Abstract:Plastic waste pollution has become a global crisis, with millions of tons of plastic expected to accumulate in landfills and in natural environments, posing a serious threat to wildlife and human health. As current recycling methods remain inefficient, there is an urgent need for innovative enzymatic solutions to break down plastics and enable a circular economy approach. In this study, we explore the plastic-degrading potential of microorganisms enriched from activated sludge (AS) sourced from a municipal wastewater treatment plant (WWTP)—a known microplastic-contaminated industrial niche. Five microbial consortia (i.e., microbiomes) were enriched under selective pressure using low-carbon conditions and high concentrations of polyester polymers, including post-consumer PET, post-consumer PLA, and virgin PLA. Enrichment was performed for 100 days at 37 °C and 50 °C, followed by microbiomes isolation and metagenomic analysis to identify plastic-active bacteria and their enzymes. The results revealed that PLA polymers, but not post-consumer PET, were effectively degraded by the microbiomes, as confirmed by nuclear magnetic resonance (NMR) and gel permeation chromatography (GPC), showing significant molecular weight reduction compared to the abiotic controls. Microbial community analysis highlighted a distinct enrichment profile driven by the polymer composition and the temperature. At 50 °C, the Bacillales order became the predominant population, whereas at 37 °C, a more diverse community within the Proteobacteria and Actinobacteria phyla were selected. Nonetheless, the enriched microbial communities at both temperatures included phyla with members known for polyester degradation. Moreover, at 50 °C, enrichment of putative PET/PLA hydrolases was also observed. These findings suggest that AS microorganisms are a reservoir of polyester-active enzymes, particularly PLA-depolymerases, and hold promise for advancing biotechnological strategies to mitigate plastic pollution through re- and up-cycling.

Deconstructed Plastic Substrate Preferences of Microbial Populations from the Natural Environment

Abstract:Over half of the world's plastic waste is landfilled, where it is estimated to take hundreds of years to degrade. Given the continued use and disposal of plastic products, it is vital that we develop fast and effective ways to utilize plastic waste. Here, we explore the potential of tandem chemical and biological processing to process various plastics quickly and effectively. Four samples of compost or sediment were used to set up enrichment cultures grown on mixtures of compounds, including disodium terephthalate and terephthalic acid (monomers of polyethylene terephthalate), compounds derived from the chemical deconstruction of polycarbonate, and pyrolysis oil derived from high-density polyethylene plastics. Established enrichment communities were also grown on individual substrates to investigate the substrate preferences of different taxa. Biomass harvested from the cultures was characterized using 16S rRNA gene amplicon sequencing and shotgun metagenomic sequencing. These data reveal low-diversity microbial communities structured by differences in culture inoculum, culture substrate source plastic type, and time. Microbial populations from the classes Alphaproteobacteria, Gammaproteobacteria, Actinobacteria, and Acidobacteriae were significantly enriched when grown on substrates derived from high-density polyethylene and polycarbonate. The metagenomic data contain abundant aromatic and aliphatic hydrocarbon degradation genes relevant to the biodegradation of deconstructed plastic substrates used here. We show that microbial populations from diverse environments are capable of growth on substrates derived from the chemical deconstruction or pyrolysis of multiple plastic types and that paired chemical and biological processing of plastics should be further developed for industrial applications to manage plastic waste. IMPORTANCE The durability and impermeable nature of plastics have made them a popular material for numerous applications, but these same qualities make plastics difficult to dispose of, resulting in massive amounts of accumulated plastic waste in landfills and the natural environment. Since plastic use and disposal are projected to increase in the future, novel methods to effectively break down and dispose of current and future plastic waste are desperately needed. We show that the products of chemical deconstruction or pyrolysis of plastic can successfully sustain the growth of low-diversity microbial communities. These communities were enriched from multiple environmental sources and are capable of degrading complex xenobiotic carbon compounds. This study demonstrates that tandem chemical and biological processing can be used to degrade multiple types of plastics over a relatively short period of time and may be a future avenue for the mitigation of rapidly accumulating plastic waste.
Multivariate analysis of enriched landfill soil consortia provide insight on the community structural perturbation and functioning during low-density polyethylene degradation

