Abstract:The Peutz-Jeghers syndrome (PJS) is an autosomal dominant hamartomatous poliposis describred in 1921. Hemminki in 1997 described the presence of LKB-1 mutation tumor-suppressor gen.The patients with PJS develop a higher cumulative incidence of gastrointestinal, pancreas and extraintestinal tumors, being occasion of a renew interest on hamartomatous polyposis syndromes regarding the clinical care, cancer surveillance treatment and long term follow-up.We report the case of a 38 years old male, diagnosed of PJS who developed a multiple adenocarcinoma in duodenum and yeyunum. Surgically treated and with a long-term free disease survival of 11 years represents the sixth case reported in the spanish literature of PJS associated with a gastrointestinal tumor.A critical review, molecular alterations and the established criteria of tumor screening and surveillance are reviewed.

Abstract:Five isolates of Bursaphelenchus xylophilus, four of which were collected from Chinese forests and one from Japan, as well as, four isolates of B. mucronatus extracted from pinewood packaging imported from Japan, Korea and from forests in China, were studied using PCR-RFLPs and sequences of the ITS and D2-D3 fragments of rDNA. RFLP analysis of the ITS region with restriction endonucleases revealed that CfoI, HaeIII, HinfI, MspI and TaqI yielded distinct patterns that separated A xylophilus and A mucronatus. Phylogenctic analyses of alignments of published and original sequences of both the ITS I region and ITS1-5.8S-ITS2 region revealed that both A xylophilus and A mucronatus are composed of two groups of populations with different ITS types. The taxonomic status of these groups is discussed.

Abstract:ABSTRACT Dramatic alteration of surface lipoprotein profiles is a key strategy that Borrelia burgdorferi , the Lyme disease pathogen, has evolved for adapting to the diverse environments of arthropod and mammalian hosts. Several of these differentially expressed lipoproteins have been shown to play important roles in the enzootic cycle of B. burgdorferi . The BBA05 protein is a previously identified putative lipoprotein (P55 or S1 antigen) that elicits antibody responses in mammals. Recent microarray analyses indicate that the BBA05 gene is differentially expressed by many environmental factors, including temperature. However, the role of the BBA05 protein in the life cycle of B. burgdorferi has not been elucidated. Here we show that expression of the BBA05 gene was exclusively induced in feeding nymphal ticks during the spirochetal transmission from ticks to mammals. Upon generating a BBA05 mutant in an infectious strain of B. burgdorferi , we showed that the BBA05 mutant remained capable of establishing infection in mice, being acquired by ticks, persisting through tick molting, and reinfecting new mammalian hosts. These results indicate that, despite being a highly conserved and regulated antigen, the BBA05 protein has a nonessential role in the transmission cycle of B. burgdorferi , at least in the animal model.

Abstract:Six populations of genus Bursaphelenchus isolated from pine wood produced in China, Japan and Korea were identified with the aid of PCR-RFLP of rDNA. It was bound that one of the populations, which had its origin in the forests of Zhenhai, Zhejiang Province, China, and the other, which was derived from imported coniferous wood packages produced in Japan were both identified as B. xylophilus. The other four populations, including populations from Fuyang of the Zhejiang Province, Korea, Hongkong and Japan, were identified as B.mucronatus. All the four restriction enzymes, namely HinfⅠ, HaeⅢ, MspⅠ, and CfoⅠ, with their special sites of cutting could be used to differentiate the two morphologically very similar species.

Abstract:Borrelia burgdorferi, the Lyme disease pathogen, dramatically alters its protein profile when it is transmitted between ticks and mammals. Several differentially expressed proteins have been shown to be critical for the enzootic cycle of B. burgdorferi. In this study, we demonstrated that expression of the surface lipoprotein-encoding gene bba07 is induced by an elevated temperature and a reduced pH during in vitro cultivation, as well as during nymphal tick feeding. Expression of bba07 is regulated by the Rrp2-RpoN-RpoS pathway, a central regulatory network that is activated during nymphal feeding. By generating a bba07 mutant of an infectious strain of B. burgdorferi, we demonstrated that although BBA07-deficient spirochetes were capable of infecting mice via needle inoculation and surviving in ticks, they were defective in infection of mammals via tick transmission. Complementation of the bba07 mutant with a wild-type copy of bba07 partially restored the transmission defect of the bba07 mutant. Based on these findings, we concluded that the surface lipoprotein BBA07 is produced during tick feeding and facilitates optimal transmission of B. burgdorferi from the tick vector to a mammalian host.

