Soy Protein Isolate-Based Hybrid Electrospun Nanofibers: An Enhanced Antimicrobial Bio-platform for Potential Wound Healing

Abstract:In this work, soy protein isolate (SPI) and N-(3-sulfopropyl) chitosan salt (SPCS) was utilized to synthesize electrospun nanofibers (NFs) for the first time. Polycaprolactone (PCL) was also added in the formulation of SPI/SPCS NFs to optimize and improve the SPI/SPCS electrospinnability, surface morphology, and mechanical stability. To confer excellent antibacterial properties in electrospun NFs, in situ incorporation of ZIF-8 NPs and Matricaria chamomilla essential oil (MCEO) by electrospinning was performed. The structural and morphological analyses of the synthesized NFs were fully characterized with different techniques, confirming their successful construction. According to the result, cell physicochemical, antibacterial activity, cell viability, biodegradability, culture data, the prepared MCEO-loaded SPI/PCL/SPCS/ZIF-8 NFs demonstrated good blood clotting (about 30%), high hemolysis rate (similar with the gauze), desired cell adhesion and noncytotoxic nature against Human skin fibroblast cells (HFF-1, over 100% cell viability). Outstandingly, they exhibited desired antibacterial efficacy toward S. aureus and E. coli with 32.3 mm and 31.2 mm inhibition zones, respectively. These results recommended that the constructed SPI-based NFs could be potential as wound healing platforms.Graphical Abstract