Abstract:Plastic-enriched sites like landfills have immense potential for discovery of microbial consortia that can efficiently degrade plastics. In this study, we used a combination of culture enrichment, high-throughput PacBio sequencing of 16 S rRNA and the ITS gene, Fourier transform infrared (FTIR), and scanning electron microscopy (SEM) to examine the compositional and diversity perturbations of bacterial and fungal consortia from landfill soils and their impact on low-density polyethylene (LDPE) film biodegradation over a 90-day period. Results showed that enrichment cultures effectively utilized LDPE as a carbon source for cellular growth, resulting in significant weight reduction (22.4% and 55.6%) in the films. SEM analysis revealed marked changes in the micrometric surface characteristics (cracks, fissures, and erosion) and biofilm formation in LDPE films. FTIR analyses suggested structural and functional group modification related to C-H (2831-2943 cm⁻¹), and CH₂ (1400 cm⁻¹) stretching, CO and CC (680-950 cm⁻¹) scission, and CO incorporation (3320-3500 cm⁻¹) into the carbon backbone, indicative of LDPE polymer biodegradation. Enrichment cultures had lower diversity and richness of microbial taxa compared to soil samples, with LDPE as a carbon source having a direct influence on the structure and functioning of the microbial consortia. A total of 26 bacterial and 12 fungal OTU exhibiting high relative abundance and significant associations (IndVal > 0.7, q < 0.05) were identified in the enrichment culture. Bacterial taxa such as unclassified Parvibaculum FJ375498, Achromobacter xylosoxidans, unclassified Chitinophagaceae PAC002331, unclassified Paludisphaera and unclassified Comamonas JX898122, and six fungal species (Galactomyces candidus, Trichosporon chiropterorum, Aspergillus fumigatus, Penicillium chalabudae, Talaromyces thailandensis, and Penicillium citreosulfuratum) were identified as the putative LDPE degraders in the enrichment microbial consortium cultures. PICRUSt2 metagenomic functional profiling of taxonomic bacterial taxa abundances in both landfill soil and enrichment microbial consortia also revealed differential enrichment of energy production, stress tolerance, surface attachment and motility pathways, and xenobiotic degrading enzymes important for biofilm formation and hydrolytic/oxidative LDPE biodegradation. The findings shed light on the composition and structural changes in landfill soil microbial consortia during enrichment with LDPE as a carbon source and suggest novel LDPE-degrading bacterial and fungal taxa that could be explored for management of polyethylene pollution.

Abstract:Wide usage of plastic coupled with mismanagement has created a humongous environmental hazard threatening entire ecosystems. To date, the potential effects of plastic debris-induced soil nutrition substance changes and the relevant microbial metabolic behavior remain unclear. Here, we studied the effect of plastic films polyethylene and polylactic acid in differential soil environments (farmland, woodland, and wetland) for 120 days. Soil enzyme activities (urease, neutral phosphatase, and catalase) and nutrition substance (NH 4 + -N, available P, available K, and soil organic matter) present obvious variations in polylactic acid groups compared to polyethylene-treated samples. 16S rRNA gene sequencing indicates that several bacteria abundance such as Bacteroidales , Actinobacteriota , Nitrososphaeraceae , Pyrinomonadalcs , Muribaculaceae , exhibited obvious up-regulation or down-regulation, and simultaneously, the carbon, nitrogen, and phosphorus cycling relevant species Bryobacte r, Bradyrhizobium , and Sphingomonas , expressed wider margin of down-regulation in abundance in plastic treatment soil samples. As a result, the abundance of metabolites including sugar, amino acid, and fatty acids, which may associated with nutrition substance metabolic pathways, were significantly altered in the stress of plastic. These findings provide valuable information on the environmental effects of plastics, and the relationships of subsequent nutrition substance changes and microbial metabolic behavior.

