Abstract:Our aim was to construct the eukaryotic expressional system of single-chain antibody (ScFv(2F9)) derived from a new clone of monoclonal antibody named ZCH-7-2F9 against human CD14 antigen, and to obtain the ScFv(2F9) protein with a high biological activity for further studies on ScFv(2F9) immunotoxin and other kinds of genetically engineered antibodies. Primers were synthesized according to the gene sequence of ScFv(2F9), 4 tandem glysines and 1 serine (Gly4Ser)3 linker and multiple cloning site(MCS) of pSectag2A vector, which included SfiI and EcoRI enzyme cleaving site, 6 x His and stop code TGA sequences. ScFv(2F9) gene was amplified through splicing by overlap extension (SOE) using the high fidelity Taq polymerase. Then the gene was cloned to pGEM-T easy and pSectag2A vectors. Positive recombinants (pSectag2A/ScFv(2F9)) were identified through enzyme cleaving and sequenced before expression. The recombinant plasmids were transfected into Chinese hamster ovary (CHO) cells for expression. The relative molecular weight (MW) and the binding activity of the interesting protein were determined by SDS-PAGE, Western-blot and flow cytometry (FCM), respectively. The scFv(2F9) V-H is a member of the IgH gene family V, subgroup and the scFv(2F9) V-L gene belongs to the Ig(k) gene family V-10 subgroup. The recombined ScFv(2F9) gene was identified to be functional by sequencing and expressing. The interesting protein could be detected in the culture supernatant of transformed CHO cells with a MW of 31 kDa. The blocking test showed that the positive cell percentages, the mean fluorescence intensity (MFI) and the peak of channel (peak Ch) were reduced by 90.02%, 63.3% and 63.38%, respectively after blockage of ScFv(2F9). The ScFv(2F9) gene was cloned and the interest protein against human CD14 antigen has been successfully expressed in eukaryotic cells with a high biological activity, which lays the foundation for further studies on its immunotoxin and other kinds of engineered antibodies.
Physical and epitope analysis of a recombinant human T-cell receptor V alpha/V beta construct support the similarity to immunoglobulin

Abstract:The genetic organization and protein structure of T-cell receptors (TCR) and immunoglobulins (Ig) are remarkably similar. Through recombinant, physical, and peptide-based immunological studies we demonstrated that rabbit antisera generated against a recombinant single-chain TCR (scTCR) react with defined peptide epitopes of their constituent TCR alpha and beta chains. These antisera cross-react with the lambda light-chain Mcg as well as with peptides duplicating its covalent structure. Conversely, rabbit antisera generated to human lambda light chains cross-reacted with the recombinant scTCR. Rabbit anti-lambda antibodies purified on an scTCR affinity column bound to T-cell lines and to T and B lymphocytes from peripheral blood. Circular dichroism analysis demonstrated plots characteristic of beta-sheets for both Mcg and recombinant scTCR. Antisera directed against TCR alpha-chain synthetic peptides reacted with scTCR, Mcg lambda light-chain protein, synthetic peptides from regions of sequence homology in beta-chains, and Mcg. Based upon this homology and the serological cross-reactions which reflect conformational determinants, we suggest that the V alpha/V beta antigen-binding domain of this particular monoclonal scTCR construct is substantially similar to the conformational structure of lambda light chains.

Abstract:OBJECTIVE:To acquire the genes of light chain variable region (VL) and heavy chain variable region (VH) of a novel clone ZCH-7-2F9(2F9) of anti-hCD14 for construction of anti-hCD14 single chain antibody(ScFv).METHODS:From the mouse hybridoma cell line 2F9 and its fusion partner murine myeloma cell line NS-1, total RNA was prepared. The VL and VH genes were amplified by RT-PCR with family specific primer pairs, respectively. The PCR products were cloned into pGEM(sound recording copyright sign)-T Easy vectors, then transfected into DH5alpha and the positive recombinants were identified and purified. After sequencing with automatic DNA sequencer the sequences were analyzed online.RESULTS:VL gene of the new clone of CD14 monoclonal antibody (McAb) 2F9 consisted of 321 bps encoding a peptide of 107 amino acid residues, and VH gene of the 2F9 antibody contained 360 bps encoding a peptide of 120 amino acid residues. According to IMMUNOGENETICS online analysis by IMGT/V-QUEST, the VL and VH genes belonged to mouse IGKV and mouse IGHV subgroups, respectively. On the position 23/88 of the light chain and 22/96 of the heavy chain genes there were cysteines, which play the key role in forming disulfo-bond in each chain. Both VL and VH chains had definitely 4 frame regions (FR) and 3 complementary determinant regions (CDR).CONCLUSION:The treatment of CML consisting of myeloablative Allo-SCT combined with Gleevec before and after transplantation is an effective and safe method for CML.

