Abstract:Changes in qualities of Red Fuji' and Golden Delicious' apples in response to different heat treatments (45 and 60 degrees C) following cold storage were investigated. Quality indices including firmness, titratable acidity, total phenolic content, antioxidant capacity, colour parameters and microstructure were measured. Results indicated heat treatment at both temperatures for 3-h hastened loss of titratable acidity in Red Fuji' and Golden Delicious' apples, but maintained firmness in both cultivars. However, heat treatment at lower temperature (45 degrees C) best maintained total phenolic compounds and antioxidant capacity in Red Fuji' apples throughout storage, whereas there was no difference between heat treatments in Golden Delicious' apples. Examination by scanning electron microscope (SEM) revealed that the effect of heat treatment at different temperatures on Red Fuji' and Golden Delicious' apples was structurally different. Apple tissue that had been heat treated at elevated temperature (60 degrees C) for 3h was associated with fractured cell walls and collapse of cells.

Abstract:The effect of 5 ℃ storage on the morphological and physiological changes were investigated in the oriental lily bulbs which harvested in mountainous area in Zhejiang province during long term storage.The inner sprouts were elongating continuously during the low temperature storage,and it was less than 1 cm reached to the top of the bulb after 60 days.The bud breaks the top after 1 month's continuous storage.During the first 2 months,starch contents decreased significantly and the outer layer scale decreased most among 3 tiers.Starch contents tend to become rising slowly in the second 2 months.The contents of the total soluble sugars and sucrose increased continuously.The contents of total soluble sugars of middle layer scale increased most,reached to 196.11%.

Abstract:Sweet potato (Ipomoea batatas (L.) Lam.) is a commercially relevant food crop with high demand worldwide. This species belongs to the Convolvulaceae family and is native to tropical and subtropical regions. Storage temperature and time can adversely affect tuberous roots’ quality and nutritional profile. Therefore, this study evaluates the effect of storage parameters using physicochemical and transcriptome analyses. Freshly harvested tuberous roots (Xingxiang) were stored at 13 °C (control) or 5 °C (cold storage, CS) for 21 d. The results from chilling injury (CI) evaluation demonstrated that there was no significant difference in appearance, internal color, weight, and relative conductivity between tuberous roots stored at 13 and 5 °C for 14 d and indicated that short-term CS for 14 d promoted the accumulation of sucrose, chlorogenic acid, and amino acids with no CI symptoms development. This, in turn, improved sweetness, antioxidant capacity, and nutritional value of the tuberous roots. Transcriptome analyses revealed that several key genes associated with sucrose, chlorogenic acid, and amino acid biosynthesis were upregulated during short-term CS, including sucrose synthase, sucrose phosphate synthase, phenylalanine ammonia-lyase, 4-coumarate-CoA ligase, hydroxycinnamoyl-CoA quinate hydroxycinnamoyltransferase, serine hydroxymethyltransferase, alanine aminotransferase, arogenate dehydrogenase, and prephenate dehydratase. These results indicated that storage at 5 °C for 14 d could improve the nutritional quality and palatability of sweet potato tuberous roots without compromising their freshness.

Abstract:The effects of 5℃ storage on the morphological and physiological changes were investigated on the dormancy in bulbs of Lilium oriental hybrids.The inner sprouts were elongating continuously during the low temperature storage,and it was indicated that sprouts are less than 1cm reached to the top of the bulb was as the index of dormancy release in lily bulbs.Starch contents decreased significantly during the low temperature storage,while contents of the total soluble sugars and sucrose increased continuously.It was sharp decrease of starch contents occurred within the initial 10 d cold treatment,both in interior and exterior scales.The contents of total soluble sugars and starch start to decline at the peaks appeared at 50 d in cold storage for cultivar 'Sorbonne'.It was suggested that the turning point of sugar contents in scales should be regarded as the beginning of sprout in lily bulbs.

Abstract:Carbohydrate contents and related enzymes activities were performed to investigate the effect of low temperature storage on the bulbs' dormancy of Oriental Lily ‘Sorbonne’. Bulbs were stored in moist peat moss for 8 weeks at 2℃, 5℃ and 8℃, respectively. The starch contents decreased continuously in interior and exterior scales, while the contents of total soluble carbohydrates and reducing sugar were increased. Activities of CAT, POD and SOD declined significantly, especially in the first week of cold storage. A peak value in α-amylase activity appeared in the 3~ rd week. Low activity levels of all enzymes presented after the 4~ th week. The results indicated that no obvious difference between the three low temperatures, and overcoming dormancy in bulbs of oriental lily ‘Sorbonne’ started in the 7~ th week.