Abstract:Microbiological degradation of polymers offers a promising approach for mitigating environmental plastic pollution. This study (i) elucidated the diversity and structure of bacterial microbiomes from distinct environments (landfill soil, sewage sludge, and river water) characterized by specific physicochemical parameters, and (ii) utilized environment-derived microbial cultures enriched with microplastics (MPs) to investigate the degradation of polymers and identify culturable bacterial strains contributing to the plastisphere. We found that alpha diversity was notably higher in river water (∼20%) compared to landfill soil and sewage sludge. Dominant phyla included Pseudomonadota in sewage sludge (39.1%) and water (23.7%), while Actinomycetota prevailed in soil (38.5%). A multistage experiment, involving successive subcultures of environmental microbiomes exposed to polypropylene (PP), polyvinyl chloride (PVC), polycarbonate (PC), and polylactic acid (PLA), facilitated the assessment of MPs degradation processes. Analysis of carbonyl indices CIs and FTIR spectra revealed substantial structural changes in the treatment PVC-landfill soil, as well as in PLA- and PC-sludge enriched cultures. Further, using enriched cultures as a source of microorganisms, the study obtained 17 strains of plastic degraders from landfill soil, 14 from sewage sludge, and 6 from river water. Remarkably, similar bacterial genera were isolated across environmental microbiomes regardless of the MPs substrate used in enriched cultures. Among the 37 identified strains, Pseudomonadota predominated (64.86%) and were accompanied by Bacteroidota (16.22%), Actinomycetota (13.51%), and Bacillota (5.41%). This study highlights the complex relationship between microbiome diversity and the biodegradation efficiency of plastics, showing the potential for using microbial communities in the plastic pollution management.
Development of plastic-degrading microbial consortia by induced selection in microcosms

Abstract:The increase in the production of highly recalcitrant plastic materials, and their accumulation in ecosystems, generates the need to investigate new sustainable strategies to reduce this type of pollution. Based on recent works, the use of microbial consortia could contribute to improving plastic biodegradation performance. This work deals with the selection and characterization of plastic-degrading microbial consortia using a sequential and induced enrichment technique from artificially contaminated microcosms. The microcosm consisted of a soil sample in which LLDPE (linear low-density polyethylene) was buried. Consortia were obtained from the initial sample by sequential enrichment in a culture medium with LLDPE-type plastic material (in film or powder format) as the sole carbon source. Enrichment cultures were incubated for 105 days with monthly transfer to fresh medium. The abundance and diversity of total bacteria and fungi were monitored. Like LLDPE, lignin is a very complex polymer, so its biodegradation is closely linked to that of some recalcitrant plastics. For this reason, counting of ligninolytic microorganisms from the different enrichments was also performed. Additionally, the consortium members were isolated, molecularly identified and enzymatically characterized. The results revealed a loss of microbial diversity at each culture transfer at the end of the induced selection process. The consortium selected from selective enrichment in cultures with LLDPE in powder form was more effective compared to the consortium selected in cultures with LLDPE in film form, resulting in a reduction of microplastic weight between 2.5 and 5.5%. Some members of the consortia showed a wide range of enzymatic activities related to the degradation of recalcitrant plastic polymers, with Pseudomonas aeruginosa REBP5 or Pseudomonas alloputida REBP7 strains standing out. The strains identified as Castellaniella denitrificans REBF6 and Debaryomyces hansenii RELF8 were also considered relevant members of the consortia although they showed more discrete enzymatic profiles. Other consortium members could collaborate in the prior degradation of additives accompanying the LLDPE polymer, facilitating the subsequent access of other real degraders of the plastic structure. Although preliminary, the microbial consortia selected in this work contribute to the current knowledge of the degradation of recalcitrant plastics of anthropogenic origin accumulated in natural environments.

Abstract:A polyethylene surrogate was utilized to enrich microbial communities capable of utilizing these compounds as a growth substrate. The graphic depicts the chain profile of a chromatogram of the polyethylene surrogate before culture, and following enrichment of a microbial community. Plastic pollution is a vast and increasing problem that has permeated the environment, affecting all aspects of the global food web. Plastics and microplastics have spread to soil, water bodies, and even the atmosphere due to decades of use in a wide range of applications. Plastics include a variety of materials with different properties and chemical characteristics, with polyethylene being a dominant fraction. Polyethylene is also an extremely persistent compound with slow rates of photodegradation or biodegradation. In this study, we developed a method to isolate communities of microbes capable of biodegrading a polyethylene surrogate. This method allows us to study potential polyethylene degradation over much shorter time periods. Using this method, we enriched several communities of microbes that can degrade the polyethylene surrogate within weeks. We also identified specific bacterial strains with a higher propensity to degrade compounds similar to polyethylene. We provide a description of the method, the variability and efficacy of four different communities, and key strains from these communities. This method should serve as a straightforward and adaptable tool for studying polyethylene biodegradation.

