Multi-Omics Analysis to the Alleviation of Aeromonas Hydrophila-Induced Intestinal-Barrier Damage by the Application of Fos in Megalobrama Amblycephala

Abstract:This study was conducted to elucidate the mitigative effects of FOS on the Aeromonas hydrophila-induced intestinal barrier and their underlying mechanisms. Megalobrama amblycephala were randomly divided into three experimental groups (Control, A. hydrophila, and A. hydrophila + FOS, respectively) with three replicates of 15 fish in each replicate. The feeding trial revealed that A. hydrophila destroyed tight-junction structure, and increased intestinal permeability, causing intestinal mucosal pathological abnormal changes in fish. Additionally, results indicated that A. hydrophila affected the secretion of the immune barrier, induced the imbalance of the intestinal flora, and disturbed the stability of the biological barrier. Dietary FOS not only ameliorated the injured intestinal structure, as control, but also in part improved the intestinal apical junction complex, however, which was partly implicated in the upregulation of the tight junction and adhesion junction protein pathways by transcriptomic analysis. Next, the study found that dietary FOS supplementation could improve the intestinal microbiota disturbed induced by A. hydrophila and be beneficial for permeability. Meanwhile, fish fed with FOS up-regulated the expressions of genes related to apoptosis, antigen presentation, and T cell-mediated immune response in intestine compared with A. hydrophila group. In this study, combined microbiome and transcriptome analyses also showed the correlation between the abundance and above-mentioned gene expressions. To our knowledge, this is the first study that explored the FOS on the intestinal barrier, and their interaction network induced by A. hydrophila. The study not only enhances the development of gut damage treatment options induced by A. hydrophila, but also assist the present understanding the role of the gut microbiome as mediator of beneficial effects of FOS diets.