The recombinant plasmids were transfected into E.coli BL21star(DE3)plysS for expression. After purification with Ni+resin and renaturation in vitro, the relative molecular mass (Mr) and the binding activity of the interesting protein were determined by SDS-PAGE and flow cytometry (FCM), respectively.RESULT:The cloned ScFv2F9 gene was identified to be functional by sequencing and expressing. The interesting protein was detected in inclusion body with a Mr of 31 000. The blocking test showed that the positive cell percentage, the mean fluorescence intensity (MFI) and the peak of channel (peak Ch) were reduced by 11.73%, 11.96% and 31.57%, respectively after once blockage by scFv2F9 protein, and 26.44 %, 21.75 % and 42.11 % after blockage twice.CONCLUSION:The ScFv2F9 against human CD14 antigen has been successfully expressed in prokaryotic cells with partial biological activity, which lays the foundation for further studies on its immunotoxin and other kinds of engineered antibodies.

Abstract:OBJECTIVE:Acute monocytic leukemia (AML)-M5 is the common type of acute myeloid leukemias in children. Studies have shown that there are abundant lipopolysaccharide (LPS) receptor (designated as CD14) molecules on the cell membrane of M5 cells and they play an important role in the diagnosis of M5, since they can be recognized and bound by mouse-anti-human CD14 monoclonal antibody (McAb). However, mouse-originated antibodies are largely not suitable for clinical application due to the severe side effects, thus "humanized antibody" is desired. As the first step for developing humanized antibody, the construction and expression of single chain antibody (scFv) with functional protein are necessary. The present study aimed to express ZCH-7-2F9 ScFv (scFv(2F9)) in eukaryotic cells and obtain the scFv(2F9) protein with a high biological activity.METHODS:Four primers were synthesized to construct the eukaryotic expressional vector, which included SfiI and EcoRI enzyme cleaving site, 6 x His and stop code TGA sequences. scFv(2F9) gene was amplified through splicing by overlap extension (SOE) using the high fidelity Taq polymerase. The blocking test showed that the positive cell percentages, the mean fluorescence intensity (MFI) and the peak of channel (peak Ch) were reduced by 90.02%, 63.30% and 63.38%, respectively after blocking with scFv(2F9), while those were 4.55%, 10.09% and 5.02% after blockage using the supernatant from the CHO cells transfected with blanked vector pSectag2A.CONCLUSIONS:The scFv(2F9) against human CD14 antigen was successfully expressed in eukaryotic cells and showed a high biological activity, which may be useful for the further studies on its humanized antibodies.

Abstract:Screening antibodies from phage displayed in vitro libraries and from affinity maturation of lead antibodies requires testing of antibody fragments (scFvs and Fabs) for function and binding affinities. Crude scFv or Fab periplasmic preparations from Escherichia coli are often not pure and/or concentrated enough for use in functional and affinity assays. We have developed an automated high-throughput approach for small and large-scale expression and purification of His-tagged scFvs and Fabs using the Qiagen BioRobot 3000 LS with optimized application software. This automated procedure enabled us to rapidly evaluate antibody fragments in functional and surface plasmon resonance (SPR) assays. We have used these procedures to make thousands of purified scFv/Fabs for several antibody maturation campaigns and significantly decreased the time needed to select the best candidates. The assay results from these purified samples were used to prioritize candidates before converting them to IgG. This protocol can process up to 300 small-scale and up to 72 large-scale scFvs or Fabs per week per full-time employee (FTE).
[Clinical evaluation of surveillance culture in cancer patients].

