Abstract:A nanofabrication method for the production of flexible core-shell structured silk elastin nanofibers is presented, based on an all-aqueous coaxial electrospinning process. In this process, silk fibroin (SF) and silk-elastin-like protein polymer (SELP), both in aqueous solution, with high and low viscosity, respectively, were used as the inner (core) and outer (shell) layers of the nanofibers. The electrospinnable SF core solution served as a spinning aid for the nonelectrospinnable SELP shell solution. Uniform nanofibers with average diameter from 301 ± 108 nm to 408 ± 150 nm were obtained through adjusting the processing parameters. The core-shell structures of the nanofibers were confirmed by fluorescence and electron microscopy. In order to modulate the mechanical properties and provide stability in water, the as-spun SF-SELP nanofiber mats were treated with methanol vapor to induce β-sheet physical crosslinks. FTIR confirmed the conversion of the secondary structure from a random coil to β-sheets after the methanol treatment. Tensile tests of SF-SELP core-shell structured nanofibers showed good flexibility with elongation at break of 5.20% ± 0.57%, compared with SF nanofibers with an elongation at break of 1.38% ± 0.22%. The SF-SELP core-shell structured nanofibers should provide useful options to explore in the field of biomaterials due to the improved flexibility of the fibrous mats and the presence of a dynamic SELP layer on the outer surface.

Abstract:Electrospinning is a very effective method to fabricate nanofibers. This technique has been achieved more attention due to its cheap cost, simple device and obtaining nanofibers similar to extracellular matrix.Silk fibroin(SF) is a natural protein with good biodegradability and low immunogenicity, Therefore, it can be widely applied in tissue engineering. In this paper, we summarize the research of electrospun silk fibrion nanofibers in regenerative medicine. Furthermore, the future research work of SF nanofibers was suggested.

Abstract:Hydrogel materials are of great value in medical field due to their similarity to human soft tissue.With silk fibroin as raw materials,organic solvent induction method can be employed to shorten the gelling time of silk fibroin to several hours.Silk fibroin/collagen composite hydrogel prepared with carbodiimide (EDC) as activation cross-linking agent has a storage modulus of up to 10 kPa.In vivo experiments indicate that silk fibroin hydrogel created by enzymatic crosslinking has good biocompatibility and biodegradability.We first explain the gelling mechanism of silk fibroin hydrogels,and then systematically introduce the research progress on preparation and modification methods of various silk fibroin hydrogels.Besides,potential applications of silk fibroin hydrogels as novel biomaterial in clinical applications are analyzed.Basic theoretical and technological issues to be solved for clinical applications of silk fibroin hydrogels are also discussed.

Abstract:Regeneration Bombyx mori(B.mori) silk fibroin(SF) nanofibers prepared by electrospinning were widely applied in in the fields of biomaterials and tissue engineering, with the advantages of high specific surface area, high porosity, and good biocompatibility. Pure regenerated SF nanofibers were fragile and poor in mechanical performance, however the composite nanofibers synthesized by blending different inorganic particles or synthetic polymers into SF has much more advantages for it can greatly improve the suitability compared with which of a single material. This paper summarized preparation of SF and inorganic particles or synthetic polymers composite nanofibers in the domestic and at abroad in recent years, which aims to improve the comprehensive performance of SF nanofibers and expand its application.

This review focuses on the preparation methods and enhanced properties of silk fibroin-nanoparticle composites, especially their biomedical applications.

Abstract:Silk fibroin (SF) has been one of promising resources of biotechnology and biomedical materials due to its unique properties. Here, different sizes of porous tubular scaffolds were fabricated from a SF aqueous solution with the addition of poly(ethylene glycol diglycidyl ether) (PGDE). The scaffolds were generally flexible and transparent at the wet state with a pore size of 81-128 mu m and porosity of 90-96%, depending on the concentrations of SF and PGDE. The mechanical properties measurement showed that the tubular SF scaffolds had satisfying tensile and compression properties, especially the excellent deformation-recovery ability. FT-IR spectra indicated that the SF in the tubular scaffolds was in a beta-sheet structure, and no PGDE characteristic band was observed, suggesting that the PGDE could be removed from the scaffolds by soaking in deionized water. The cell compatibility of scaffolds was evaluated, and no obvious cytotoxicity to mouse L-929 fibroblasts was detected. (C) Koninklijke Brill NV, Leiden, 2009

Abstract:The silk fibroin nanofiber materials fabricated by electrospinning have the advantages of large specific surface area,high void ratio,and good biocompatibility etc. So,they have wide application potential in the field of biological medicine. However,the low crystallinity and poor mechanical properties of silk fibroin limit its application. Modification of the silk fibroin nanofibers has been a hot issue in the field. This paper mainly reviews the research status of pure silk fibroin nanofibers prepared with different solvents for electrostatic spinning and the research progress of modification technology of silk fibroin nanofibers in recent years. The potential applications for these nanofiber materials have also been expected.

