Abstract:Peroxynitrite (ONOO-) has been proven to participate in various physiological and pathological processes, and may also be a contributing factor in many diseases. In view of this, there is a need to develop detection tools for unambiguously tracking a small amount of endogenous ONOO- to reveal its exact mechanisms. In this paper, a colorimetric and red-emitting fluorescent probe Red-PN, based on a rhodamine-type fluorophore and hydrazide reactive site is described. The probe Red-PN possesses the advantages of rapid response (within 5 s), visual color change (from colorless to pink), preeminent sensitivity (detection limit = 4.3 nM) and selectivity. Because of these outstanding performances, it was possible to accurately detect endogenous ONOO-. It was encouraging that the probe Red-PN could be used effectively for tracking the relatively low levels of endogenous and exogenous ONOO-in living cells and zebrafish. Thus, it is envisioned that the probe Red-PN would have promising prospects in applications for imaging ONOO- in a variety of biological settings.

Abstract:A growing body of evidence shows that peroxynitrite (ONOO-) is involved in a large number of diseases, signal conduction, and immunoregulation in living systems. The determination of intracellular ONOO- attracts increasing attention. In this paper, a novel dicyanoisophorone-derived borate-containing near-infrared (NIR) fluorescent probe NIR-PN for sensitive detection of ONOO- was designed. Probe NIR-PN holds ultrasensitivity (DL = 5 nM), high specificity, and fast response (<3 s) for ONOO-. In addition, based on the significant advantage of dicyanoisophorone-derived fluorophore, probe NIR-PN exhibited NIR emission (lambda(em) = 660 nm) and a large Stokes shift (similar to 150 nm). Importantly, the biological application potential of probe NIR-PN was effectively verified by visualizing ONOO- levels in living cells and zebrafish.

Abstract:Peroxynitrite (ONOO−), as an important reactive oxygen species (ROS), holds great potential to react with a variety of biologically active substances, leading to the occurrence of various diseases such as cancer and neurodegenerative diseases. In this work, we developed a novel mitochondria-localized fluorescent probe, HDBT-ONOO−, which was designed as a mitochondria-targeting two-photon fluorescence probe based on 1,8-naphthylimide fluorophore and the reactive group of 4-(bromomethyl)-benzene boronic acid pinacol ester. More importantly, the probe exhibited good biocompatibility, sensitivity, and selectivity, enabling its successful application in imaging the generation of intracellular and extracellular ONOO−. Furthermore, exogenous and endogenous ONOO− products in live zebrafish were visualized. It is greatly expected that the designed probe can serve as a useful imaging tool for clarifying the distribution and pathophysiological functions of ONOO− in cells and zebrafish.

Dual NIR-channel Fluorescent Probe for Detecting ONOO− in Vitro and Vivo

Abstract:As one of endogenous reactive oxygen species (ROS), peroxynitrite (ONOO-) performs various functions in both pathological and physiological mechanisms. In this work, an optical and near-infrared (NIR) fluorescent probe (NX), which based on 3-dihydro-1H-xanthene and 2-dicyanomethylene-3-cyano-4,5,5-trimethyl-2,5-dihydrofuran (TCF) group was designed and prepared to detect ONOO-. This probe revealed an obvious optical and a fluorescent response when ONOO- was present and it exhibited higher selectivity over other ROS. Especially, the dual NIR fluorescence changes at 660 and 800 nm allowed quantitative detection of ONOO- in the range of 15-40 mu M, and the detection limit was 82 nM. Finally, the probe was effectively employed to visualize exogenous and endogenous ONOO- in HepG2 cells and zebrafish, respectively. All the results indicated the dual NIRchannel probe could serve as a potent detecting tools in studying ONOO- in vitro and in vivo.

Abstract:As a reactive nitrogen species (RNS), peroxynitrite (ONOO-) is known for its crucial functions in biological systems. Excessive formation of ONOO- is very closely related to the pathogenesis of a series of diseases, and thus, effective methods are needed for highly sensitive detection of ONOO-. Herein, a simple fluorescent probe RHHP-PN was proposed to detect ONOO-, and manifested obvious color changes from colorless to red in less than 10?s. Besides, it is not only suitable for quantitative detection of ONOO-, but also satisfactory in distinguishing ONOO- from other related species. What is particularly noteworthy is the fluorescence bioimaging for tracing endogenous and exogenous ONOO- in living cells and zebrafish. Therefore, probe RHHP-PN could be used as an imaging tool to visualize intracellular ONOO- in biological systems.

Abstract:Peroxynitrite (ONOO-), an endogenous reactive nitrogen species, plays an important role in maintaining intracellular homeostasis. Abnormal levels of ONOO- in cells could cause protein oxidation which is confirmed that related with Alzheimer's diseases, so accurate monitoring of ONOO- in cells is crucial. Herein, a novel fluorescent probe (XPC) based on dicyanomethylene-4H-benzothiopyran was developed by regulating its intramolecular charge transfer (ICT) effect to detect ONOO-. Once reaction with ONOO-, the fluorescence of XPC was turned on and the emission wavelength could reach up to 750 nm. Furthermore, XPC exhibited satisfactory performances for ONOO- such as large Stokes shift (200 nm), good sensitivity (Limit of detection = 13 nM), high selectivity to ONOO- over other a reactive nitrogen species (RNS)/reactive oxygen species (ROS). More importantly, XPC was successfully applied for monitoring the fluctuations of ONOO- in living cells.