Abstract:Bioplastic waste may play a key role in shaping the microbial communities in composting environment. The present study aims to determine the possible influence of bioplastics on microbial structure of aerobic organic waste treatments and to assess the evolution of microbial community during thermophilic and maturation phases of composting, comparing microorganisms in compost and attached on bioplastics surface. Composting process was simulated in a 2-month lab-scale test. Mater-Bi®, PBAT, PLA and LDPE were considered as benchmark. A qualitative screening was done with DGGE, and then the bacterial community profile was disclosed with 16S rRNA amplicon analysis. The 16S rRNA profile in bioplastics-associated communities was found to largely differed from that of the initial compost (positive control). Moreover, as expected, during the thermophilic phase there was a prevalence of better heat-resistant phyla compared to the mesophilic phase. Some specialists were found to be plastic nature dependent, in particular Streptomyces , Pseudomonas , Aeribacillus , Schlegellela and Cohnella were among the most abundant genera. Finally, to select bacterial species capable of growing on the tested bioplastics, an enrichment method was applied. The method disclosed the possibility to culture some specific bioplastics degrading species, opening the frontiers for bioaugmentation practice in industrial composting.

Abstract:As the global threat of plastic pollution has grown in scale and urgency, so have efforts to find sustainable and efficient solutions. Research conducted over the past few years has identified gut environments within insect larvae, including Tenebrio molitor (yellow mealworms), as microenvironments uniquely suited to rapid plastic biodegradation. However, there is currently limited understanding of how the insect host and its gut microbiome collaborate to create an environment conducive to plastic biodegradation. In this work, we provide evidence that T. molitor secretes one or more emulsifying factor(s) (30-100 kDa) that mediate plastic bioavailability. We also demonstrate that the insect gut microbiome secretes factor(s) (<30 kDa) that enhance respiration on polystyrene (PS). We apply these insights to culture PS-fed gut microbiome enrichments, with elevated rates of respiration and degradation compared to the unenriched gut microbiome. Within the enrichment, we identified eight unique gut microorganisms associated with PS biodegradation including Citrobacter freundii, Serratia marcescens, and Klebsiella aerogenes. Our results demonstrate that both the mealworm itself and its gut microbiome contribute to accelerated plastic biodegradation. This work provides new insights into insect-mediated mechanisms of plastic degradation and potential strategies for cultivation of plastic-degrading microorganisms in future investigations and scale-up.

Abstract:Abstract Waste plastic presently accumulates in landfills or the environment. While natural microbial metabolisms can degrade plastic polymers, biodegradation of plastic is very slow. This study demonstrates that chemical deconstruction of polyethylene terephthalate (PET) with ammonium hydroxide can replace the rate limiting step (depolymerization) and by producing plastic-derived terephthalic acid and terephthalic acid monoamide. The deconstructed PET (DCPET) is neutralized with phosphoric acid prior to bioprocessing, resulting in a product containing biologically accessible nitrogen and phosphorus from the process reactants. Three microbial consortia obtained from compost and sediment degraded DCPET in ultrapure water and scavenged river water without addition of nutrients. No statistically significant difference was observed in growth rate compared to communities grown on DCPET in minimal culture medium. The consortia were dominated by Rhodococcus spp., Hydrogenophaga spp., and many lower abundance genera. All taxa were related to species known to degrade aromatic compounds. Microbial consortia are known to confer flexibility in processing diverse substrates. To highlight this, we also demonstrate that two microbial consortia can grow on similarly deconstructed polyesters, polyamides, and polyurethanes in water instead of medium. Our findings suggest that microbial communities may enable flexible bioprocessing of mixed plastic wastes when coupled with chemical deconstruction.

