3 (J3) is a special transcriptional regulator in flowering time control, but the molecular mechanism of J3 in regulating flowering time has not been thoroughly revealed in B. juncea which is one important oilseed and vegetable crop. In this study, J3 gene was cloned from B. juncea (BjuJ3). Phylogenetic relationship analysis showed that the BjuJ3 had high amino acid sequence similarity (>93%) with other Brassica plants. The BjuJ3-transgenic tobacco plants exhibited early flowering, suggesting that BjuJ3 was an activator of flowering time. The qRT-PCR analysis found that BjuJ3 could be ubiquitously induced by the long-day and vernalization treatments in all the tissues of B. juncea. Yeast two-hybrid assays and GST pull-down experiments revealed that BjuJ3 could not directly interact with BjuSOC1, BjuSVP and BjuAGL24. Whereas, yeast one-hybrid and Dual-Glo® Luciferase assays found that BjuJ3 could not interact with BjuAGL24 promoter but could specifically bind to BjuSOC1-1 which is one of truncated fragments of BjuSOC1 promoter. Our research will provide valuable information for unraveling regulatory mechanisms of flowering time in B. juncea.
Interactions of Flowering Promoting Factor AGL19 with Integrator Factors AGL24 and SOC1 in Brassica Oleracea Var.italica

Abstract:In order to clarify the protein interaction mechanisms of flowering promoting factor AGL19 with another two floral integrator factors (AGL24 and SOC1) in flowering pathways,AGL19,AGL24 and SOC1 genes were cloned from Brassica oleracea var.italica,and encoded 221,221 and 214 amino acids,respectively.AGL19,AGL24 and SOC1 were MIKC-type proteins and similar to Brassica oleracea,B.rapa and Chinese cabbage.AGL19 and SOC1 were members of TM3/SOC1 subfamily.Yeast two-hybrid assays showed that the fused strains could grew with blue colonies on QDO/X/A plates,indicating that AGL 19 interact with AGL24 and activated the expression of the reporter genes A UR1-C,HIS3,ADE2 and MEL1.However,AGL19 could not interact with SOC1.These results suggested that AGL24 not SOC 1 was the direct target protein ofAGL 19.Additionally,SOC1 also could interact with the AGL24,suggesting that AGL 19 can indirectly interact with SOC 1 through AGL24.
Inheritance and Genetic Mapping of Late-Bolting to Early-Bolting Gene, BrEb-1, in Chinese Cabbage (brassica Rapa L.)

Abstract:Chinese cabbage (Brassica rapa L.) is one of the most important and highly nutritious vegetables in China belonging to the Brassicaceae family. Flowering or bolting is one of the most critical developmental stages in flowering plants. For the spring-sown Chinese cabbage, late-bolting is desirable over early-bolting according to consumer preferences. We determined the inheritance pattern of the late-bolting trait using F-1 and F-2 generated from a cross between 'SY2004' (late-bolting) and 'CX14-1' (early-bolting). The genetic analysis revealed that the late-bolting to early-bolting trait was controlled by an incomplete dominant gene that we named BrLb-1. Furthermore, we performed bulked segregant analysis (BSA) via whole genome re-sequencing and the results showed that this gene was harbored on the chromosome A07 at the intersections of 20,070,000 to 25,290,000 bp and 20,330,000 to 25,220,000, an interval distance of 4.89 Mb. In this candidate interval, totals of 2321 and 1526 SNPs with non-synonymous mutations, and 229 and 131 InDels with frameshift mutations, were found between the parents and the bulked pools, respectively. Furthermore, we identified three putative candidate genes for the late-bolting trait, including BraA07g029500, BraA07g029530 and BraA07g030360, which code for the AGAMOUS-like MADS-box protein AGL12, a pentatricopeptide repeat-containing protein and NAC transcription factor 29, respectively; however, further functional analysis is required. These genetic variants could be utilized for the further development of molecular markers for marker-assisted breeding in Chinese cabbage.
Brassica rapa orphan gene BR1 delays flowering time in Arabidopsis

Abstract:Orphan genes are essential to the emergence of species-specific traits and the process of evolution, lacking sequence similarity to any other identified genes. As they lack recognizable domains or functional motifs, however, efforts to characterize these orphan genes are often difficult. Flowering is a key trait in Brassica rapa , as premature bolting can have a pronounced adverse impact on plant quality and yield. Bolting resistance-related orphan genes, however, have yet to be characterized. In this study, an orphan gene designated BOLTING RESISTANCE 1 ( BR1 ) was identified and found through gene structural variation analyses to be more highly conserved in Chinese cabbage than in other available accessions. The expression of BR1 was increased in bolting resistant Chinese cabbage and decreased in bolting non-resistant type, and the expression of some mark genes were consist with bolting resistance phenotype. BR1 is primarily expressed in leaves at the vegetative growth stage, and the highest BR1 expression levels during the flowering stage were observed in the flower buds and silique as compared to other tissue types. The overexpression of BR1 in Arabidopsis was associated with enhanced bolting resistance under long day (LD) conditions, with these transgenic plants exhibiting significant decreases in stem height, rosette radius, and chlorophyll content. Transcriptomic sequencing of WT and BR1 OE plants showed the association of BR1 with other bolting resistance genes. Transcriptomic sequencing and qPCR revealed that six flowering integrator genes and one chlorophyll biosynthesis-related gene were downregulated following BR1 overexpression. Six key genes in photoperiodic flowering pathway exhibited downward expression trends in BR1 OE plants, while the expression of floral repressor AtFLC gene was upregulated. The transcripts of these key genes were consistent with observed phenotypes in BR1 OE plants, and the results indicated that BR1 may function through vernalization and photoperiodic pathway. Instead, the protein encoded by BR1 gene was subsequently found to localize to the nucleus. Taken together, we first propose that orphan gene BR1 functions as a novel regulator of flowering time, and these results suggested that BR1 may represent a promising candidate gene to support the selective breeding of Chinese cabbage cultivars with enhanced bolting resistance.