Abstract:In this study, we present a novel near-infrared (NIR) fluorescent probe Nile-ONO designed for the selective and sensitive detection of ONOO − . The probe Nile-ONO employed Nile red as the fluorophore, with diphenylphosphinate serving as the reaction site. In the presence of ONOO − , the probe Nile-ONO exhibits remarkable fluorescence enhancement at 659 nm, with a response time of less than 20 min and a low detection limit of 0.32 μM. Importantly, MTT assays demonstrate low cytotoxicity in living cells. Furthermore, Nile-ONO has excellent imaging capabilities for endogenous ONOO − . Overall, this work introduces a valuable new method for the rapid detection of ONOO − in biological systems.

Abstract:Peroxynitrite (ONOO − ) is one of the important active nitrogen/reactive oxygen species that plays various roles in biological processes, such as inducing apoptosis and necrosis. Recent studies have shown that a significant increases in ONOO − content during tumor development, which is closely related to the level of oxidative stress within the tumor. It has been found that herbicide paraquat (PQ) can significantly increase the level of ONOO − in cells. Therefore, accurate monitoring abnormal changes in ONOO − caused by environmental hazardous materials and tumors is helpful in promoting the diagnosis and treatment of oxidative stress diseases (tumors), evenly environmental detection. Currently, traditional fluorescent probes for ONOO − detection have background interference. To address this, we developed a chemiluminescent probe ( CL-1 ) and a fluorescent probe ( Flu-1 ), using diphenyl phosphonate as a recognition group. CL-1 shows extremely sensitivity (9.8 nM), a high signal-to-noise(S/N) ratio (502), and excellent bioimaging capabilities compared to fluorescent probe ( Flu-1 ). We have successfully used CL-1 to detect ONOO − produced by PQ stimulated cells, as well as endogenous ONOO − in tumor cells, mice, and human liver cancer tissues. Therefore, CL-1 can not only be a valuable tool for visualizing tumor and studying the role of ONOO − in tumor pathology, but the probe has the potential to be a powerful molecular imaging tool for exploring the complex biological role of ONOO − in a variety of biological Settings.

Abstract:Developing a reliable tool for monitoring peroxynitrite (ONOO-) in the complex organisms is critical to uncover its detailed cellular functions in numerous physiological and pathological processes. Thus, a novel ratiometric fluorescent probe was reasonably designed for ONOO-, in which the rhodol-HBT conjugate dye was chosen as a fluorophore, and hydrazine moiety was adopted as recognition receptor of ONOO-. The probe exhibited outstanding sensitivity, specificity, and a fast response towards ONOO-. More importantly, the probe with a large emission shift (Delta lambda(em)= 127 nm) has been proven to be capable of tracking ONOO- with high resolution in live cells and zebrafish. This work provides a simple and reliable tool for detecting and imaging ONOO- in the living systems, and thus we anticipate that the designed probe could be used as a powerful imaging tool in unraveling biological functions of ONOO-.

Abstract:Peroxynitrite is a short-lived endogenous reactive species and plays important roles in many physiological and pathological processes. In this work, we synthesized a near-infrared probe based on the structure of benzothiazole derived cyanine for determination of peroxynitrite (ONOO-). The designed probe specifically reacted with ONOO- through oxidative cleavage of conjugated C˭C double bonds and generating the non-fluorescent product. Meanwhile, the characteristic absorption of the probe at 630 nm greatly decreased after reaction with ONOO-, accompanied by drastic color change from bright blue to green yellow, which exhibited a distinct visual feature. It was demonstrated that the probe could be used to measure ONOO- in a dose-response manner and had a detection limit lower as 26 nM. Furthermore, the probe Cy-SN was applied for the imaging of endogenous ONOO- in living cells by confocal microscopy, which showed good cell permeability and low cytotoxicity. Successful application of probe for exogenous colorimetric detection and endogenous fluorescence imaging of ONOO- is suggesting its great potential applications in biological analysis.

Abstract:Peroxynitrite (ONOO−) is a transient and reactive oxidant with significant roles in numerous biological processes. Research indicated the correlation between excessive mitochondrial ONOO− production and various diseases. We developed a mitochondria-targeting, fluorescence-based ratiometric probe using a boronate group for ONOO− recognition and coumarin as the fluorophore. The probe exhibited a 615 nm far-red emission, and a new fluorescence emission at 475 nm developed upon adding ONOO−.The probe possessed high sensitivity and selectivity for ONOO− detection with distinct ratiometric fluorescent output and a low detection limit of 11 nM. Significantly, Probe 1 could monitor ONOO− level fluctuations in biological systems due to its superior spectral properties and low toxicity. Notably, probe 1 has been effectively used for imaging in live HepG2 cells, zebrafish, Arabidopsis thaliana, and liver injury mouse tissues.