Abstract:BACKGROUND: Bamboo shoot is sensitive to chilling, and low-temperature injury is a major limitation to its long-term storage. Studies on the causes of postharvest deterioration in bamboo shoot are few. The aim of the present study was to evaluate the effects of ethylene and 1-methylcyclopropene (1-MCP) treatments on the physiological and biochemical responses of bamboo shoot and their ability as a postharvest tool to alleviate chilling injury and lignification of bamboo shoot during storage at 2 degrees C.RESULTS: Disease incidence, electrical conductivity (EC), respiration and ethylene production of control bamboo shoot increased after about 2 - 3 weeks of cold storage, associated with higher chilling injury (CI) index. Increased shoot firmness during cold storage was positively correlated with higher lignin and cellulose accumulation, and this accumulation of lignin in flesh tissue was also positively correlated with the activities of phenylalanine ammonia lyase (PAL), cinnamyl alcohol dehydrogenase (CAD) and peroxidase (POD). Ethylene treatment was associated with higher disease incidence, CI index, EC, respiration and ethylene production, enhanced lignin and cellulose accumulation and accelerated the activities of PAL, CAD and POD. In contrast, 1-MCP treatment was associated with lower respiration, ethylene production, CI index and EC, reduced lignin and cellulose accumulation and retarded the activities of PAL, CAD and POD.CONCLUSION: The present findings throw light on the role of ethylene in postharvest physiological disorders such as chilling injury and lignification and suggest that 1-MCP could be used commercially to control these disorders in bamboo shoot during cold storage. Abstract:Bamboo shoots are highly nutritious but possess poor shelf life after harvest due to rapid decline of tenderness mostly result from lignification processes. Present investigation was performed to evaluate the potential of gamma radiation as a postharvest technology to preserve bamboo shoots. The effects of 0.5 kGy gamma radiation on quality and lignification of bamboo shoots during storage at 2 degrees C were investigated. The results showed that gamma radiation reduced the decay rate of bamboo shoots by 71% at the end of storage. It also significantly retarded ethylene production through controlling the activities of ACC synthase (ACS) and ACC oxidase (ACO), which may contribute to inhibition of lignin synthesis. Meanwhile, gamma radiation decreased the activities of phenylalanine ammonia-lyase (PAL), cinnamyl alcohol dehydrogenase (CAD) and peroxidase (POD), which are associated with lignin accumulation, thus reduced lignin content by 12.5% than that of the control after twenty-eight days of storage. These results demonstrated the potential of the gamma radiation as a promising postharvest technique to maintain the quality and inhibit the lignification of bamboo shoots. (C) 2015 Elsevier B.V. Abstract:A method of clonal germplams preservation utilizing dehydrated somatic embryos and cool temperature storage conditions was demonstrated. Somatic embryos of grapevine (Vitis vinifera L) Autumn Seedless and Chardonnay were produced from suspension cultures. After washing twice with sterile water mature somatic embryos were blot-dried and placed on sterile filter paper in an open Petri dish in a laminar flow hood until they reached about 25% of their initial weight. Approximately 300 dried embryos were placed in each sterile 90×15 mm Petri dish, which was tightly sealed with two layers of ParafilmTM. Sealed dishes were stored in the dark at 4°C in a standard refrigerator. Samples of 25–60 individual dehydrated somatic embryos were periodically tested for viability by placing them on solidified MS medium for germination and plant regeneration. After 42 mo. of dehydrated storage, 90% of the somatic embryos regenerated into plants. To further test utility, of this storage method dehydrated embryos stored for 12 and 26 mo. were shipped from Florida to Washington where 75 and 87.5% regenerated into plants, respectively. Cool temperature storage of dehydrated somatic embryos is a simple and inexpensive method of clonal, germplasm preservation when compared to alternatives such as cryopreservation.
Vitrification Cryo-Foil for Apple Cryopreservation and the Seesaw Effect Between Shoot Recovery and Virus Eradication

