Abstract:Novel biodegradable chemically crosslinked polymers, poly(5-hydroxylevulinic acid-co-alpha,omega-diol)s (PHLA-diols), were synthesized from 5-hydroxylevulinic acid and alpha,omega-diols and characterized by Fourier transform infrared spectroscopy, differential scanning calorimetry, thermogravimetric analysis, and dynamic mechanical analysis. The gel content, swelling ratio, tensile properties, and hydrolytic degradation behaviors were also measured and assessed. The glass-transition temperature of the PHLA-diols could be adjusted within a wide range (-50 to 30 degrees C) by the type and feed ratio of the diol. Because of the low glass-transition temperature and crosslink structure, they exhibited certain elastic properties. The tensile modulus, strength, and elongation at break measured at 37 C were 1.4-6.3 MPa, 0.8-1.6 MPa, and 10-25%, respectively. These polymers could be hydrolytically degraded. Abstract:Biodegradable macroporous hydrogels were prepared using a biodegradable crosslinker based on chitosan derivatives. The chitosan crosslinker was synthesized through the amidating reaction of amino groups in the chitosan oligosaccharide (CSO) chains and carboxyl groups in acrylic acid (AA) under the catalysis of 1-ethyl-3-(3-dimethylaminopropyl)-carbodiimidehydro-chloride (EDC) and N-hydroxysuccinimide (NHS). The amidating reaction is able to provide polymerizable carbon-carbon double bonds for CSO. The chemical structure of the resulting compound chitosan oligosaccharide-graft-acrylic acid (CSO-g-AA) was characterized by Fourier transform infrared spectroscopy (FTIR) and H-1 nuclear magnetic resonance (H-1 NMR) spectroscopy. The content of double bonds was measured by titration analysis. The measurement showed a grafting ratio of AA on CSO of about 3.5% (on average two acryoyl groups per CSO-g-AA chain) when the applied molar ratio of AA to CSO was 1.0. Polyacrylamide (PAM) cryogels were synthesized by the free radical polymerization of acrylamide using a modified freezing polymerization method crosslinked by CSO-g-AA. The obtained cryogels were macroporous hydrogels which could be degraded into solutions of linear polymer chains when treated with appropriate enzymes. Then we used snailase as a model enzyme to study the biodegradation process. The degradation was monitored by morphological studies using a confocal laser scanning microscope (CLSM) and scanning electron microscope (SEM), mechanical strength, and swelling ratio. The duration of the degradation process was adjustable from one month to two months when using different concentrations of snailase solutions.

Abstract:A new hydrogel based on a substituted phenyl glucoside with a Schiff base in the aglycon was synthesized, and the self-assembling characteristics was studied. FTIR spectra, UV-vis absorption spectra and X-ray diffraction (XRD) revealed that pi-pi interactions between the Schiff base moieties, hydrogen bonds, and the interdigitated interactions between hydrophobic chains had effects on the formation of the self-assembling hydrogel. Scanning electron microscopic (SEM) and transmission electron microscopic (TEM) observation showed that the three-dimensional hydrogel network was constructed from nanotubes with inner diameters of ca. 75 nm and wall of ca. 20 nm. Abstract:A well-defined poly(ethylene glycol) based hyperbranched thermoresponsive copolymer with high content of acrylate vinyl groups was synthesized via a "one-pot and one-step" deactivation enhanced atom transfer radical polymerization approach, which provided an injectable and in situ crosslinkable system via Michael-type thiol-ene reaction with a thiol-modified hyaluronan biopolymer. The hyperbranched structure, molecular weight, and percentage of vinyl content of the copolymer were characterized by gel permeation chromatography and (1)H NMR. The lower critical solution temperature of this copolymer is close to body temperature, which can result in a rapid thermal gelation at 37 °C. The scanning electron microscopy analysis of crosslinked hydrogel showed the network formation with porous structure, and 3D cell culture study demonstrated the good cell viability after the cells were embedded inside the hydrogel. This injectable and in situ crosslinking hybrid hydrogel system offers great promise as a new class of hybrid biomaterials for tissue engineering.