Abstract:Plastic pollution of the soil is a global issue of increasing concern, with far-reaching impact on the environment and human health. To fully understand the medium- and long-term impact of plastic dispersal in the environment, it is necessary to define its interaction with the residing microbial communities and the biochemical routes of its degradation and metabolization. However, despite recent attention on this problem, research has largely focussed on microbial functional potential, failing to clearly identify collective adaptation strategies of these communities. Our study combines genome-centric metagenomics and metatranscriptomics to characterise soil microbial communities adapting to high polyethylene and polyethylene terephthalate concentration. The microbiota were sampled from a landfill subject to decades-old plastic contamination and enriched through prolonged cultivation using these microplastics as the only carbon source. This approach aimed to select the microorganisms that best adapt to these specific substrates. As a result, we obtained simplified communities where multiple plastic metabolization pathways are widespread across abundant and rare microbial taxa. Major differences were found in terms of expression, which on average was higher in planktonic microbes than those firmly adhered to plastic, indicating complementary metabolic roles in potential microplastic assimilation. Moreover, metatranscriptomic patterns indicate a high transcriptional level of numerous genes in emerging taxa characterised by a marked accumulation of genomic variants, supporting the hypothesis that plastic metabolization requires an extensive rewiring in energy metabolism and thus provides a strong selective pressure. Altogether, our results provide an improved characterisation of the impact of microplastics derived from common plastics types on terrestrial microbial communities and suggest biotic responses investing contaminated sites as well as potential biotechnological targets for cooperative plastic upcycling.

Abstract:Despite increasing efforts across various disciplines, the fate, transport, and impact of synthetic plastics on the environment and public health remain poorly understood. To better elucidate the microbial ecology of plastic waste and its potential for biotransformation, we conducted a large-scale analysis of all publicly available meta-omic studies investigating plastics (n = 27) in the environment. Notably, we observed low prevalence of known plastic degraders throughout most environments, except for substantial enrichment in riverine systems. This indicates rivers may be a highly promising environment for discovery of novel plastic bioremediation products. Ocean samples associated with degrading plastics showed clear differentiation from non-degrading polymers, showing enrichment of novel putative biodegrading taxa in the degraded samples. Regarding plastisphere pathogenicity, we observed significant enrichment of antimicrobial resistance genes on plastics but not of virulence factors. Additionally, we report a co-occurrence network analysis of 10 + million proteins associated with the plastisphere. This analysis revealed a localized sub-region enriched with known and putative plastizymes—these may be useful for deeper investigation of nature's ability to biodegrade man-made plastics. Finally, the combined data from our meta-analysis was used to construct a publicly available database, the Plastics Meta-omic Database (PMDB)—accessible at plasticmdb.org. These data should aid in the integrated exploration of the microbial plastisphere and facilitate research efforts investigating the fate and bioremediation potential of environmental plastic waste.

Abstract:Plastics derived from fossil fuels are used ubiquitously owing to their exceptional physicochemical characteristics. However, the extensive and short-term use of plastics has caused environmental challenges. The biotechnological plastic conversion can help address the challenges related to plastic pollution, offering sustainable alternatives that can operate using bioeconomic concepts and promote socioeconomic benefits. In this context, using soil from a plastic-contaminated landfill, two consortia were established (ConsPlastic-A and -B) displaying versatility in developing and consuming polyethylene or polyethylene terephthalate as the carbon source of nutrition. The ConsPlastic-A and -B metagenomic sequencing, taxonomic profiling, and the reconstruction of 79 draft bacterial genomes significantly expanded the knowledge of plastic-degrading microorganisms and enzymes, disclosing novel taxonomic groups associated with polymer degradation. The microbial consortium was utilized to obtain a novel Pseudomonas putida strain (BR4), presenting a striking metabolic arsenal for aromatic compound degradation and assimilation, confirmed by genomic analyses. The BR4 displays the inherent capacity to degrade polyethylene terephthalate (PET) and produce polyhydroxybutyrate (PHB) containing hydroxyvalerate (HV) units that contribute to enhanced copolymer properties, such as increased flexibility and resistance to breakage, compared with pure PHB. Therefore, BR4 is a promising strain for developing a bioconsolidated plastic depolymerization and upcycling process. Collectively, our study provides insights that may extend beyond the artificial ecosystems established during our experiments and supports future strategies for effectively decomposing and valorizing plastic waste. Furthermore, the functional genomic analysis described herein serves as a valuable guide for elucidating the genetic potential of microbial communities and microorganisms in plastic deconstruction and upcycling.
Environmental factors control microbial colonization of plastics in the North Sea