Abstract:To clarify usefulness of surveillance culture (SC) as a predictor of infection, we retrospectively studied 70 episodes of bacteremia in 271 cancer patients between April, 1984 and April, 1988. SC was obtained from the throat swabs, stool and urine on admission and then once weekly during hospitalization. Of 76 aerobic strains from blood, 18 (24%) were isolated from SC prior to bacteremia. Seven strains were first identified from SC within 4 days before bacteremia occurred. Of 63 cases of bacteremia, excluding polymicrobial bacteremia, 14 (22%) were initially detected by SC. An antibiotic therapy was altered within 5 days after bacteremia in 19 bacteremic patients with negative SC and 5 with positive SC. Six bacteremic patients with negative SC and 3 with positive SC died within 5 days after the onset of bacteremia. No significant differences were observed in antibiotic therapy (chi 2 test, p less than 0.05) or mortality (p greater than 0.05) between patients who had positive and negative SC. Routine SC is unlikely to influence an initial diagnosis or antimicrobial management for cancer patients with fever.

Abstract:This paper proposes a parallel structure of a feedback controller for the ball-screw drive systems which have significant dynamic variations during operations. The controller consists of two controllers with two different objectives. One controller aims at moving the workpiece table so that its position tracks reference trajectories with high speed and high accuracy. The other controller is for suppressing the structural vibration caused by the cutting force disturbance around system's resonant frequencies. To take into account the resonant modes' variations which are peculiar to ball-screw drives, a gain-scheduled controller is utilized as the vibration suppression controller. Advantages of the proposed controller structure in the ball-screw drive control application are discussed. It is experimentally demonstrated that the controller with the parallel structure is effective in achieving high performance in both trajectory tracking and cutting force disturbance rejection.

Abstract:γδ T cells, especially Vγ9Vδ2 T cells, play an important role in mycobacterial infection. We have identified some Vγ9Vδ2 T cells that recognize protein/peptide antigens derived from mycobacteria, which may induce protective immune responses to mycobacterial infection. To clarify the structural basis of the molecular recognition mechanism, we tried many methods to express the Vγ9Vδ2 T-cell receptor (TCR). The Vγ9Vδ2 TCR was not expressed well in a prokaryotic expression system or a baculovirus expression system, even after extensive optimization. In a mammalian cell expression system, the Vγ9Vδ2 TCR was expressed in the form of a soluble heterodimer, which was suitable for crystal screening. Reduced-temperature cultivation (cold shock) increased the yield of the recombinant TCR. The recombinant purified TCR was used for crystal trials, and crystals that could be used for X-ray diffraction were obtained. Although we have not yet determined the crystal structure of the Vγ9Vδ2 TCR, we have established a procedure for Vγ9Vδ2 TCR expression and purification, which is useful for basic research and potentially for clinical application.

Abstract:The functional decryption of the human proteome is the challenge which follows the sequencing of the human genome. Specific binders to every human protein are key reagents for this purpose. In vitro antibody selection using phage display offers one possible solution that can meet the demand for 25,000 or more antibodies, but needs substantial standardisation and minimalisation. To evaluate this potential, three human, naive antibody gene libraries (HAL4/7/8) were constructed and a standardised antibody selection pipeline was set up. The quality of the libraries and the selection pipeline was validated with 110 antigens, including human, other mammalian, fungal or bacterial proteins, viruses or haptens. Furthermore, the abundance of VH, kappa and lambda subfamilies during library cloning and the E. coli based phage display system on library packaging and the selection of scFvs was evaluated from the analysis of 435 individual antibodies, resulting in the first comprehensive comparison of V gene subfamily use for all steps of an antibody phage display pipeline. Further, a compatible cassette vector set for E. coli and mammalian expression of antibody fragments is described, allowing in vivo biotinylation, enzyme fusion and Fc fusion.

Abstract:Single-chain variable fragment (scFv) is a low-molecular-mass recombinant antibody and is usually expressed as inclusion bodies in Escherichia coli. Highly efficient purification and refolding methods are required to provide enough active proteins for therapeutic or diagnostic use. In the present study, an anti-TNF alpha (tumour necrosis factor alpha) scFv (TNF-scFv) was constructed and expressed in E. coli BL21 (DE3) star as inclusion bodies, and a convenient procedure of one-step on-column purification and refolding was provided for it. Briefly, denatured TNF-scFv was firstly captured by immobilized metal (Ni) affinity chromatography, and then non-denaturing detergent (Triton X-100)-containing and beta-cyclodextrin-containing solutions were loaded in turn on to the column to perform 'artificial chaperone-assisted refolding' after removing impurities. More than 77% of denatured TNF-scFv protein was refolded successfully with a purity of more than 95%. Activity assays showed that refolded TNF-scFv could bind to rhTNF alpha (recombinant human TNF alpha) specifically with high affinity. It could inhibit rhTNFa from binding to TNF receptors and neutralize the cytolytic activity of rhTNF alpha against L929 cells effectively. A conclusion was obtained that this refolding method is time-saving and suitable for industrial production. It may also be applicable to other scFvs or other recombinant proteins.
Construction and Expression of Single Chain Variable Fragments (scfv) Against Human CD19 Antigen