Abstract:Abstract Improvements of existing cryopreservation protocols are necessary to facilitate long-term preservation of plant germplasm and the cryotherapy-effect of pathogen eradication. This study reported a vitrification (V) cryo-foil/plate methods for cryopreservation of shoot tips and cryotherapy effect in ‘Pink Lady’ apple. In V cryo-foil/plate protocols, shoot tips were first attached onto aluminum foils/plates using calcium alginate before other procedures. Shoot tips cryopreserved by V cryo-foil required 6.1 weeks to fully recover and 53% of shoot regrowth was obtained, comparable to the Dv cryopreservation. Similar regrowth levels were produced between applying V cryo-foil and Dv cryopreservation to another 4 Malus genotypes. Histological observations in shoot tips cryopreserved by Dv and V cryo-foil found only those with surviving apical dome and leaf primordia (LPs) could recover after cryopreservation. In apical meristem of shoot tips cryopreserved by Dv and V cryo-foil, higher surviving probability was detected from the V cryo-foil protocol, and the young LPs showed the highest level of surviving. Virus detection in cryo-derived plants showed apple stem grooving virus and apple chlorotic leaf spot virus were all preserved after cryopreservation, and higher eradication efficiency of apple stem pitting virus (70%) was produced by Dv than the 55% of V cryo-foil. These results supported applying V cryo-foil as an improvement to the widely applied Dv method in shoot tip cryopreservation, and also revealed a seesaw mode between shoot recovery and cryotherapy effect. Once the seesaw moves to increase the recovery after cryopreservation, the cryotherapy-effect on the other side would be decreased.
Plant Regeneration from Leaf and Stem Segments of Shoots Raised in vitro from Mature Apple Trees

Abstract:Plants were regenerated from leaf segments of shoots raised in vitro from mature apple trees of the cultivars Åkerö, McIntosh, McIntosh Wijcik, Gravenstein and the apple rootstock M26.The regeneration capacity was influenced by wounding, macroelements, light conditions, polarity and maturity of the leaves. The highest number of shoots per leaf segment was formed using young expanding leaves on a medium based on N6 macroelements plus 22 µM BAP and 1 µM NAA. Dark treatment during the first 3 weeks of culture and placing the leaf segments on the medium with the abaxial surface uppermost enhanced shoot formation. The morphogenetic response for stem segments was very low since only McIntosh Wijcik formed shoots and at a low frequency. The histological studies revealed both embryoidlike structures and adventitious shoots. Shoots regenerated from both leaves and meristems were rooted and placed in the greenhouse for comparison. Trees of the cultivars McIntosh, McIntosh Wijcik and M26 were planted in the field for further evaluation of growth habit and fruit setting.

Abstract:Establishment of a new method for improved shoot tip cryopreservation is crucial to facilitate the long-term preservation of plant germplasm as well as the use of cryotherapy for pathogen eradication. The present study reported a vitrification (V) cryo-foil method for shoot tip cryopreservation and virus eradication in apple. Shoot tip regrowth levels after cryopreservation were comparable among V cryo-foil (53%), V cryo-plate (46%) and conventional droplet vitrification (Dr-vi, 48%). The V cryo-foil is more efficient to perform than Dr-vi as more shoot tips can be cryopreserved by one person. In the histological study applying an image-overlaying strategy, shoot tips cryopreserved by V cryo-foil showed a higher survival chance in the youngest leaf primordia than in the apical dome. When V cryo-foil was tested for virus eradication, fifty-five percent (55%) of cryo-derived shoots were free of the apple stem pitting virus (ASPV), while none and less than 10% were free of the apple stem grooving virus (ASGV) and the apple chlorotic leaf spot virus (ACLSV), respectively. Thus, these two viruses were efficiently preserved by V cryo-foil cryopreservation. Noticeably, although the shoot regrowth level was reduced to 27%, a higher frequency (81%) of ASPV eradication was achieved when a reduced duration of cryoprotectant exposure was applied in V cryo-foil, supporting the use of insufficient cryoprotection for improved virus eradication.

Abstract:By comparing nine fresh broad bean cultivars,Sucan No.2,Haimen Daqingpi,Shangyu Tianjiqing, American Horsebean,Lingxi Yicun,Tongxian No.6,Japanese Dabaipi,Cixi Dabaican,independent developed new line(long pod broad bean),as experimental materials to conduct low temperature preservation test. The results indicated that:in terms of pod storage,Shangyu Tianjiqing kept the best preservation effect,Cixi Dabaican,Japanese Dabaipi and long pod broad bean took the second place under the storage conditions of 2℃ and 28 days;in terms of peeled-bean storage,Japanese Dabaipi and Cixi Dabaican preserved the best under the storage conditions of 2℃ and 21 days.
Recent advances in the cryopreservation of shoot-derived germplasm of economically important fruit trees of Actinidia, Diospyros, Malus, Olea, Prunus, Pyrus and Vitis