Abstract:Japanese meagre (Argyrosomus japonicus) is well-known for its high nutritional and economic value. In recent years, with the rapid expansion and intensification of aquaculture, Japanese meagre culture has suffered serious economic losses due to infectious diseases. Toll-like receptors (TLRs) can recognize highly conserved structural motif pathogen-associated microbial patterns (PAMPs) and danger-associated molecular patterns (DAMPs), playing a key role in resisting diseases by initiating host innate immune responses. Here, a total of 12 members of the TLR gene family (TLR1, TLR2, TLR2b, TLR3, TLR5M, TLR5S, TLR7, TLR9, TLR13, TLR14, TLR22, and TLR23) were identified in the genome of Japanese meagre. The TLR genes in Japanese meagre were distributed on 10 of 24 chromosomes. The evolutionary analyses showed that 12 different TLR genes were divided into different subfamilies, including the TLR1, TLR3, TLR5, TLR7, and TLR11 subfamilies. The conserved motifs were compared with three other teleost species. The results revealed that the conserved motifs in the same subfamilies were highly consistent. The identity and similarity of the TLR-TIR domain of Japanese meagre demonstrated that the regulatory mechanisms of each of the subfamilies were similar. qRT-PCR assays showed different expression patterns of TLRs in the liver, spleen, head kidney, trunk kidney, brain, and heart of Japanese meagre, but the expression levels in immune tissues were higher. Double-stranded RNA virus (dsRNA) is a common etiological agent of diseases in aquaculture species. Therefore, a synthetic double-stranded RNA called poly (I:C) was used as an injection to understand the immune system function and disease control of Japanese meagre. Following poly (I:C) stimulation, seven of twelve ArgTLR genes, including ArgTLR2, ArgTLR2b, ArgTLR3, ArgTLR7, ArgTLR13, ArgTLR22 and ArgTLR23, were significantly upregulated. In summary, our results not only play a guiding role in disease prevention and control and vaccine development in the future, but also provide basic data for future study of the TLR gene family of Japanese meagre.

Abstract:Objective To investigate the genotype and hereditary feature of prevalent strains of Rickettsia orientalis (Ro) in certain area.Methods Gene fragment,encoding Sta50 protein in samples of patients and mice infected by Ro,was detected by real-time fluorescent quantitative polymerase chain reaction assay (qPCR).Positive samples were analyzed for genotype and sequence,and the results were compared with nest PCR.The homology was analyzed by comparison between sequencing results and nucleotide sequences of Ro registered in GenBank.Results The 523 bp fragments of Gilliam genotype could be amplified in samples of patients and mice,with homology of 96.3% with Kawasaki strain.The 318 bp fragment of Gilliam genotype could be amplified in samples of mice infected by Ro,with homology of 78.5% with TA686 strain.The homology of nucleotide sequences of Ro in samples from patients and mice was 100.0%.Conclusion Gilliam genotype and Kawasaki quasispecies could be detected among Ro in this area,and TA686 quasispecies could be detected in mice.Natural infection of Ro might exist in human and mice in this area,and natural foci of tsutsugamushi disease have formed.

Abstract:Thirty-two strains of Borrelia burgdorferi sensu lato were isolated from Ixodes persulcatus ticks collected from northeastern China from May to June in 2004 and 2005. Restriction fragment length polymorphism (RFLP) analysis and sequence analysis of 5S-23S rRNA intergenic spacer revealed that 29 (90.6%) belonged to Borrelia garinii, demonstrating B, C, and a unique pattern. The remaining three isolates (9.4%) were Borrelia afzelii with pattern D. The phylogenetic analysis based on 5S-23S rRNA intergenic spacer showed that B. garinii and B. afzelii genospecies clustered into two separate lineages. B. garinii strains were classified into three different branches: All the strains with RFLP pattern C were in the same branch, strain VH10 with a unique RFLP pattern clustered with strains VH9 and MDH2 with pattern B, and the rest of the strains with pattern B constitute another branch. These findings demonstrate the genetic diversity of B. burgdorferi sensu lato isolates from northeastern China.
The first molecular record of Borrelia afzelii, B. garinii, B. valaisiana, B. burgdorferi s.s. and B. bavariensis in Bosnia and Herzegovina