Abstract:The genes encoding for the light and heavy chain variable regions were cloned by RT-PCR from a murine monoclonal hybridoma cell line, which could produce monoclonal antibody to recognize CD19 antigen on human B lymphocyte. Then fused the light and heavy chain variable regions together by a short peptide linker containing 15 amino acid (Gly4Ser)3 using splice-overlap extensive PCR. The recombinant anti-CD19- ScFv was subcloned into the expression vector pET28a and induced to be expressed by IPTG in E. coli BL21. SDS-PAGE and Western blot analysis showed that the recombinant anti-CD19-ScFv gene was expressed in E. coli BL21. ScFv expression was in the form of an inclusion bodies and the purified fusion protein was obtained after a series of purification steps including cell break, inclusion body solubilization, Ni2+ metal affinity chromatography and protein refolding. Flow cytometry analysis showed that the ScFv can react with human CD19 antigen. In conclusion, recombinant anti-CD19-ScFv gene has been successful constructed and expressed in E. coli BL21, which could provide a basic study for the future target therapy to the B lymphoid leukemia and B lymphoma.
Isolation of ScFv antibodies of rP27 kip 1 from phage display libraries constructed from immunized and non-immunized repertoires

Abstract:Through mRNA extract, RT and a series of PCR, phage antibody libraries were constructed from rP27 kip 1 -immunized and non-immunized mice. After only one round of selection with rP27 kip 1 , clones from each library were chosen randomly and digested by Tag I and Hinf I. 11 of 64 clones from the immunized animal had consistent restriction pattern, while none of the 64 clones from the non-immunized animal had, except that one had the same fragments pattern as that of the 11 clones. The 12 fragments were expressed in E. coli BL21(DE3)/pET-28b(+) system. ELISA showed that some of the fragments could bind to rP27 kip 1 specifically. AU these results implied that specific antibody can be obtained by genetic engineering without hybridoma or many rounds of growth and panning selection.
Effect of Converting Enzyme Inhibitor (SQ14,225) on Myocardial Hypertrophy in Spontaneously Hypertensive Rats

Abstract:SUMMARY Tbe potent conrerting enzyme inhibitor (CEI) SQ14.225, which is known to prevent the formation of angiotensin II (All) has been used to evaluate the role of AH in tbe development and reversal of cardiac hypertrophy. The present study describes tbe effect of CEI on blood pressure (BP) and myocardial hypertrophy (prevention and reversal) in tbe spontaneously hypertensive rat (SHR). A group of 3-weekand 8-week-old male SHR was treated with CEI (30 mg/kg in drinking water) for 6 weeks. An additional group of SHR was also treated with a combination of CEI and a diuretic (hydrochlorothiazide, 500 mg/Uter). Heart weight, BP, deoxyrtbonudeic add (DNA), ribonudeic add (RNA), hydroxyproline, myocardial catecboiamlnes, and plasma rente activity (PRA) were determined. In the prevention study, we found a significant reduction in tbe ratio of heart weight to body weight along with the prevention of hypertension (200 vs 145 mm Hg, p < 0.001). Similar reductions in BP and heart weights were obtained with the reversal group. A better BP control was noted in the CEI and hydrochlorotbiazide group. The reduction of heart weight was associated with a reduction in RNA and hydroxyproline content. In all groups, we found a significant Increase in PRA (p < 0.001) and a slight increase in tissue catecholamlne concentration. No change in kidney weight was found in any group. Data dearly showed that oral administration of CEI prevented and reversed cardiac hypertrophy in SHR. Reversal was associated with a decrease in myocardial collagen content. These data indicate that prevention of All formation in combination with BP control can prevent and reverse cardiac hypertrophy in SHR. Of course, whether or not CEI acts only through the renin angiotension system is still speculative.