Abstract:Abstract In this study, poly(ɛ-caprolactone- g -2-hydroxy ethyl methacrylate) [P(CL- g -HEMA)] graft copolymer hydrogels were successfully synthesized through multi-step reactions. For this purpose, firstly, hydroxyl-terminated poly(ɛ-caprolactone) (PCL-OH) was obtained by ring-opening polymerization (ROP) method of ɛ-caprolactone using 3-chlor-1,2-propanediol initiator, which is suitable for ring-opening polymerization method. Then, from the reaction of synthesized PCL-OH and 3-bromopropionyl chloride, a new brominated poly(ɛ-caprolactone) (PCL-Br) was synthesized for use as a functionalized atom transfer radical polymerization (ATRP) initiator. Poly(ɛ-caprolactone- g -2-hydroxy ethyl methacrylate) [P(CL- g -HEMA)] graft copolymer hydrogels were synthesized by “grafting from” atom transfer radical polymerization (ATRP) of 2-hydroxy ethyl methacrylate (HEMA) presence the new synthesized functionalized ATRP initiator (PCL-Br) and hydrogel properties were investigated. The synthesized functionalized initiators and graft copolymer hydrogel were characterized by spectroscopic methods such as 1 H-NMR, FT-IR, TGA, DSC and SEM. The observation of two different decomposition temperatures, respectively, from the TGA analysis results may support the formation of the biblock graft copolymer. A glass transition temperature ( Tg ) of the graft copolymer hydrogel was found by DSC, and this value is between the Tg values of the homopolymers forming the graft copolymer hydrogel. Water swelling values of graft copolymer hydrogels were measured and calculated every 24 h in pure water with pH = 7 at from + 4 to 65 °C. Considering the weight of dry graft copolymer hydrogels, it was seen that water was absorbed at most at + 4 °C. As the temperature increased, the water absorption or swelling of the hydrogel decreased.

Abstract:Hydrogels, promising biological materials, need to have both strong mechanical properties and also inherent self-healing properties. In this work a double cross-linked network (DN) hydrogel was designed and prepared by combining a Diels-Alder click reaction and coordination effects. This DN hydrogel had good thermodynamic properties, anti-EDTA performance and self-healing properties. In addition, the mechanical properties, swelling properties and surface morphology of DN hydrogels can be controlled by adjusting the ratio of Fe3+-catechol. The adjustment of pH value can change the color, crosslinking mode and mechanical properties of the DN hydrogel. This smart hydrogel created from DA click chemistry and coordination effects has significance for guiding the design of new hydrogels with good mechanical properties, self-healing properties and controlled cross-link density.

Abstract:The development of hydrogels based on dextrans, pullulan and lentinan to be used in biomedical applications including tissue engineering is reported. Despite the fact that selected polysaccharides such as hyaluronic acid are well established, little is known, how these polysaccharides can be chemically modified to create hydrogels under controlled conditions. In this study we present a small library of chemically modified polysaccharides which are used for a divergent approach to achieve biomedical relevant hydrogels. In this case the crosslinking is based on thio ether formation between thiol modified donor and vinylsulfone or maleimide modified acceptor components. Successful synthesis of the linker systems and coupling at the polysaccharides, hydrogel formation takes place under physiological conditions. We extended the study by coupling small molecules like adhesion factors for increasing cell compatibility as well as a dye for further studies. The different hydrogels were studied to their rheological properties, water uptake, their permeability, biodegrability and their cytotoxicity.

Abstract:Hydrogel composite was synthesized by radical copolymerization of polyethylene glycol(PEG) and 2-hydroxyethyl methacrylate (HEMA), with azobisisobutyronitrile (AIBN) as an initiator, N,N’-methylene bis(acryl-amide) (Bis) as crosslinking agent. The intermolecular interaction of hydrogel was studied with Fourier transform infrared spectroscopy (FTIR). FTIR investigation illustrates that there was strong interaction between PEG and HEMA based upon intermolecular hydrogen bonds.

Abstract:A novel biocompatible polysaccharide-based hydrogel with double cross-linked network (DN) was designed and developed by combining dynamic imine bond and Diels-Alder (DA) click reaction. Our results showed that the dynamic covalent imine bond led to the hydrogel's self-healing property, while the DA reaction gave the superior thermodynamic property and acid resistance to the hydrogel. At the same time, the double cross-linked structure improved the mechanical properties of hydrogels. Moreover, the DN hydrogel had good biocompatibility, as confirmed by three-dimensional cell encapsulation. On the basis of these excellent properties, the present prepared hydrogel holds great potential in the field of biomaterials.

Abstract:Double crosslinked hydrogels based on a biodegradable polymer were prepared by photocopolymerization of methacrylated hydroxypropyl guar (HPG-MA) and 3-acrylamidophenylboronic acid. Along with irreversible strong covalent crosslinks by methacrylic groups, these hydrogels contained labile boronate crosslinks formed as a result of the interaction of boronic acid with cis-diol moieties of HPG. These hydrogels demonstrated higher elasticity than HPG-MA hydrogels with only irreversible covalent crosslinks. Labile boronate crosslinks not only strengthened the hydrogels but also imparted to them pronounced responsive properties. It was demonstrated that the mechanical properties, the swelling behavior, as well as the uptake and release of some substances from the double crosslinked hydrogel were pH controlled. For instance, the hydrogels could release cationic disinfectant at a rate regulated by pH. Such photocrosslinkable in situ forming hydrogels are very promising for the production of smart coatings that release targeted substances at the desired rate.