Abstract:This paper presents the advances made over the last decade in cryopreservation of economically important vegetatively propagated fruit trees. Cryopreservation protocols have been established using both dormant buds sampled on field-grown plants and shoot tips sampled on in vitro plantlets. In the case of dormant buds, scions are partially dehydrated by storage at -5 °C, and then cooled slowly to -30 °C using low cooling rates (c.a. 1 °C/h) before immersion in liquid nitrogen. After slow rewarming and rehydration of samples, regrowth takes place either through grafting of buds on rootstocks or excision of apices and inoculation in vitro. In the case of shoot tips of in vitro plantlets, the cryopreservation techniques employed are the following: controlled rate cooling procedures involving slow prefreezing followed by immersion in liquid nitrogen or vitrification-based procedures including encapsulation-dehydration, vitrification, encapsulation-vitrification and droplet-vitrification. The current status of cryopreservation for a series of fruit tree species including Actinidia, Diospyros, Malus, Olea, Prunus, Pyrus and Vitis is presented. Routine application of cryopreservation for long-term germplasm storage in genebanks is currently limited to apple and pear, for which large cryopreserved collections have been established at NCGRP, Fort Collins (USA), using dormant buds and in vitro shoot tips, respectively. However, there are a growing number of examples of pilot scale testing experiments under way for different species in various countries. Progress in the further development and application of cryopreservation techniques will be made through a better understanding of the mechanisms involved in the induction of tolerance to dehydration and cryopreservation in frozen explants.
Optimization of Short-Term Hot-Water Treatment of Apples for Fruit Salad Production by Non-Invasive Chlorophyll-Fluorescence Imaging

Abstract:For fresh-cut salad production, hot-water treatment (HWT) needs optimization in terms of temperature and duration to guarantee a gentle and non-stressing processing to fully retain product quality besides an effective sanitation. One major initial target of heat treatment is photosynthesis, making it a suitable and sensitive marker for HWT effects. Chlorophyll fluorescence imaging (CFI) is a rapid and non-invasive tool to evaluate respective plant responses. Following practical applications in fruit salad production, apples of colored and of green-ripe cultivars ('Braeburn', 'Fuji', 'Greenstar', 'Granny Smith'), obtained from a local fruit salad producer, were hot-water treated from 44 to 70 °C for 30 to 300 s. One day after HWT and after 7 days of storage at 4 °C, CFI and remission spectroscopy were applied to evaluating temperature effects on photosynthetic activity, on contents of fruit pigments (chlorophylls, anthocyanins), and on various relevant quality parameters of intact apples. In 'Braeburn' apples, short-term HWT at 55 °C for 30 to 120 s avoided any heat injuries and quality losses. The samples of the other three cultivars turned out to be less sensitive and may be short-term heat-treated at temperatures of up to 60 °C for the same time. CFI proved to be a rapid, sensitive, and effective tool for process optimization of apples, closely reflecting the cultivar- or batch-specificity of heat effects on produce photosynthesis.

Abstract:In vitro cultivation and cryopreservation techniques are essential tools for genetic diversity conservation and pathogen-free plant propagation of horticultural crops. The optimisation of cryopreservation protocols typically focuses on minimising the negative effects of pretreatment with cryoprotectors (CPs), cryogenic freezing (CF) treatment, and recovery procedures on explants. However, the impact of in vitro and CF techniques on plant-associated microbiota remains poorly understood, and their potential to improve plant adaptation after cryopreservation is underexplored. The aim of the present study was to investigate in vitro shoot culture and cryopreservation-induced changes in the endophytic bacterial diversity of two sweet cherry cultivars and to assess the potential of an inoculum of bacterial isolates to improve the growth of shoot culture after CF.

Abstract:Apples are seasonal fruits, and it is important to prepare them adequately for storage and ensure proper storage conditions. In this study, we used ten different apple cultivars: ‘Alva’, ‘Auksis’, ‘Connell Red’, ‘Cortland’, ‘Ligol’, ‘Lodel’, ‘Noris’, ‘Rubin’, ‘Sampion’, and ‘Spartan’. We studied the qualitative and quantitative composition of phenolic compounds in the apple and apple extracts antioxidants activity before placing them in the controlled atmosphere chambers and again at the end of the experiment, eight months later. Different concentrations of O2, CO2, and N2, constant temperature, relative humidity, and removal of endogenous ethylene were continually maintained. HPLC analysis showed that the highest amount of 2265.7 ± 152.5 μg/g of chlorogenic acid was found in apple samples of the ‘Auksis’ cultivar stored under variant IV conditions. Different concentrations of gas in the controlled atmosphere chambers caused changes in antioxidant activity in whole apple and apple peel extracts. In our study, we found that the antioxidant activity of apple extracts varied between samples of different apple cultivars and depended on the composition of the controlled atmosphere. Determining the optimal storage conditions is beneficial to providing the consumers with apples that have a known and minimally altered chemical composition of phenolic compounds and the strongest antioxidant activity, which determine the use of apples in the healthy food chain.
Metabolomic Analysis of the Effect of Freezing on Leaves of Malus sieversii (Ledeb.) M.Roem. Histoculture Seedlings