Abstract:Recombinant antibody single-chain variable fragments (scFv) are difficult to purify homogeneously from a protein complex mixture. The most effective, specific and fastest method of purification is an affinity chromatography on Protein L (PpL) matrix. This protein is a multi-domain bacterial surface protein that is able to interact with conformational patterns on kappa light chains. It mainly recognizes amino acid residues located at the VL FR1 and some residues in the variable and constant (CL) domain. Not all kappa chains are recognized, however, and the lack of CL can reduce the interaction. From a scFv composed of IGKV10-94 according to IMGT®, it is possible, with several mutations, to transfer the motif from the IGKV12-46 naturally recognized by the PpL, and, with the single mutation T8P, to confer PpL recognition with a higher affinity. A second mutation S24R greatly improves the affinity, in particular by modifying the dissociation rate (kd). The equilibrium dissociation constant (KD) was measured at 7.2 10(-11) M by surface plasmon resonance. It was possible to confer PpL recognition to all kappa chains. This protein interaction can be modulated according to the characteristics of scFv (e.g., stability) and their use with conjugated PpL. This work could be extrapolated to recombinant monoclonal antibodies, and offers an alternative for protein A purification and detection.

Abstract:Objective:Isolation and identification of single chain antibodies against surface molecules of hematopoietic progenitor cells via phage display antibody library.Methods:An individual KG1a+ phage-scFv library for the binding specificity to eight hematopoietic progenitor cell lines by indirect immunoflurorescein were identified. Then,the monoclonality of the scFvs was proved by testing specific binding to antigen proteins by Western blot.Results:64 KG1a+ phage-scFvs were identified. They showed three kinds of cell binding repertoire. The unique primary structures of two phage-scFv clones were proved by DNA sequence. Their recombinant scFv-SA fusion proteins bound to the single band of proteins from KG1a cells with molecular weights of 34 kD and 22 kD,respectively.Conclusion:Reliable methods to identify phage-scFv clones against cell surface molecules have been established and obtained two unique phage-scFv clones against hematopoietic progenitor cell surface molecules from an anti-KG1a cell phage-scFv library.
Reactivity and epitope mapping of single-chain T cell receptors with monoclonal antibodies

Abstract:To examine further the structure of the T cell receptor (TCR) and the specificity of mAbs generated against the native protein, the TCR was expressed in Escherichia coli as a single chain in which the variable regions of the alpha and beta chains are joined by a 25 amino acid linker. Five single-chain TCR that have different alpha and/or beta variable (V) regions were examined with the anti-V beta 8 region mAbs KJ16 and F23.1 and the anti-V alpha 8 mAbs KT50, KT65 and B21.14. Each of the mAbs reacted with one or more of the single-chain receptors. Western blot analysis demonstrated that the intrachain disulfide bonds were required for proper epitope conformation and recognition of the TCR by the antibodies. KT50, KT65 and B21.14 antibodies distinguished between two related V alpha regions that differed at only six residues. A model of the V regions of the TCR based on immunoglobulin (Ig) structure suggests that three of these six variant residues are in the putative CDR1 of the receptor and possibly accessible to antibody. To test this possibility, site-directed mutagenesis of the unreactive V alpha region demonstrated that the combination of all three residues restored binding by the anti-V alpha 8 antibodies. In addition, these three complimentarity determining regions (CDR) residues are likely to be in close proximity to the putative CDR3 which also influenced binding of the antibodies. The epitopes recognized by the V alpha-specific antibodies are thus predicted to reside closer to the putative binding site than the epitopes previously determined to be recognized by the anti-V beta 8 antibodies, KJ16 and F23.1. Finally, the specificities of KT50 and KT65 as determined with the E. coli expression system suggests an explanation for previous observations about the differences in the T cell populations that are recognized by these antibodies.
An optimal sequence of tasks for autonomous learning systems

Abstract:In this paper, we consider an optimal sequence of tasks for systems that improve their performances due to autonomous learning (learning-by-doing). In particular, we focus on a problem of determining sequence of performed tasks for the autonomous learning systems to minimize the total weighted completion times of tasks. Fundamental for the presented approach is that schedule (a sequence of tasks) allows to efficiently utilize learning abilities of the system to optimize its objective, but it does not affect the system itself. To solve the problem, we prove an eliminating property that is used to construct a branch and bound algorithm and present some fast heuristic and metaheuristic methods. An extensive analysis of the efficiency of the proposed algorithms is also provided.