Abstract:Borrelia burgdorferi sensu lato complex comprises 20 species, from which B. afzelii , B. garinii , B. burgdorferi sensu stricto, B. bavariensis , and B. spielmanii are directly associated with Lyme borreliosis, while B. bissettiae , B. lusitaniae , and B. valaisiana were detected in individual cases. Their main vector in Europe is the hard tick species Ixodes ricinus . To date, two species, B. spielmanii and B. lusitaniae , have been molecularly detected in Bosnia and Herzegovina. To test for the presence of other Borrelia species, we performed nested PCR targeting intergenic region rrf (5S) – rrl (23S) on DNA isolates from 49 ticks collected from vegetation by flagging method and 43 removed from humans in The Center for Emergency Medical Assistance of the Sarajevo Canton and The Healthcare Centers of the Sarajevo Canton. Borrelia species were detected by one-directional Sanger sequencing of the amplified region using the same forward primer as in PCR. Out of six Borrelia species detected in the present study, this is the first record of B. afzelii , B. garinii , B. burgdorferi s.s . B. bavariensis , and B. valaisiana in Bosnia and Herzegovina.

Abstract:Background: The taxonomic status of the relapsing fever spirochete Borrelia hermsii in western North America was established in 1942 and based solely on its specific association with the soft tick vector Ornithodoros hermsi . Multilocus sequence typing (MLST) of the 16S rRNA , flaB , gyrB , glpQ , and 16S-23S rRNA intergenic spacer of B. hermsii isolates collected over many years from various geographic locations and biological sources identified two distinct clades designated previously as B. hermsii Genomic Group I (GGI) and Genomic Group II (GGII). To better assess the taxonomic relationship of these two genomic groups to each other and other species of Borrelia , DNA sequences of the entire linear chromosome were determined. Materials and Methods: Genomic DNA samples were prepared from 11 spirochete isolates grown in Barbour-Stoenner-Kelly-H medium. From these preparations, DNA sequences of the entire linear chromosome of two isolates of B. hermsii belonging to each genomic group and seven additional species were determined. Results: Chromosomal sequences of four isolates of B. hermsii contained 919,212 to 922,307 base pairs. DNA sequence identities between the two genomic groups of B. hermsii were 95.86–95.99%, which were more divergent than chromosomal sequences comparing Borrelia parkeri and Borrelia turicatae (97.13%), Borrelia recurrentis and Borrelia duttonii (97.07%), and Borrelia crocidurae and B. duttonii (97.09%). The 3′ end of the chromosome of the two GGII isolates also contained a unique intact oppA gene absent from all other species examined. Conclusion: Previous MLST and the chromosomal sequences presented herein support the division of the B. hermsii species complex into two species, B. hermsii sensu stricto ( = GGI) and Borrelia nietonii sp. nov. ( = GGII). We name this unique relapsing fever spirochete in honor of our late friend and colleague Dr. Nathan Nieto for his outstanding contributions to our understanding of tick-borne relapsing fever.

Abstract:Utilizing whole genome sequencing data for 141 Eurasian distributed Borrelia isolates, we show that the PFam54 gene array is highly variable among and within Borrelia species, including the absence of genes of known function during infection. Further analysis highlighted though that understudied paralogs most likely are important to completing the Borrelia life cycle, although with a yet unknown function, requiring further research. Taken together, the results display the importance of studying individual variation in Borrelia species from a population genetics perspective to provide novel hypotheses regarding Borrelia or vector‐borne pathogen evolution. Lyme borreliosis (LB) is the most common vector‐borne disease in the Northern Hemisphere caused by spirochetes belonging to the Borrelia burgdorferi sensu lato (Bbsl) complex. Borrelia spirochetes circulate in obligatory transmission cycles between tick vectors and different vertebrate hosts. To successfully complete this complex transmission cycle, Bbsl encodes for an arsenal of proteins including the PFam54 protein family with known, or proposed, influences to reservoir host and/or vector adaptation. Even so, only fragmentary information is available regarding the naturally occurring level of variation in the PFam54 gene array especially in relation to Eurasian‐distributed species. Utilizing whole genome data from isolates (n = 141) originated from three major LB‐causing Borrelia species across Eurasia (B. afzelii, B. bavariensis, and B. garinii), we aimed to characterize the diversity of the PFam54 gene array in these isolates to facilitate understanding the evolution of PFam54 paralogs on an intra‐ and interspecies level. We found an extraordinarily high level of variation in the PFam54 gene array with 39 PFam54 paralogs belonging to 23 orthologous groups including five novel paralogs. Even so, the gene array appears to have remained fairly stable over the evolutionary history of the studied Borrelia species. Interestingly, genes outside Clade IV, which contains genes encoding for proteins associated with Borrelia pathogenesis, more frequently displayed signatures of diversifying selection between clades that differ in hypothesized vector or host species. This could suggest that non‐Clade IV paralogs play a more important role in host and/or vector adaptation than previously expected, which would require future lab‐based studies to validate.

