Purpurin-18 imide derivative synthesis and functionalization for the photodynamic cancer therapy

Abstract:As part of our research in the PEIRENE laboratory, we are interested in the development and vectorization of photosensitizers for photodynamic cancer therapy applications. In this approach, a new stable photosensitizer, with an absorption band in the near infrared was designed from chlorophyll a. Hemisynthesis of purpurin-18 was realized according to a method described in the literature1. After opening the anhydride exocycle with an amine carrying an adamantyl group, the derivative of chlorin-p6 obtained reacted with acetic anhydride and led to the purpurin-18 imide derivative (Figure 1). The adamantyl group will be allow a subsequent encapsulation by hydrophobic interaction in cyclodextrins carried by cellulose nanocrystals to effect its delivery to tumor cells by EPR effect and increase its solubility. In an other hand, a triphenylphosphonium moiety was grafted onto the photosensitizer; indeed, this group is described in the literature as a mitochondrial targeting unit and should increase the selectivity and efficiency of PDT treatment on cancer cells2.