Abstract:Large quantities of plastic enter the oceans each year providing extensive attachment surfaces for marine microbes yet understanding their interactions and colonization of plastic debris remains limited. We investigated microbial colonization of various plastic types (polyethylene, polystyrene, polyethylene-terephthalate, and nylon) in ex-situ incubation experiments. Plastic films, both UV-pretreated and untreated, were exposed to seawater from a coastal and an offshore location in the North Sea. 16S rRNA amplicon sequencing was employed to assess microbial community structures after 5, 10, 30, and 45 days of incubation. Our findings show the significant influence of time, seawater origin and plastic type on microbial community succession. We also identified several genera associated with hydrocarbon or plastic degradation potential as well as genera selecting for specific plastics such as Ketobacter and Microbacterium. Our results highlight potential role of microorganisms in plastic biodegradation and support the idea that microbial colonizers on marine plastics debris seemingly select distinct substrate types.

Abstract:With the benefits of coming at low-cost, being light-weight and having a high formability and durability, conventional plastics are widely used in both industry and daily life. However, because of their durability and extensive half-life with poor degradability and the low recycling rate, large amounts of plastic waste are accumulated in various environments, posing a significant threat to organisms and ecosystems. Compared to conventional physical and chemical degradation, biodegradation of plastic might become a promising and environmentally friendly solution for this problem. One of the aims of this review is to briefly describe the impact of plastics (especially microplastics). To facilitate rapid advancements in the area of plastic biodegradation, this paper provides a comprehensive review of the candidate organisms capable of biodegrading plastics and originating from four categories including natural microorganisms, artificially derived microorganisms, algae and animal organisms. In addition, the potential mechanism during plastic biodegradation and associated driving factors are summarized and discussed. Furthermore, the recent biotechnological progress (e.g. synthetic biology, systems biology, etc.) is highlighted as being key for future research. Finally, innovative research avenues for future studies are proposed. Concluding, our review is addressing the practical application of plastic biodegradation and the plastic pollution, thus necessitating more sustainable developments.

Abstract:The incomplete degradation of bio-based and biodegradable plastics (BBPs) in soils causes multiple threats to soil quality, human health, and food security. Plastic residuals can interact with soil microbial communities. We aimed to link the structure and enzyme-mediated functional traits of a microbial community composition that were present during poly (butylene succinate-co-butylene adipate (PBSA) decomposition in soil with (PSN) and without (PS) the addition of nitrogen fertilizer ((NH4)2SO4). We identified bacterial (Achromobacter, Luteimonas, Rhodanobacter, and Lysobacter) and fungal (Fusarium, Chaetomium, Clonostachys, Fusicolla, and Acremonium) taxa that were linked to the activities of ß-glucosidase, chitinase, phosphatase, and lipase in plastic-amended soils. Fungal biomass increased by 1.7 and 4 times in PS and PSN treatment, respectively, as compared to non-plastic amended soil. PBSA significantly changed the relationships between soil properties (C: N ratio, TN, and pH) and microbial community structure; however, the relationships between fungal biomass and soil enzyme activities remained constant. PBSA significantly altered the relationship between fungal biomass and acid phosphatase. We demonstrated that although the soil functions related to nutrient cycling were not negatively affected in PSN treatment, potential negative effects are reasoned by the enrichment of plant pathogens. We concluded that in comparison to fungi, the bacteria demonstrated a broader functional spectrum in the BBP degradation process.

Abstract:Gut microbial communities of plastic-munching worms provide novel insights for the development of plastic-processing biotechnologies. Considering the complexity of worm maintenance and the gut microbial communities, it is challenging to apply the worms directly in plastic processing. Harnessing the power of microbial communities derived from the worm gut microbiomes in vitro may enable a promising bioprocess for plastic degradation. Here, we established stable and reproducible plastic-associated biofilm communities derived from the gut microbiome of a superworm, Zophobas atratus, through a two-stage enrichment process: feeding with plastics (HDPE, PP, and PS) and in vitro incubation of gut microbiomes obtained from the plastic-fed worms. Plastic feeding exhibited marginal influence on bacterial diversity but substantially changed the relative abundance of different bacterial groups, and intriguingly, enriched potential plastic degraders. More prominent shifts of microbial communities were observed during the in vitro incubation of the gut microbiomes. Taxa containing plastic-degrading strains were further enriched, while other taxa represented by lactic acid bacteria were depleted. Additionally, the plastic characterization confirmed the degradation of the incubated plastics by the plastic-associated microbial communities. Community functional inference for both gene abundance and community phenotype suggested that the in vitro incubation enhanced plastic-degrading potential. Deterministic ecological effects, in particular, selection processes, were identified as the main driving force of the observed community shifts. Our findings provide novel insights into plastic-munching-worm-inspired bioprocessing of plastic wastes.