Abstract:The capacity for Borrelia burgdorferi to cause disseminated infection in humans or mice is associated with the genotype of the infecting strain. The cytokine profiles elicited by B. burgdorferi clinical isolates of different genotype (ribosomal spacer type) groups were assessed in a human PBMC co-incubation model. RST1 isolates, which are more frequently associated with disseminated Lyme disease in humans and mice, induced significantly higher levels of IFN-α and IFN-λ1/IL29 relative to RST3 isolates, which are less frequently associated with disseminated infection. No differences in the protein concentrations of IFN-γ, IL-1β, IL-6, IL-8, IL-10 or TNF-α were observed between isolates of differing genotype. The ability of B. burgdorferi to induce type I and type III IFNs was completely dependent on the presence of linear plasmid (lp) 36. An lp36-deficient B. burgdorferi mutant adhered to, and was internalized by, PBMCs and specific dendritic cell (DC) subsets less efficiently than its isogenic B31 parent strain. The association defect with mDC1s and pDCs could be restored by complementation of the mutant with the complete lp36. The RST1 clinical isolates studied were found to contain a 2.5-kB region, located in the distal one-third of lp36, which was not present in any of the RST3 isolates tested. This divergent region of lp36 may encode one or more factors required for optimal spirochetal recognition and the production of type I and type III IFNs by human DCs, thus suggesting a potential role for DCs in the pathogenesis of B. burgdorferi infection.

Abstract:Background Borrelia miyamotoi is a spirochete species transmitted via hard ticks. Following its discovery in Japan, this pathogen has been detected around the world, and is increasingly confirmed as a human pathogen causing febrile disease, namely relapsing fever. Its presence has been confirmed in the northeast China. However, there is little information regarding the presence of B. miyamotoi and other hard-tick-borne relapsing fever spirochetes in southern China including Yunnan Province, where tick and animal species are abundant and many people both inhabit and visit for recreation. Methods For the present study, we collected samples of ticks, wildlife, and domestic animal hosts from different counties in Yunnan Province. Nucleic acids from samples were extracted, and the presence of B. miyamotoi and other relapsing fever spirochetes was confirmed using polymerase chain reaction (PCR) for the 16 s rRNA specific target gene fragment. The positive samples were then amplified for partial genome of the flaB and glpQ genes. Statistical differences in its distribution were analyzed by SPSS 20 software. Results A total of 8260 samples including 2304 ticks, 4120 small mammals and 1836 blood of domestic animal hosts were collected for screening for infection of B. miyamotoi and other relapsing fever spirochetes. Cattle and sheep act as the main hosts and Rhipicephalus microplus, Haemaphysalis nepalensis , H. kolonini and Ixodes ovatus were identified as the important vector host with high prevalence or wide distribution. Only one Mus caroli (mouse) and one Sorex alpinus (shrew) were confirmed positive for relapsing fever spirochetes. Evidence of vertical transmission in ticks was also confirmed. Sequence of partial 16S rRNA, flaB and glpQ genome were analyzed and phylogenetic trees were constructed. Two known strains of B. miyamotoi and one novel relapsing fever spirochetes, B. theileri -like agent, were confirmed and described with their host adaptation, mutation, and potential risk of spreading and spillover for human beings. Conclusions Our results provide new evidence of relapsing fever spirochetes in vector and animal hosts in Yunnan Province based on large sample sizes, and offer guidance on further investigation, surveillance and monitoring of this pathogen.

