Abstract:Upon influenza A virus (IAV) infection, DAI is activated, recruits downstream proteins to assemble a multiprotein platform (PANoptosome), and then triggers cell death. Until now, the protein composition and assembly mechanism of the PANoptosome during IAV infection had not been elucidated.

What problem does this paper attempt to address?
The problem that this paper attempts to solve is to reveal the molecular mechanism of cell death during Influenza A Virus (IAV) infection, especially the role of Sperm - Associated Antigen 9 (SPAG9) in this process and its relationship with the c - Jun N - Terminal Kinase (JNK) signaling pathway. Specifically, the research aims to: 1. **Identify new proteins involved in PANoptosome assembly**: By using proximity - labeling technology and mass spectrometry analysis technology, the researchers discovered SPAG9 as a new component of PANoptosome and confirmed that SPAG9 can promote IAV - induced cell death, which is mainly achieved by enhancing the interactions among RIPK1, RIPK3 and DAI. 2. **Clarify the role of SPAG9 in cell death caused by IAV infection**: The study shows that SPAG9 promotes the occurrence of DAI - mediated apoptosis, necroptosis and pyroptosis by interacting with DAI, and these processes are all achieved through the DAI/SPAG9/JNK axis. 3. **Explore the role of the JNK signaling pathway in IAV - induced cell death**: By using different inhibitors, the study found that the JNK signaling pathway specifically regulates IAV - induced DAI - mediated cell death, including apoptosis, necroptosis and pyroptosis. In addition, the study also revealed the enhancing effect of the JNK signaling pathway on the interactions among RIPK1, RIPK3 and DAI, which is crucial for the formation of PANoptosome. In summary, this study not only enhances the understanding of the molecular mechanism of cell death caused by IAV infection, but also provides potential targets for the development of treatment strategies against IAV infection.