Abstract:A new method for the Synthesis of hyperbranched polymers involving the use of AB, macromonomers containing linear units have been investigated. Two types of novel hyperbranched polyurethanes have been synthesized by a one-pot approach. The structures of monomers and polymers were characterized by elemental analysis, H-1 NMR, C-13 NMR, Fourier transform infrared spectroscopy, gel permeation chromatography, and thermogravimetric analysis. The hyperbranched polymers have been proven to be extremely soluble in a wide range of solvents. Polymer electrolytes were prepared with hyperbranched polymer, linear polymer as the host, and lithium perchlorate (LiClO4) as the ion source. Analysis of the isotherm conductivity dependence of the ion concentration indicated that these hyperbranched polymers could function as a "solvent" for the lithium salt. Abstract:A structurally dense hydrogel, with strong hydrogen bonding networks, was formed from poly(vinyl alcohol), sodium alginate, and oligomeric proanthocyanidins, using a combination of freeze–thaw cycles and calcium ion cross-linking. The structure of the hydrogel was characterized by scanning electron microscopy and Fourier transform infrared spectroscopy. Mechanical testing and thermogravimetric analysis showed that incorporation of proanthocyanidins enhanced both the mechanical properties and the thermal stability of the hydrogel. The hydrogel was also demonstrated to have excellent ultraviolet resistance and antioxidant properties. The hydrogel was further shown that this hydrogel is also capable of generating electrochemical reactions, which strongly suggests that this hydrogel has exciting potential in many fields.

Abstract:Perfluroalkyl-terminated hyperbranched polymer was synthesized based on perfluorooctanoyl chloride grafting on hyperbranched polyglycerol and characterized by NMR, IR, GPC, EA and DSC. Hyperbranched polyglycerol and perfluorooctanoyl chloride were synthesized by cationic ring-opening polymerization of glycidol and by the reaction of perfluorooctanoic acid and SOCl_2 respectively. In addition, this kind of polymer has potential applications in biomedical materials field due to its good biologic compatibility and special structure with the core hydrophilic and the shell hydrophobic and lipophobic.

Abstract:Novel biodegradable hydrogels by photo-cross-linking macromers based on polyphosphoesters and poly(ethylene glycol) (PEG) are reported. Photo-cross-linkable macromers were synthesized by ring-opening polymerization of the cyclic phosphoester monomer 2-(2-oxo-1,3,2-dioxaphospholoyloxy) ethyl methacrylate (OPEMA) using PEG as the initiator and stannous octoate as the catalyst. The macromers were characterized by 1H NMR, Fourier transform infrared spectroscopy, and gel permeation chromatography measurements. The content of polyphosphoester in the macromer was controlled by varying the feed ratio of OPEMA to PEG. Hydrogels were fabricated by exposing aqueous solutions of macromers with 0.05% (w/w) photoinitiator to UV light irradiation, and their swelling kinetics as well as degradation behaviors were evaluated. The results demonstrated that cross-linking density and pH values strongly affected the degradation rates. The macromers was compatible to osteoblast cells, not exhibiting significant cytotoxicity up to 0.5 mg/mL. "Live/dead" cell staining assay also demonstrated that a large majority of the osteoblast cells remained viable after encapsulation into the hydrogel constructs, showing their potential as tissue engineering scaffolds.

Abstract:A poly(vinyl alcohol) hydrogel was prepared by coupling poly(vinyl alcohol) with epichlorohydrin as the cross-linking agent. The structure of the hydrogel was characterized by FTIR and GPC techniques. Various amounts of water were added into the dry gel to swell it, and the quantity of water in various states in the partially swollen hydrogel was determined by DSC technique. The analytical results indicate that the water introduced into the dry gel first combines with the hydrophilic groups of the network chains through hydrogen bond forming non-freezable water. The weight ratio of the non-freezable water to dry gel in the hydrogels is about 0.20. After the non-freezable water is saturated, the additional water penetrates the network space and exists simultaneously both in the freezable and free water states until reaching equilibrium swelling.

Abstract:Different dense and porous biodegradable matrices based on solely atelocollagen, or with different atelocollagen and hyaluronic acid derivative ratios, were obtained by varying feeding formulations, cross-linking reaction parameters, and preparative protocols. The compositions and methods for forming hydrogels through a combination of physical and chemical cross-linking processes are provided. The chemical cross-linking was mainly mediated by a synthetic component, a poly(ε-caprolactone) reactive derivative, aiming the development of new hybrid hydrogels with tailored characteristics by an appropriate use of the advantages offered by the included natural and synthetic components and the selection of the preparative procedure. The structure and morphology of the 3D hybrid materials were comparatively investigated by means of Fourier-transform infrared spectroscopy (FTIR), differential scanning calorimetry (DSC), X-ray diffraction (XRD), and environmental scanning electron microscopy (ESEM). FTIR and XRD analysis showed no signs of collagen denaturation during the formation of 3D structures. The influence of various factors, such as the chemical composition of the resulted hydrogels and their morphology, on water uptake and water vapor sorption, mechanical behavior, as well as on in vitro degradation characteristics, was systematically investigated. The experimental results point on the advantage offered by the high and modular physicochemical stability of the ternary hydrogels cross-linked by combined approaches. All newly developed materials show no hemolytic effect, which recommends them for potential biomedical applications.