Abstract:Malus sieversii (Ledeb.) M.Roem. is the ancestor of cultivated apples, and is an excellent germplasm resource with high resistance to cold. Artificial refrigerators were used to simulate the low temperature of -3 °C to treat Malus sieversii (Ledeb.) M.Roem. histoculture seedlings. Observations were performed to find the effects of freezing stress on the status of open or closed stomata, photosystems, and detection of metabolomic products in leaves of Malus sieversii (Ledeb.) M.Roem. histoculture seedlings. The percentage of closed stomata in the Malus sieversii (Ledeb.) M.Roem. histoculture seedlings increased, the maximum fluorescence (Fm') excited by a strong light (saturating pulse) was weakened relative to the real-time fluorescence in its vicinity, and the quantum yield of unregulated energy dissipation was increased in PSII under freezing stress. The metabolites in the leaves of the Malus sieversii (Ledeb. M.Roem.) histoculture seedlings were analyzed by ultra-performance liquid chromatography-tandem mass spectrometry using CK, T12h, T36 h, and HF24h. Results demonstrated that cold stress in the Malus sieversii (Ledeb.) M.Roem. histoculture seedlings led to wilting, leaf stomatal closure, and photosystem damage. There were 1020 metabolites identified as lipids (10.2%), nucleotides and their derivatives (5.2%), phenolic acids (19.12%), flavonoids (24.51%), amino acids and their derivatives (7.75%), alkaloids (5.39%), terpenoids (8.24%), lignans (3.04%), organic acids (5.88%), and tannins (0.88%). There were 110 differential metabolites at CKvsT12h, 113 differential metabolites at CKvsT36h, 87 differential metabolites at T12hvsT36h, 128 differential metabolites at CKvsHF24h, 121 differential metabolites at T12hvsHF24h, and 152 differential metabolites at T36hvsHF24h. The differential metabolites in the leaves of the Malus sieversii (Ledeb.) M.Roem. seedlings grown under low-temperature stress mainly involved glycolysis, amino acid metabolism, lipid metabolism, pyrimidine metabolism, purine metabolism, and secondary metabolite metabolism. The Malus sieversii (Ledeb.) M.Roem. seedlings responded to the freezing stress by coordinating with each other through these metabolic pathways. The metabolic network of the leaves of the Malus sieversii (Ledeb.) M.Roem. histoculture seedlings under low temperature stress was also proposed based on the above pathways to deepen understanding of the response of metabolites of Malus sieversii (Ledeb.) M.Roem. to low-temperature stress and to lay a theoretical foundation for the development and utilization of Malus sieversii (Ledeb.) M.Roem. cultivation resources.
Synthetic seeds for in vitro preservation of Asparagus officinalis L.

Abstract:Abstract Background Asparagus ( Asparagus officinalis ) is a perennial vegetable of economic importance for its high nutritional and medicinal value. Male plants are more desirable because of their higher spear yield. Therefore, the aim of this study was to evaluate the efficiency of the gibberellin inhibitors; paclobutrazol (PBZ) and cycocel (CCC) for in vitro preservation by encapsulation of in vitro-derived shoot tips of the superior germplasm of the first generation of asparagus male hybrid; cultivar Mary Washington 500W. Methods An efficient technique for synthetic seeds production was achieved, consisting of 3% (w/v) Na-alginate dissolved in water, Murashige and Skoog (MS) medium, MS medium with the growth inhibitors; PBZ and CCC at different concentrations, and 0.1 M calcium chloride (CaCl 2 .2H 2 O). Synthetic seeds were stored at 4 °C and then cultured after different storage durations (0, 2, 4, 8, 12, 16, 20 and 24 weeks) on MS medium supplemented with 1 mg l −1 kinetin, 0.2 mg l −1 naphthalene acetic acid and 0.5 mg l −1 gibberellic acid for shoot tip multiplication; then, recovered shoots were transferred to MS medium supplemented with 1 mg l −1 indole butyric acid and 0.5 mg l −1 PBZ for rooting. Results The beads containing 1 mg l −1 PBZ considered the optimum for producing complete well-developed plantlets of Asparagus officinalis from recovered shoot tips after 24 weeks of storage that successfully acclimatized in the greenhouse. Conclusions This protocol is efficient for in vitro preservation by encapsulation of shoot tips and regeneration of Asparagus officinalis F1 male hybrid (Mary Washington 500w), using anti-gibberellin plant growth regulators within the beads to prolong the duration of storage and provide a continuous supply of the plant.
