Abstract:Background: In both human and murine models, aging is associated with worsening metabolism (e.g.insulin resistance, hyperlipidemia, increased adiposity) and disruption of circadian rhythms.The gut microbiome changes cyclically which is important to maintain healthy metabolism and is required for normal circadian rhythms.Disruption in these cyclical changes leads to dysmetabolism.While differences in the both metabolism and the gut microbiome have been observed with aging, the relationship between cyclical microbiome dynamics and its association with these changes is still poorly understood.We hypothesize that aging causes disruption of microbe-host circadian dynamics which could underlie ageassociated dysmetabolism.Methods: C57BL/6 mice >80 weeks (aged) and >8 weeks (young) were assessed for insulin sensitivity, body composition, feeding behavior, activity, and respiratory exchange ratio.To assess changes in luminal dynamics, stool was collected every 4 hours for 24 hours and DNA was extracted for 16S analysis.Results: Aged mice had reduced insulin sensitivity at 15-and 30-minutes during an insulin tolerance test (p= 0.05 and p=0.003, respectfully).Aged mice also had increased gonadal white adipose tissue (p= 0.0017).These metabolic changes occurred in the setting of reduced food intake and loss of diurnal feeding pattern (i.e.aged mice ate a higher percentage of their food during the daylight period as opposed to nocturnal period).Aged mice also had observed changes in the microbe-host circadian dynamics.Young and aged mice at each observed time point over 24-hours had different microbiomes (p= 0.001, q= 0.001).Along with these compositional differences, altered luminal cycling of the gut microbiome was seen between the groups.Conclusions: Aging alters microbe-host circadian dynamics with shifts in feeding and cyclical change in microbiome composition.These factors may underlie the dysmetabolic phenotype associated with aging, including insulin resistance, hyperlipidemia, and increased adiposity.Restoring luminal dynamics, such as with time-restricted feeding, may be a promising new treatment option for age-related morbidity.
