Abstract:Powdery mildew is an apple disease caused by the obligate trophic fungus Podosphaera leucotricha. Basic helix–loop–helix (bHLH) transcription factors play important roles in plant development and stress responses, and they have been widely studied in model plants such as Arabidopsis thaliana. However, their role in the stress response of perennial fruit trees remains unclear. Here, we investigated the role of MdbHLH093 in the powdery mildew of apples. The expression of MdbHLH093 was significantly induced during the infection of apples with powdery mildew, and the allogenic overexpression of MdbHLH093 in A. thaliana enhanced the resistance to powdery mildew by increasing the accumulation of hydrogen peroxide (H2O2) and activating the salicylic acid (SA) signaling pathway. The transient overexpression of MdbHLH093 in apple leaves increased the resistance to powdery mildew. Conversely, when MdbHLH093 expression was silenced, the sensitivity of apple leaves to powdery mildew was increased. The physical interaction between MdbHLH093 and MdMYB116 was demonstrated by yeast two-hybrid, bi-molecular fluorescence complementation, and split luciferase experiments. Collectively, these results indicate that MdbHLH093 interacts with MdMYB116 to improve apple resistance to powdery mildew by increasing the accumulation of H2O2 and activating the SA signaling pathway, as well as by providing a new candidate gene for resistance molecular breeding.

What problem does this paper attempt to address?
The problem that this paper attempts to solve is the resistance mechanism of apple powdery mildew. Specifically, the researchers focused on a transcription factor named MdbHLH093 and explored its role in enhancing the resistance of apples to powdery mildew (caused by the fungus Podosphaera leucotricha). By promoting the activation of the salicylic acid (SA) signaling pathway and the accumulation of hydrogen peroxide (\( \text{H}_2\text{O}_2 \)), MdbHLH093 can enhance the resistance of apples to powdery mildew. In addition, the study also found that MdbHLH093 has a physical interaction with another transcription factor MdMYB116, and they are both involved in the defense response of apples to powdery mildew. ### Research Background - **Apple Powdery Mildew**: Apple powdery mildew is caused by the obligate parasitic fungus Podosphaera leucotricha, which seriously affects the growth of apple trees and the quality of fruits. - **bHLH Transcription Factors**: The bHLH (basic Helix - Loop - Helix) transcription factor family plays an important role in plant development and stress responses, but its role in perennial fruit trees is still unclear. ### Research Objectives - **Identify the Function of MdbHLH093**: Through heterologous expression and transient transformation experiments, verify whether MdbHLH093 can enhance the resistance of apples to powdery mildew. - **Reveal the Molecular Mechanism**: Explore how MdbHLH093 improves the disease - resistance ability of apples by activating the SA signaling pathway and increasing the accumulation of \( \text{H}_2\text{O}_2 \). - **Provide Candidate Genes**: Provide candidate genes for breeding apple varieties resistant to powdery mildew, and lay the foundation for further research on the disease - resistance mechanism of apples. ### Main Findings - **Expression Pattern Analysis**: MdbHLH093 is significantly up - regulated after apple leaves are infected with powdery mildew, and is mainly highly expressed in leaves. - **Subcellular Localization**: MdbHLH093 is located in the nucleus. - **Heterologous Expression**: Heterologous expression of MdbHLH093 in Arabidopsis can significantly enhance the resistance to powdery mildew, manifested as a reduced infection rate, a decreased spore number, and an increased H2O2 accumulation. - **Transient Transformation**: Transient over - expression of MdbHLH093 in apple leaves can also significantly improve the resistance to powdery mildew. - **Molecular Mechanism**: MdbHLH093 enhances the resistance of apples to powdery mildew by promoting the expression of genes related to the SA signaling pathway (such as AtEDS1, AtICS1, AtPR2, AtPR5) and increasing the accumulation of H2O2. ### Conclusion MdbHLH093 enhances the resistance of apples to powdery mildew by promoting the SA signaling pathway and H2O2 accumulation. This finding not only provides new insights into understanding the disease - resistance mechanism of apples, but also provides potential candidate genes for breeding apple varieties resistant to powdery mildew.