Abstract:As a member of the reactive nitrogen species (RNS) family, peroxynitrite (ONOO−) as an oxidant and nitrating mediator plays a significant role in some physiopathologic processes. The excessive production of peroxynitrite anion in a drug-damaged liver is a culprit of hepatotoxicity. The detection of peroxynitrite is of vital importance for the treatment of some diseases including cancer and liver injury. In this study, a novel turn-on fluorescent probe IC-ONOO with new chalcone fluorophore was designed and synthesized for the detection of in vitro and in vivo. The probe responded rapidly towards ONOO− (only within 15 min did the fluorescent intensity maximize), and was endowed with high sensitivity and excellent selectivity. Given the fact that the linear correlation between the fluorescent intensity at 560 nm and the concentrations of the probe ranged from 0 to 9 μM, the limit of detection (LOD) was calculated to be 3.1 × 10−8 M. With all the merits, probe IC-ONOO was qualified as a robust tool to monitor peroxynitrite anion under physiopathologic condition. Moreover, it was successfully applied in the imaging of endogenous peroxynitrite in living MCF-7 cells (Human breast carcinoma cells) and mouse drug-damaged liver tissue with low cytotoxicity. Given all the extraordinary merits, great potential has been seen in its application to other peroxynitrite related diseases.

Abstract:Peroxynitrite (ONOO-), owing to its high oxidative and nitrating stress, is associated with several physiological processes in addition to various pathological processes, including those related to neurodegenerative diseases and cancer. Detection of ONOO- at the cellular level is of great significance to understand its pathogenesis. To this end, a variety of fluorescent probes based on small molecules and nanoparticles (NPs) have been engineered and applied as excellent tools for imaging of ONOO- in cells as well as in their diverse biological applications. In this review, we highlight representative cases of fluorescent probes based on recognition mechanism and emphasize their response type (ratiometric, two-photon, long-wavelength/near-infrared, and targeting) in ONOO- detection in the last five years. We further discuss their design strategy, sensing mechanism, and application in bio-imaging and describe NP-based probes according to diverse nanoplatforms.

Abstract:The fluorescent probe, GXY-ADP-2, with xanthene structure as the fluorescent core was designed and prepared for the selective detection of peroxynitrite (ONOO−). ONOO− can be produced endogenously and exogenously and is a strong oxidant with a short half-life. Oxidative modifications of biomolecules, that can be attributed to the formation of ONOO−, occur in the reactions of biomolecules with secondary ONOO−-derived radical oxidants. Therefore, it is very important to develop a specific fluorescent probe for detecting ONOO− to monitor oxidative stress state. The excitation wavelength and emission wavelength of the probe are 689 nm and 739 nm respectively. In the process of co-incubation with ONOO−, generate a new substance with two internal conjugated structures through a special reaction mechanism, one giving the fluorescence with the excitation wavelength of 347 nm and the emission wavelength of 484 nm with the detection limit of 0.12 μM, and the other that with the excitation wavelength of 433 nm and the emission wavelength of 583 nm with the detection limit of 0.077 μM. The linear dynamic range of the probe is 0–5 μM. Its response is not affected by the other reactive oxygen species, thus can sensitively detect ONOO−. In bioimaging experiments with HepG2 cells, the green and blue cell fluorescence signals (583 nm and 433 nm, respectively) were increased, while the red one (739 nm) was significantly reduced, under lipopolysaccharide (LPS) induced oxidative stress, proving that the probe could sensitively detect ONOO− in living cells. This work provides a new tool for the dynamic changes of ONOO− and oxidative stress processes in biological systems.

Abstract:Peroxynitrite (ONOO- ), a potent biological oxidant, which has a short half-life in physiological conditions, is related to many diseases. Accurate peroxynitrite determination with superior selectivity and sensitivity is important for understanding biological roles of peroxynitrite in different health and disease tissues. Autofluorescence is an inevitable interference in luminescence biodetection and bioimaging, which often reduces signal-to-noise ratio during detection. In this work, a phosphorescent peroxynitrite nanoprobe (MSN-ONOO) which displays two emission bands is prepared by immobilizing two long-lived phosphorescent iridium(III) complexes that are peroxynitrite-activable and -inert, respectively, into water-dispersible mesoporous silica nanoparticles. Owing to the fast response rate, excellent sensitivity and outstanding selectivity of the nanoprobe toward peroxynitrite, it is further used for peroxynitrite determination in vitro and in vivo via ratiometric photoluminescence imaging. More notably, taking advantage of the long-lived phosphorescence of MSN-ONOO, in vivo elevated peroxynitrite is imaged with diminished autofluorescence interference and improved signal-to-noise ratio via time-resolved photoluminescence imaging. As far as it is known, this is the first time for endogenous peroxynitrite detection in vivo via the time-resolved photoluminescence imaging. Furthermore, the production of peroxynitrite in inflamed tissues is visualized.