Electrospinning of reconstituted silk fiber from aqueous silk fibroin solution

Abstract:Silk nanofibers were successfully prepared by electrospinning an aqueous solution of regenerated silk fibroin (RSF, from Bombyx mori) with higher molecular weight. Many factors, such as concentration, electrical conductivity of the fibroin solution and applied electric field were found to influence the morphology of these nanofibers. The conformation of RSF nanofibers was transformed from random coil/helical to β-sheet after the post treatment with pure ethanol. Under optimal conditions, the as-spun non-woven mats achieved good mechanical properties. The apparent stress and strain at break were 11.1±0.7 MPa and 10.2±1.6%, respectively, which is important for the application of such a unique fibrous protein.

Abstract:Thermoplastic polyurethane/silica nanocomposite fibers with good mechanical properties were prepared by electrospinning, using colloidal silica as the source of silica and dimethyl formamide as the solvent. The fiber morphology was examined by field emission scanning electron microscopy. The average fiber diameter is about 0.8 mu m with 0-10 wt % silica, and silica nanoparticles were observed on all fiber surfaces. X-ray photoelectron spectroscopy analysis of Si in combination with transmission electron microscopy observation suggest that silica nanoparticles have a fairly uniform distribution in the fibers rather than enriching on the fiber surfaces. Tensile tests show that the incorporation of silica nanoparticles can bring about a significant reinforcing effect without decreasing the ductility. The reinforcing effect is further confirmed by dynamic mechanical analysis. The thermoplastic polyurethane/silica composite fiber mats can adsorb gold nanoparticles after further treatment with 3-aminopropyltriethoxysilane, demonstrating that the composite fibers could be used as functional fibers by using the properties of silica nanoparticles. Abstract:The electrospinning of regenerated silk fibroin(SF) and gelatin blended with the composition ratio 70∶30 was performed with 98% formic acid as spinning solvent.The influence of mass concentration and ethanol treatment on the microstructure,mechanical properties and solubility of electrospun SF/gelatin nanofibers was investigated.The pore diameter and porosity of SF/gelatin fiber membrane with different mass concentrations and different thickness were measured.The mouse fibroblasts and human umbilic vein endothelium cells were seed on SF/gelatin nanofibers scaffold.The results showed that: along with the mass concentrations of spinning solution increasing and ethanol treating,the content of β-sheet of SF in electrospun SF/gelatin nanofibers was increased,crystallinity increased,water-dissolved rate declined,and tensile strength enhanced of electrospun SF/gelatin nanofibers;as the mass concentration of spinning solution increased,the pore diameter of electrospun SF/gelatin nanofibers was decreased and the porosity was increased;both the pore diameter and porosity were decreased with the increase of the thickness of electrospun SF/gelatin nanofibers.The SF/gelatin nanofibers scaffold may be a good candidate for both L929 and HUVECs spreading and proliferating.

Abstract:The high crystallinity and sophisticated hierarchical architecture of native animal silk endow it with comprehensive mechanical properties that are superior to those of most synthetic fibers. However, these features also make the direct exfoliation of silk nanofibrils (SNFs) highly challenging. On the other hand, silk-based materials prepared by conventional method (i.e., through silk fibroin aqueous solution) are usually weak, so the preparation methods based on SNFs have attracted much attention in recent years. Herein, a facile and environmentally friendly route is developed to directly exfoliate SNFs from natural Bombyx mori silkworm silk via ammonium persulfate oxidation followed by ultrasonication. The obtained SNFs have a high yield (nearly 40%) and are well dispersed in water in a wide pH range, so they can act as a good starting material to prepare subsequent silk-based materials. The main application of the SNFs demonstrated in this article is a curcumin (Cur)/SNFs hydrogel as wound dressing. In vivo experimental results show that the Cur/SNFs hydrogel significantly enhanced the healing rate of wounds on diabetic mice. Therefore, the preparation method developed in this study provides an efficient way to produce SNFs, which have great potential for a wide range of applications, including as wound dressings for diabetics. A facile and environmentally friendly route is developed to directly exfoliate silk nanofibrils from natural silkworm silk via ammonium persulfate oxidation followed by ultrasonication.