Abstract:Flowering time is an important agronomic trait in cabbage (Brassica oleracea L. var. capitata), but the molecular regulatory mechanism underlying flowering time regulation in cabbage remains unclear. In this study, transcriptome analysis was performed using two sets of cabbage materials: (1) the early-flowering inbred line C491 (P1) and late-flowering inbred line B602 (P2), (2) the early-flowering individuals F2-B and late-flowering individuals F2-NB from the F2 population. The analysis revealed 9508 differentially expressed genes (DEGs) common to both C491_VS_ B602 and F2-B_VS_F2-NB. The Kyoto Encyclopedia of Genes and Genomes (KEGGs) analysis showed that plant hormone signal transduction and the MAPK signaling pathway were mainly enriched in up-regulated genes, and ribosome and DNA replication were mainly enriched in down-regulated genes. We identified 321 homologues of Arabidopsis flowering time genes (Ft) in cabbage. Among them, 25 DEGs (11 up-regulated and 14 down-regulated genes) were detected in the two comparison groups, and 12 gene expression patterns closely corresponded with the different flowering times in the two sets of materials. Two genes encoding MADS-box proteins, Bo1g157450 (BoSEP2-1) and Bo5g152700 (BoSEP2-2), showed significantly reduced expression in the late-flowering parent B602 compared with the early-flowering parent C491 via qRT-PCR analysis, which was consistent with the RNA-seq data. Next, the expression levels of Bo1g157450 (BoSEP2-1) and Bo5g152700 (BoSEP2-2) were analyzed in two other groups of early-flowering and late-flowering inbred lines, which showed that their expression patterns were consistent with those in the parents. Sequence analysis revealed that three and one SNPs between B602 and C491 were identified in Bo1g157450 (BoSEP2-1) and Bo5g152700 (BoSEP2-2), respectively. Therefore, BoSEP2-1 and BoSEP2-2 were designated as candidates for flowering time regulation through a potential new regulatory pathway. These results provide new insights into the molecular mechanisms underlying flowering time regulation in cabbage.

Abstract:Heading is one of the most important agronomic traits for Chinese cabbage crops. During the heading stage, leaf axial growth is an essential process. In the past, most genes predicted to be involved in the heading process have been based on leaf development studies in Arabidopsis. No genes that control leaf axial growth have been mapped and cloned via forward genetics in Chinese cabbage. In this study, we characterize the inward curling mutant ic1 in Brassica rapa ssp. pekinensis and identify a mutation in the OCTOPUS (BrOPS) gene by map-based cloning. OPS is involved in phloem differentiation in Arabidopsis, a functionalization of regulating leaf curvature that is differentiated in Chinese cabbage. In the presence of brassinosteroid (BR) at the early heading stage in ic1, the mutation of BrOPS fails to sequester brassinosteroid insensitive 2 (BrBIN2) from the nucleus, allowing BrBIN2 to phosphorylate and inactivate BrBES1, which in turn relieves the repression of BrAS1 and results in leaf inward curving. Taken together, the results of our findings indicate that BrOPS positively regulates BR signaling by antagonizing BrBIN2 to promote leaf epinastic growth at the early heading stage in Chinese cabbage.

Abstract:MADS-box protein SUPPRESSOR OF OVEREXPRESSION OF CONSTANS1 (SOC1) plays an important role in the regulation of flowering by integrating multiple flowering signals in Arabidopsis thaliana. SOC1-like genes have been isolated from many plants and their functions received wide attention. To investigate SOC1 homologs in bamboo, a SOC1 homolog, referred to as PvSOC1, was isolated from Phyllostachys violascens, and characterized. PvSOC1 encoded a protein of 226 amino acids containing a highly conserved SOC1 motif. PvSOC1 transcript was expressed in all examined tissues but preferentially in leaves. Ectopic expression of PvSOC1 in both Arabidopsis and rice caused early flowering, abnormal floral organs and leaves. A P-box, the gibberellin response element existed in the promoter region of PvSOC1 but not in that of SOC1 and OsSOC1. Upon gibberellic acid treatment, PvSOC1 transcript was upregulated in the leaves of Ph. violascens seedlings, suggesting a positive response to gibberellin acid. Thus, our experiments suggest PvSOC1 to be a multifunctional gene, which not only regulates flowering, but also affects leaf development.