Abstract:Plastic polymers with different properties have been developed in the last 150 years to replace materials such as wood, glass and metals across various applications. Nevertheless, the distinct properties which make plastic desirable for our daily use also threaten our planet's sustainability. Plastics are resilient, non-reactive and most importantly, non-biodegradable. Hence, there has been an exponential increase in plastic waste generation, which has since been recognised as a global environmental threat. Plastic wastes have adversely affected life on earth, primarily through their undesirable accumulation in landfills, leaching into the soil, increased greenhouse gas emission, etc. Even more damaging is their impact on the aquatic ecosystems as they cause entanglement, ingestion and intestinal blockage in aquatic animals. Furthermore, plastics, especially in the microplastic form, have also been found to interfere with chemical interaction between marine organisms, to cause intrinsic toxicity by leaching, and by absorbing persistent organic contaminants as well as pathogens. The current methods for eliminating these wastes (incineration, landfilling, and recycling) come at massive costs, are unsustainable, and put more burden on our environment. Thus, recent focus has been placed more on the potential of biological systems to degrade synthetic plastics. In this regard, some insects, bacteria and fungi have been shown to ingest these polymers and convert them into environmentally friendly carbon compounds. Hence, in the light of recent literature, this review emphasises the multifaceted roles played by microorganisms in this process. The current understanding of the roles played by actinomycetes, algae, bacteria, fungi and their enzymes in enhancing the degradation of synthetic plastics are reviewed, with special focus on their modes of action and probable enzymatic mechanisms. Besides, key areas for further exploration, such as the manipulation of microorganisms through molecular cloning, modification of enzymatic characteristics and metabolic pathway design, are also highlighted.

Abstract:Plastic pollution has emerged as a significant environmental issue in marine ecosystems. The prolonged presence of plastics in mangrove sediments can have a substantial impact on the carbon and nitrogen cycles of these vital environments. We conducted in situ burial exposure experiments to assess microbial colonization and activity on biodegradable and refractory plastics in mangrove sediments, with a focus on their influence on nitrogen cycling. Variations in microbial community succession and metabolic processes on plastic surfaces were primarily determined by the type of plastic polymer. After 60 days of exposure, the abundance of denitrifying bacteria and denitrification-related genes (narG, nirS, nosZ, and norB) was higher on biodegradable plastics. After 360 days, however, a high abundance of ammonia-oxidizing archaea and nitrification-related genes (pmoA-amoA, pmoBamoB, pmoC-amoC, and hao) were found on refractory plastics. Consequently, plastic pollution in mangrove sediments could disrupt the nitrogen cycling equilibrium in these ecosystems, underscoring the critical necessity to regulate and alleviate the detrimental impacts of plastic pollution in crucial coastal areas.

Abstract:Plastics, particularly polystyrene (PS) and polyethylene (PE), are pervasive in ecosystems and degrade slowly, leading to the formation of microplastics and nanoplastics, which pose environmental and health risks. The gut microbiota of Tenebrio molitor larvae (mealworms) can degrade plastics, making them a potential model for studying microbial plastic digestion. This study investigated how rearing T. molitor on PS- or PE-supplemented diets over three generations affected gut microbiota composition and plastic degradation. Larvae were reared on wheat bran (control), PS, or PE diets, and microbial communities from their guts were cultured to assess plastic metabolism. Results showed that the PE-supplemented diet enhanced bacterial plastic degradation, while the PS-supplemented diet did not lead to similar improvements. Microbes from PE-fed larvae exhibited increased plastic metabolic activity compared to controls, and several bacterial strains with plastic-degrading capabilities were identified. However, microbial digestion of PS was less efficient, and potential dysbiosis due to the presence of Dickeya phage in PS-fed larvae may have influenced the results. Further research is needed to clarify the mechanisms driving microbial plastic digestion and to determine the applicability of these findings to environmental plastic remediation, particularly for micro- and nanoplastics.