Abstract:Ticks are one of the most important vectors that can transmit pathogens to animals and human beings. This study investigated the dominant tick-borne bacteria carried by ticks and tick-borne infections in forestry populations in Arxan, Inner Mongolia, China. Ticks were collected by flagging from May 2020 to May 2021, and blood samples were collected from individuals at high risk of acquiring tick-borne diseases from March 2022 to August 2023. The pooled DNA samples of ticks were analyzed to reveal the presence of tick-borne bacteria using high-throughput sequencing of the 16S rDNA V3–V4 region, and species-specific polymerase chain reaction (PCR) related to sequencing was performed to confirm the presence of pathogenic bacteria in individual ticks and human blood samples. All sera samples were examined for anti-SFGR using ELISA and anti- B. burgdorferi using IFA and WB. A total of 295 ticks (282 Ixodes persulcatus and 13 Dermacentor silvarum ) and 245 human blood samples were collected. Rickettsia , Anaplasma , Borrelia miyamotoi , and Coxiella endosymbiont were identified in I. persulcatus by high-throughput sequencing, while Candidatus R. tarasevichiae (89.00%, 89/100), B. garinii (17.00%, 17/100), B. afzelii (7.00%, 7/100), and B. miyamotoi (7.00%, 7/100) were detected in I. persulcatus , as well the dual co-infection with Candidatus R. tarasevichiae and B. garinii were detected in 13.00% (13/100) of I. persulcatus . Of the 245 individuals, B. garinii (4.90%, 12/245), R. slovaca (0.82%, 2/245), and C. burnetii (0.41%, 1/245) were detected by PCR, and the sequences of the target genes of B. garinii detected in humans were identical to those detected in I. persulcatus . The seroprevalence of anti-SFGR and anti- B. burgdorferi was 5.71% and 13.47%, respectively. This study demonstrated that Candidatus R. tarasevichiae and B. garinii were the dominant tick-borne bacteria in I. persulcatus from Arxan, and that dual co-infection with Candidatus R. tarasevichiae and B. garinii was frequent. This is the first time that B. miyamotoi has been identified in ticks from Arxan and R. solvaca has been detected in humans from Inner Mongolia. More importantly, this study demonstrated the transmission of B. garinii from ticks to humans in Arxan, suggesting that long-term monitoring of tick-borne pathogens in ticks and humans is important for the prevention and control of tick-borne diseases.

Abstract:Ixodid ticks were collected from medium-sized to large mammals in Ehime Prefecture, Shikoku, Japan. Ten species of ticks (Amblyomma testudinarium, Dermacentor taiwanensis, Haemaphysalis flava, H. formosensis, H. hystricis, H. longicornis, H. megaspinosa, Ixodes nipponensis, I. ovatus, and I. tanuki) were collected from a total of 29 mammals comprising 11 species. Haemaphysalis hystricis, a possible vector of Japanese spotted fever in Ehime prefecture, was collected from Canis lupus familiaris (domestic dog), Martes melampus melampus, and Sus scrofa leucomystax. This is a first report of H. hystricis from the domesticated dog in the endemic area of Japanese spotted fever. This suggests that it is necessary to pay attention to dogs as a host of the vector ticks for Japanese spotted fever control. Nyctereutes procyonoides and Ma. melampus are new hosts for A. testudinarium. Nyctereutes procyonoides, Mustela itatsi, and Lepus brachyurus are new hosts for H. formosensis. Martes melampus is a new host for H. hystricis.

Abstract:Abstract The ability of guanine-rich sequences to form quadruplex structures in telomeres for example is important in a number of biological processes such as aging, carcinogenesis and gene regulation. Ionizing radiation can cause damage to guanine moieties that can affect the stability or formation of the guanine quadruplex structures. In addition, the mechanisms of formation of these radiation damages in quadruplex structures may be different from those that occur in single- or double-stranded conformations. We have studied the quantitative aspects of the radiation induced formation of 8-hydroxy-2′-guanine base modifications and unaltered guanine base release in single-, double- and four-stranded conformations of polyriboguanylic acid as a model of guanine-rich sequences in telomere-like structures. The results show that the strandedness of guanine-rich sequences is an important variable in the observed yields of these base damages and suggests that telomere-like structures with G-quadruplexes will be relatively more radiosensitive than the other regions of duplex DNA. Hydroxyl radicals are the major reactive species that produce the DNA damage, although the presence of oxygen significantly reduces their radiation yields for all conformations of polyriboguanylic acid and changes the proportions of the yields of the various damages among the polymer conformations.
